Abstract Allium cepa L. is a commonly consumed vegetable that belongs to the Amaryllidaceae family and contains nutrients and antioxidants in ample amounts. In spite of the valuable food applications of onion bulb, its peel and outer fleshy layers are generally regarded as waste and exploration of their nutritional and therapeutic potential is still in progress with a very slow progression rate. The present study was designed with the purpose of doing a comparative analysis of the antioxidant potential of two parts of Allium cepa, i.g., bulb (edible part) and outer fleshy layers and dry peels (inedible part). Moreover, the inhibitory effect of the onion bulb and peel extracts on rat intestinal α-glucosidase and pancreatic α-amylase of porcine was also evaluated. The antioxidant potential of onion peel and bulb extracts were evaluated using 2,2-diphenyl- 1-picryl hydrazyl (DPPH), ferric-reducing antioxidant power assay (FRAP), 2,2’-azino-bis- 3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging assay, H2O2 radical scavenging activity and Fe2+ chelating activity. Total flavonoids and phenolic content of ethanolic extract of onion peel were significantly greater as compared to that of onion bulb. Ethanolic extract of onion peel also presented better antioxidant and free-radical scavenging activity as compared to the ethanolic extract of bulb, while the aqueous extract of bulb presented weakest antioxidative potential. Onion peel extract’s α-glucosidase inhibition potential was also correlated with their phenolic and flavonoid contents. The current findings presented onion peel as a possible source of antioxidative agents and phenolic compounds that might be beneficial against development of various common chronic diseases that might have an association with oxidative stress. Besides, outer dry layers and fleshy peels of onion exhibited higher phenolic content and antioxidant activities, compared to the inner bulb. The information obtained by the present study can be useful in promoting the use of vegetable parts other than the edible mesocarp for several future food applications, rather than these being wasted.

The current investigation is intended to evaluate the content of phytochemical constituents and antioxidant potential of hydroalcoholic extracts of stem and root of Grewia serrulata DC (HAESGS & HAERGS) and leaf and bark of Grewia Nervosa (Lour.) panigrahi (HAELGN & HAEBGN). Initially, all the extracts at different concentrations were estimated for their total phenolic content and total flavonoid content. The study was further extended for their antioxidant potential evaluation using various in vitro methods such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical scavenging assays. The total phenolic content (mg gallic acid equivalent per gram of extract) was high in HAELGN (170.82±0.19) and HAERGS (123.00±0.48) than HAESGS (111.2±0.26) and HAEBGN (119.60±0.23). The total flavonoid content (mg quercetin equivalent per gram) is greater in HAERGS (71.24±0.50) and HAESGS (65.68±0.27) than HAELGN (55.82±0.35) and HAEBGN (62.38±0.45). The IC50 values (µg/ml) of different plant extracts inferred that DPPH radical scavenging activity is greater in HAELGN (42.91±0.88) and HAEBGN (53.87±0.35) than HAESGS (126.73±1.20) and HAERGS (88.87±1.25). However, hydroxyl and superoxide radical scavenging activity is more in HAERGS (135.41±1.19 & 88.00±1.42) and HAELGN (172.28±1.91 & 108.163±1.09) than HAESGS (237.3±1.65 & 110.074±0.87) and HAEBGN (204.7±1.04 & 125.54±1.07). The results of present comprehensive analysis demonstrated that both the plants Grewia serrulata DC and Grewia Nervosa (Lour.) panigrahi possess high phenolic, flavonoid contents and potential antioxidant activity, and could be used as a valid source of natural antioxidants and might be utilized for pharmacological screening of various therapeutic activities. Objective: To investigate the antioxidant activity of ethanolic extract of Terminalia bellerica fruit pulp (TBFP), seed (TBS) and bark (TBB), and identification of phytochemical constituents of said extracts.Methods: The antioxidant potential of T. bellerica (TB) parts was evaluated by free radical scavenging activity (FRSA), superoxide anion radical scavenging activity (SARSA), hydroxyl radical scavenging activity (HRSA) and compared with reference standard quercetin. Lipid peroxidation (LPO), ferric thiocyanate activity (FTC) and reducing power (RP) of the plant extracts were also examined. The phytochemical constituents of said extracts have been quantified by gas chromatography-mass spectrometer (GC-MS) method.Results: Ethanolic extract of TBFP exhibited high phenolic content (254.33) followed by TBS (227.33) and TBB (185) mg/g of GAE. TBFP showed lowest IC50 for FRSA (62 µg/ml), SARSA (39 µg/ml) and HRSA (27 µg/ml) and highest RP (3.39 ascorbic acid equivalents/ml). TBB showed lowest IC50 for LPO (48 µg/ml) whereas TBS showed lowest IC50 for FTC (109 µg/ml). Predominant phyto-compounds present in TB extracts were quinic acid, gallic acid, ethyl galate, 9, 12 octadecadienoic acid and glucopyranose in varying concentrations as analyzed by GC-MS.Conclusion: On the basis of correlation of antioxidant studies with total phenolic content (TPC) and GC-MS analysis of different part of TB, it may be concluded that high concentration of total phenolics and other bioactive phytochemicals of TB extracts make it a potential source of nutraceutical antioxidants.

