Dynamics of phagocytic activity of neutrophils in rats under oxidative stress and the action of liposomal preparation

The article gives the results of the research on the influence of a developed complex liposomal preparation on the dynamics of morphological and biochemical indicators of the blood of rats subjected to modeled oxidative stress caused by intermuscular injection of 50% carbon tetrachloride at a dose of 0.25 ml per 100 g of body mass. We found that in conditions of intoxication by carbon tetrachloride, the physiological level of hematological indicators of the researched animals’ body was disrupted. This is indicated by the reduction in the amount of erythrocytes, hemoglobin content, concentration of hemoglobin per erythrocyte, increase in the amount of leucocytes, mass of hemoglobin per erythrocyte and increase of colour indicator. Also, we detected suppression of protein synthesis function of the liver. The levels of total protein and albumin fraction were below the normal physiological level. High indicators were observed in the levels of creatinine, urea and total bilirubin. In order to normalize the functional state of the liver under oxidative stress it is reasonable to use a liposomal preparation which contains butafosfan, interferon, thistle and vitamins. In order to normalize morphological and biochemical indicators of the blood of rats subject to intoxication of carbon tetrachloride it is reasonable to use a liposomal preparation that contains butafosfan, selenium, thistle, methionine and vitamins. Using the liposomal preparation Butaselmevit for rats under oxidative stress the morphological and biochemical indicators in the blood normalized. On the 14th day the indictors of the number of erythrocytes, the hemoglobin content, the white blood cell count and red blood indexes compared to control came within the physiological indicators, indicating a recovery of hematopoietic function of the bone marrow and normalized indicators of the functional state of the liver.

The results of research on the influence of the elaborated complex immunotropic drug containing butafosfan, interferon, thistle and fat-soluble vitamins A, D3, E in the form of a liposomal emulsion on the activity of T- and B-cell chains of immunity in rats under the conditions of action on the body of oxidative stress are presented. It has been established that the introduction of 50% tetrachloromethane into the rats of the first and second experimental groups, with a dose of 0.25 ml per 100 g of body weight, causes oxidative stress in them which negatively affects the cellular immunity and functional activity of immunocompetent blood cells. Immunosuppressive effects of oxidative stress were manifested by a decrease in the blood of rats in the first and second experimental groups of the number of T- and B-lymphocytes and their regulatory subpopulations mainly on the 2nd and 5th day of the study. At the same time, in the blood of rats of the first experimental group in all research periods a decrease in the relative number of common, active and theophylline-resistant T-lymphocytes, as well as B-lymphocytes was observed with a noticeable increase in the number of their undifferentiated forms. At the same time, the obtained data suggest the positive effect of butafosfan, interferon, thistle and vitamins A, D3, E in the liposomal preparation on the relative amount of T- and B-lymphocytes and on the redistribution of avidity in the direction of strengthening the receptor field of plasma membranes immunocompetent cells. It was found that the normalization on the 2nd day of blood level in the second experimental group of common T-lymphocytes occurred due to the secondary forms of the blood and active T-lymphocytes by changes in the number of low-avid forms. In addition to the indicators characterizing the cellular immunity of rats, the components of the liposomal preparation showed regulatory influence on the humoral link of the immune response. In particular, on the 10th day of research in blood of rats of the second experimental group a tendency towards an increase in the relative number of B-lymphocytes and an increase in the number of cells with low and medium density of receptors was found, which, under the conditions of oxidative stress, indicates an increase in the body's ability to actively synthesize protective antibodies

This research studied the interrelationship between the immune and oxidative-antioxidant systems in a group of individuals who had lived for a long time in areas contaminated with radionuclides after the Chernobyl catastrophe and as a result experienced prolonged exposure to small doses of ionizing radiation. We have examined a group of 100 students aged 18–24, where 50 of them formed the control group and the remaining 50 belonged to the experimental group as they arrived from the territories of enhanced radioecological control (IV radiation zone, density of soil contamination by isotope 137Cs 3.7 x 104 – 18.5 x 104 Bq/m2). Here we determined the level of cortisol, leukocytes and their populations, the levels of lymphocyte subpopulations with phenotypes CD3+, CD5+, CD4+, CD8+, CD16+, CD72+, immunoregulatory index CD4+/CD8+, indicators of phagocytic activity of neutrophils and monocytes, IgG (H), IgM (H), IgA (H), malondialdehyde (MDA), ceruloplasmin (CP), transferrin (Tr), sulfhydryl (SH); and also calculated the oxidative stress index (OSI). We performed the analysis twice: in the absence/presence of additional emotional stress such as an examination session. The studies showed an increase in the oxidative stress index in the group examined from the experimental cluster, especially in terms of emotional stress. At the same time, the neutrophil level increased, but phagocytic activity of neutrophils and monocytes, the relative and absolute number of lymphocytes with phenotypes CD3+, CD5+, CD4+, CD4+/CD8+, and IgG levels decreased. Consequently we revealed the negative correlation between the indexes of oxidative stress in the group of examined (the oxidative stress index (ISO)/the level of malonic dialdehyde (MDA)) and the parameters of phagocytic activity of monocytes, the immunoregulatory index CD4+/CD8+, and the number of lymphocytes with the CD16+ phenotype. In this study we demonstrated the decrease in the participation of ceruloplasmin (CP) as an important antioxidant factor in maintaining the immune homeostasis of the group examined from radiation-contaminated areas compared with control group. The evidence of this is the lack of reliable correlation between ceruloplasmin level and immune system parameters. Moreover we found that radiation-induced intensification of oxidative processes in the experimental group grew in conditions of additional stresses of an emotional nature. Besides, it was accompanied by a significant correlation in the level of oxidative stress and phagocytic activity parameters. Reducing phagocytic activity and the CD4+/CD8+ index on the background of oxidative stress increase can be considered as a sign of immune system ageing, while a decrease in the number of lymphocytes with the CD16+ phenotype is a sign of antitumor defense inhibition. Thus, we draw the conclusion that the inhabitants of the territories of strengthened radioecological control, undergoing exposure to small doses of ionizing radiation from birth, show a significant imbalance of redox homeostasis, which creates the preconditions for immunoreactivity pathology development at the level of both innate and acquired immunity.

