The Old Kladruber horse is the oldest original Czech horse breed &ndash; gene resource, with pedigree records spanning three centuries. Because the population is closed, there is a concern about the loss of genetic variation. The genetic diversity within and amongst sire lines was evaluated using microsatellite markers and based on pedigree information. The DNA analysis, covering 16 microsatellite loci, revealed that the genetic diversity (observed heterozygosity = 0.637, genetic diversity = 0.678 and the mean number of alleles = 8.25) for the Old Kladruber horse is consistent with other populations of horses. Inter-line genetic difference in sire lines reached the mean value (the F<sub>ST</sub> values ranged from 0.020 to 0.017). Differences amongst the sire lines were identified using genetic distances and principal component analysis. One gene cluster comprised black variation sire lines, while the second cluster included the sire lines of the grey variation. In the subjects monitored, the average inbreeding coefficient of 0.076 was estimated on the basis of pedigree information. The highest mean values of the coefficient of relationship were estimated within the colour variations. When comparing the genetic distance and the average coefficient of relationship, differences were found out. These results are useful for the development of breeding strategies which consider classical horse breeding as well as recent achievements of population and conservation genetics.

In cultivated commercial crop species, genetic diversity tends to decrease because of the extensive breeding processes. Therefore, germplasm of commercial crop species, such as Brassica napus L. should be evaluated and the genotypes, which have higher genetic diversity index, should be addressed as potential parental cross materials in breeding programs. In this study, the genetic diversity was analysed by using randomly amplified polymorphic DNA analysis (RAPD) technique in nine Turkish commercial rapeseed varieties. The RAPD primers (10-mer oligonucleotides) produced 51 scorable loci, 31 loci of which were polymorphic (60.78%) and 20 loci (39.22%) were monomorphic The RAPD bands were scored as binary matrix data and were analysed using POPGENE version 1.32. At locus level, the values of genetic diversity within population (Hs) and total (HT) were 0.15 and 0.19 respectively. The genetic differentiation (GST) and the gene flow (Nm) values between the populations were 0.20 and 2.05 respectively. The mean number of alleles (na), the mean number of effective alleles (nae), and the mean value of genetic diversity (He) were 2.00, 1.26, and 0.19 respectively. According to Pearson’s correlation, multiple regression and principal component analyses, eco-geographical conditions in combination had significant effect on genetic indices of commercial B. napus L. varieties were discussed.

China had the largest population of raising donkeys in the world, however the number of Chinese indigenous donkey decreased dramatically due to the increase of agriculture mechanization in the last century. The species has still been important in China because of its edible and medical value, therefore the survey on its genetic diversity in China is necessary for its conservation and utilization. In this study, 15 microsatellite markers were used to evaluate genetic diversity and population structure of five Chinese indigenous donkey breeds. The mean values of expected heterozygosity, allelic richness, and total number of alleles for all the tested Chinese donkeys were 0.70, 6.04, and 6.28 respectively, suggesting that the genetic diversity of Chinese indigenous donkeys is rich. The Bayesian analysis and principal component analysis plot yielded the same clustering result, which revealed that Guanzhong donkey was the most differentiated breed in all detected samples, and Jinnan (JN) and Guangling (GL) were genetically closed together. Additionally, our results indicated that the heterozygote deficit was severe in two Chinese indigenous donkey breeds (GL and JN), and it warned us that animal conservation activities on this species should be considered carefully in near future.

