Background: Dengue Virus Infektion is major public health problem in Indonesia. Aedesaegypti is widespread in both urban and rural areas, where multiple virus Serotype are circulating. On 2013 outbreak of dengue virus infection occur in East Java. Therefore study seroepidemiology in Bangkalan and Lombok had been done. Aim: to find a mutated strain of Dengue Virus in 4 cities of Indonesia. Method: On 2011 and 2012 seroepidemiology study had been done in Dr. Soetomo Surabaya and Soerya Sidoarjo Hospital; and on  2013 study had been done in Surabaya, Bangkalan and Lombok Hospital . Diagnosis of Dengue Virus Infection was based on Criteri WHO - 2009. Virus isolation in Surabaya, Sidoarjo, Bangkalan and Lombok had been done. Result: a total of 349 isolate were obtained from dengue patients sera collected in Surabaya and Sidoarjo, 2011–2012 showed that Den V1 (182), Den V2 (20) Den V4 (1) were found in Surabaya on 2011 and Den V 1 (79) , Den V 2 (7) were found in Surabaya on 2012; Den V1 (40), Den V 2 (3) were found in Sidoarjo on 2011 and Den V 1 (17) were found in Sidoarjo on 2012; Virus isolation in Surabaya on 2013, January: 237 serum sample were collected, found Den V 1 (8), Den V 3 (2) and Den V 4 (5). And PCR stereotyping of isolated viruses in Madura found Den V 1 (1) and Den V 4 (23). In Lombok found Den V 4 (4).It is possible to shift predominant strain in Surabaya , Genotype or Serotype shift might increase the number of dengue patients. Conclusion: there were shift predominant strain in Surabaya especially Den V 1. Therefore to continuous surveillance of circulating viruses is required to predict the risk of DHF and DF.

Abstract: Dengue is a major public health problem that can be found throughout the tropical and subtropical countries around the world. Spectrum of clinical manifestations of dengue virus infection varies greatly. In addition to clinical symptoms, the diagnosis of dengue virus infection need to be supported by blood tests such as hematocrit and erythrocyte sedimentation rate. This study aimed to determine the hematocrit value and erythrocyte sedimentation rate (ESR) in children with dengue virus infection in Manado. This study used a cross-sectional design and was conducted from Desember 2014 to January 2015 at GMIM Pancaran Kasih, Advent, and Robert Wolter Mongisidi hospitals in Manado. There were 37 patients that fulfilled the inclusion criteria, consisted of 17 males and 20 females. The results showed that of 37 patients, there were 36 with hematocrit within normal limits. Moreover, of 37 patients only 6 patients performed the ESR examination; only 1 child had a rapid ESR result (> 15mm/h). Conclusion: In this study, most of the pediatric patients with dengue virus infection in Manado had normal hematocrit. Of 6 patients who had performed ESR test, only 1 had rapid ESR.Keywords: dengue, hematocrit, erythrocyte sedimentation rateAbstrak: Dengue merupakan masalah kesehatan masyarakat yang utama dan dapat ditemui diseluruh daerah tropis dan subtropis di seluruh dunia. Spektrum manifestasi klinis infeksi virus dengue sangat bervariasi. Selain gejala klinis, diagnosis infeksi virus dengue perlu ditunjang hasil uji darah di laboratorium antara lain hematokrit dan laju endap darah (LED). Penelitian ini bertujuan untuk mengetahui nilai hematokrit dan LED pada anak dengan infeksi virus dengue di Manado. Penelitian ini menggunakan desain potong lintang, dilakukan pada bulan Desember 2014 sampai Januari 2015 di RSU GMIM Pancaran Kasih Manado, RS Advent Manado, dan RSAD Robert Wolter Mongisidi Teling17 anak laki-laki dan 20 anak perempuan. Hasil penelitian memperlihatkan 36 dari 37 pasien mempunyai hematokrit normal. Dasri 37 pasien, hanya 6 anak yang menjalani pemeriksaan LED; hanya 1 anak dengan LED cepat (> 15mm/jam). Simpulan: Pada studi ini, sebagian besar pasien anak dengan infeksi virus dengue di Manado mempunyai nilai hematokrit normal. Dari 6 pasien yang dilakukan pemeriksaan LED, 1 anak mempunyai hasil LED cepat.Kata kunci: dengue, hematrokrit, laju endap darah.

