Kokoleceran (Vatica bantamensis) is an endemic plant of Banten which is only in Ujung Kulon, and has been designated as identity of Banten province. Now the existence of Kokoleceran has been endangered. Based on data from the IUCN, since 1998 there has been no research. Kokoleceran initiation needs technology for preventing from the extinction. One of the mass propagation is through the technique culture in vitro. This research aimed to get the precise medium and BAP concentration for Kokoleceran, and knowing the response of Kokoleceran growth in in vitro. This research was done in November 2015 until March 2016 at the Laboratory of Biotechnology, Department of Agroecotechnology, Faculty of Agriculture, University of Sultan Ageng Tirtayasa, Serang, Banten. This research used Completely Randomized Design (CRD) two factors. The first factor was medium that were MS and WPM media. The second factor was BAP concentration which consisted of four levels namely 0 mg L-1, 1 mg L-1, 2 mg L-1, and 3 mg L-1. The results showed that the use of different media and BAP concentrations had no impact on the time appear of shoot, shoot number, and root number. There was effect from both treatments on medium color, and growing of callus on the explants.

The  objective of the study was to obtain the combination of sucrose and kinetin concentration reducted in the best effect on Dendrobium explant. This research was conducted in laboratory of tissues culture in Balai Benih Induk Dinas Pertanian and Peternakan Provinsi Riau Pekanbaru from April to June 2015. The experiment was designed using completely randomized design (CRD), factorial consisting of two factors with three replications, the first factor was sucrose with four level respectively 0, 25, 50,75 g L-1 medium. The second factor was kinetin with four  level : 0, 0,1, 1,0, 10 mg L-1. The optimum interaction between effect of sucrose and kinetin for percentage of life explants, age bud formation, percentage of explant forming buds, the number of shoot, a percentage of explant producing root  was 50 g L-1 sucrose and 1,0 mg L-1  kinetin.  Application of sucrose significantly effect age of bud formation, percentage of explant forming buds, number of shoot, shoots high, percentage of explant producing  root, number of root, root high, root and shoot ratio (T/R Ratio). Sucrose at 50 g L-1 medium being the best consentration and Kinetin 1,0 mg L-1   being the best consentration.

Research with the title shoot multiplication shoots tomatoes (Lycopersicumesculentum mill) using the Benzyl Amino Purine (BAP) dan Naphtalene Acetic Acid(NAA) in vitro have been conducted at the Laboratory of Biotechnology Faculty ofAgriculture, Islamic University of Riau, Pekanbaru. This research has been carried outfor three months carried out startingfrom November 2010 to February 2011. This studyaims to determine the effect of a single interaction between the administration and plantgrowth regulators BAP and NAA on shoot multiplication of in tomato shoots vitro.Rancangan used in this study was completely randomized design (CRD) in factorialwhich consist of two factors. The first factor is factor B (concentration of BAP) withfour standard treatments are: B0 (0 ppm), B1 (1 ppm), B2 (2 ppm), B3 (3 ppm).Thesecond factor is the factor (the concentration of NAA) with four standard treatments,including: N0 (0 ppm), N1 (0.1 ppm), N2 (0.5 ppm), and N3 (1 ppm), to obtain 16combined treatment with three replications. Parameters observed, namely: age emergedshoots, number of shoots, shoot height, the percentage of growing shoots, roots andgrowing percentage of the number of explants forming callus. The data was statisticallyanalyzed the results of observations, when the F calculated is greater than the F table,followed by a further test of honest real difference (HRD) 5%. From the results ofresearch in the interaction of BAP and NAA effect on the parameters of high-shoots bytreatment tebaik B1N0 namely 6.16 cm. BAP singly significantly affect the parametersage appears buds (days) and the percentage grows shoots with the best treatment B2 (2ppm), shoot height with the best treatment B0 (0 ppm), the number of shoots (the fruit)with the best treatment B3 (3 ppm). singly whereas NAA significantly affect theparameters age appears shoots (day) and high-shoots with the best treatment N0 (0ppm), and the percentage grows roots with the best treatment N3 (1 ppm).

Abstract Background A need for research searching for alternative rumen enhancers warrants immediate attention. The in vitro fermentation experiment was conducted using factorial arrangement of two factors of roughage to concentrate and seven level of red amaranth leaf powder percentage of total substrate in a Completely randomized design (CRD). Two factors, namely Factor A was two ratio of roughage (R) to concentrate (C) at 60:40 and 40:60 and Factor B was level of red amaranth (Amaranthus cruentus, L) leaf powder (RALP) supplementation at 0, 2, 4, 6, 8, 10, and 12% of total dietary substrate. Results Red amaranth leaf powder (RALP) contained phytonutrients both condensed tannins and saponins in addition with high macro minerals (Ca, K, and Mg). This experiment revealed innovations of the RALP supplementation by enhancing rumen propionate (C3) production, reducing acetate (C2) to (C3) ratio, reducing protozoal population and mitigating methane (CH4) production. Furthermore, rumen dry matter degradation percentages were remarkably enhanced (P < 0.001) by increasing RALP supplementation. Conclusion Plants rich in phytonutrients and minerals such as red amaranth leaf powder (RALP) have a vital and promising role in modulating rumen fermentation, mitigating methane production, as well as increasing substrate degradability.

