Abstract: The digital image analysis of seedlings has become largely employed in seed quality-control programs due to its feasibility, objectivity and fast results. Despite these advantages, no studies have yet demonstrated the efficiency of this technique for analyzing coffee seed vigor. The present study aimed at evaluating the efficiency of the Seed Vigor Imaging System (SVIS®) in determining the vigor of coffee seeds (Coffea arabica L.) from digital images of seedlings. Also, the results of these analyses were contrasted with conventional vigor tests. Six seed lots from each cultivar, Bourbon and Catucaí 20/15, were used. The research was conducted in two experimental times, and the seed vigor was determined by the tests of first germination count, accelerated aging, electrical conductivity, primary root protrusion speed, as well as by the analysis of scanned images of seedlings, made possible by the SVIS® software. For the Bourbon and Catucaí 20/15 cultivars, the SVIS® analysis was able to discriminate lots of both high and low vigor (vigor indexes of 317 to 752, and 181 to 703, respectively). A similar outcome was obtained from the conventional tests appraised in this research. Eventually, it was possible to conclude that the digital analysis of 20-day-old seedlings by SVIS® is an efficient method to evaluate coffee seed vigor.

The assessment of physiological potential is essential in seed quality control programs. This study compared the sensitivity of different procedures for evaluating super sweet corn seed vigor, focusing on the primary root protrusion test. Six seed lots, each of the SWB 551 and SWB 585 hybrids, were used. Seed physiological potential was evaluated by germination and vigor tests (speed of germination, traditional and saturated salt accelerated aging, cold test, seedling length, seedling emergence and primary root protrusion). Primary root protrusion was evaluated every 12 hours at 15 °C, 20 °C and 25 °C using two criteria (primary root protrusion and seedlings at the 2 mm root stage). It was concluded that the primary root protrusion test at 15 °C can evaluate super sweet corn seed vigor by counting the number of seedlings at the 2 mm root stage.

Seed vigor testing is an important component of quality control programs adopted by seed industry. The software Seed Vigor Imaging System (SVIS) has been successfully used for seed vigor assessment in different species. The objective of this research was to verify the SVIS efficiency to assess okra seed vigor in comparison to other vigor tests used for this species. Five seed lots of 'Clemson Americano' and four of 'Santa Cruz' were submitted to germination (speed and percentage), cold germination (speed and percentage), traditional and saturated salt accelerated aging (41 ºC/72 and 96 h) and seedling emergence tests during 12 months storage. Vigor index, uniformity of growth and seedling length were determined by the software Seed Vigor Imaging System (SVIS). Results showed that the vigor index and seedling length determined by the SVIS analyses are efficient to determine okra seed vigor as well as the accelerated aging test (41 ºC/72 h).

Standardized seed vigor tests must be developed for greenhouse-grown flower species. Current vigor tests used to evaluate large-seeded agronomic crops are generally not useful for evaluating smaller-seeded flower species. One alternative is to use radicle length in seedlings grown under controlled environments as an indicator of seed vigor. For that purpose, a seed vigor test was developed that uses digital images taken using a flat bed scanner to measure radicle length in small-seeded flower species. A novel, cellulose substrate was used for germinating seeds. It provided similar moisture-holding properties to standard germination blotters used by commercial seed analysts, but is clear. This has allowed for quick image acquisition without removing seedlings from the petri dish. Correlations were made between seedling growth (radicle length, total seedling length, and total seedling area) with other vigor tests (saturated salts accelerated aging) and greenhouse plug flat emergence. For several seed lots of impatiens that varied in initial seed quality, radicle length after 4 days showed good correlations (>R2 = 0.79) with other measures of seed vigor for describing seed quality. This system is an improvement over other attempts to use computer-aided assessment of digital images because it provides digital images that do not vary due to external lighting; it uses software that can evaluate radicle length in a petri dish assay that does not require a slant-board for straight radicle growth; it relies on standard germination technics used by every seed lab; it uses a clear substrate to replace the opaque blotter to allow digital images to be taken within the petri dish; and accurate measurements of seedling parts is performed in under 2 min per petri dish.