O objetivo do trabalho foi de caracterizar a diversidade e a estrutura genética de 82 indivíduos de P. adamantinum, espécie de campos rupestres e de altitude do Cerrado brasileiro, na região central de Minas Gerais, divididos em 02 populações de 41 indivíduos. Foram utilizados 08 primers ISSR que amplificaram 135 locos, com 108 (80%) locos polimórficos entre as populações e média de 16,9 locos por primer. O valor médio de PIC foi de 0,375. Os valores médios do índice de Shannon (I) e da diversidade genética de Nei (He) foram de 0,491 e 0,341, respectivamente. O índice da proporção da diversidade genética (GST = 0,047) demonstrou que a variabilidade entre e dentro das populações contribuiu com 4,7% e 95,3% da heterozigosidade total (HT), respectivamente. O fluxo gênico observado foi alto (Nm = 10,213), o que contrapõe os efeitos da deriva genética. Foi observada a ocorrência de baixa estruturação genética espacial (EGE) (Sp= 0,058) e coancestria positiva nas duas populações (Fij = 0,016; P < 0,123 em POP01 e Fij = 0,018; P < 0,057 em POP02), ambas avaliadas até a segunda classe de distância. Mesmo com a presença de fatores favoráveis à endogamia, P. adamantinum apresentou altos índices de diversidade genética. In Belgium, a total of three Vaccinium species with a shrub or dwarf shrub growth form can be found: Vaccinium myrtillus L., Vaccinium vitis-idaea L., and Vaccinium uliginosum L. Vaccinium uliginosum is the only one of these for which the extent of clonality is unknown. Therefore, the clonal structure of two V. uliginosum populations was inferred from random amplified polymorphic DNA (RAPD) markers. Among the 47 sampled patches, 61 clones were identified. The mean values of the number of clones divided by the number of samples (G/N), the Simpson's index (D), and the genotypic evenness (E) were 0.28, 0.95, and 0.96, respectively. Vaccinium uliginosum exhibited a typical phalanx growth strategy that resulted in structured populations at the ramet level, that is, ramets belonging to the same clone were closely associated and formed distinct clumps. However, at the clone level, populations were not structured, that is, genetic distances between pairs of clones were not correlated with the spatial distances between the clones within a population. Genetic diversity was as high as that in nonclonal species (mean value of Shannon's diversity index (Ho) = 0.647). In accord with the life history traits of V. uliginosum (long-lived species with a mixed breeding system and potentially high seed dispersal), most of the genetic variation was found within populations.Key words: genetic diversity, genetic structure, seedling recruitment, vegetative propagation, Ericaceae.

Seasonal variations in the stable isotopic composition of snow and meltwater were investigated in a sub-arctic, mountainous, but non-glacial, catchment at Okstindan in northern Norway based on analyses of δ18O and δD. Samples were collected during four field periods (August 1998; April 1999; June 1999 and August 1999) at three sites lying on an altitudinal transect (740–970 m a.s.l.). Snowpack data display an increase in the mean values of δ18O (increasing from a mean value of −13.51 to −11.49‰ between April and August), as well as a decrease in variability through the melt period. Comparison with a regional meteoric water line indicates that the slope of the δ18O–δD line for the snowpacks decreases over the same period, dropping from 7.49 to approximately 6.2.This change points to the role of evaporation in snowpack ablation and is confirmed by the vertical profile of deuterium excess. Snowpack seepage data, although limited, also suggest reduced values of δD, as might be associated with local evaporation during meltwater generation. In general, meltwaters were depleted in δ18O relative to the source snowpack at the peak of the melt (June), but later in the year (August) the difference between the two was not statistically significant. The diurnal pattern of isotopic composition indicates that the most depleted meltwaters coincide with the peak in temperature and, hence, meltwater production.

The food needs for increasing population, climatic changes, urbanization and industrialization, along with the destruction of forests, are the main challenges of modern life. Therefore, it is very important to evaluate plant genetic resources in order to cope with these problems. Therefore, in this study, a set of ninety-one walnut (Juglans regia L.) accessions from Central Anatolia region, composed of seventy-four accessions and eight commercial cultivars from Turkey, and nine international reference cultivars, was analyzed using 45 SSR (Simple Sequence Repeats) markers to reveal the genetic diversity. SSR analysis identified 390 alleles for 91 accessions. The number of alleles per locus ranged from 3 to 19 alleles with a mean value of 9 alleles per locus. Genetic dissimilarity coefficients ranged from 0.03 to 0.68. The highest number of alleles was obtained from CUJRA212 locus (Na = 19). The values of polymorphism information content (PIC) ranged from 0.42 (JRHR222528) to 0.86 (CUJRA212) with a mean PIC value of 0.68. Genetic distances were estimated according to the UPGMA (Unweighted Pair Group Method with Arithmetic Average), Principal Coordinates (PCoA), and the Structure-based clustering. The UPGMA and Structure clustering of the accessions depicted five major clusters supporting the PCoA results. The dendrogram revealed the similarities and dissimilarities among the accessions by identifying five major clusters. Based on this study, SSR analyses indicate that Yozgat province has an important genetic diversity pool and rich genetic variance of walnuts.

