Purple nonsulfur bacteria are a group that has so much biotechnological applications, particularly in producing of functional food rich with unsaturated fatty acids. A purple nonsulfur bacterium (named HPB.6) was chosen based on its strong growth, high lipid and synthesis of unsaturated fatty acid (omega 6,7,9). Studying on basic biological characteristics showed that the cells of HPB.6 were observed as ovoid-rod shape, none motility, Gram negative staining. The diameter of single bacterium was about 0.8-1.0 µm. The cells divide by binary fission and had bacteriochlorophyll a (Bchl a). This bacterium grew well on medium with carbon and nitrogen sources such as acetate, succinate, pyruvate, butyrate, glutamate, arginine, leucine, tyrosine, alanine, methionine, threonine, glutamine, yeast extract and NH4Cl. This selected strain grew well on medium with salt concentrations from 1.5 - 6.0% (optimum 3%), pH from 5.0 to 8.0 (optimum at pH 6.5) and could withstand Na2S at 4.0 - 5.2 mM. Based on morphological, physiological properties and 16S rRNA analysis received demonstrated that HPB.6 strain belongs to the species Rhodovulum sulfidophilum.

The objective was to get beef that contain unsaturated fatty acids (especially omega 3 and 6), so as to improve intelligence, physical health for those who consume. The study design using CRD with 3 treatments, each treatment used 4 Bali cattle aged approximately 1.5 years. Observations were made 8 weeks. Pasta mixed with ginger provided konsentrat. P1 (control); P2 (6% saponification lemuru fish oil, olive oil 1%; rice bran: 37.30%; corn: 62.70%; KLK: 7%, ginger paste: 100 g); P3 (lemuru fish oil saponification 8%, 2% olive oil; rice bran; 37.30; corn: 62.70%; KLK: 7%, ginger paste: 200 g). Konsentrat given in the morning as much as 1% of the weight of the cattle based on dry matter, while the grass given a minimum of 10% of the weight of livestock observation variables include: fatty acid composition of meat. Data the analyzies qualitative. The results of the study showed that the composition of saturated fatty acids in meat decreased and an increase in unsaturated fatty acids, namely linoleic acid (omega 6) and linolenic acid (omega 3), and deikosapenta deikosaheksa acid.Keywords :

Minyak ikan patin mengandung kadar asam lemak omega-9, tetapi rendah asam lemak omega 3 Minyak hati ikan hiu memiliki kandungan asam lemak omega 3 relatif tinggi. Penelitian ini bertujuan untuk menentukan karakteristik dan sifat kimia minyak ikan patin dan profil asam lemak kombinasi minyak ikan patin dengan hati kan hiu. Metode penelitian eksperimen dengan melakukan ekstraksi lemak perut ikan patin secara dry rendering dihasilkan minyak kasar dan dimurnikan. Minyak ikan patin murni dilakukan pencampuran dengan minyak hati ikan hiu. dengan perbandingan 1:1, 2:1 dan 2:1. Parameter analisis terdiri atas karakteristik kimia (bilangan asam lemak bebas (ALB),bilangan  asam, peroksida, iod,  p-anisidin, penyabunan, total oksida/totox), dan profil asam lemak. Hasil penelitian menunjukkan bahwa kombinasi  minyak patin dan hati ikan hiu dihasilkan karakteristik kimia terdiri atas asam lemak bebas sebesar 1,59-2,19%, bilangan asam 2,86-3,00 mg KOH/g, peroksida 15,50-18,32 meq/kg, iod 63,46-84,42 gI2/100g), p-anisidin 21,53-24,16, penyabunan 142,21-162,28 mg KOH/g dan TOTOX 53,33-69,43. Hasil analisis komposisi asam lemak jenuh (saturated fatty acid/SAFA) dihasilkan  asam palmitat tertinggi,  asam lemak tak jenuh tunggal  (mono unsaturated fatty acid/MUFA)  asam oleat,  dan asam lemak tak jenuh ganda (polyunsaturated fatty acid/PUFA)  asam linoleat. Hasil analisis karakteristik kimia kombinasi  minyak ikan patin dan hati ikan hiu sesuai dengan standar IFOMA. Kombinasi minyak hati ikan hiu dan ikan patin dengan perbandingan (2:1) dihasilkan kandungan omega 3 dan omega 6 sebesar 2,59 % dan 29,75%.

