The success of beef cattle reproduction development in Indonesia very much depends on the quality of the semen produced by the Indonesian Institute for Artificial Insemination. Good quality bull will benefit farmers, frozen semen producers, and the country in large. This study aimed to determine effect of age and breed on the libido and quality of bull semen collected in the Regional Artificial Insemination Center (RAIC). This study used PO, Simmental, and Brahman Bull in RAIC Yogyakarta, with a total amount of 15, aged 4-7 years old. Semen was collected using an artificial vagina, semen evaluation used a spectrophotometer, and eosin-nigrosin staining method used for sperm viability and abnormalities examination. The result of this study indicated that bull breed did not affect the libido value and the semen quality of bulls (P>0,05). Volume, concentration, motility, and viability in various age groups did not show significant differences (P>0,05), but age groups had a significant effect on sperm abnormalities (P<0,05). The group of bulls aged 5-6 years showed the best quality compared to other groups, with the volume 4,58 ± 1,37 ml, concentration 1,91 ± 0,36 x109/ml, motility 3,80 ± 1,30, viability 89.83 ± 10.69%, and sperm abnormalities 2.51 ± 0.81%. It can be concluded that breed did not affect the bull libido and semen quality, while age had an effect on bull sperm abnormalities.

The indigenous Madura cattle semen production and quality were evaluated. Total of 2275 semen collection records from 11 bulls were analyzed to evaluate the genetic potency of semen productions. The genetic evaluation was described as an estimation of bull semen volume, sperm concentration and sperm individual motility. Data were analyzed by HGLM-REML with environmental factors of age, seasons, interval of semen collections and frequency of ejaculations as fixed effect. The result showed that the age and the ejaculation frequency affected to all variables; an interval of semen collections affected to both semen volume and sperm concentrations; and seasons only affected to sperm concentrations. Repeatability estimate of semen volume, sperm concentration and sperm individual motility were 0.376, 0.445 and 0.567 respectively. It can be concluded that the quality of ejaculate was less affected by environmental factors, supported by the good adaptation of indigenous Madura cattle under the local climatic conditions and management in the artificial insemination station.

Objective: The aim of this study was to ascertain the effects of adding green tea extract (GTE) to skim milk-egg yolk (SM-EY) extender on both the quality of post-thawed bull semen and the pregnancy rates of the recipient cows. Methods: Twelve ejaculates from four Simmental bulls, aged 3 to 5 years and weighing 900 to 950 kg, were diluted SM-EY extender, added with 0, 0.05, 0.1, and 0.15 mg GTE/100 mL extender and then frozen. After four weeks storage in liquid nitrogen, the sperm were thawed and evaluated for viability, motility, intact plasma membrane (IPM), and DNA fragmentation. Meanwhile, the estrus cycles of 48 recipient cows were synchronized by intramuscular administration of a single injection of 5 mg prostaglandin F2α. Estrus cows were divided into four equal groups and inseminated artificially 18 to 20 h after the onset of estrus by using semen from each extender group. Pregnancy was diagnosed by measuring serum progesterone levels at 21 days, followed by transrectal palpation 90 days after insemination. Results: The findings revealed that adding 0.1 mg of GTE/100 mL extender produced the highest percentages of sperm viability (70.67%±1.75%), motility (69.17%±1.47%), and IPM (69.23%±1.21%) and the lowest percentage of DNA fragmentation (3.00%±0.50%). The pregnancy diagnosis revealed that all cows (36/36) inseminated using frozen semen in GTE addition extender were pregnant (pregnancy rate 100%), whereas the pregnancy rate of the control group was 83.33% (10/12).Conclusion: It may be concluded that 0.1 mg GTE/100 mL extender yields the best quality of spermatozoa and that all variants doses of GTE in extender produce a higher pregnancy rate among recipient cows.

ABSTRACT The aim of this research was to determine the proportion, viability and motility of X and Y ram spermatozoa separated with egg white albumin. Sperm samples derived from Garut ram, which was collected by using an artificial vagina. Observations were made on spermatozoa fraction above and below each medium fraction treatment. There are treatment egg white albumin as separation medium, each medium consisting of fractions top and bottom fraction with different concentration: 1) P0 = sperma before separation (control); 2) P1 = 10% above fraction + 30% lower fraction; P2 = 25% + 45%; P3 = 25% + 75%. Data proportion of X and Y, viability and motility were analyzed statistically by Completely Randomized Design patern in the direction followed by Duncan’s Multiple Range Test for data with a real difference. Separation with egg white albumin affect significantly increased the proportion of spermatozoa X and Y (P≤0.05), but tends to decrease the viability and motility of spermatozoa. The proportion of spermatozoa X and Y was highest in treatment P3,76.76% of spermatozoa X (fraction above 25%) and 79.81% spermatozoa Y (75% lower fraction), with an average viability obtained respectively 68,9% (fraction above) and 59,7% (bottom fraction), motility 77,5% (fraction above) dan 84,0% (bottom fraction). It was concluded that the egg white albumin is very effective in changing the proportions of X and Y ram sperm with the quality of spermatozoa after separation feasible for applications insemination or processed into frozen semen.  (Keywords: Garut ram, White egg albumin, Spermatozoa X and Y)

The purpose of this study was to determine the effects of apigenin and astragalus polysaccharides on the cryopreservation of bovine semen. Apigenin, astragalus polysaccharides, or their combination were added to a frozen diluent of bovine semen. Afterwards, Computer Assisted Semen Analysis (CASA), membrane functionality, acrosome integrity, mitochondrial integrity, CAT, SOD, GSH-Px, MDA, and ROS detection were conducted. The results showed that adding 0.2 mmol/L AP or 0.5 mg/mL APS could improve the quality of frozen sperm. Compared to 0.2 mmol/L AP alone, the combination of 0.2 mmol/L AP and 0.3 mg/mL APS significantly increased the total motility (TM), average path distance (DAP), straight line distance (DSL), average path velocity (VAP), curvilinear velocity (VCL), wobble (WOB), and sperm CAT and SOD levels (p < 0.05), while reducing the ROS and MDA levels (p < 0.05). These results indicated that the addition of 0.2 mmol/L AP or 0.5 mg/mL APS alone has a protective effect on the freezing of bovine semen. Compared to the addition of 0.2 mmol/L AP, a combination of 0.2 mmol/L AP and 0.3 mg/mL APS could further improve the quality of frozen semen.
