Low density lipoprotein on poor quality mithun (Bos frontalis) semen preservation

Low density lipoproteins (LDL) extracted from hens egg yolk (EY) has been studied over EY based extender for liquid storage of mithun semen with the objective to explore the use of LDL in place of EY. Physio-morphological attributes (PMAs) and mobility and velocity parameters were measured by computer assisted sperm analyser (CASA). Leakage of intracellular enzymes, activity of total antioxidants and lipid peroxidation following liquid storage (5oC) of mithun semen were studied. Fifty ejaculates were collected through transrectal massage method from matured mithun bulls and based on the mass activity and individual motility; the semen samples were splited into good and poor quality and diluted with the tris citrate glycerol (TCG) extender and were splited into three equal aliquots: Group I: Control, EY; Group II and Group III contained 8 and 10% LDL (w/v), respectively. PMAs, intracellular enzymatic leakage and biochemical profiles were evaluated at 5°C following 10hrs incubation. Result revealed a significant (p less than 0.05) improvement in PMAs, CASA parameters and cholesterol content of spermatozoa as well as reduction in leakage of intracellular enzymes, oxidative stress in Group II than control and other treatment group. It was concluded that addition of 8% LDL holds a clear advantage over EY or 10% LDL in liquid preservation of mithun semen.

Virgin coconut oil (VCO) is a saturated fat with promising antidiabetic properties but its ameliorative effect on lipid profiles in diabetics is rarely reported. Therefore, in this study, a total of fifteen (15) male rats weighing 200–250 g were divided into 3 experimental groups (n=5). Group I (control) and Group II (diabetic control group) were fed a normal rat chow while Group III (diabetic test group) was fed a 10% VCO diet for 3 weeks. Group II and Group III were made diabetic by intraperitoneal injection of 150 mg/kg of alloxan. After 72 hours of injection, blood glucose was tested to confirm diabetes mellitus. After 3 weeks, the animals were sacrificed to collect blood samples for lipid profile analysis. The results showed a significant increase in concentrations of triglyceride, total cholesterol, low density lipoprotein, and very low density lipoprotein and decrease in concentration of high density lipoprotein in Group II when compared to Group I. Also, the concentrations of triglyceride, total cholesterol, low density lipoprotein, and very low density lipoprotein except high density lipoprotein significantly reduced in Group III when compared to Group II (P<0.01, 0.001). VCO consumption can be claimed to ameliorate lipid levels in diabetes mellitus.

Background: Objective was to study lipid profile in patients of subclinical hypothyroidism in tertiary centre in Kolkata, West Bengal and compare the same with matched controls.Methods: This observational study (single exposure) was conducted May 2019 to Nov 2019 at Department of Medicine, KPC Medical College, Jadavpur, Kolkata in subjects diagnosed with Sub Clinical Hypothyroidism [defined as normal T3 or FT3, normal T4 or FT4, and with increased TSH. Sixty patients with subclinical hypothyroidism were selected after careful exclusion; lipid profile was compared with matched controls.Results: Between the two groups (group I - controls vs. group II - cases), the values were as follows: Mean serum total T3 value was 114.03±29.22 ng/dl vs. 106.15±36.24 ng/dl (p = 0.3476); mean total T4 was 7.07±1.69 μg/dl vs. 6.86±1.31 μg/dl (p = 0.535); mean TSH was 3.17±1.28 μIU/ml vs. 9.77±4.19 μIU/ml (p <0.0001). Lipid profile pattern (group I vs. group II) was as follows: Mean total cholesterol (TC) 125.50±9.18 mg/dl vs. 162.07±42.32 mg/dl (p <0.0001), mean triglycerides (TG) is 136.65±14.82 mg/dl vs. 148.90±65.27 mg/dl (p = 0.3236), low-density lipoprotein (LDL)-cholesterol is 62.17±7.40 mg/dl vs. 98.81±33.26 mg/dl (p <0.0001), high-density lipoprotein (HDL)-cholesterol 39.17±6.62 mg/dl vs. 34.27±9.63 mg/dl (p = 0.0702), very low-density lipoprotein (VLDL) levels are 34.54±15.38 mg/dl vs. 32.08±13.21 mg/dl (p = 0.5245)..Conclusions: Subclinical hypothyroidism is associated with increased serum total cholesterol and LDL-Cholesterol levels. Therefore, there is a potential association between Subclinical hypothyroidism and atherosclerosis.