Conventionally, the strength of toe plantar flexion (STPF) is measured in a seated position, in which not only the target toe joints but also the knee and particularly ankle joints, are usually restrained. We have developed an approach for the measurement of STPF which does not involve restraint and considers the interactions of adjacent joints of the lower extremities. This study aimed to evaluate this new approach and comparing with the seated approach. A thin, light-weight, rigid plate was attached to the sole of the foot in order to immobilize the toe area. Participants were 13 healthy young women (mean age: 24 ± 4 years). For measurement of STPF with the new approach, participants were instructed to stand, raise the device-wearing leg slightly, plantar flex the ankle, and push the sensor sheet with the toes to exert STPF. The sensor sheet of the F-scan II system was inserted between the foot sole and the plate. For measurement with the seated approach, participants were instructed to sit and push the sensor with the toes. They were required to maintain the hip, knee, and ankle joints at 90°. The mean values of maximum STPF of the 13 participants obtained with each approach were compared. There was no significant difference in mean value of maximum STPF when the two approaches were compared (new: 59 ± 23 N, seated: 47 ± 33 N). The coefficient of variation of maximum STPF was smaller for data obtained with the new approach (new: 39%, seated: 70%). Our simple approach enables measurement of STPF without the need for the restraints that are required for the conventional seated approach. These results suggest that the new approach is a valid method for measurement of STPF.

Background:Boolean remission criteria is one most popular and stringent criteria in treating patient with rheumatoid arthritis (RA), because it may guarantees a stable clinical course after attaining remission.Objectives:Impact of time span from initiation to achieving Boolean remission on maintaining disease activity, daily activities, and quality of life after attaining Boolean remission was investigated from daily clinical practice data.Methods:685 patients with RA since August 2010 under the T2T strategy were treated. They were monitored for their TJC, SJC, PGA, EGA, CRP, and disease activity indices such as CDAI, SDAI, DAS28, and Boolean criteria at every visit. HAQ-DI score, pain score using visual analog scale (PS-VAS), and EQ-5D were also monitored, and the quality of life score (QOLS) calculated from EQ-5D was determined at every visit from the time of diagnosis (baseline).Of 685 patients, 465 patients had achieved Boolean remission >1 times, and were consecutively followed up for >3 years. These patients were enrolled in the study. Time span from the first visit to first Boolean remission was calculated. The relationship between the time span and each of background parameters, and the relationship between the time span and each of the mean values of the SDAI score, HAQ score, PS-VAS, SHS, and QOLS at the first Boolean remission and thereafter was evaluated statistically.Patients were subsequently divided into the G ≤ 6 and G > 6 groups based on the achievement of first Boolean remission within two groups: time span G ≤ 6 months and G > 6 months. The two groups were compared with regard to the SDAI score, HAQ score, PS-VAS, SHS, and QOLS at first visit and at the time of first Boolean remission, and the mean values of these parameters after remission were evaluated statistically. Moreover, changes of these parameters and the mean Boolean remission rate after the first remission, and SDAI remission rate at the first Boolean remission to thereafter were compared between the two groups statistically.Results:Out of 465 patients, females comprised 343 (73.7%), and the mean age was 67.8 years (range, from 21–95 years). The mean disease duration at first visit was 6.1 years (range, from 1 months–45 years). The mean follow up length was 88.1 months (range: 36–122 months; median: 85 months) and mean time span from the first visit to the first Boolean remission was 8.1 months. The mean SDAI score, HAQ score, PS-VAS, and the QOLS at first visit were 13.3, 0.467, 33.2, and 0.834, respectively. Among the study parameters, PS-VAS and QOLS were significantly correlated with the time span. For parameters at the first Boolean remission, HAQ-DI score, PS-VAS, and QOLS demonstrated significant correlation with the time span, whereas SDAI, HAQ-DI score, PS-VAS, SHS, and QOLS after the Boolean remission demonstrated significant correlation with the time span.The comparison between the G ≤ 6 and the G > 6 groups revealed that the disease duration, HAQ score, and PS-VAS at baseline in the G > 6 were significantly higher than that in the G ≤ 6 group, and QOLS in the G ≤ 6 group was significantly higher than that in the G > 6 group at baseline. Similarly, the HAQ score and PS-VAS at the first Boolean remission in the G > 6 group were significantly higher than that in the G ≤ 6 group, whereas QOLS in the G ≤ 6 group demonstrated no significant difference compared with that in the G > 6 group.The mean value of the SDAI score after the first Boolean remission in the G > 6 group was significantly higher than that in the G ≤ 6 group. Similarly, the SDAI score, HAQ score, and PS-VAS after the first Boolean remission in the G > 6 group were also significantly higher than those in the G ≤ 6 group, and the mean value of the QOLS in the G ≤ 6 group were significantly higher than that in the G > 6 group. The Boolean remission rate and SDAI remission rate after the first Boolean remission were significantly higher in the G ≤ 6 group than those in the G > 6 group.Conclusion:Attaining Boolean remission ≤ 6 months for RA has significant benefit for more stable disease control, that leads good maintenance of ADL.Disclosure of Interests:None declared