Cells of Staphylococcus aureus, strain S-6, can grow in the presence of 100 μg of cerulenin/ml if the basal medium is supplemented with certain saturated or unsaturated fatty acids. The production of enterotoxin B (SEB) is markedly influenced by both the ratio of saturated to unsaturated fatty acid and by the melting point of the unsaturated fatty acid supplement. The results presented suggest that a certain degree of membrane fluidity promotes maximum SEB production and that greater or lesser degrees of membrane fluidity prohibit substantial SEB formation but fail to affect final growth density.

Rats were fed diets that differed in fatty acid composition or in the proportion of energy derived from fat to determine if alteration of dietary fat intake influences the structural lipid composition of liver plasma membrane and the expression of an associated hormone-receptor-mediated function. Weanling rats were fed 9% (w/w) or 20% (w/w) low-erucic acid rape-seed oil or 9% (w/w) soya-bean oil for 24 days. Plasma membranes were isolated and the effect of diet fat on the fatty acid composition of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, phosphatidylinositol and sphingomyelin was determined. Diet fat significantly altered total saturated and (omega-9) and (omega-6)-unsaturated fatty acid composition in addition to the (omega-6)- to (omega-3)-unsaturated fatty acid ratio in these polar lipids. Feeding the high-fat diet increased the (omega-6)- to (omega-3)-unsaturated fatty acid ratio and the (omega-9)-unsaturated fatty acid content in all lipids except sphingomyelin. Assay of glucagon-stimulated adenylate cyclase activity at both high and low glucagon concentrations indicated that high-fat intake also decreased cyclic AMP formation. In a second experiment the fat intake was held constant (40% of energy) and oleic acid was substituted for linoleic acid by blending high- and low-linoleic acid-type safflower oils. This experiment established that a dose-response relationship exists between dietary intake of fatty acid and the fatty acid composition of plasma-membrane phospholipids. Specific diet-induced transitions in membrane phospholipid fatty acid composition were paralleled by changes in glucagon-stimulated adenylate cyclase activity. This study suggests that transitions in dietary fat intake can alter a hormone-receptor-mediated enzyme function in vivo by changing the surrounding lipid environment.

ABSTRACT The fabA and fabB genes are responsible for anaerobic unsaturated fatty acid formation in Pseudomonas aeruginosa. Expression of the fabAB operon was repressed by exogenous unsaturated fatty acids, and DNA sequences upstream of the translational start site were used to affinity purify DesT. The single protein interaction with the fabAB promoter detected in wild-type cell extracts was absent in the desT deletion strain, as was the repression of fabAB expression by unsaturated fatty acids. Thus, DesT senses the overall composition of the acyl-coenzyme A pool to coordinate the expression of the operons for the anaerobic (fabAB) and aerobic (desCB) pathways for unsaturated fatty acid synthesis.

Abstract Medicinal plants have a significant role in preventing and curing several diseases, and Tanacetum L. is one of these plants. The aim of the present study is to determine the fatty acid, lipid-soluble vitamin, sterol, phenolic content, and antioxidant capacity of Tanacetum densum subsp. laxum and Tanacetum densum subsp. amani, to compare the effect of altitude on the biochemical content and to compare systematically by using fatty acids and phenolics. This study showed that palmitic acid (C16:0) and stearic acid (C18:0) are major sources of saturated fatty acid and oleic acid (C18:1 n9), and linoleic acid (18:2 n6c) and a-linolenic acid (C18:3 n3) are the principal unsaturated fatty acids in the two endemic Tanacetum densum taxa. Also, this study found that the unsaturated fatty acid content (60.11±1.61%) of Tanacetum densum subsp. laxum was higher than the unsaturated fatty acid content (44.13±1.28%) of Tanacetum densum subsp. amani. And also, the ω6/ω3 ratio of Tanacetum densum subsp. laxum (1.74) and Tanacetum densum subsp. amani (1.60) was found to be similar. However, this study determined that the lipid soluble vitamin and sterol content of two endemic Tanacetum taxa are low except for stigmasterol. Present study showed that catechin is principal phenolic in the Tanacetum densum taxa. This study also found that Tanacetum densum subsp. laxum and Tanacetum densum subsp. amani had the highest levels of catechin, vanillic acid, and caffeic acid content though the phenolic amounts, particularly catechin and quercetin, were dissimilar in the T. densum taxa. This study suggested that ecological conditions such as altitude may affect the biochemical content of two endemic Tanacetum densum taxa. Furthermore, the current study determined that two endemic Tanacetum L. taxa had potent radical scavenging capacities and found a correlation between total phenolics and antioxidant activity.

