Syzygium filiforme var. filiforme is a plant variety from dicotyledonous plant family (Myrtaceae). Phytochemical studies on S. filiforme var. filiforme stem bark have successfully isolated and characterized arjunolic acid (1), alphitolic acid (2), betulinic acid (3), ursolic acid (4), ursolic acid 3-methyl ester (5), β-sitosterol (6) and stigmasterol (7). The inhibitory activities against free radical, starch, and bacteria for major compounds were tested by using DPPH, α-glucosidase and minimum inhibitory and bacterial concentration assays, respectively. No promising antioxidant activity was shown on tested samples except methanolic crude extract. For antidiabetic activity, methanolic and dichloromethane crude extracts displayed potent activity compared to 1-deoxynojirimycin. Minimum inhibitory concentration (MIC) and minimum bacterial concentration (MBC) assays for antibacterial activity were evaluated on Escherichia coli, Staphylococcus aureus, and Bacillus subtilis. All crude extracts and major compounds displayed weak and no promising activities for MIC method, respectively. Meanwhile, for MBC method, hexane crude extract and compound 1 showed inhibition against B. subtilis.

The methanol extract of Acacia ferruginea (Family- Mimosaceae) was studied for possible antidiabetic activity on normoglycaemic, OGTT and alloxan induced diabetic rats at doses of 100, 200, and 400 mg/kg, p.o. The acute toxicity studies were carried out on Swiss albino mice to determine the LD50 values. The experiments were performed as per OECD guidelines. The results of the normoglycaemic, OGTT and hyperglycaemic studies revealed that the extract exhibited reduction in blood glucose concentration in a dose dependant manner as compared to the standard drug metformin (250 mg/kg, p.o.). The acute toxicity studies revealed no signs of mortality in animals treated with a single dose of 2000 mg/kg body weight. Preliminary phytochemical studies of the methanol extract revealed presence of alkaloids, steroids, triterpenoids, saponins, flavonoids, tannins and phenolic compounds, carbohydrates, gums and mucilages, proteins and amino acids. The present study justifies the use of the plant for treating diabetes as suggested in folklore remedies.

Abstract The development of host resistance to anthelmintics and the increasing cost of commercial anthelmintics have encouraged the need for the in vitro anthelmintic evaluation of crude extract and fractions of Hymenodictyon pachyanta plant as alternative drugs against Haemonchus contortus. H. contortus is one of the most prevalent and highly pathogenic parasitic nematodes in small ruminant farming globally. H. pachyanta stem bark is a prospective plant used by the local and indigenous farmers of Nsukka, Enugu state, Nigeria. The stem bark of H. pachyanta were collected, dried, pulverized and extracted with 80 % methanol. The purpose of this study was to investigate the in vitro anthelmintic effects of these crude extract and fractions against H. contortus in sheep and goats. The two extracts (crude and fractions) of H. pachyanta were tested by the egg hatch assay (EHA) and the larval development inhibition assays (LDIA) and to compared the results with albendazole (as the positive control). The concentrations for the crude extract and albendazole used for this study were 0.78, 1.56, 3.125, 6.25 and 12.5 mg.ml−1. The results demonstrated that the crude extracts, fractions and albendazole all at the concentration doses of 12.5 mg.ml−1 produced 100 % inhibition of egg hatching and larval development. Statistically, there was no significant difference (P > 0.05) in the mean percentage inhibition of egg hatching and larval development inhibition of the crude extracts and fractions when compared with albendazole. However, a significant difference (P < 0.05) was observed with n-butanol fraction which inhibited 96.17 % of egg hatchability. All of the extracts and albendazole showed ovicidal and larvicidal effects and were able to induce over 50 % of the egg hatching and mortality of larvae at the concentration ranges of 0.78—12.5 mg.ml−1. The results obtained from our study suggest that H. pachyanta had ovicidal and larvicidal activity against H. contortus and that the bioactive plants compounds responsible for this effect could be attributed to the presence of tannins, alkaloids and the saponins contained in the crude extracts.