Chryssanthenum is ornamental plant with variety of shape and color which are unique and appealing. So that, it is in great demand in the community. The production of Chrissanthenum conventionally hampered by availability and quality of seeds. Thus, it needs research through tissue culture. The aim of this research is to know interaction between coconut water and thiamine and to determine the best coconut water and thiamine concentration toward Chryssanthenum micro cuttings. This research used laboratory experimental method by using completely randomized design with two factor. The 1st factor was coconut water concentration consisted of three level which were 5%, 10% and 15%. The 2nd factor was thiamine concentration consisted of three level which were 1mg/L, 2 mg/L and 3 mg/L. From the two factors, those were found that, there were nine combination of treatments and repeated 3 times. The variety  of data was analyzed by using Analysis of Variance (ANOVA) with level of α=5%, and continued by examining Duncan’s Multiple Range Test (DMRT) with level of α=5%. The result indicated that the interaction of coconut water concentration was 5% and thiamine was 1 mg/L on the parameters when growing shoot. There was also interaction on coconut water concentration which was 10 % and thiamine was 1 mg/L on the parameters in the number of shoots. interaction of coconut water combination was 15% and thiamine was 2 mg/L on  fresh weight. The addition of 10% coconut water and 1 mg/L thiamine showed the best result on shoot length.

The propagation of Cavendish (Musa acuminata L.) seedling conventionally relatively took a longer time, not uniformed in quality and possibly has a disease that is derived from unhealthy mother stock. This research aimed to find the effect of IBA and BA on difference concentrations on the growth of plantlet, also to determine the optimum concentration of IBA and BA on the growth of Cavendish bud plantlets in vitro. This research had been carried out started from April to July 2018 in Tissue Culture Laboratory, Research Institute of Horticultural Seed Development of Pasir Banteng, Sumedang, West Java. Indonesia. The method used in this research was Completely Randomized Design with two factors. The first factor was IBA (i0 = 0 mg L-1, i1= 0,75 mg L-1, dan i2= 1,5 mg L1), and the second factor was BA (b1= 1 mg L-1 b2= 3 mg L-1 b3= 5 mg L-1 b4= 7 mg L-1), replicated three times. Duncan Multiple Range Test (DMRT) was used as a further test. The result showed an interaction between IBA and BA on the parameters of shoot growth and the number of leaves at 6 WAC. IBA showed an independent effect on the number of buds. The addition of 1,5 mg L-1 IBA and 3 mg L-1 BA was optimum for the growth of Cavendish bud explants.

This study aimed for knowing and obtaining the best concentration of kinetin and NAA interaction effects in influencing the shoot induction, knowing how the plant growth regulators in induction mediastill affect the shoot additionin the MS0media and also knowing the largest number of roots in rooting media for shallot by in vitro. The experiment was conducted at Laboratory of Tissue Culture Seed Technology, Faculty of Agriculture, Padjadjaran University, during January 2011 until May 2011. This experiment divided in 3 stages, namely shoot induction stage, shoot subculture to MS0 media stage and shoot subculture to rooting media stage. Experimental method used in the shoot induction stage was factorial Completely Randomized Design with three replications. The first factor was the kinetin with four levels,0, 1, 2, and 3 mg L-1. The second factor was the NAA with three levels, as 0, 0.01, and 0.1 mg L-1. Basic media used for each treatment was MS. The experiment result showed there was an interaction between kinetin and NAA on shoot induction stagewith the plantlet height, leaf number, and shoot addition. The best result for leaf number was gained from interaction with 2 mg L-1 kinetin without NAA,while the treatment of 2 mg L-1 kinetin with 0.01 mg L-1 NAA gave a better interaction for theshoot addition variable.

