Evaluate the fresh and Post Thawing Quality of Holstein Friesian Bull semen

TThe   study   was  carried   out   during the yearr  2017   to   determine   the   fresh   and   post-thaw   quality   of   four   Holstein   Friesian   bulls.   Semen   samples   were   examined   for ejaculate   volume,   pH,   mass   activity,   sperm   concentration   and   sperm   motility and   sperm   membrane   integrity   (HOST).   The   ejaculated   semen   volume   of   Holstein   Friesian   bulls   was   in   the   range   of   6-8ml   and   differences   in   ejaculates   were   significant   (P<0.01).   The   volume   was   significantly   (P<0.05)   higher   (7.75ml)   when   collected   on   5th   July,   slightly   decrease   in   volume   (7.25ml)   when   collected   on   12th   May   and   26th   July.   Semen   pH   was   higher   (6.65)   for   26th   July   ejaculation   and   lowest (6.55ml)   for   24th   May   ejaculation.   The   results   indicated   that   the   semen   of   Holstein   Friesian   bulls   did   not   have   considerable   variation   in   pH   during   May   –   July.   Most   of   the   semen   samples   were   creamy   white and   yellow   in   colour,   while   few   samples   were   milky   white   and   watery.   Mass   activity   score   of   semen   samples   indicated   vigorous   movement with   moderate rapid   waves   and   eddies.   The   sperm   motility   of   fresh   vs   post-thaw   semen   was   obtained   72.80   vs   48.75   percent   (12th   May),   75.00   vs   48.75   percent   (24th   May),   76.25   vs   55.00   percent   (2nd   June),   73.75   vs   52.50   percent   (21st   June),   75.00   vs   56.25   (5th   July)   and   75.00   vs   50.00   percent   (26th   July).   In   case   of   post-thaw   semen,   highest   sperm   motility   of   56.25   percent   was   recorded   in   semen   ejaculated   on   5th   July,   while   lowest   post-thaw   sperm   motility   of   48.75   percent   was   observed   in   semen   ejaculated   on   12th   May   and   24th   May.   Highest   sperm   concentration   of   1549.75x106   was   determined   in   semen   ejaculated   on   21.06.2017   and   lowest   concentration   (1259.50x106)   in   26.07.2017   collected   samples.   It   was   concluded   that   sperm   motility   was   significantly   (P<0.01)   affected   by   semen   types   (fresh   and   post-thaw),   while   sperm   concentration   was   also   significantly   (P<0.01)   affected   by   ejaculation   date   and   bulls.   The   membrane   integrity   (HOST)   of   the   pooled   data   on   fresh   semen   samples   over   a   period   of   three   months   was   58.37   percent   against   post-thaw   membrane   integrity   of   44.79   percent.

This study was conducted to investigate the effect of docosahexanoic acid (DHA) supplementation in BioXcell extender on the quality of frozen–thawed bull semen. Twenty-four ejaculates were collected from three bulls (eight from each bull). Ejaculates with motility ≥70% and normal morphology ≥80% were extended into BioXcell extender to which 0 (control), 3, 5, 10 or 15 ng mL–1 DHA was added. The supplemented semen samples were incubated at 37°C for 15 min for DHA uptake by spermatozoa. Later, samples were cooled for 2 h at 5°C and packaged into 0.25-mL straws, frozen in liquid nitrogen for 24 h and subsequently thawed for evaluation. Results are presented as percentages ± s.e.m. Supplementation with DHA at 3 ng mL–1 significantly improved sperm functional parameters including sperm motility, normal morphology, viability, acrosome integrity and membrane integrity when compared with other supplemented groups and the control. Lipid peroxidation increased as the incorporation of DHA supplementation increased. In conclusion, 3 ng mL–1 concentration of DHA resulted in superior quality of frozen–thawed bull spermatozoa and is suggested as the optimum level of DHA to be added into BioXcell extender.

Boar semen is voluminous and ejaculated as jets or fractions of pre-sperm, sperm rich (SRF) and post-sperm rich fractions. Recent studies have reported more resilient characteristics of sperm in initial portions of SRF towards cold shock and cryopreservation. The present study was conducted to assess the quality of specific fractions of SRF, namely, first 10mL of SRF (F1) and rest of SRF (F2) in Large white Yorkshire (LWY) boar semen. Ejaculates were collected using gloved-hand technique and were subjected to quality assessments of volume, pH, sperm progressive motility, concentration, plasma membrane integrity, abnormality, acrosome integrity and sperm membrane cholesterol. Upon statistical analysis, significant differences were noticed in volume, pH, sperm concentration and sperm membrane cholesterol between fractions of the ejaculate.