The emissions of carbon monoxide, carbon dioxide, and hydrocarbon from four stroke-powered motorcars and two stroke-powered motorcycles and tricycles in Southwest Nigeria were examined using an automotive 4-gas analyer. Results show that tricycles produced more hydrocarbon and carbon monoxide emissions than motorcycles, while motorcycles emitted more of these pollutants than the gasoline fueled motor cars. (The gasoline fueled motorcars produced lowest hydrocarbon and carbon monoxide while the tricycles produced the highest hydrocarbon and carbon monoxide emissions). On the contrary, motor cars had the highest mean value of carbon dioxide followed by the motorcycles, while tricycles had the least. This could be attributed to the presence of the catalytic converters in some of the motor cars oxidizing carbon monoxide to carbon dioxide. The mean values of hydrocarbon, carbon monoxide and carbon dioxide emissions from motorcars are 630ppm, 10200ppm and 59900ppm. This is much higher than the NESREA (National Environmental standards and Regulations Enforcement Agency) standards as well as Euro II and Euro III (European standards) for vehicular emission. The mean values for hydrocarbon, carbon monoxide and carbon dioxide emissions from motorcycles and tricycles are (2150ppm, 21530ppm and 31200ppm) and (2820ppm, 24880ppm and 38710ppm) respectively. These results do not comply with Nigeria and European emission standards for hydrocarbon, and carbon monoxide. Tricycles and motorcycles account for higher concentrations of hydrocarbon and carbon monoxide pollutants from mobile sources, while they emit carbon dioxide minimally.

Genetic divergence of 40 parental lines comprising 30 restorer and 10 maintainer lines were studied through Mohalanobis's D2 and principal component analysis for eleven characters. Genotypes were grouped into five different clusters. Cluster V comprised maximum number of genotypes (thirteen) followed by cluster I and II. The inter-cluster distance was maximum between clusters I and V (13.495) indicating wide genetic diversity between these two clusters followed by the distance between cluster I and 11 (9.489), cluster IV, and cluster V (8.969) and cluster I and cluster III (8.039). The minimum inter-cluster distance was observed between cluster II and cluster III (3.034) followed by cluster 111 and cluster IV (3.834) and cluster II and cluster V (4.945) indicating that the genotypes of these clusters were genetically close. The intra cluster distance in the entire five clusters was more or less low which indicated that the genotypes within the same cluster were closely related. Among the characters panicle weight contributed most for divergence in the studied parental lines. Difference in cluster means existed for almost all the characters studied. Highest mean value for number of effective tillers (7.8), days to 50% flowering (95.5), panicles/m2 (192.6), panicle weight (2.9), spikelet fertility (84.8), number of grains/panicle (177.8), days to maturity (123.6), and grain yield/plot (1065.5) were observed in cluster I indicated the parental lines fallen in this cluster having the genetic potentiality to contribute better for yield maximization of hybrid rice.