Abstrak: Brotowali mengandung antidioksidan (flavonoid, dan tanin), yang diharapkan dapat menghambatperkembangan lesi aterosklerosis. Salah satu faktor resiko penyebab aterosklerosis yaitu makanan yang mengandung asam lemak trans seperti margarin. Mengonsumsi lemak trans berlebihan dapat mengganggu funsi endotel sehingga lipoprotein berdensitas rendah Low Density Lipoprotein (LDL) dapat masuk dan menjadi yang teroksidasi. Makrofak menangkap Ox-LDL dan menjadi lesi dini ateroklerosis yang secara mikrokopik di sebut dengan sel busa. Tujuan: Mengetahui gambaran histologi aorta tikus wistar dengan diet margarin tanpa pemberian ekstrak batang brotowali, diet margarin bersaamaan pemberian ektrak batang brotowali, setelah diet margarin dilanjutkan pemberian ekstak batang brotowali. Metode: Penelitian eksperimental , 14 ekor tikus wistar dibagi menjadi 4 kelompok, yaitu kelompok I tanpa  perlakuan (kontrol negatif), kelompok II (dkelompok positif), kelompok III diet margarin bersamaan dengan pemberian ekstrak brotowali selama 14 hari (Perlakuan kelompok I) dan kelompok IV (28 hari) 14 hari diet margarin dilanjutkan pemberian ekstrak brotowali. (Perlakuan kelompok II). Hasil: Penelitian ini menunjukkan gambaran histologi aorta tikus wistar:kelompok I normal, kelompok II dan III terdapatnya sel-sel busa, kelompok IV masih terdapat sel busa, namun dalam jumlah lebih sedikit dibandingkan kelompok II dan III. Simpulan: Tikus wistar dengan diet margarin selama 14 hari didapatkan gambaran histologi terdapat sel-sel busa pada tunika intima dan tunika media, kemudian dilanjutkan dengan pemberian ekstrak brotowali hari ke 15 sampai hari ke 28, sel-sel busa berkurang pada tunika intima dan tunika media. Kata kunci: brotowali, margarin, sel-sel busa, tikus wistar.   Abstract: Brotowali contains antidioksidan (flavonoids, and tannins), which is expected to inhibit the development of atherosclerotic lesions. One of the causes of atherosclerosis risk factors which are foods that contain trans fatty acids like margarine. Consuming excessive trans fats can interfere with endothelial function is so low density lipoprotein Low Density Lipoprotein (LDL) can enter and become oxidized. Makrofak capture Ox-LDL and into early lesions of atherosclerosis that are mikrokopik called foam cells. Objective: To determine the aorta histology Wistar rats with diet margarine without brotowali extract, diet margarine brotowali concurrent administration of extract, diet margarine followed after giving brotowali extract. Methods: Experimental research, on 14 Wistar rats were divided into 4 groups: group I with no treatment (negative control), group II (positive group), group III in conjunction with diet margarine brotowali extract for 14 days (treatment group I) and the group IV (28 days) 14 days diet margarine continued brotowali extract. (Treatment group II). Results: This study shows histologic Wistar rat aorta: normal group I, group II and III the presence of foam cells, there is still a group IV foam cells, but in much smaller amounts than group II and III. Conclusion: Wistar rats with diet margarine for 14 days obtained histology contained foam cells in the tunica intima and tunica media, and the continuation of the extract brotowali day 15 to day 28, reduced foam cells in the intima and tunica media . Abstract Background Pemafibrate, a selective PPARα modulator, has the beneficial effects on serum triglycerides (TGs) and very low density lipoprotein (VLDL), especially in patients with diabetes mellitus or metabolic syndrome. However, its effect on the low density lipoprotein cholesterol (LDL-C) levels is still undefined. LDL-C increased in some cases together with a decrease in TGs, and the profile of lipids, especially LDL-C, during pemafibrate administration was evaluated. Methods Pemafibrate was administered to type 2 diabetes patients with hypertriglyceridemia. Fifty-one type 2 diabetes patients (mean age 62 ± 13 years) with a high rate of hypertension and no renal insufficiency were analyzed. Pemafibrate 0.2 mg (0.1 mg twice daily) was administered, and serum lipids were monitored every 4–8 weeks from 8 weeks before administration to 24 weeks after administration. LDL-C was measured by the direct method. Lipoprotein fractions were measured by electrophoresis (polyacrylamide gel, PAG), and LDL-migration index (LDL-MI) was calculated to estimate small, dense LDL. Results Pemafibrate reduced serum TGs, midband and VLDL fractions by PAG. Pemafibrate increased LDL-C levels from baseline by 5.3% (− 3.8–19.1, IQR). Patients were divided into 2 groups: LDL-C increase of > 5.3% (group I, n = 25) and < 5.3% (group NI, n = 26) after pemafibrate. Compared to group NI, group I had lower LDL-C (2.53 [1.96–3.26] vs. 3.36 [3.05–3.72] mmol/L, P = 0.0009), higher TGs (3.71 [2.62–6.69] vs. 3.25 [2.64–3.80] mmol/L), lower LDL by PAG (34.2 [14.5, SD] vs. 46.4% [6.5], P = 0.0011), higher VLDL by PAG (28.2 [10.8] vs. 22.0% [5.2], P = 0.0234), and higher LDL-MI (0.421 [0.391–0.450] vs. 0.354 [0.341–0.396], P < 0.0001) at baseline. Pemafibrate decreased LDL-MI in group I, and the differences between the groups disappeared. These results showed contradictory effects of pemafibrate on LDL-C levels, and these effects were dependent on the baseline levels of LDL-C and TGs. Conclusions Pemafibrate significantly reduced TGs, VLDL, midband, and small, dense LDL, but increased LDL-C in diabetes patients with higher baseline TGs and lower baseline LDL-C. Even if pre-dose LDL-C remains in the normal range, pemafibrate improves LDL composition and may reduce cardiovascular disease risk.