This article reviews Dengue, a common viral disease in humans and is an emerging public health problem in Tarai Region of Nepal. The most affected are among the poorest populations living in remote, rural areas and urban slums who have even no access for medical treatment, acquired by bite of infected mosquito. Aedes Aegypti infected with dengue virus is the major source of infections for humans and cannot be transmitted from person-to-person because human are the dead end host. DENV-1 was first isolated by Ren Kimura and Susumu Hotta in Japan in 1943. An epidemic of DF involving at least 200,000 cases had occurred between 1942 and 1944 during World War II in Japanese port cities such as Nagasaki, Kobe, and Osaka. First case of dengue was reported in 2004 in Nepal. The seroprevalence study were done in different part of Nepal by IgM antibody capture ELISA and positive rate was highest (50.0%) in Biratnagar, and lowest (19.6%) in Chitwan male to female ratio was 2:1. IgM-positive rate was 29.0% at ages 21-30, 25.4% at ages 11-20 and 23.6% at ages 0-10, but 10.9% at ages 31-40, and ages over 40. There was not significant association between occupation of the patients and positive rate among farmer, labour, service, business and student. Introduction: Detection of dengue virus infection as soon as possible is critical for management of dengue virus infected patients. Immuno-chromatographic (ICT) tests are easy, cost effective method for dengue virus antigen detection.The sensitivity and specificity of ICT should compare with a gold standard test like RT-PCR. Aim of this study was to compare two test methods (ICT and RT-PCR), observe dengue serotype and seasonal impact on dengue infection.Methodology & result: The patients of Ibn Sina Medical College Hospital from October 2015 to October 2017 were tested for dengue NS1 antigen by ICT method. Out of 3201 sample tested 32.39% were found positive and 89 of which were re-tested for RT-PCR for comparison. Eighty eight of 89 NS1 positive cases showed positive by RT-PCR method giving an accuracy of 98.87%. Among the RT-PCR positive cases 45 were further analyzed for serotype. DEN-1, DEN-2 or both DEN- 1 and DEN-2 were found in 21, 23 and 1cases respectively. No cases of DEN-3 or DEN-4 were detected.Conclusion: This study showed that easily available and cost effective dengue NS1 antigen detection method (ICT) is as effective as molecular test (RT-PCR). DEN-1 and DEN-2 serotype were prevalent during last few years in Bangladesh. Continuous monitoring of dengue virus serotype is important for prevention and control of sudden epidemic by other serotype. INTRODUCTION: Dengue fever (DF) is an emerging mosquito borne viral disease and important public health problem in low land Terai region which is also moving towards hilly region Nepal. This study was designed to determine the sero-prevalence of dengue virus infection in patients visiting hospitals of Nepal. MATERIALS AND METHODS: This study was conducted during period (June-November) of 2010 in Nepalese patients with fever visiting hospitals of Birganj, Damouli, Biratanagar, Dhading Besi and Chitwan. The sero-prevalence of dengue virus specific IgM was determined by enzyme linked immunosorbent assay (ELISA). Serum samples were collected from 289 patients visiting hospitals with history of fever and clinically suspected dengue fever. RESULTS: The anti-dengue IgM positivity was found to be 8.99%. The positive dengue cases were higher in male (10.8%) as compared to female (7.1%) though it was not statistically significant (P>0.05). Among different age groups, the highest positive cases (12.3%) were from age group below 15 years followed by above 50 years 8.3%. Out of 5 hospitals, the highest positive cases were in Tanahu hospital, Damouli (23.8%) followed by Bharatpur hospital and Chitwan (22.2%). Age and gender were found to be independent predictors. The highest numbers of dengue positive cases were in occupation group business (13.3%) followed by agriculture (12.7%). CONCLUSIONS: Prevalence of dengue virus infection is increasing and proper control measure should be provided. IgM capture ELISA was used for laboratory analysis and remains as a reliable and inexpensive method for the diagnosis of dengue. Hence, the IgM capture ELISA has become the most accepted technique for the diagnosis of dengue in developing countries like Nepal. Dengue Virus infection is an emerging mosquito-borne disease. It is a global health problem and its expanding endemicity towards new territories is a serious concern. Relatively a new disease in Nepalese context, dengue abruptly appeared as massive outbreak in 2010, merely four years after its first introduction. It is a nagging public health problem in the low lands of Terai, expanding to new areas of Nepal in recent years.  A cross-sectional study was conducted to determine anti-Dengue IgM positive rate in Lumbini, Dhading and Chitwan district. The study was carried from June 2012 to November 2012. The total number of Serum samples was collected from 275 patients visiting hospitals with history of fever, headache and suspected DF. The samples were examined by ELISA. The anti-Dengue IgM positivity was found to be 29.09 %. The positive rate was highest in Dhading (70.37%) followed by Bharatpur (37.6%) and Lumbini (11.38%). The Dengue positive cases were higher in males (32.5 %) than female (24.8 %). The highest positive cases (41.6%) were from age group less than 15 years. Dengue has substantial expansion in Western and Far Western Terai region of Nepal which was limited to the middle Terai region in the past and mostly infects older people.DOI: http://dx.doi.org/10.3126/ijasbt.v1i4.9135  Int J Appl Sci Biotechnol, Vol. 1(4): 224-227

Here, we report the complete genome sequence of a chikungunya virus coinfection strain isolated from a dengue virus serotype 2-infected patient in Malaysia. This coinfection strain was determined to be of the Asian genotype and contains a novel insertion in the nsP3 gene.