For the functional food applications, antioxidant properties and the bioactive compounds of the 23 Curcuma species commercially cultivated in Thailand were studied. Total phenolic content and DPPH radical scavenging activity were determined. The concentrations of eight bioactive compounds, including curcumin (1), demethoxycurcumin (2), bisdemethoxycurcumin (3), 1,7-diphenyl-(4E,6E)-4,6-heptadien-3-ol (4), germacrone (5), furanodienone (6), zederone (7), and ar-turmerone (8), were determined from the Curcuma by HPLC. While the total phenolic content of C. longa was highest (22.3 ± 2.4 mg GAE/g, mg of gallic acid equivalents), C. Wan Na-Natong exhibited the highest DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) radical scavenging activity. Twenty-three Curcuma species showed characteristic distributions of the bioactive compounds, which can be utilized for the identification and authentication of the cultivated Curcuma species. C. longa contained the highest content of curcumin (1) (304.9 ± 0.1 mg/g) and C. angustifolia contained the highest content of germacrone (5) (373.9 ± 1.1 mg/g). It was noteworthy that 1,7-diphenyl-(4E,6E)-4,6-heptadien-3-ol (4) was found only from C. comosa at a very high concentration (300.7 ± 1.4 mg/g). It was concluded that Thai Curcuma species have a great potential for the application of functional foods and ingredients.

Objective: We aimed to investigate the free radical scavenging, antioxidant and hepatoprotective potential of M. concanensis Nimmo leaves.Methods: Free radical scavenging activity was evaluated by employing various accepted in vitro­ systems, such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), hydroxyl (OH) and nitric oxide (NO) radical. Antioxidant potential of M. concanensis Nimmo extract was assessed against H2O2 in goat liver by determination of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione S-transferase (GST), glucose-6-phosphatase dehydrogenase (G-6-PDH), total reduced glutathione (GSH), vitamin C, vitamin E activity, and lipid peroxidation (LPO).Results: Results showed that the amount of plant extract of M. concanensis Nimmo required to scavenge 50% of the DPPH radicals was 401.80 μg/ml, ABTS radical was 353.14 μg/ml, OH radical was 433.71 μg/ml and NO radical was 371.24 μg/ml. Also, the pre-treatment of ethanolic extract of M. concanensis Nimmo leaves in goat liver showed a significant protection against H2O2 induced oxidative stress by retaining of antioxidants of SOD (4.76±0.25), CAT (26.81±0.25), GPx (6.41±0.34), GST (2.31±0.10), G-6-PDH (0.84±0.02), GSH (37.30±1.12), vitamin C (3.01±0.07), vitamin E (14.43±0.45) within normal range.Conclusion: In conclusion, promising free radical scavenging, the antioxidant activity of M. concanensis Nimmo leaves can be able to treat various diseases caused by free radicals.