The objective of this study was to investigate the effect of using different extenders <italic>viz.</italic> Androhep, Safecell Plus and SUS during cryopreservation on quality of frozen-thawed boar semen. Semen volume, sperm motility, sperm concentration, percentage of morphologically abnormal spermatozoa, total number of spermatozoa per ejaculate and activity of the enzyme aspartate aminotransferase (AST) were assessed in fresh semen collected from 39 fertile AI boars. Semen from each boar was divided into three portions and diluted 1:1.5 in extender Androhep, Safecell Plus and SUS and keep at 17°C for 15-h holding time before cryopreservation. Then sperm was cryopreserved. Straws were thawed in a water bath at 38°C for 40s and post-thaw sperm motility with AST activity was assessed. Significant difference in post-thaw sperm motility was found between extender Androhep and Safecell Plus (P<0.05). AST activity did not differ significantly between tested extenders (P>0.05). In conclusion, the results of the study indicate that using Safecell Plus extender during holding period before cryopreservation significantly affected post-thaw sperm motility.

Repeatability for seminal traits and their phenotypic relationships with testes measurements and performance traits in Black Bengal bucks were estimated from a total of 116 repeated observations on 15 young Black Bengal bucks. Performance traits included age, body condition and body weight of bucks. Testes measurements were testes length, breadth and volume, and scrotal circumference, while seminal traits were ejaculate volume, semen density, mass activity, sperm motility, sperm concentration, total sperm per ejaculation and percent of normal sperm. High positive correlations (ranging from 0.81 to 0.90) were found between body weight and testes measurements. Semen volume and percent of normal sperm were positively correlated with age, body condition and body weight of bucks (ranging from 0.24 to 0.60). The testes measurements were strongly and positively correlated with semen volume and total sperm per ejaculation (ranging from 0.53 to 0.61), while the correlations between testes measurement and percent of normal sperm were moderate (ranging from 0.34 to 0.44). Among seminal traits, strong correlations were found between semen volume and total sperm per ejaculation (0.81) and, between mass activity and sperm motility (0.82). Repeatability of ejaculate volume was higher (r = 0.78) which indicated that selection or culling for semen ejaculate volume could be practiced from single or few observations. It could be concluded that bucks’ age, body weight and body condition along with testes volume should be considered as selection criteria for improving semen quality and semen production of breeding bucks. Abstract The objective of the study was to determine the effect of high hydrostatic pressure (HHP) on quality of cryopreserved semen of young bulls. Semen for this study was collected from 8 bulls aged between 13 and 18 months at monthly intervals, from June to September. After collection, semen was diluted in a commercial Bioxcell® extender (one part at 1:1 and a second part to give a sperm concentration of 20 million/0.2 mL), filled into straws and treated with HHP at 30 MPa for 90 min. After HHP treatment, pre-diluted semen (1:1) was diluted to a sperm concentration 20 million/0.2 mL and filled into straws. In addition, part of the semen diluted to a concentration of 20 million/0.2 mL was not treated with HHP (control). All of it was held at +4°C and frozen in a freezer after 2.5-h equilibration. Semen was thawed in a water bath at 38°C and subjected to estimation of the percentage of motile sperm both subjectively and using a computer-assisted semen analyzer and cytometric assessment of sperm cell membrane integrity. Subjective motility and fast progressive motility were significantly higher with pre-diluted (1:1) and HHP treated semen compared to control (P<0.05). No significant differences were observed in percentage of membrane-intact spermatozoa between control and experimental groups. Additionally, the influence of HHP on the sperm of individual bulls was assessed. In bull number 2, the HHP treatment after semen pre-dilution significantly improved progressive motility from 54.1 to 63.4 percent (P <0.05). In bull number 4, the HHP treatment after semen pre-dilution significantly improved subjective motility, rapid motility and progressive motility by 12.5, 16.8 and 16.3 percent, respectively (P<0.05). No effect was seen for 6 bulls. It is concluded that for some bulls, the application of HHP before semen freezing may improve the cryopreservation outcome. However, this requires further research in this area, also to determine the fertilizing capacity of bull semen exposed to high hydrostatic pressure.

The indigenous Madura cattle semen production and quality were evaluated. Total of 2275 semen collection records from 11 bulls were analyzed to evaluate the genetic potency of semen productions. The genetic evaluation was described as an estimation of bull semen volume, sperm concentration and sperm individual motility. Data were analyzed by HGLM-REML with environmental factors of age, seasons, interval of semen collections and frequency of ejaculations as fixed effect. The result showed that the age and the ejaculation frequency affected to all variables; an interval of semen collections affected to both semen volume and sperm concentrations; and seasons only affected to sperm concentrations. Repeatability estimate of semen volume, sperm concentration and sperm individual motility were 0.376, 0.445 and 0.567 respectively. It can be concluded that the quality of ejaculate was less affected by environmental factors, supported by the good adaptation of indigenous Madura cattle under the local climatic conditions and management in the artificial insemination station.

