Background: Carbonate hydroxyapatite (CHA) is used as a scaffold to repair bone resorption. Alveolar bone resorption in periodontitis caused by an infection requires the presence of an antibacterial to support bone regeneration. Human β-defensin 3 (HBD3) is an antimicrobial peptide. The local application of the HBD3 antimicrobial is beneficial to inhibiting drug resistance and protecting tissue regeneration against invasive bacteria. Purpose: This study aims to investigate the effect of the administration of a combination of   CHA with HBD3 on the collagen density of periodontitis rats (Sprague Dawley). Methods: This study was a true experimental study with a post-test control group design. Thirty-two Sprague Dawley animal models were randomly blind selected placed under anaesthetic, then a 2-mm silk ligature was attached as a ligation to the mandibular incisors for 14 days in order to generate periodontitis. The study subjects were divided into two groups, the group with CHA and CHA loaded HBD3 (CHA + HBD3) implantation. On days 7, 14, 21 and 28, four rats were taken randomly from each group for decapitation, followed by histological processing and examination with trichome Mallory staining. The data was analysed using the Kruskal Wallis test (p<0.05). Results: An increase in collagen density during the healing process was found. There was a significant difference between CHA and CHA+HBD (p=0.004 and p=0.008; p<0.05) in collagen density between the groups. Conclusion: The combination of CHA and HBD3 can enhance the collagen density in periodontitis Sprague Dawley rats, compared to CHA only groups.

Gelam honey has been shown to exhibit antioxidant and anti-inflammatory activities in animal model. The aim of this study was to determine the effects of Gelam honey (Melaleuca cajuputi) on alveolar bone level in experimental periodontitis. Thirty male Sprague-Dawley rats were used in this study and randomly divided into four groups: ligated saline (LS), ligated honey (LH), nonligated saline (NLS), and nonligated honey (NLH). Fifteen days after supplementation with Gelam honey (3 g/kg), the rats were sacrificed and alveolar bone level was determined by radiography and histomorphometry. The number of osteoclasts was also calculated for all groups. Both radiographic and histomorphometric analyses showed that alveolar bone resorption was severely induced around the ligated molar in the LS and LH groups. There was no significant difference in alveolar bone level between the LS and LH groups. However, there was a nonsignificant reduction of osteoclast number by 15.2% in LH group compared to LS group. In the NLH group, there was less alveolar bone resorption and the number of osteoclasts was reduced by 13.2% compared to NLS group. In conclusion, systemically supplemented Gelam honey was shown to have the potential of reducing osteoclast activity in the experimental periodontitis rats, even though the effect on alveolar bone level was not well demonstrated and it warrants further research.