This study is aimed at investigating the antiplasmodial activity and acute toxicity of the methanol (MeOH) extracts of the leaves and roots, and the dichloromethane (DCM) extracts of the stem bark, leaves, and roots of Goniothalamus lanceolatus. Phytochemical analysis was then carried out on the most active extract. In vivo antiplasmodial activity was assessed using the 4-day suppressive test against Plasmodium berghei ANKA (PbANKA) in mice. The plant extracts were administered intraperitoneally (i.p.) as a single dose (30 mg/kg) starting 4 h after infection. At a dose level of 30 mg/kg (i.p.), the DCM extracts of the stem bark and leaves, and the MeOH root extracts, prolonged the survival period of infected mice compared to that of the negative control. In addition, all crude extracts, except for the DCM root extract, exhibited parasitemia suppressive activity. The highest level of parasitemia suppression was recorded in mice treated with the DCM stem bark extract at 66.3%. No mortality was observed in mice treated with the DCM extracts of the stem bark and leaves, and the MeOH extract of the leaves, indicating that the LD50 is greater than 300 mg/kg. On the other hand, both the MeOH and DCM extracts of the roots showed toxic effects at a dose of 300 mg/kg (i.p) with an 83.3% mortality rate. The results obtained indicate that the stem bark of G. lanceolatus (DCM crude extract) possesses good antiplasmodial activity against PbANKA infected mice without causing acute toxicity. Five known styrylpyrone derivatives namely goniodiol 1, 8-epi-9-deoxygoniopypyrone 2, 9-deoxygoniopypyrone 3, digoniodiol 4 and goniothalamin 5 have been isolated from the bark of Goniothalamus lanceolatus (DCM crude extract). The structures and stereochemistry of all compounds were elucidated by interpretation of spectroscopic data. This study provides a scientific basis to support the traditional use of the plant as a remedy for malaria.

Background: Aspergillus Versicolor is a marine-derived fungus isolated from Hyrtios Erectus Red Sea sponge. Methods: The aim of this study was to carry out a pharmacological screening and investigation for the in vitro biological activity (antioxidant, cholinergic, antidiabetic and anticancer) of Aspergillus Versicolor crude extract’s active compounds by using different qualitative and quantitative methods. Results: The present study results showed that Aspergillus Versicolor crude extracts contain 0.6 mg total phenolic/mg crude extract. Aspergillus Versicolor also showed a potent antioxidative capacity by decreasing the oxidation of ABTS. The anticancer and inhibitory effects of Aspergillus Versicolor crude extracts on PTK and SHKI were found to be 75.29 % and 80.76%; respectively. The AChE inhibitory assay revealed that Aspergillus Versicolor extracts had an inhibitory percentage of 86.67%. Furthermore, the anti-inflammatory activity using COX1, COX2, TNF, and IL6 was 77.32, 85.21 %, 59.83%, and 56.15%; respectively. Additionally, the anti-viral effect using reverse transcriptase enzyme showed high antiviral activity with 92.10 %. Conclusion: The current study confirmed that the Aspergillus versicolor crude extract and its active constituents showed strong effects on diminishing the oxidative stress, neurodegenerative damage, antiinflammatory, anti-cancer and anti-viral, suggesting their beneficial role as a promising fermented product in the treatment of cancer, oxidative stress, Alzheimer's, anti-inflammatory and anti-viral diseases.

Cordia dichotoma Forst. (F. Boraginaceae) has been traditionally used for the management of a variety of human ailments. In our earlier work, the antidiabetic activity of methanolic bark extract of C. dichotoma (MECD) has been reported. In this paper, two flavonoid molecules were isolated (by column chromatography) and identified (by IR, NMR and mass spectroscopy/spectrometry) from the MECD with an aim to investigate their antidiabetic effectiveness. Molecular docking and ADMET studies were carried out using AutoDock Vina software and Swiss ADME online tool, respectively. The isolated flavonoids were identified as 3,5,7,3′,4′-tetrahydroxy-4-methoxyflavone-3-O-L-rhamnopyranoside and 5,7,3′-trihydroxy-4-methoxyflavone-7-O-L-rhamnopyranoside (quercitrin). Docking and ADMET studies revealed the promising binding affinity of flavonoid molecules for human lysosomal α-glucosidase and human pancreatic α-amylase with acceptable ADMET properties. Based on computational studies, our study reports the antidiabetic potential of the isolated flavonoids with predictive pharmacokinetics profile.