1. The fatty acid composition of the ole-1 and ole-1 petite mutants of Saccharomyces cerevisiae was manipulated by growing the organism in the presence of defined supplements of Tween 80 or by allowing cells that had first been grown in the presence of Tween 80 to deplete their unsaturated fatty acids by sequent growth in the absence of Tween 80. 2. The transition temperature of Arrhenius plots of mitochondrial ATPase (adenosine triphosphatase) increases as the unsaturated fatty acid content is lowered. 3. Cells require larger amounts of unsaturated fatty acids to grow on ethanol at lower temperatures. 4. Cells that stop growing owing to unsaturated fatty acid depletion at low temperatures are induced to grow further by raising the temperature and this results in a further depletion of unsaturated acids. This is due to a higher rate, but not a greater efficiency, of mitochondrial ATP synthesis. 5. Arrhenius plots of the passive permeability of mitochondria to protons between 4 and 37°C are linear. The rate and the Arrhenius activation energy of proton entry increase greatly as the unsaturated fatty acid content is lowered. 6. Unsaturated fatty acid depletion has the same effects on the proton permeability of ole-1 petite mitochondria, indicating that the mitochondrially synthesized subunits of the ATPase are not involved in the enhanced rates of proton entry. 7. The adenylate energy charge of depleted ole-1 cells is greatly decreased by growth on ethanol medium. 8. The adenylate energy charge of isolated mitochondria is also lowered by unsaturated fatty acid depletion. 9. The results confirm that unsaturated fatty acid depletion uncouples oxidative phosphorylation in yeast both in vivo and in vitro, and is a consequence of changes in the lipid part of the membrane.

The objective of this work was to evaluate the meat lipid profile from Devon beef steers finished in pearl millet (Pennisetum americanum) pasture and fed at different rates of concentrate supplementary diet. Twelve steers weighing 270 kg, at 12‑month‑average initial age, were randomly distributed into three treatments: pearl millet pasture; and pearl millet pasture plus a concentrate equivalent at 0.5 or 1.0% of body weight, with two replicates. Total contents of saturated and unsaturated fatty acids, the polyunsaturated:saturated ratio and other relevant fatty acids as the vaccenic acid, conjugated linoleic acid, omega‑3, and omega‑6 were not affected by the consumption of a concentrate supplement at 0.5 or 1.0% live weight. However, the 0.5% supplementation level reduced the concentration of dihomo‑γ‑linolenic fatty acid (C20: 3 n‑6), while the 1.0% supplementation level elevated the content of docosahexaenoic (DHA) (C22: 6 n‑3) fatty acid, and the omega‑6:omega‑3 ratio in meat. Consumption of up to 1.0% energy supplementation increases the omega‑6:omega‑3 ratio in meat from Devon steers grazing on pearl millet pasture.

AbstractTwo experiments were carried out with growing-fattening bulls, offered a fattening diet based on concentrate and supplemented with soya oil or steam-flaked linseed. Soya oil (200 g/kg) was mixed with middlings (800 g/kg). Linseed (500 g/kg) and whole barley (500 g/kg) were steam flaked. The fat incorporation resulted in an increase of the ether extract content from 26·5 g/kg dry matter to about 45 g/kg. The incorporation of soya oil increased the average daily gain (P> 0·05). The saturated: unsaturated fatty acid ratio was reduced in perirenal fat due to an increase of oleic acid and a reduction of stearic acid. The inclusion of steam-treated linseed resulted in a lower slaughter weight (P< 0·05) and a shorter fattening period (P> 0·05). The fatty acid composition of perirenal fat was affected significantly: the concentrations of oleic, linoleic and linolenic acid were increased and the concentrations of myristic, palmitic and stearic acids were reduced. This resulted in a reduced saturated: unsaturated fatty acid ratio.In a third experiment, samples of the two fat concentrates were incubated in nylon bags in the rumen. The ether extract disappearance was high with both concentrates. Furthermore, the fatty acid composition was altered to a large extent for soya oil while large proportions of unsaturated fatty acids were still present with steam-flaked linseed.
