Abstract. The aim of the study was to determine the goat milk quality from animals grown in a mountainous area in Bulgaria based on physicochemical and microbiological parameters. The study was carried out in a farm that breeds local goats and goats of the Bulgarian White Dairy Goat (BWD). Individual milk samples were taken on a monthly basis from morning milking. A total of 100 individual and 10 bulk milk samples were examined for fat, solids non fat, protein and density. A total of 62 samples were collected at a time to determine the microbiological characteristics of milk. For the period May-September 2017, the percentage of fat in the milk of local goats averaged 3.61% and of goats from BWD goat – 3.54%. The solids non fat were 8.27% and 8.19%, total protein – 3.13% and 3.10%, and the dry matter – 11.89% and 11.74%, respectively. For the period May-August 2017 the individual constituents of milk changed to varying degrees with the most variable being milk fat (decrease of 0.97% in local goats’ milk and 1.09% in milk from BWD goat) followed by solids non fat (0.56% and 0.7%, respectively). The slightest change was in protein – 0.21% and 0.26%, respectively. Coagulase-negative staphylococci were the predominant bacterial species in the goats’ milk samples.

The objective of the investigations described in this work was to determine the energy status and to make recommendations for correcting the cow diet at a farm of high-yield dairy cows, on the grounds of values for the concentration of organic components of milk and their ratios in individual milk samples. A total of 147 cows were examined, including 97 in the first and 50 in the second lactation. Average concentrations of milk fat and urea were within the physiological values. Namely, the milk fat concentration in cows in the first lactation was 38.88?5.07 g/l, and it was 36.47?4.82 g/l in cows in the second lactation, while the urea concentration in cows in the first lactation 3.16?0.58 mmol/l and it was 3.72?0.64 mmol/l in cows in the second lactation. The protein concentration in both groups of cows was below the physiological values, being 30.33?2.35 g/l in cows in the first lactation and 30.17?2.27 g/l in cows in the second lactation. Based on the ratio of urea and protein concentrations, as well as of fat and proteins in the individual milk samples, it was concluded that in most examined cows, both in those in the first and those in the second lactation, there is a deficit of energy, along with a deficit or relative surplus of proteins. On the grounds of the obtained results, recommendations were given for correcting the feed rations in the coming period. .

In May 2003 differences in milk fat composition in two main dairy breeds in the Czech Republic, Czech Pied cattle and Holstein cattle, were studied in two uniform groups, each containing eight cows. The groups were housed together and received the same daily diet. Fatty acids were determined in mean milk samples from the individual cows as their methyl esters using a gas chromatography procedure. The groups of the fatty acids, namely saturated (SAFA), monounsaturated (MUFA) and polyunsaturated (PUFA) ones, were examined together with the individual acids. The milk fat of Czech Pied cattle was found to contain significantly less SAFAs than the fat of Holstein cows (60.78 and 63.62% of total acids; P &lt; 0.05). Determined mean MUFA contents (27.64 and 25.76%) and total levels (34.31 and 32.11%) of all the unsaturated acids (MUFAs and PUFAs) were insignificantly elevated in the milk fat of Czech Pied cattle. The contents of the most of the individual fatty acids did not differ considerably between the breeds. In Holstein cows, significantly higher contents (P &lt; 0.05) of capric acid (C<sub>10:0</sub>) and stearic acid (C<sub>18:0</sub>) 3.30 and 4.45%, respectively, as compared with 2.69 and 2.61% for Czech Pied cows, were observed. The milk fat of Czech Pied cows had significantly higher contents of oleic acid (C<sub>18:1</sub>) 23.60% (P &lt; 0.05) and of an isomer of octadecatrienic acid (C<sub>18:3n4</sub>) 0.16% (P &lt; 0.001) as compared with 21.68 and 0.10%, respectively, in the fat ofHolstein cows. &nbsp;

Six hundred and thirty clinically-normal milk samples from dairy goat flocks comprising a mixed population of German Alpine, Toggenburg, Saanen and Galla crosses were exam-ined over a 3-month period to determine the prevalence of bacterial organisms. Bacteria were isolated in 28.7 % of the milk samples (181/630) either singly (92.8 %) or in combination (7.2 %). The most prevalent bacterial organisms were Staphylococcus spp. (60.3 %), followed by Micrococcus spp. (17.7 %), Acinetobacter spp. (5 %), Actinomyces spp. (5%) and Streptococcus spp. (1.1 %). The Staphylococcus spp. were mainly coagulase negative (64.3 %). Coagulase-- negative staphylococci and coagulase-positive staphylococci accounted for 37.5 % and 22.7 % respectively of the total bacteria isolated. The isolation of bacteria, some of which are important in clinical and subclinical mastitis, in apparently normal caprine milk, indicates that particular attention should be given to the management of these dairy goat flocks in order to avoid the development of cases of clinical mastitis.