Abstract INTRODUCTION: Dengue virus (DENV) causes a wide range of diseases in humans, from acute febrile illness Dengue fever (DF) to life-threatening Dengue hemorrhagic fever (DHF) or Dengue shock syndrome (DSS). Factors believed to be responsible for spread of Dengue virus infection include explosive population growth, unplanned urban overpopulation with inadequate public health systems, poor standing water and vector control, climate changes and increased international recreational, business, military travel to endemic areas. All of these factors must be addressed to control the spread of Dengue and other mosquito-borne infections. The detection of Dengue virus RNA by reverse transcriptase PCR (RT-PCR) in human serum or plasma samples is highly indicative of acute Dengue fever. Moreover, the method is able to identify the Dengue virus serotype by demonstrating defined sequence homologies in the viral genomic RNA. METHODS AND RESULTS: During the nine year period of this study analysis, 6767 strongly suspected cases were tested by RT-PCR. 1685 (24.9%) were Dengue PCR positive and confirmed as Dengue cases. Observations on the seasonality were based on the nine year's data as the intensity of sampling was at its maximum during monsoon season. Dengue typing was done on 100 positive samples after storage of Dengue RNA at – 80°C. Dengue serotypes were detected in 69 samples of which Dengue 2 was most predominant. 576 samples were processed for NS1 antigen and PCR simultaneously. 19/576 were positive (3.3 %) for NS1 as well as by PCR. 23/576 samples were negative for NS1 antigen, but were positive by RT-PCR. The remaining 534 samples which were negative for NS1 antigen were also negative by Dengue RT-PCR. CONCLUSION: In this study we sought to standardize rapid, sensitive, and specific fluorogenic probe-based RT-PCR assay to screen and serotype a representative range of Dengue viruses that are found in and around Mumbai. Qualitative Dengue virus TaqMan assays could have tremendous utility for the epidemiological investigation of Dengue illness and especially for the study of the viremic response with candidate live-attenuated dengue virus vaccines.

Dengue virus infection is associated with the upregulation of metabolic pathways within infected cells. This effect is common to infection by a broad array of viruses. These metabolic changes, including increased glucose metabolism, oxidative phosphorylation and autophagy, support the demands of viral genome replication and infectious particle formation. The mechanisms by which these changes occur are known to be, in part, directed by viral nonstructural proteins that contact and control cellular structures and metabolic enzymes. We investigated the roles of host proteins with overarching control of metabolic processes, the transcriptional regulators, cyclin-dependent kinase 8 (CDK8) and its paralog, CDK19, as mediators of virally induced metabolic changes. Here, we show that expression of CDK8, but not CDK19, is increased during dengue virus infection in Huh7 human hepatocellular carcinoma cells, although both are required for efficient viral replication. Chemical inhibition of CDK8 and CDK19 with Senexin A during infection blocks virus-induced expression of select metabolic and autophagic genes, hexokinase 2 (HK2) and microtubule-associated protein 1 light chain 3 (LC3), and reduces viral genome replication and infectious particle production. The results further define the dependence of virus replication on increased metabolic capacity in target cells and identify CDK8 and CDK19 as master regulators of key metabolic genes. The common inhibition of CDK8 and CDK19 offers a host-directed therapeutic intervention that is unlikely to be overcome by viral evolution.

PCR (polymerase chain reaction) is a routinely used tool in every diagnostic and research laboratory. This technique has been used in detection of mutations and pathogens, forensic investigation, and even is the base tool for human genome sequencing. A modification of PCR technique, real time PCR, allows the quantification of nucleic acids with higher sensibility, specificity and reproducibility. This article is intended to clarify the foundations of real-time PCR, using an application model for virology. In the actual work, it was quantified the viral load of dengue virus serotype 2 produced from infected murine macrophages; the obtained results in this work established that murine strain BALB/c presents a greater susceptibility to dengue virus infection, which establishes BALB/c murine strain as a best model of study for investigation of dengue virus infection physiopathology.

AbstractLack of a dengue hemorrhagic animal model recapitulating human dengue virus infection has been a significant impediment in advancing our understanding of the early events involved in the pathogenesis of dengue disease. In efforts to address this issue, a group of rhesus macaques were intravenously infected with dengue virus serotype 2 (strain 16 681) at 1 × 107 PFU/animal. A classic dengue hemorrhage developed 3 to 5 days after infection in 6 of 6 animals. Blood chemistry appeared to be normal with exception of creatine phosphokinase, which peaked at 7 days after infection. A modest thrombocytopenia and noticeable neutropenia concomitant with slight decrease of hemoglobin and hematocrit were registered. In addition, the concentration of D-dimer was elevated significantly. Viremia peaked at 3 to 5 days after infection followed by an inverse relationship between T and B lymphocytes and a bimodal pattern for platelet-monocytes and platelet-neutrophil aggregates. Dengue virus containing platelets engulfed by monocytes was noted at 8 or 9 days after infection. Thus, rhesus macaques inoculated intravenously with a high dose of dengue virus produced dengue hemorrhage, which may provide a unique platform to define the early events in dengue virus infection and help identify which blood components contribute to the pathogenesis of dengue disease.
