Effect of genetic and environmental factors on semen productionand quality of Madura cattle in Indonesia

The indigenous Madura cattle semen production and quality were evaluated. Total of 2275 semen collection records from 11 bulls were analyzed to evaluate the genetic potency of semen productions. The genetic evaluation was described as an estimation of bull semen volume, sperm concentration and sperm individual motility. Data were analyzed by HGLM-REML with environmental factors of age, seasons, interval of semen collections and frequency of ejaculations as fixed effect. The result showed that the age and the ejaculation frequency affected to all variables; an interval of semen collections affected to both semen volume and sperm concentrations; and seasons only affected to sperm concentrations. Repeatability estimate of semen volume, sperm concentration and sperm individual motility were 0.376, 0.445 and 0.567 respectively. It can be concluded that the quality of ejaculate was less affected by environmental factors, supported by the good adaptation of indigenous Madura cattle under the local climatic conditions and management in the artificial insemination station.

The aim of this study was to analyze production and quality of semen in boars in an insemination centre. Experimental material consisted of 8,052 semen samples collected from 146 boars. During the period of 5 years, an analysis of semen parameters was conducted. The obtained data were divided according to age of boars at the time of semen collection. Results showed that boars between 8 months and 3 years of age reached higher levels of semen volume, percentage of abnormal spermatozoa and number of spermatozoa. Older boars had lower sperm concentration levels. The largest total number of spermatozoa and corrected number of spermatozoa were observed in boars between 13 and 36 months of age. Later, the production of spermatozoa decreased. Based on the results of the presented study, the age category of 13 – 36 months can be recommended to dominate in the herd of boars in the insemination centre.

This study was carried out in Artificial Insemination Center of Iraq to revealed FMD disease effect on some seminal attributer parameters of 14 imported Holstein bulls divided to three groups according to different reproductive efficiency (four High, five medium and five weak). Results showed that FMD disease had significant (P < 0.05) adverse effect on most seminal attributer parameters, mass, individual motility and sperm concentration / ml during post disease in first of two, four, all months of high, medium and weak semen quality bulls respectively .but semen volume didn’t influenced significantly with this disease. So semen collection should be suspended until resume normal fertility of sperm, after two, four month of high and medium bulls respectively, and must be revealed weak bulls when disease happen to avoid the failure of conception from artificial insemination and there is no economic benefit to use or keep weak bulls

In poultry breeding program the success of artificial insemination (AI) is highly influenced by  the quality of semen. The aim of the present research was to evaluate the variations in semen characteristics of three phenotypes of chicken of the same specie raised in Akko, Nigeria. A total of 9 cocks from threedifferent phenotypes; Red feathered (n = 3), White feathered (n = 3) and Black feathered (n =3). The semen was evaluated for macroscopic (i.e. semen volume and colour) and microscopic (i.e. sperm concentration, motility and morphology) criteria after being collected by abdominal massage method. There were phenotypes variation (P > 0.05)effects on semen motility, number of live/dead sperm and sperm abnormalities. No differences(P>0.05) were observed on volume, colour, concentration, mass motility and pH of semen. The observed sperm progressive motility was ranged from 90.5 ±1.21% to 95.09±0.82%. The White feathered strain had the highest sperm progressive motility (P<0.05)and highest value for live and normal sperm (93.5±0.63% and 87.90±0.25%), while the black feathered had the least and (92.4±0.73% and 85.5±0.50%) respectively. The Red feathered strain had the highest value for both dead and sperm head defect (9.2±0.33%and 7.0±0.52%). Whilst, the Black feathered strain scored the highest percentage of both sperm tail and neck defects with values ranged from 11.4±0.43%to 14.2±1.08% and 19.4±0.53% to 23.6±0.69%, respectively. This study suggests that there are large variations present in semen characteristics of different phenotypes of cocks; White feathered strain is likely have better semen characteristics compared to Red and Black feathered strains. Therefore, White feathered strain can potentially be used in artificial insemination (AI) for chicken production and improvement.