The present study was to analyze the antioxidant activity and the responsible chemical constituents in Cephalotaxus oliveri Mast. aerial parts. DPPH (2,2-diphenyl-1-picryl hydrazyl), ABTS [2,2?-azinobis(3-ethylbenzothiazoline- 6-sulfonic acid)], reducing power and total phenolic content assays indicate that the methanol extract of the aerial parts is the most potent radical-scavenger and reducing agent with the highest level of phenolic content among the test extracts. The highly positive linear correlations implicate that the four assays have similar capacity to predict the antioxidant potential of the aerial parts and phenolic compounds present contribute significantly to DPPH and ABTS radical scavenging activity and reducing power of the extracts. The plant also displayed considerable superoxide anion radical scavenging activity. LC-MS/MS and GC-MS analyses resulted in the identification of 22 compounds in the methanol extract, among which are 15 phenolic compounds. The total amount of the phenolic compounds in the methanol extract determined by HPLC method was more than 5622.39 ?g/g dry weight. The considerable antioxidant potential and a high content of phenolic antioxidants suggest that C. oliveri aerial parts are a potential source of natural antioxidants.

Objective: Protection from oxidative damage to the tissues is provided by natural or synthetic antioxidants. Because of the multiple mechanisms of actions and presence of various phytochemicals, plant-based antioxidants are preferred. Dioscorea is well known as a traditional edible tuber. The present study was undertaken to explore and highlights the nutritional benefits and to evaluate the in vitro antioxidant and free radical scavenging capacity of ethanolic extract of three different underutilised Dioscorea species (D. alata, D. pentaphylla and D. oppositifolia) with a common cultivated crop, Plectranthus rotundifolius.Methods: Different processing methods are adopted to assess their influence on nutritive value and antioxidant properties. Tubers were studied for proximate composition using standard analytical methods. Mineral elements were analysed using ICP-MS (Inductively Coupled Plasma-Mass Spectrometer). Antioxidant potential of raw and processed samples was determined using a range of in vitro models.Results: The presence of essential minerals such as Na, K, Ca, P, Zn and Mn were observed. High radical scavenging activity was observed in the tubers of Dioscorea. Results indicated that phenolic compounds are the significant contributors to the antioxidant activity.Conclusion: Current study suggests that Dioscorea might be a promising natural antioxidant and could be very useful health food, as they enhance the antioxidant defence system. It also proposes that underutilized tubers can efficiently contribute to the nutritional requirement and food security.

A series of diazenyl chalcones was prepared by base catalyzed Claisen-Schmidt condensation of synthesized hydroxy substituted acetophenone azo dye with various substituted aromatic/ heteroaromatic aldehydes. The structural conformation of synthesized chalcones was done by a number of physicochemical and spectral means like FTIR, UV-visible, mass, NMR spectroscopy and CHNS/O analysis. These diazenyl chalcones were assessed for their in vitro antimicrobial potential against several Gram-negative, Gram-positive bacterial and fungal strains by serial tube dilution method. The fluconazole and cefadroxil were used as standard drugs. The target compounds were also evaluated for their antioxidant potential by DPPH assay. (2E)-3-(2,4-Dichlorophenyl)-1-(4-((2,6- dihydroxyphenyl)diazenyl)phenyl)prop-2-en-1-one (C-7) had shown very good antimicrobial potential with MIC ranges from 3.79 to 15.76 μg/ml against most of the tested microorganisms. Most of the synthesized diazenyl chalcones were found to be active against B. subtilis. The (2E)-1-(5-((2-Chloro- 4-nitrophenyl)diazenyl)-2-hydroxyphenyl)-3-(2-hydroxynaphthalen-1-yl)prop-2-en-1-one (C-10) had shown high free radical-scavenging activity when compared with the ascorbic acid as the reference antioxidant.

AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.
