Acute hypoxia influence on cardio-respiratory system and new possibilities of pharmacological prophylactics of hypoxia in experiment

Cardiorespiratory system activities have been studied on rats in experiment after oral introduction of the new selenium-containing metal-complex substance πQ1983 in dose 100 mg/kg before and under action of acute hypercapnic hypoxia (AH + Hc). The substance was introduced 90 min. before (incubation period) placement of animals into hypoxic chambers with 1.0 L free volume. During each experiment as well as during AH+Hc an electrical activity of myocardium (ECG) and respiration curves called pneumobarogramm (PBG) were recorded simultaneously. It has been established that the substance πQ1983 made cardiodepressive effect and decreases parameters of lungs ventilation in animals. According to ECG and PBG dynamics, rats protected by the substance demonstrated a high resistance level to the aroused acute hypoxia with hypercapnia, that expressed by weakening of early reactions from cardiorespiratory system under hypercapnia action, and by twice longer life span of animals in hypoxic experiment.

Changes of external respiration parameters have been studied in experimental rats after oral introduction of new selenium-containing metal-complex substance πQ1983 in dose 100 mg/kg before and under action of acute hypercapnic hypoxia (AH+Hc). A substance for comparison was metabolic antihypoxant amthizole administered in similar way and dosage. The substances were introduced 90 min before (incubation period) placement of animals into hypoxic chambers with 1.0 L free volume. During each experiment as well as during AH+Hc a respiration curve called pneumobarogramm (PBG) was recorded continually. It has been established that the substance πQ1983 decreases parameters of lungs ventilation in the rats. According to PBG dynamics, rats protected by the substance demonstrated a high resistance level to the aroused acute hypoxia with hypercapnia, that expressed by weakening of early reactions from respiratory system under hypercapnia action, twice longer life span of animals in hypoxic experiment, and by rising of possibility to withstand low oxygen concentration. It has been proved that the substance πQ1983 significantly surpass a substance for comparison amthizole in both antihypoxic activity and influence on external respiration parameters.

Fine changes of electrical activities in rat myocardium were studied after oral introduction of new selenium-containing metal-complex substance πQ1983 in dose 100 mg/kg before and after an acute hypercapnichypoxia (AH + Hc) development. A substance for comparison — metabolic antihypoxant amthizole was used by same way and dose. The substances were introduced 90 min before (incubation period) placing of animals into hypoxic chambers with 1.0 L free volume. ECG was recorded continually during both an incubation period and a hypoxic experiment. It has been established that the inhibitory action of the substance πQ1983 on electrical activity of rat myocardium manifests itself like a significant bradycardia. According with ECG data, rats protected with substance πQ1983 show the resistance level to the AH + Hc higher than ones that intake the antihypoxant amthizole. It was proved by peculiarities of early myocardial hypercapnic reactions, parameters of lifespan, and length of so called relative wellbeing period.

PURPOSE: To study the effect of amtizol, 2-aminobenzthiazole (2-ABT) and 2-amino-4-acetylthiazolo[5,4-b]indole (BM-606) on the resistance of male outbred mice to acute hypoxia with hypercapnia under conditions of isolated functioning of one from the hemispheres, as well as both hemispheres of the brain. METHODS: A model of acute hypoxia with hypercapnia (canned hypoxia) was used in mice of the same mass, the lifespan of all animals was determined. Temporary shutdown of the cortex of one of the hemispheres or both hemispheres was achieved by epidural application of filter paper moistened with 25% potassium chloride solution, creating a spreading depression according to Leao. Amtizol, 2-aminobenzthiazole (2-ABT) and 2-amino-4-acetylthiazolo[5,4-b]indole (BM-606) at equimolar doses of 25, 32.5, and 50 mg/kg, respectively were used as pharmacological analyzers, the compounds were injected intraperitoneally 30 min before the hypoxic episode. RESULTS: It was shown that, in contrast to amtizol, 2-ABT and VM-606 increase the life time of experimental animals when any hemisphere is turned off. The use of drugs when both hemispheres were turned off revealed that amtizol has approximately equal effect on the brain and the rest of the body, in 2-ABT antihypoxic activity is 1/3 associated with the brain, in VM-606 exclusively with the brain. CONCLUSION: The experimental model used in this work makes it possible to quite easily evaluate the effect of either one drug or compare several drugs, their role in the functioning of the cerebral hemispheres, on which part of the sample highly resistant or low resistant to hypoxia they have the greatest effect.

The value of standard energy metabolism had measured on rats after intragastral (oral) administration of the new selenium containing metal-complex substance πQ1983 in dose 100 mg/kg. The well-known antihypoxant amthizol was used as a substance of comparison in a same dose. The substance πQ1983 administered 90 min before study was shown to decrease the activity of energy processes in organism from 194.4 ± 0.7 kcal/day/kg to 74.5 ± 0.5 kcal/day/kg but effect of amthizol was not reliable. It was revealed also that both substances (πQ1983 significantly, amthizol slightly) decreased the oxygen consumption rate during rising acute exogenous hypoxia that could form high resistance level to oxygen insufficiency in rats. Probably, the decrease of oxygen consumption is due to the inhibitory effects of the substances studied, especially substance πQ1983, on active energetic processes in the organism.

