Background: Dental caries and periodontal disease can be caused by bacteria that attack hard and soft tissues in the oral cavity like Staphylococcus aureus and Streptococcus mutans bacteria One way to prevent infections caused by the bacteria are by using mouthwash. Currently, mouthwash that is widely used is mouthwash containing chlorhexidine, which has side effects in the form of tooth staining when used prolonged. The need for alternative medicines from nature that effectively cope with polymicrobial infections. One of the marine products that has pharmaceutical properties is squid, especially the ink it produces.Methods: This research is a pure experimental study (true experimental design) with a randomized pretest-posttest control group design. Squid ink Loligo sp. extract, obtained by extracting squid ink Loligo sp., with maceration method using 96% ethanol solvent. This research uses serial dilution method with spectrophotometric testing method.Results: After measuring the turbidity value in each treatment tube, it was found that at a concentration of 1.56% squid ink Loligo sp. extract began to inhibit the growth of Staphylococcus aureus and Streptococcus mutans bacteria.  This is proven because the absorbance value after and before incubation is fixed.Conclusions: The minimum inhibitory concentration of squid ink Loligo sp. extract on the growth of Staphylococcus aureus and Streptococcus mutants at a concentration of 1.56%.

Abstract: Noni fruit (Morinda cittrifolia L.) is an herbal plant that contains antibacterial substances, inter alia acubin, lasperuloside, alizarin, and some antraquinons, that have been proved to inhibit the growth of Streptococcus mutans. This study was aimed to obtain the minimum inhibitory concentration (MIC) of noni fruit extract (Morinda citrifolia L.) against the growth of Streptococcus mutans. This was a true experimental study with a randomized pretest-posttest control group design. This study used a serial dilution method with turbidimetry and spectrophotometry tests. Noni fruit was extracted by using maceration method with 96% ethanol. Streptococcus mutans bacteria were obtained from pure stock of Microbiology Laboratory of Pharmacy Program Study, University of Sam Ratulangi Manado. The results showed that the MIC of noni fruit extract against the growth of Streptococcus mutans was at 50% of concentration. Jenis penelitian ialah eksperimental murni dengan randomized pretest-posttest control group design. Metode yang digunakan yaitu metode serial dilusi dengan metode pengujian turbidimetri dan spektrofotometri. Buah mengkudu diekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Bakteri Streptococcus mutans diambil dari stok bakteri murni Laboratorium Mikrobiologi Program Studi Farmasi Fakultas MIPA Universitas Sam Ratulangi. Hasil penelitian menunjukkan bahwa KHM ekstrak buah mengkudu terhadap pertumbuhan Streptococcus mutans pada konsentrasi 50%. Cinnamon (Cinnamomum burmannii) is widely used by the public as a food ingredient and contains chemical compounds such as alkaloids, flavonoids, polyphenols, saponins, and terpenoids which function as an antibacterial against Staphylococcus aureus. This study aims to determine the Minimum Inhibitory Concentration and Minimum Bactericidal Concentration of cinnamon ethanol extract on Staphyloccocus aureus. This research is experimental with posttest only control group design through the tube dilution method. The results of the Minimum Inhibitory Concentration (MIC) study showed no clarity at concentrations of 30% and 40%. The results of the Minimum Bactericidal Concentration (MBC) obtained the number of colonies at a level of 10% by 51 CFU / plate, 20% by 27 CFU / plate, 30% by 6 CFU / plate and 40% by 0 CFU / plate. Based on the results of this study concluded that MIC cinnamon ethanol extract was 30%, and MBC cinnamon ethanol extract was 40%.