The software Seed Vigor Imaging System (SVIS®), has been successfully used to evaluate seed physiological potential by automated analyses of scanned seedlings. In this research, the efficiency of this system was compared to other tests accepted for assessing cucumber (Cucumis sativus L.) seed vigor of distinct seed lots of Supremo and Safira cultivars. Seeds were subjected to germination, traditional and saturated salt accelerated aging, seedling emergence, seedling length and SVIS analyses (determination of vigor indices and seedling growth uniformity, lengths of primary root, hypocotyl and whole seedlings). It was also determined whether the definition of seedling growth/uniformity ratios affects the sensitivity of the SVIS®. Results showed that analyses SVIS have provided consistent identification of seed lots performance, and have produced information comparable to those from recommended seed vigor tests, thus demonstrating a suitable sensitivity for a rapid and objective evaluation of physiological potential of cucumber seeds. Analyses of four-days-old cucumber seedlings using the SVIS® are more accurate and growth/uniformity does not affect the precision of results.

Seed deterioration causes losses to seed companies during the storage. For researches, the challenge has been to develop techniques which improve the seed longevity as well as laboratory analysis methods required for evaluating the evolution of deterioration in storage. This study had the objective of determining the effectiveness of the accelerated aging test with a saturated NaCl solution and Seed Vigor Imaging System (SVIS) analysis in checking soybean seed physiological in comparison with the recommended vigor tests. Soybean seeds from the cultivars BRS 184 and M-Soy 7908 RR were used, each of them were represented by six lots and stored for six months under two environment conditions. Seed physiological quality was checked every two months in storage through germination evaluations, accelerated aging (different methods), tetrazolium test (vigor and viability) seedling emergence, and SVIS analysis. The tetrazolium test is the most advisable method for evaluating soybean seed quality during storage. The results of the saturated salt accelerated aging test with NaCl at 41 ºC for 72 or 96 hours and the SVIS evaluation are also both efficient for estimating the physiological potential of seeds.

During the past 2 decades, automated plug production in the flower seed industry has created important requirements by growers for high-quality flower seeds. Using computerized imaging technology, a new seed vigor testing system, Seed Vigor Imaging System (SVIS), was developed at The Ohio State University. By analyzing the digital images of seedlings, it can detect and measure the length of hypocotyls and radicles separately, and then generate a value for the growth and uniformity each. This system provides a fast, labor-saving and objective approach to measuring seed quality. In this study, its capacity and correlation with field performance was studied and compared with other traditional tests, i.e. standard germination test, germinate rate, and accelerated aging test. Five species (dianthus, cleome, rudbeckia, salvia, and lettuce) were selected and their quality was tracked continuously by SVIS and other mentioned tests. It was found that stressed test (ageing test) was able to detect the quality deterioration earlier than others under ideal conditions, but SVIS could generate much more information, such as the growth, uniformity, and vigor level of the seed lot. Therefore, SVIS following 3-day ageing was developed and shown to be the most sensitive and comprehensive vigor test for those ornamental species mentioned above. Being fast and objective, this system will also benefit the global seed trade by providing a unique quality standard. In addition, it can also be of great usage to seed companies and germplasm centers worldwide for the routine quality track during shipment/storage and inventory management.

Seed production usually requires fast decisions to improve the efficacy of seed handling during harvesting, processing and storage. Seed technologists have focused on the development or improvement of procedures which allow rapid and consistent identification of higher quality seed lots. This research verified the effectiveness of the potassium leachate test on the evaluation of the physiological potential of maize seeds in comparison to recommended seed vigor tests. Five seed lots of the hybrid Avant were submitted to the potassium leachate, standard germination, first count, accelerated aging, electrical conductivity, field emergence and cold tests. The amount of potassium leachate was determined after seed imbibition for 30, 60, 90, 120, 150 and 180 minutes, at 25°C; a flame photometer adjusted to 50 mug K+ mL-1 pattern and reading 50 was used to determine the amount of leached potassium. The potassium leachate test consistently ranked the seed lots according to their physiological quality in comparison to other tests, and is a new alternative test for maize seed quality control programs.