TThe   study   was  carried   out   during the yearr  2017   to   determine   the   fresh   and   post-thaw   quality   of   four   Holstein   Friesian   bulls.   Semen   samples   were   examined   for ejaculate   volume,   pH,   mass   activity,   sperm   concentration   and   sperm   motility and   sperm   membrane   integrity   (HOST).   The   ejaculated   semen   volume   of   Holstein   Friesian   bulls   was   in   the   range   of   6-8ml   and   differences   in   ejaculates   were   significant   (P<0.01).   The   volume   was   significantly   (P<0.05)   higher   (7.75ml)   when   collected   on   5th   July,   slightly   decrease   in   volume   (7.25ml)   when   collected   on   12th   May   and   26th   July.   Semen   pH   was   higher   (6.65)   for   26th   July   ejaculation   and   lowest (6.55ml)   for   24th   May   ejaculation.   The   results   indicated   that   the   semen   of   Holstein   Friesian   bulls   did   not   have   considerable   variation   in   pH   during   May   –   July.   Most   of   the   semen   samples   were   creamy   white and   yellow   in   colour,   while   few   samples   were   milky   white   and   watery.   Mass   activity   score   of   semen   samples   indicated   vigorous   movement with   moderate rapid   waves   and   eddies.   The   sperm   motility   of   fresh   vs   post-thaw   semen   was   obtained   72.80   vs   48.75   percent   (12th   May),   75.00   vs   48.75   percent   (24th   May),   76.25   vs   55.00   percent   (2nd   June),   73.75   vs   52.50   percent   (21st   June),   75.00   vs   56.25   (5th   July)   and   75.00   vs   50.00   percent   (26th   July).   In   case   of   post-thaw   semen,   highest   sperm   motility   of   56.25   percent   was   recorded   in   semen   ejaculated   on   5th   July,   while   lowest   post-thaw   sperm   motility   of   48.75   percent   was   observed   in   semen   ejaculated   on   12th   May   and   24th   May.   Highest   sperm   concentration   of   1549.75x106   was   determined   in   semen   ejaculated   on   21.06.2017   and   lowest   concentration   (1259.50x106)   in   26.07.2017   collected   samples.   It   was   concluded   that   sperm   motility   was   significantly   (P<0.01)   affected   by   semen   types   (fresh   and   post-thaw),   while   sperm   concentration   was   also   significantly   (P<0.01)   affected   by   ejaculation   date   and   bulls.   The   membrane   integrity   (HOST)   of   the   pooled   data   on   fresh   semen   samples   over   a   period   of   three   months   was   58.37   percent   against   post-thaw   membrane   integrity   of   44.79   percent.

THE EFFECT OF FALSE MOUNTING ON SEMEN QUALITY OF LIMOUSIN AND SIMMENTAL BULL IN LEMBANG ARTIFICAL INSEMINATION CENTER. The purpose of this study was to determine the effect of false mounting on the quality of Limosin and Simental bull semen at the Lembang Artificial Insemination Center. The 55 individu of Limousin and 55 Simmental bull aging 5-9 years old have been used in thos study. The data have been collected directly in the Artificial Insemination Center Lembang - West Java Province. The data obtained in the form of macroscopic and microscopic observations on Limousin and Simmental bull include volume, pH, mass movement, spermatozoa motility and spermatozoa concentration. Data were analyzed descriptively by calculating the standard deviations, correlations coefficients and the T test. The variables measured in this study were macroscopic and microscopic quality of semen. The results of this study indicate that the effect of false mounting on the average semen volume of Limousin and Simmental bull is maximum at 3, 4, and 5 times false mounting but there was not significantly difference between the amount of false mounting, as well as the average volume, pH, mass movement, motility of spemetozoa and concentration spermatozoa are not significantly different. It was concluded that false mounting in Limousin and Simmental bulls in Lembang Artificial Insemination Center had no significantly effect on the quality of macroscopic and microscopic semen. Keywords: False Mounting, Cement Quality, Limousin Cattle, Simmental Cows.

The purpose of this research was find out the effect of age and ejaculation frequency on quality of Aceh bull spermatozoa.. This research was conducted at Frozen Semen Prossesing Laboratorium, Saree, Aceh Besar from Januari to April 2013. Six healthy aceh bulls were divided into two groups of age, those were 3.0-4.0 years old; and 4.0-5.0 years old. Three numbers of bull at each group were assumed as the replication. Semen was collected once a week using artificial vagina and collected until third semen ejaculation. After collecting, volume, pH, color, consistency, concentration, motility, mortality, and abnormality of spermatozoa were immediately measured. This data was analyzed by analysis of variance (ANOVA) two-way pattern, followed by Duncant test. The result showed that the evaluation of bull semen at 3.0-4.0 years old and 4.0-5.0 years old affect significantly (P0.05) on semen volume and spermatozoa concentration and had no contribute significantly on motility, mortality and abnormality of spermatozoa,while ejaculation frequency effect significantly (P0.05) on semen volume, concentration, and abnormality spermatozoa, but had no effect significantly on motility and mortality (P0.05). It is concluded that aged and ejaculation frequency effect significantly on quality of aceh bull fresh spermatozoa.Key words: ejaculation frequency, quality of spermatozoa, aceh bull, age

The objective of this study was to determine the semen cryopreservation capacity of BBB bulls in Hanoi-Vietnam. Research conducted on the fresh semen collected from 05 BBB bulls. Results showed that semen color was normal (milky white, ivory white, ivory yellow), semen volume ranged from 6.35 mL to 7.48 mL (P <0.05), initial motility of semen ranged from 80.53% to 82.92% (P <0.05), sperm concentration in semen  ranged from 1.02 x 109 sperms/ml to 1.12 x 109 sperms/mL (P <0.05), abnormal sperm ratio ranged from 6.45% to 8.12% (P <0.05), alive sperm ratio ranged from 76.34% to 82.97% (P <0.05), sperm motility after thawing from straw semen ranged from 71.33% to 75.92% (P<0.05). In conclusion, successfully semen collection from 05 breeding BBB bulls at Hanoi Cattle Breeding Joint Stock Company, semen samples had normal color and good quantity and quality, suitable for production of frozen semen; and semen cryopreservation of straws of the 05 bull BBB semen mentioned at -196oC, sperm motility after freezing-thawing reached the economic and technical norms of 675/2014 of the Ministry of Agriculture and Rural Development.