Abstract: Noni (Morinda cittrifolia L.) leaves contain antraquinon, atsiri oil, saponin, tannin, alkaloid, flavonoid, polifenol, and sterol that have been proved can inhibit the growth of Candida albicans. This study was aimed to establish the minimum inhibitory concentration (MIC) of noni leaf extract against Candida abicans. This was a true experimental study with a randomized pretest-posttest control group design. We used serial dilution method with turbidimetry and spectrophotometry tests. Noni leaves were extracted by using maceration with 96% ethanol. Candida albicans fungi were obtained from Microbiology Laboratory of Faculty of Mathematics and Natural Sciences, Sam Ratulangi University. The turbidimetry test using three repetitions showed that the MIC of noni leaf extract against Candida albicans was 6.25% meanwhile the spectrophotometry test established 12.5% as the MIC of noni leaf extract. Hasil penelitian menunjukkan bahwa pengujian turbidimetri dengan tiga kali perlakuan mendapatkan KHM pada konsentrasi 6,25% sedangkan pengujian spektrofotometri mendapatkan KHM pada konsentrasi 12,5%. Abstract: Inadequate oral and dental hygiene can cause plaques containing various kinds of bacteria; one of them is Streptococcus mutans which is the main cause of dental caries. Binahong leaf (Anredera cordifolia Steenis) is a medicinal herb that contains antibacterial compounds namely flavonoids, alkonoid, terpanoid, and saponins. This study aimed to determine the minimal inhibitory concentration (MIC) of binahong leaf extract to the growth of Streptococcus mutans. This was a true experimental study with a randomized pretest-posttest control group design. The method used in this study was serial dilution method with turbidimetry and spectrophotometry as the test methods. Binahong leaves were taken from Tempok village and were extracted with maceration method using ethanol 96%. Streptococcus mutans bacteria were obtained from a pure bacterial stock in Microbiology Laboratory of Pharmacy Study Program, Faculty of Mathematics and Natural Science University of Sam Ratulangi Manado. Abstract: Impatiens balsamina L. is an herbal plant which contains flavonoids that has been proven to inhibit the growth of Streptococcus mutans. This study was aimed to determine the minimum inhibitory concentration (MIC) of the balsamina leaf extract on the growth of Streptococcus mutans. This was a true experimental study with a randomized pretest-postest control group design. The method used was serial dilution and tested with the spectrophotometer and turbiditry measurements. Balsamina leaf was extracted by using maceration method with 96% ethanol. The Streptococcus mutans bacteria were obtained from pure stock bacteria at Microbiology Laboratory of Pharmacy Faculty of Mathematics, University of Sam Ratulangi. ABSTRACTBackground: Aggressive periodontitis is a disease that is mostly suffered by young patients with minimal local factors. The prevalence of this disease is approximately 8% in Indonesia, where Aggregatibacter actinomycetemcomitans bacteria are presented as dominant bacteria in this disease. Aggregatibacter actinomycetemcomitans is a gram-negative bacterium that is sensitive to flavonoids. Ramania leaves are natural substances which contain flavonoids. Flavonoids in ramania leaves can be applied as an antibacterial substance. Objectives: The objectives of this research are to determine and analyze the minimum inhibitory concentration and the minimum bactericidal concentration of flavonoids extract from ramania leaves at the concentration of 0.1%, 0.3%, and 0.5%, and analyze the antibacterial effectiveness on Aggregatibacter actinomycetemcomitans. Methods: True experimental design with pre and post-test with control group design was applied in this study. Antibacterial test using broth and agar dilution method was performed in a total of 5 treatment groups with 6 repetitions. Results:  All exract at 0.1%, 0.3% and 0.5% concentration demonstrated the ability to inhibit bacterial growth, and the minimum bactericidal concentration was found at 0.5% concentration. Result test for One way ANNOVA on minimum inhibitory concentration and Kruskal wallis test on minimum bactericidal concentration exhibited significant value of less than 0.05 presenting significant differences in some groups. Conclusion:  Minimum inhibitory concentration of ramania leaves flavonoid extract is effective on Aggregatibacter actinomycetemcomitans at the concentration of 0.1% and minimum bactericidal concentration of ramania leaves flavonoid extract is more effective at the concentration of 0.5%.Keywords: Aggregatibacter Actinomycetemcomitans, Aggressive Periodontitis, Dilution Method, Flavonoids, Ramania Leaves.

Background: Moringa oleifera and Gongronema latifolium have many nutritional values that contain bioactive components such as alkaloids, saponin, tannin, steroids and terpenoids, as  antimicrobials.            Objective: The general purpose of this study was to evaluate the minimum inhibitory concentration (MIC) and Minimal bactericidal concentration(MBC) of both Moringa oleifera and Gongronema latifolium  extracts against Staphylococcus aureus, Salmonella typhi and Escherichia coli. Method: This is laboratory experimental research involving post test only control group design using the Kirby Bauer dilution method. With treatment combination of Moringa oleifera and Gongronema latifolium extracts concentration ranging from 50-200 mg/ml and positive control (gentamycin). Result: Mininum bacterial concentration was found between 6.25 mg /ml – 12.5mg /ml for Moringa oleifera and Gongronema latifolium extracts while minimum inhibitory concentration of Moringa oleifera and Gongronema latifolium was found between 12.5 mg / ml – 25.0 mg/ml respectively. U-Mann whitney results showed that the value of p < 0.05 were significant. In this study, the Staphylococcus aureus showed the highest inhibition zone diameter compared to other isolates. Conclusion: The results from this work affirms the use of Moringa oleifera and Gongronema latifolium as antimicrobials and emplores the attention of pharmaceutical companies to exploit production of antibiotics for treatment of infections resulting from Salmonella typhi and Staphylococcus aureus isolates.

ABSTRACTBackground: Mauli banana stem and basil leaves are natural ingredients that have the potential to be used as a treatment for Candida albicans infections. The methanol extract of mauli banana stem with 25% concentration is merely fungistatic which is still not capable to induce fungicidal effect, while the 100% concentration is known to be less effective than 0.2% chlorhexidine gluconate. Adding basil leaves extract to mauli banana stem extract is expected to increase antifungal activities against Candida albicans. Purpose: to analyze the antifungal activity of mauli banana stem and basil leaves extract mixture with concentrations of 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90% and 100% against Candida albicans by dilution method. Method: This research used a true experimental method with a pretest–posttest control group design consisting of 11 treatments to obtain Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC). Result: The 10% concentration was identified as the Minimum Inhibitory Concentration (MIC), and no concentration could be classified as the Minimum Bactericidal Concentration (MBC) because all treatment group still exhibited Candida albicans growth on the SDA medium. Conclusion: Mauli banana stem and basil leaves extract mixture has an MIC value of 10% and does not demonstrate any value for the MBC. Keywords: Basil leaf, Candida albicans, Mauli banana stem, MBC, MIC