The stability (longevity of activity) of three crude urease extracts was evaluated in a laboratory study as part of an effort to reduce the cost of urease for applications that do not require high purity enzyme. A low-cost, stable source of urease will greatly facilitate engineering applications of urease such as biocementation of soil. Inexpensive crude extracts of urease have been shown to be effective at hydrolyzing urea for carbonate precipitation. However, some studies have suggested that the activity of a crude extract may decrease with time, limiting the potential for its mass production for commercial applications. The stability of crude urease extracts shown to be effective for biocementation was studied. The crude extracts were obtained from jack beans via a simple extraction process, stored at room temperature and at 4 ℃, and periodically tested to evaluate their stability. To facilitate storage and transportation of the extracted enzyme, the longevity of the enzyme following freeze drying (lyophilization) to reduce the crude extract to a powder and subsequent re-hydration into an aqueous solution was evaluated. In an attempt to improve the shelf life of the lyophilized extract, dextran and sucrose were added during lyophilization. The stability of purified commercial urease following rehydration was also investigated. Results of the laboratory tests showed that the lyophilized crude extract maintained its activity during storage more effectively than either the crude extract solution or the rehydrated commercial urease. While incorporating 2% dextran (w/v) prior to lyophilization of the crude extract increased the overall enzymatic activity, it did not enhance the stability of the urease during storage.

Phytochemical study on the stem bark of P. arborescens has isolated six secondary metabolites from the crude extracts by using column chromatography, while identification was performed by using Gas Chromatography - Mass Spectrometry (GC-MS), Nuclear Magnetic Resonance (NMR) and Fourier Transform Infrared (FTIR). The isolated metabolites were identified as caryophyllene oxide (1), α-bisabolol (2), benzamide 2-(methylamino) (3), 2-ethylpiperidine (4), piperine (5) and methyl eugenol (6). Toxicity test on the four crude extracts of P. arborescens shows a high cytotoxicity against Artemia salina brine shrimp with LC50 values ranging from 13.12 to 58.70 µg/mL. Greater cytotoxicity of the crude extracts of P. arborescens indicated the presence of potent cytotoxic components in this Piper spp. Antioxidant assay of P. arborescens against 2-diphenyl-1-picrylhydrazyl (DPPH) indicated moderate antioxidant activities of methanol, dichloromethane, chloroform and hexane crude extracts with EC50 values of 21.68, 23.82, 32.88 and 36.88 µg/mL, respectively.

Analyses of the constituents of the crude extracts obtained from young and mature moss species namely: Archidium ohioense and Philonotis hastata were conducted with a view to investigating the effects of maturity stages on their bioactive constituents. The mosses collected from their natural population were air dried at ambient temperature in the laboratory, extracted with methanol and the crude extracts subjected to gas chromatographymass spectrometry (GC-MS) analysis. The results of the analyses showed the presence of 20 compounds in young A. ohioense with n-hexadecanoic acid [26.60%], bis(2-ethylhexyl) phthalate [12.47%], bicyclo (3.1.1)heptane 2,6,6-trimethyl-[1r-(1.alpha.,2.beta.,5.alpha.)]- [11.59%] and phytol [9.69%] forming the prominent components while in the mature A. ohioense, 13 compounds were present, from which n-hexadecanoic acid [51.25%], hexadecanoic acid 2-hydroxy-1-(hydroxymethyl)-ethyl ester [9.90%] and n-propyl 9-octadecenoate [7.47%] formed the prominent components. In P. hastata, 20 compounds were identified in the young stage sample with n-hexadecanoic acid [22.46%], bis(2-ethylhexyl) phthalate [20.95%] and phytol [18.14%] as the prominent components while 9 compounds were identified in the mature sample with n-hexadecanoic acid[51.84%], hexadecanoic acid 2-hydroxy-1-(hydroxymethyl)ethyl ester [18.12%] and bis(2-ethylhexyl) phthalate [9.11%] which formed the prominent components. The study indicated that, maturity stages at collection of the mosses affected their bioactive compositions, with the young stage mosses showing more bioactive compounds than the mature ones.