Four bulls of mixed breeds (Sahiwal × Zebu, Sindhi × Zebu, Jersey × Zebu and Holstein-Friesian × Zebu )were examined physically and their semen was evaluated in three seasons: I (Rainy): May - November, II (Winter): December - February, III (Summer): March- April. The semen volume, sperm motility, sperm concentration and percentage of dead sperm ranged from 4.1 to 7.6 ml, 56.6 to 76%, 1010.3 to 1290.7 × 106/ml and 18.4 to 24.7%, respectively. All these attributes varied significantly (p<0.05) between breeds and seasons. The variations of semen quality between breeds were not significant (p>0.05). Aim: This study aimed to analyze the individual factors influencing the sperm quality of Bali bulls at Baturiti Artificial Insemination (AI) center. Materials and Methods: Semen that was ejaculated from nine Bali bulls was collected using artificial vaginas (n=5/bull). Semen ejaculates were evaluated immediately after collection to measure the quality of the fresh semen, including semen volume, sperm concentration, progressive motility, membrane integrity (MI), and abnormal morphology. Frozen semen was evaluated for progressive sperm motility, concentration, viability, MI, abnormal morphology, and deoxyribonucleic acid (DNA) fragmentation. Other secondary data, focusing on semen quantity (semen volume and sperm concentration), were also collected from frozen the semen production data of the Baturiti AI center from 2017 to 2019. Data were analyzed statistically using a completely randomized design, and one-way analysis of variance was applied to find differences among individual bulls. Results: Significant differences (p<0.05) were found among the bulls in semen volume, sperm motility, concentration, and MI of the fresh semen. Significant differences (p<0.05) were also found among the bulls in sperm motility, viability, MI, abnormal morphology, and DNA fragmentation of the frozen semen. Conclusion: Individual variation in all the tested sperm parameters of the fresh semen of Bali bulls, except sperm viability and abnormalities, was noted. Similarly, individual variation in all the tested sperm parameters in frozen semen, except sperm concentration, was noted. Therefore, individual factors can be used for selecting a superior bull in Bali cattle.

The aim of this study was to compare the effect of two semen extenders and four permeating cryoprotectants on post-thaw sperm quality of Andalusian donkeys. First, 32 ejaculates were pooled, split and frozen in either Gent B or INRA 96 with egg yolk and glycerol. Second, 12 pooled semen samples were simultaneously frozen in Gent B (glycerol) or Gent A containing ethylene glycol (EG; 1 or 1.5%) or dimethyl sulfoxide (DMSO; 1.5 or 2%). Finally, nine pooled samples were simultaneously cryopreserved in Gent A containing 1% EG (as control), dimethylformamide (DMFA; 1 or 2.5%) or a combination of 1% EG and 1.5% DMFA. Gent B yielded a higher (P < 0.01) post-thaw sperm motility than modified INRA96. EG 1% increased the sperm membrane integrity (P < 0.001), whereas DMSO affected sperm motility and membrane integrity (P < 0.001). DMFA 2.5% yielded higher (P < 0.001) values for sperm motility and membrane integrity. We concluded that Gent B improves in vitro post-thaw sperm quality of donkey spermatozoa, but the replacement of glycerol with 1% EG or 2.5% DMFA increased sperm protection against cryodamage. The use of DMSO for freezing donkey semen was unsuccessful and a toxic effect is suspected. These extenders should be included in the pre-freeze test for each donkey.

ABSTRACT This study aimed to evaluate the effects of Zinc (Zn) supplementation on the quality and quantity of striped catfish sperm. Experimental design for this study was a complete randomized design with five treatments and five replications. Male broods fed with Zn supplementation for eight weeks. The Zn supplemented into the fish diet at different concentrations (0, 50, 100, 150 and 200 mg/kg of feed). The results showed that Zn supplementation could improve the quality and quantity of striped catfish sperm. The treatments also showed significant effects on semen volume, sperm motility, sperm viability, and sperm concentration (P<0.05). Zn supplementation at a dose of 200 mg/kg feed demonstrated the best result has indicated by enhancement of quality and quantity of striped catfish sperm, increasing 51% of the volume, 11.6% of motility, 5.81% of viability, 54.1% of concentrations. The results suggested that Zn played an important role in improving reproductive performances of male striped catfish reproduction. Keywords: quality of sperm, a quantity of sperm, striped catfish, supplementation zinc                                                                                            ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi Zinc (Zn) terhadap kualitas dan kuantitas sperma ikan patinPangasianodon hypophthalmus. Penelitian ini menggunakan rancangan acak lengkap dengan  lima perlakuan danlima ulangan. Induk jantan diberi pakan dengan suplementasi Zn selama 8 minggu. Zn disuplementasikan dengan dosis berbeda (0, 50, 100, 150 dan 200 mg/kg pakan). Hasil penelitian menunjukkan bahwa suplementasi Zn dapat meningkatkan kualitas dan kuantitas sperma ikan patin sehingga berpengaruh signifikan terhadap volume semen, motilitas, viabilitas dan konsentrasi sperma (P<0,05). Suplementasi Zn pada dosis pakan 200 mg/kg menunjukkan hasil terbaik yang ditunjukkan oleh peningkatan kualitas dan kuantitas sperma ikan patin 51% volume; 11,6% motilitas; 5,81% viabilitas; 54,1% konsentrasi sperma. Hasil penelitian ini menunjukkan bahwa Zn memainkan peran penting dalam meningkatkan reproduksi ikan patin. Kata kunci: kualitas dan kuantitas sperma, Ikan patin, suplementasi Zn