Background: Lactational mastitis constitutes a significant cause of premature weaning. However, its etiology, linked to the presence of pathogenic microorganisms, has been scarcely reported. Research aim: The aim of this study was to describe the microbial diversity in milk samples from women suffering from lactational mastitis and to identify more accurately a collection of isolates belonging to coagulase-negative staphylococci, streptococci, and coryneform bacteria. Methods: This is a cross-sectional descriptive one-group study. A total of 5,009 isolates from 1,849 mastitis milk samples was identified by culture, biochemical, and/or molecular methods at the species or genus level. A more precise identification of a collection of 211 isolates was carried out by 16S rRNA gene sequencing. Results: Mean total bacterial count in milk samples was 4.11 log10 colony-forming units/ml, 95% confidence interval [4.08, 4.15]. Staphylococcus epidermidis was the most common species being isolated from 91.56% of the samples, whereas Staphylococcus aureus was detected in 29.74%. Streptococci and corynebacteria constituted the second (70.20%) and third (16.60%) most prevalent bacterial groups, respectively, found in this study. In contrast, Candida spp. was present in only 0.54% of the samples. Sequencing of the 16S rRNA gene revealed a high diversity of bacterial species among identified isolates. Conclusion: Many coagulase-negative staphylococci, viridans group streptococci, and corynebacteria, usually dismissed as contaminant bacteria, may play an important role as etiologic agents of mastitis. Proper diagnosis of mastitis should be established after performing microbiological testing of milk based on standardized procedures. A reliable analysis must identify the mastitis-causing pathogen(s) at the species level and its(their) concentration(s).

Abstract In this study, milk samples (n=52) from nine healthy female raccoons at 3–45 days of lactation were manually collected at a regular morning hour from all active teats and used to investigate the changes of lipid globules size, fatty acids (FAs) profile and fat content.The results indicated that raccoon milk is characterized by a high fat content. Small lipid globules sizing up to 6 µm prevailed with their greatest share in milk during lactation phases I and III. The milk fat content was increasing with the proceeding lactation, whereas the content of free FAs had a decreasing tendency. Totally eighteen FAs were identified in raccoon milk. The unsaturated long-chain C18–C20 FAs were dominating (over 60%). The individual FAs contents in raccoon milk did not exceed 1%, except for palmitic, vaccenic and linoleic acids representing over 20% of the total FA content.The study results can be used for establishing the energy requirements during the suckling period for proper growth and development of puppies.

This study is based on analyzing physico-chemical and microbiological characteristics of ewe milk from three different regions, which is used as a raw material in dairy industry of North Macedonia. All milk samples (n=123) were collected on dairy reception points of dairy industry in Pelagonia region, in a period of six (6) months. The average values for chemical composition of milk were in the range as follows: 6.83±0.22% to 7.68±0.36% for milk fat, 5.52±0.09% to 6.06±0.23% for proteins, 4.62±0.08% to 4.79±0.05% for lactose, 11.16±0.11% to 1.51±0.24% for non-fat dry matter (NFDM), -0.561 to -0.543oC for freezing point and 8.61±0.4 oSH to 8.88±0.47oSH for titrable acidity. The microbiological and hygienic status of examined ewe milk samples was of poor hygienic quality. The average value of the examined samples for somatic cell count ranged from 753.333 cfu/mL to 1.125.000 cfu/mL and the average number of total bacteria count was from 532.444 cfu/mL to 1.986.222 cfu/mL.