The purpose of the work was to study the parameters of the functional and antioxidant state of the liver in rats under conditions of oxidative stress and at the actions of the liposomal drug “Butaselmevit”. The research was carried out on young white laboratory male rats of the Vistar line with a body weight of 180-200 g, which were kept in the standard conditions of the institute vivarium of the State Scientific Research Control Institute of Veterinary Preparations and Feed Additives. Animals were divided into three groups of 20 animals in each: 1st group (K) intact animals; 2nd group (D1) – rats, affected with tetrachloromethane; group 3 (D2) – rats, affected with tetrachloromethane and treated with Botselmevit liposomal preparation. Toxic lesion of rats was caused by intramuscular administration of 50% tetrachloromethane at a dose of 0.25 ml per 100 g of animal body weight for the first and third day of the study. Animals of the experimental group D2 on the first and third day of the study, an hour after the introduction of tetrachloroethane, additionally administered a liposomal preparation at a dose of 2 ml per 1 kg of body weight of the animal. The composition of this preparation includes the following substances: butafosfan, selenium, methionine, thistle injection and vitamins A, E and D3. The development of oxidative stress in rats caused by intramuscular administration of tetrachloromethane was accompanied by suppression of protein synthesis of the liver, as evidenced by the low level of total protein and the decrease in the albumin-globulin factor. Low levels of albumin and high levels of globulins in the blood of diseased rats indicate an albumin-globulin disproportion. The indicators of liver function were quite high, namely: the level of creatinine increased by 46%, urea by 74% and total bilirubin by 34%. When applying the liposomal drug "Butaselmevit" to rats, under conditions of oxidative stress during research, the normalization of biochemical parameters occurs in the blood. It has been proven that modeling of the stress response in rats of the experimental group leads to a probable decrease in the level of reduced glutathione and the activity of glutathione peroxidase in the second and fifth days of the experiment. When applying the liposomal drug Butaselmevit to rats, under conditions of oxidative stress during the studies, activation of the glutathione linkage of the antioxidant system occurs in the blood, indicating an increase in the level of reduced glutathione and the activity of glutathione peroxidase in the blood of animal data. At the 14th day of the research, the level of reduced glutathione and the activity of glutathione peroxidase in blood of the experimental group D2 were the highest. The results of the research indicate the antioxidant properties of the new liposomal drug “Butaselmevit”.

This article presents the results of research into the influence of the complex liposomal preparation Butaintervit on protein synthesis function in the livers of rats under the influence of carbon tetrachloride poisoning. Intramuscular injection of carbon tetrachloride into rats at a dose of 0.25 ml per100 gof body weight causes antigenic load on the body and leads to disruption of protein synthesis function in the liver. This is shown by reduction in blood levels of total protein and its fractions. Thus, the level of albumin in the serum of rats under the conditions of oxidative stress was 70% lower than in clinically healthy animals. However, the level of total protein in the serum was only 10% lower. This is because, along with the decrease of albumin content in the serum, the levels of globulin protein fraction increased by 8.8%. This has led to albumin/globulin disparities in the serum of sick animals. As a result, the value of A/G coefficient was 0.28 ± 0.03, compared to 0.52 ±0.02 inclinically healthy rats. For the normalization of functional state of the liver under oxidative stress it is advisable to apply the liposomal preparation Butaintervite, which in its structure contains butafosfan, interferon, thistle and vitamins A, D and E. Under conditions of oxidative stress and under the action of the liposomal preparation in the rats from the second experimental group we have found significant increase in the levels of total protein and albumins and a decrease in serum globulin in the animals on the fifth and tenth days of the experiment. On the fourteenth day of the experiment under the conditions of oxidative stress and under the action of the liposomal preparation in the rats from the second experimental group the normalization of protein synthesis function in the liver was observed. The level of indicators of total protein, albumin, globulin and the coefficient of albumin/globulin compared with the control group of animals were within normal values.

The study of the mechanism of oxidative stress and regulation of oxygen-dependent processes is important, as the establishment of a complex interaction between oxidative stress, oxidative protein damage and the body's antioxidant system makes it possible to clarify metabolic pathways of disease pathogenesis. In our study, we concluded that in patients with chronic non-specificinflammatory diseases of the genitals of men and women, the phagocytic activity of polymorphonuclear leukocytes is reduced mainly by the oxygen-dependent mechanism, and tissue - by the oxygen-independent. As an exception in patients with chlamydial infection, the phagocytic activity of tissue polymorphonuclear leukocytes was also reduced by an oxygen-dependent mechanism.