Two glycopeptides (A and B) were isolated from pronase-digested vitellenin, the protein moiety of the low-density lipoprotein of hen's egg yolk. Aspartic acid was the only N-terminal amino acid of both glycopeptides but only A contained N-acetylneuraminic acid. A contained 55% hexose (mannose), 14% hexosamine, 12% N-acetylneuraminic acid, 0.71% amide nitrogen, and its molecular weight was 2.3 × 103. The corresponding values for B were 64, 17, 0.0, 0.75, and 2.0 × 103. Chemical analyses showed that B (and probably A) occurs in vitellenin with the heteropolysaccharide group bound N-glycosidically via the β-amide group of an asparaginyl residue. The indicated structure is R∙(NH)Asp∙Thr∙Ser∙(Ala, Gly, Val)∙Ile, where R, the heteropolysaccharide group, contains 2 hexosamine and 8 hexose residues.

The purpose of this research was to know the effect of insemination time on motility and viability of spermatozoa in egg yolk tris diluter post insemination of goat. In this research 20 female goats was used and divided into two groups and synchronized with PGF2 alfa. If sign of oestrus appeared, then goats in group I, were inseminated with semen from buck diluted with egg yolk tris at the beginning of oestrus and group II inseminated with semen from buck diluted with egg yolk tris at the mid oestrus. Semen was collected from cervical canal of goats at one, two, 3, 6, or 24 hours after insemination for evaluating its motility and viability. Implication of this research is indicate that artificial insemination can be conducted in the early also the mid oestrus of goat.

ABSTRACT This study investigated in vitro the efficacy of four different extenders (TES-TRIS and TRIS with LDL low-density lipoprotein at concentrations of 10 or 5%) on the longevity of buffalo sperm in the refrigeration process at 5ºC. Sperm motility was assessed every 24 hours up to 72 hours of incubation using computer assisted sperm analysis and sperm membrane integrity was examined by the hypoosmotic test (HOST) at T1, T24, T48 and T72 hours. Eleven buffaloes (1 ejaculate per buffalo) of the Murrah breed were used, ranging in age from 4 to 5 years. Immediately after collection, each ejaculate was fractionated into 4 aliquots, and each aliquot was diluted in one of four diluents to obtain 50x106SPTZ/mL. The samples were packed in 0.5mL straws and refrigerated (-0.25°C/min) to 5°C and maintained at this temperature until evaluation. Prior to evaluation the samples were heated at 37°C for 30 seconds. The statistical package used for analysis was STATA 12.0 "Statistical Analysis Software" and means were compared by the Friedman test (P<0.05). The results of sperm kinetics and HOST indicate that the TRIS diluent with 10% LDL could be a promising alternative for semen refrigeration at 5ºC, to be used in conventional and fixed time artificial insemination.