[SWEET CORN EXTRACT AND COCONUT WATER EFFECT TO INDUCE GARLIC (Allium sativum L.) ADVENTIST SHOOTS BY IN-VITRO CULTURE]. Garlic (Allium sativum L.) is one of the horticultural plants used daily for various uses. This study aims to determine the best extract concentration of sweet corn and coconut water and its interaction for induction garlic Adventist shoots (Allium sativum L.) var. Lumbu Hijau. The study was conducted at the Biotechnology Laboratory, Agriculture Faculty, UPN ”Veteran” East Java, in February – April 2021. The experimental design used was a factorial completely randomized design (CRD). The first factor is four levels of extract of sweet corn concentration (0, 50, 100, and 150 g/L), while the second factor is four levels of coconut water concentration (0, 50, 100, and 150 mL/L). The results showed that the extract of sweet corn concentration-time treatment increased the growing adventist shoots percentage, adventist shoots number, and adventist shoots size. The section of sweet corn concentration of 150 g/L increased the rate of growing adventist shoots, adventist shoots number, and adventist shoots size. The treatment of coconut water concentration and its interaction with the extract of sweet corn concentration is still not able for induction garlic adventist shoots (Allium sativum L.) of the var. Lumbu Hijau.

Conventionally, cultivations of Nepenthes are conducted by using seeds, cutting, and filial separation. However, there are many obstacles come both from time and technical aspect. In vitro culture is an alternative way for cultivating  N. ampullaria (Jack,). One of a technique of in vitro culture is micro cutting. BAP (6-benzyl amino purine) growth regulator could be added to optimize the growth of  N. ampullaria microcutting. The purpose of this research was to determine the effect of  BAP on the growth of Nepenthes microcutting. This research was done experimentally using Completely Randomized Design (CRD). BAP treatment consisted of 5 concentrations: 0; 0.5; 1; 1.5; 2 (ppm), each treatment were multiplied 4 times. The parameters observed were: a time of bud initiation, time of root initiation, total of leaves, total of new buds, total of roots, length of leave, length of root, and height of bud. The data obtained were analyzed with ANOVA (Analisis of Variance) and continued with 5% and 1% LSD (Least Significant Different)  test. The result showed that addition of BAP affected the growth of  N. ampullaria microcutting in total leaves, length of leave, and total of buds. LSD test proved that 0.57 ppm of BAP was optimal concentration to increase total buds, whit the value reached of 3.86. Here, we found that BAP can be utilized to enhanche N. ampullaria growth on in vitro culture. The benefit of this study is to conserve N. ampullaria in vitro using BAP at  0.57 ppm.

This study aims to determine the effect of growth regulators Hormonik against Parthenocarpy process on three major varieties of pepper plants and to determine the effect of plant growth regulator that is optimum for the process Hormonik Parthenocarpy in three major varieties of chili. This research was conducted in Tanjung Selor, in January to May 2016. The design used in this study is completely randomized design (CRD) with two factors and nine replications. The first factor is the treatment Hormonik consisting of four levels ie 0 cc, 1 cc, 2 cc, 3 cc, the second factor is the variety Arielle, Darmais and Pilar. From the analysis of variance showed that the treatment Hormonik to varieties of very significant effect on the parameters of heavy fruit crop, the number of fruit crops, fruit length persampel and the number of seeds persampel first harvest, after a further test using LSD 5%, by weight of fruit crops and the amount of fruit planting the first crop was not significantly different, but significantly different with persampel fruit length and number of seeds persampel first harvest.

The aim of research to determine the effect of urea and SP-36 fertilizer as well as their interaction on the graftedrubber seedling growth, and to find appropriate doses of urea and SP-36 fertilizers for better growth of grafted rubber seedlings.The research was conducted from February 2015 until May 2015 in the village of Empas, Sub District of Melak, West Kutai Regency, East Kalimantan Province.The study design used was completely randomized design (CRD) in a factorial 4 x 4, and repeat 3 times, which comprises two factors research. The first factor was Urea (N), consisted of four levels, namely: no urea application or control (n0), 2,5g/polybag (n1), 5g/polybag (n2), and 7,5g/polybag (n3).  The second factor was SP-36 fertilizer (P), consisting of 4 levels, namely: no SP-36 fertilizers application or control (p0), 2,5g/polybag (p1), 5g/polybag (p2), and 7,5g/polybag (p3).The results showed that the treatment of urea (N) affects highly significant on the plant height at ages of 2, 3 and 4 months after grafting, number of leaf at 4 months after grafting and stem diameter at 3 and 4 months after grafting. It affected significantly on the number of leaf at 3 months after grafting.  But it did not affect significantly on the number of leaf at 2 months after grafting, and stem diameter at 2 months after grafting.SP-36 fertilizer treatments (P) affected very significantlyon the stem diameter at 3 and 4 months after grafting.  It affected significantly on the plant height at 4 months after grafting. But it did not affect significantly on the seedling height at 2 and 3 months after grafting, number of leaves at 2, 3 and 4 months after grafting, and stem diameter at 2 months after grafting.The interaction treatment (NXP) affected significantly on the stem diameter at 4 months after grafting.  But it did not affect significantly on the seedling height at 2, 3 and 4 months after grafting, number of leaves at 2, 3 and 4 months after grafting, and stem diameter at 2 and 3 months after grafting