<p>ABSTRACT<br />Information of seed quality during storage can be determined through the actual storage and storability vigor estimation. This study aimed at comparing effective accelerated aging method<br />between physical and chemical, and studying the seed deterioration during storage in ambient (T =28-30 0C, RH=75-78%) and AC (T =18-20 0C, RH =51-60%) condition with three levels of initial moisture content (8-10%, 10-12%, and 12-14%) for 20 weeks. The final objective of this research<br />was to develop model for storability vigor of papaya seed. Two experiments, accelerated aging and seed storage were conducted at Seed Laboratory, Department of Agronomy and Horticulture, Bogor Agricultural University from October 2015 to May 2016. A completely randomized design with nested factors and four replications was applied to both experiments. The results showed that physical accelerated aging using IPB 77-1 MMM machine was more effective than chemical accelerated aging using IPB 77-1 MM machine for papaya seed. The viability of seed stored in AC condition remained high until the end of the storage period, whereas it declined at 16 week storage period in the ambient condition. The viability of seed with initial moisture content of 12-14% declined faster than that of initial moisture content of 8-10% after 18 week storage periode. The model used to estimate the storability vigor of papaya seed accurately was the equation y = a + b expcx where y : storability vigor estimation, x : aging time and a,b,c : constant value. Penelitian ini bertujuan untuk membandingkan metode<br />pengusangan cepat yang efektif antara fisik dengan kimia serta mempelajari pola penurunan viabilitas benih selama penyimpanan aktual pada kondisi simpan kamar (suhu =28-30 0C, RH =75-78%) dan AC (suhu =18-20 0C, RH =51-60%) dengan tiga tingkat kadar air awal (8-10%, 10-12%, dan 12-14%) selama 20 minggu. Tujuan akhirnya adalah membangun model vigor daya simpan benih pepaya. Penelitian pengusangan cepat dan penyimpanan dilakukan pada bulan Oktober 2015 sampai Mei 2016 di Laboratorium Benih, Departemen Agronomi dan Hortikultura, Institut Pertanian<br />Bogor. Kedua penelitian menggunakan rancangan acak lengkap tersarang dengan empat ulangan. Viabilitas benih yang disimpan pada kondisi AC tetap tinggi hingga akhir periode simpan, sedangkan pada kondisi kamar penurunan viabilitas dimulai pada periode simpan 16 minggu. Benih yang disimpan dengan tingkat KA awal sebesar 12-14% lebih cepat mengalami penurunan viabilitas mulai periode simpan 18 minggu dibandingkan dengan benih dengan KA awal 8-10%. Hasil penelitian juga menunjukkan terdapat korelasi yang erat antara pola kemunduran benih pada pengusangan cepat dan penyimpanan aktual, sehingga model pendugaan vigor daya simpan (y) berdasarkan waktu pengusangan (x) dapat disusun dengan persamaan y = a + b expcx. International research on vegetable seed vigor is not at the same level attained for grain crops species. This study was conducted to identify reliable procedures for the accelerated aging and controlled deterioration tests to rank onion (Allium cepa L.) seed lots according to their physiological potential. Six seed lots of the cultivars Aurora and Petroline were evaluated in the laboratory for germination, first count, seedling vigor classification, traditional and saturated salt accelerated aging (41ºC / 48 and 72 h), controlled deterioration (24% of water / 45ºC / 24 h) and seedling emergence tests. Seed moisture content after the saturated salt accelerated aging test was lower and uniform, which is considered an important advantage in comparison to the traditional procedure. The saturated salt accelerated aging (41ºC / 48 and 72 h) and controlled deterioration (moisture content adjusted to 24% / 45ºC / 24 h) tests were the best procedures to assess the physiological potential of onion seeds, and are indicated for use in quality control programs.

Barley (Hordeum vulgare L.) planted area has increased in Brazil after a decline from 2005‒2009, but the country continues to import malt for the brewing industry. The physiological quality of the seed lot is essential for plant establishment and for screening for high yield grain. However, a uniform accelerated aging (AA) procedure has not been developed for testing barley seeds. Thus, this study aimed to determine the best AA method for measuring barley seed vigor. Seeds of three barley cultivars (BRS Cauê, BRS Brau, and MN 6021) were subjected to four AA treatment (T1: 41 °C for 72 h; T2: 42 °C for 48 h; T3: 42 °C for 60 h; and T4: 43 °C for 48 h). The following variables were evaluated using traditional vigor tests: seed viability in the first and last count of the germination test, number of seminal roots, shoot length, and radicle length. In addition, digital images of seedlings were acquired with the Seed Analysis System (SAS®) to compute seedling vigor, growth, and uniformity. AA methods T2 and T3 were effective in separating barley cultivars into vigor categories at day four of the germination test. Computer analysis of digital images of barley seedlings is a valuable tool for testing barley seed vigor. Accelerated aging methods T2 and T3 also gave the best separation in seed lot vigor and seedling growth using the SAS® index, respectively.