Abstract: The main problem in dental health is dental caries that occurs due to the fermentation by Streptococcus mutans bacteria. Gedi leaf (Abelmoschus manihot L. Medical) is one of the common plants in Northern Celebes that contains antimicrobial compounds namely flavonoid, alkaloid, steroid, and saponin; all of them have been proved to inhibit the growth of S. mutans. This study was aimed to determine the minimal inhibitory concentration (MIC) of gedi leaf extract (Abelmoschus manihot L. Medical) to S. mutans growth. This was a true experimental study with a randomized pretest-posttest control group design. The method used in this study was serial dilution method with turbidimetry and spectrophotometry. Samples of gedi leaves were obtained at Paal 2 Manado, and were extracted with maceration method using ethanol 96%. S. mutans bacteria were obtained from the pure bacterial stock at Microbiology Laboratory of Pharmacy Study Program, Faculty of Mathematics and Natural Science University of Sam Ratulangi Manado. The turbidimetry test showed that the tube content became clearer at 25% of extract concentration. Moreover, the UV-Vis spectro-photometer showed a decrease of OD value for the first time at 25% of extract concentration. Conclusion: The minimal inhibitory concentration (MIC) of gedi leaf extract (Abelmoschus manihot L. Abstract: Dental caries is an oral cavity disease which becomes a health problem in Indonesia. Bacteria that play some important role in caries formation are Streptococcus mutans. One of the nature material that can be used to restrain Streptococcus mutans is seaweed Eucheuma cottonii. This study aimed to determine the Minimum Inhibitory Concentration (MIC) of seaweed Eucheuma cottonii extract against Streptococcus mutans. This was an experimental laboratory study with a post test only control group design. MIC test was done by serial dilution, turbidimetry, and spectrophotometry methods, as well as bactericidal test using aa calibration method. Eucheuma cottonii were taken from the sea around Naen island and then were extracted with maceration method using ethanol 96%. The results showed that the MIC of Echeuma cottonii extract was in the concentration of 6.25% against Streptococcus mutans. Growth inhibition of Streptococcus mutans was significant in the concentration of 6.25%, 12.5%, 25%, 50%, and 100%. MIC of Eucheuma cottonii extract was 6.25% but the MBC or bactericidal effect was not found.The higher concentration of Eucheuma cottonii used, the greater ability to inhibit the growth of Streptococcus mutans. Conclusion: Seaweed Echeuma cottonii extract has bacteriostatic abilities but do not have bactericide abilities to against Streptococcus mutans.Keywords: streptococcus mutans, mic, eucheuma cottonii, alternative medicine, coastal communitiesAbstrak: Karies gigi merupakan penyakit rongga mulut yang paling banyak diderita masyarakat dan menjadi salah satu masalah kesehatan di Indonesia. Bakteri yang paling berperan dalam pembentukan karies adalah Streptococcus mutans. Cara alternatif untuk menanggulangi Streptococcus mutans adalah dengan menggunakan bahan alami yaitu rumput laut (Eucheuma cottonii).Tujuan penelitian ini adalah untuk mengetahui Minimum Inhibitory Concentration (MIC) rumput laut (Eucheuma cottonii) terhadap Streptococcus mutans. Penelitian ini merupakan penelitian eksperimental laboratorium menggunakan desain post-test only control group design. Uji MIC dilakukan dengan metode serial dilusi, turbidimetri dan spektrofotometri, serta uji kalibrasi untuk uji sifat bakteriosid. Sampel diambil dari perairan pulau Naen kemudian diekstraksi dengan metode maserasi menggunakan pelarut etanol 96%. Hasil penelitian menunjukkan bahwa MIC ekstrak rumput laut (Eucheuma cottonii) pada konsentrasi 6.25% terhadap Streptococcus mutans. Hasil analisis dan pengolahan data menunjukkan daya hambat ekstrak rumput laut (Eucheuma cottonii) terhadap pertumbuhan Streptococcus mutans yang paling signifikan ialah konsentrasi 6,25%, 12,5%, 25%, 50%, dan 100%. MIC ekstrak rumput laut (Eucheuma cottonii) 6,25% sedangkan MBC atau sifat bakteriosid tidak ditemukan. Semakin tinggi konsentrasi ekstrak rumput laut (Eucheuma cottonii) yang digunakan, maka semakin besar kemampuan menghambat pertumbuhan Streptococcus mutans. Simpulan: Ekstrak rumput laut (Eucheuma cottonii) memiliki sifat bakteriostatik namun tidak memiliki sifat bakteriosid terhadap Streptococcus mutans.Kata kunci: streptococcus mutans, mic, eucheuma cottonii, pengobatan alternatif, masyarakat pesisir pantai