Although Corynebacterium bovis and coagulase-negative staphylococci are frequently the most commonly isolated bacteria from milk samples submitted for identification of pathogens causing intramammary infection, the individual quarter somatic cell count (SCC) from those samples is most often low. The present study aimed at evaluating the difference in bacteriology results from milk sampled by the standard technique (as recommended by the National Mastitis Council) and by the use of a teat cannula surpassing the teat canal, since C. bovis is often only found in the teat canal. Single quarter milk samples were collected in duplicate from 132 dairy cows on a commercial dairy farm using the standard milk sampling technique and also using a cannula introduced into the teat. Two groups of quarters were sampled: a group that was selected randomly at cow and quarter level and a group that was selected based on having SCC >200 000 cells/ml at the previous milk recording at cow level and on California mastitis test result at quarter level. Bacteriological culture performed on the samples yielded 29 Corynebacterium spp. isolates from the samples collected with the standard technique and 6 isolates from the samples collected with a cannula. Bacteriological culture yielded 73 and 100 culture negative samples respectively with the standard and the alternative sampling technique. A significant difference between the two sampling techniques was observed for recovery of Corynebacterium spp. and for no-growth samples. There was no significant difference in the isolation of Corynebacterium spp. or other bacterial species when using the standard technique before or after sampling with the cannula; thus the observed difference in bacteriology results could not be attributed to a particular sampling order. No significant change was observed overall in individual quarter SCC measured on the sampling day and 7 d later. Our results agree with several studies showing that Corynebacterium bovis often colonizes the teat canal, without causing true intramammary infection.

In order to determine the safety of milk produced by smallholder dairy goat farms, a farm-based research study was conducted on commercial dairy goat farms to compare the microbiological quality of milk produced using 3 different types of dairy goat production systems (intensive, semi-intensive and extensive). A survey of dairy goat farms in and around Pretoria carried out by means of a questionnaire revealed that most of the smallholder dairy goat farms surveyed used an extensive type of production system. The method of milking varied with the type of production system, i.e. machine milking; bucket system machine milking and hand-milking, respectively. Udder half milk samples (n=270) were analysed, of which 31.1 % were infected with bacteria. The lowest intra-mammary infection was found amongst goats in the herd under the extensive system (13.3 %), compared with 43.3 % and 36.7 % infection rates under the intensive and semi-intensive production systems, respectively. Staphylococcus intermedius (coagulase positive), Staphylococcus epidermidis and Staphylococcus simulans (both coagulase negative), were the most common cause of intramammary infection with a prevalence of 85.7 % of the infected udder halves. The remaining 14.3 % of the infection was due to Staphylococcus aureus. Bacteriology of bulk milk samples on the other hand, showed that raw milk obtained by the bucket system milking machine had the lowest total bacterial count (16 450 colony forming units (CFU)/mℓ) compared to that by pipeline milking machine (36 300 CFU/mℓ) or handmilking (48 000 CFU/mℓ). No significant relationship was found between the somatic cell counts (SCC) and presence of bacterial infection in goat milk. In comparison with the herds under the other 2 production systems, it was shown that dairy goat farming under the extensive production system, where hand-milking was used, can be adequate for the production of safe raw goat milk.

Reliable identification of the aetiological agent is crucial in mastitis diagnostics. Real-time PCR is a fast, automated tool for detecting the most common udder pathogens directly from milk. In this study aseptically taken quarter milk samples were analysed with a real-time PCR assay (Thermo Scientific PathoProof Mastitis Complete-12 Kit, Thermo Fisher Scientific Ltd.) and by semi-quantitative, in-depth bacteriological culture (BC). The aim of the study was to evaluate the diagnostic performance of the real-time PCR assay in routine use. A total of 294 quarter milk samples from routine mastitis cases were cultured in the national reference laboratory of Finland and examined with real-time PCR. With BC, 251 out of 294 (85·7%) of the milk samples had at least one colony on the plate and 38 samples were considered contaminated. In the PCR mastitis assay, DNA of target species was amplified in 244 samples out of 294 (83·0%). The most common bacterial species detected in the samples, irrespective of the diagnostic method, was the coagulase negative staphylococci (CNS) group (later referred as Staphylococcus spp.) followed by Staphylococcus aureus. Sensitivity (Se) and specificity (Sp) for the PCR assay to provide a positive Staph. aureus result was 97·0 and 95·8% compared with BC. For Staphylococcus spp., the corresponding figures were 86·7 and 75·4%. Our results imply that PCR performed well as a diagnostic tool to detect Staph. aureus but may be too nonspecific for Staphylococcus spp. in routine use with the current cut-off Ct value (37·0). Using PCR as the only microbiological method for mastitis diagnostics, clinical relevance of the results should be carefully considered before further decisions, for instance antimicrobial treatment, especially when minor pathogens with low amount of DNA have been detected. Introducing the concept of contaminated samples should also be considered.