Aim. Comparative analysis of the efficiency of metal-complex (πQ1983, πQ2721) and aminothiol (Amtizole, Sunazole) antihypoxants in experiments on rats exposed to acute hypoxia with hypercapnia. Methods. Experiments performed on 182 male rats of Wistar line weighing 150-170 g. The study of antihypoxic activity of substances was carried out on the model the AH+Hc. The condition of acute hypoxia in rats was formed by placing them in glass airtight containers with a free volume of 1.0 L. Antihypoxic effect was evaluated by the life expectancy of animals in the described conditions. Substances πQ2721, πQ1983, Amtizole and Sunazole was administered once intraperitoneally at doses of 25, 50 and 100 mg/kg. Previously each substance was dissolved in 0.9% NaCl (1.0 ml). Testing the effectiveness of the substances on AH+Hc model was carried out after 1 h after administration of the substances and after 24 h. Animals of control groups were injected with 1.0 ml of 0.9% NaCl. In animals exposed to test AH+Hc in 1 h after administration were performed measurements of the rectal temperature before the experiment and through 1 h after administration, i.e. before AH+Hc. In animals selected for 24-hour observation, rectal temperature was measured before the experiment, and then after 1, 3, 6, 12, 18 and 24 h of observation, after which they were exposed to AH+Hc. Results. The antihypoxic effect of a selenium-containing substance πQ2721 based on Zn2+ was confirmed in experiments on rats. In a number of substances for comparison the πQ2721 proved himself not only as equally effective. It is found that after 1 h after administration at a dose of 50 mg/kg πQ2721 superior to all studied compounds, including antihypoxant with succinate Sunazole. Crassocephalum crepidioides on Cornfields in Dairi Regency had been reported tobecome more difficult to control using paraquat. The objective of the research was todetermine the characteristics and the distribution of C.crepidioides resistant to paraquatin cornfields. The experiment was carried out in two steps, the first step was screeningthe population of C. crepidioides with paraquat at the recommended dose, and the secondstep, dose-response experiment for the resistance level of C. crepidioides population withdose 0, 76, 152, 304,5, 609, 1218, and 2436 g.ai /ha. In the first step experiment, paraquatdichloride was applied at 280 g.ai/ha. The treatments were arranged in a randomized blockdesign with 3 replication. The second step experiment was that the resistant populationsconfirmed in the first experiment were sprayed for their dose-response. The treatmentswere arranged in a randomized complete block design (CRBD). The results showed thatof 30 populations of C. crepidiodes, 19 populations (63.3%) were categorized to beresistant with the mortality ranging from 10.84% to 52.08%, and 11 populations (36.7%),was categorized as high resistance with mortality of 0% to 9.21%. The level ofresistance (R/S) of R-C25, R-C27, and R-C30 populations of C. crepidioides were 12,3,14,86, and 24,83 times consecutively, compared with the susceptible population. Thenumber of C. crepidioides chlorophyl leaves in susceptible populations was significantlylower than that of a resistant populations.

Bos taurus cattle with high resistance to the tick Boophilus micro plus, whether free-grazing or in covered pens, had significantly more arteriovenous anastomoses (A VA) in their skin than did animals of low resistance. These differences in number of A VA associated with resistance level were most marked above the level of the sebaceous gland in the neck region, an area favoured for tick feeding. In this skin layer, the number of AVA in low-resistance animals (4�0�0�4 per 2�1 mm) was significantly lower than in animals of high resistance (12�3 � 2�2 per 2�1 mm) while the mean value for the naive animals (8�2 � 1 �9 per 2�1 mm) was intermediate. No differences in morphology of AVA were detectable between the three groups using light microscopy.

ABSTRACT The Mycobacterium tuberculosis mmpL7 gene, encoding a hypothetical resistance nodulation division transporter, confers a high resistance level to isoniazid when overexpressed in Mycobacterium smegmatis. The resistance level decreased in the presence of the efflux pump inhibitors reserpine and CCCP (carbonyl cyanide m-chlorophenylhydrazone). Energy-dependent efflux of isoniazid from M. smegmatis cells expressing the mmpL7 gene was observed.

The experiments were performed in male albino outbred mice kept in a group and under the conditions of long-term social isolation. The changes in the monoaminergic systems of the left and right hemispheres of the brain after acute hypoxia with hypercapnia have been studied. The levels of dopamine (DA), serotonin (5-HT) and their metabolites – dioxyphenylacetic (DOPAC), homovanillic (HVA), and 5-hydroxyindoleacetic (5-HIAA) acids – were determined by HPLC in the cerebral cortex, hippocampus and striatum of the right and left sides of the brain. In the control mice kept both in the group and under the conditions of social isolation, a higher content of DA in the cortex of the left hemisphere has been found. In the other brain structures the monoamine content was symmetric. In the cerebral cortex of the mice in the group, acute hypoxia with hypercapnia led to a right-sided increase in the DA and 5HT levels. At the same time, the DOPAC content decreased in the left cortex. In mice in the group, under the hypoxia with hypercapnia conditions, the DA level in the left hippocampus increased. In the striatum, the content of monoamines and their metabolites did not change significantly. In animals kept for a long time under the conditions of social isolation, hypoxia with hypercapnia no statistically significant changes in the monoamines and their metabolites levels were found. It has been concluded that the preliminary maintenance under the conditions of prolonged social isolation changes the reaction of central monoaminergic systems to acute hypoxia with hypercapnia.