Cedera sel yang luas pada luka bakar melepaskan banyak mediator inflamasi. Respon tubuh yang berlebihan pada keadaan tersebut akan memperberat kerusakan sel yang sudah terjadi. Nurtrisi (lemak) yang cukup seperti omega 3 dapat membantu proses penyembuhan luka serta mencegah infeksi pada luka supaya tidak menjadi luka kronis. Maka dari itu peneliti memilih judul penelitian tentang efektifitas kombinasi suplementasi omega 3 dengan restriksi kalori terhadap penyembuhan luka bakar pada tikus Sprague Dawley, ditinjau dari VEGF dan Re-epitelisasi. Untuk mengetahui tingkat efektifitas suplementasi Omega 3 dan retriksi kalori pada penyembuhan luka bakar. Peneliti dalam melakukan penelitian menggunakan metode penelitian eksperimental dengan menggunakan 1 kelas kontrol dan 3 kelas perlakuan tikus Sprague Dawley, yang diinduksi luka bakar kemudian diberi perlakuan, dan diamati secara berkala. Hasil penelitian didapatkan perbedaan yang signifikan antara kelompok kontrol dan kelompok perlakuan yang di uji dengan one way ANOVA mempunyai nilai p = 0,000 (p < 0,05), dan uji Mann-Whitney didapatkan perbedaan yang signifikan antara kelompok perlakuan 1, 2, 3, dan kelompok kontrol p = 0,025; 0,004; 0,004; 0,004; 0,004, nilai p < 0,05. Dan uji Games-Howell ditunjukkan pada kolom rerata ± SD jika memuat huruf yang  berbeda berarti ada perbedaan yang bermakna (p-value <0.05) K1 = 23,3587a ± 0,57556; P1 = 17,1015b ± 2,60108; P2 = 124,1503c ± 15,30168; P3 = 237,5053d ± 0,78263. Kesimpulan yang peneliti dapat bahwa kombinasi suplementasi omega 3 dengan retriksi kalori berpengaruh signifikan terhadap penyembuhan luka bakar pada tikus Sprague Dawley. It’s led to excessive body response and cause aggravate cell damage that has already occurred. Adequate nutrients (fat) like omega 3 can help the healing process and prevent infection in wounds to become chronic. Therefore the researchers chose the title of the study of the effectiveness combination omega-3 supplementation with calorie restriction for burns in Sprague Dawley rats, in terms of VEGF and Re-epithelialization. To determine the effectiveness of Omega 3 supplementation and calorie restriction when healing burns. The researchers using experimental research methods with 1 control and 3 training classes of Sprague Dawley mice, which were induced by burns were then observed periodically. The results showed a significant difference between the control group and the treatment group tested by one way ANOVA having a value of p = 0,000 (p <0.05), and the Mann-Whitney test found a significant difference between the treatment groups 1, 2, 3, and the control group p = 0.025; 0.004; 0.004; 0.004; 0.004, p value <0.05. Osteoporosis is a bone disease characterized by reduced bone mass and damage to the microarchitecture of bone tissue. One of osteoporosis types is primary type-1 osteoporosis or postmenopausal osteoporosis due to estrogen deficiency. Calcium is an important mineral that plays a central role in the formation of bone strength structure. Jackfruit seeds (Artocarpus heterophyllus) contain isoflavones that can replace the role of estrogen in the body. Fortification of jackfruit seeds and high calcium milk can be a potential alternative for the treatment of postmenopausal osteoporosis. The purpose of this study was to determine the effect of drinking high calcium milk fortified with jackfruit seed extract (Artocarpus heterophyllus) on alveolar bone collagen fiber density of menopause induced rats. Twenty-five Sprague dawley rats were included in this study. Twenty Sprague dawley rats were ovariectomized, while 5 rats were left untreated (normal control group). Seven days after ovariectomy was performed, 5 rats were treated with fortification of jackfruit seed extract and high calcium milk, 5 rats were treated with jackfruit seed extract, 5 rats were treated with high calcium milk, and 5 rats were left untreated (ovariectomized control group). The treatments were done for 28 days. The rats were euthanized and the alveolar bone was taken and processed for a histological specimen. Alveolar bone collagen fibers were observed under a binocular microscope with 100 magnification. Kruskal-Wallis result showed there was a significant difference in the alveolar bone collagen fiber density among these groups (p<0.05). Mann-Whitney U Test showed a significant difference (p<0.05) in the alveolar bone collagen fiber density of the rats treated with fortification of jackfruit seed extract and high calcium milk compared to the group treated with jackfruit seed extract, that with high calcium milk, or ovariectomized control group. No significant difference in alveolar bone collagen fiber density was found between the group treated with fortification of jackfruit seed extract and high calcium milk and the normal control group (p>0.05). In conclusion, consuming high calcium milk fortified with jackfruit seed extract increases the density of alveolar bone collagen fibers.

Background: Tooth extraction can cause alveolar resorption, and will reduce the denture retention. The process of bone resorption looks like the process of osteoporosis. Calcium and vitamin D supplementation is the rational therapy for minimizing bone loss. Miacalcic is the drug of choice for osteporotic patient. Purpose: This study is aimed to know whether the combination of miacalcic, calcium lactate, and vitamin C are effective in inhibiting post extracted alveolar resorption. Methods: Thirty three healthy postmenopausal women were chosen as samples and they were classified randomly into control group (without treatment), 1st experiment group (treatment was started 3 months post extraction), and 2nd experiment group (treatment was started at the 2nd day post extraction). The treatment was done by giving miacalcic nasal spray, calcium lactate 500 mg and vitamin C 100 mg tablets every morning in 10 days every month for 3 months. X-ray photo of the post extracted area were taken an hour, 3 months, and 6 months post-extraction. Results: After 6 month, there was significant difference in buccolingual thickness decreasing among three groups (p<0.05). The maximum mean difference of buccolingual thickness decreasing was 0.72 mm, between control and 2nd experiment groups. There was no significant difference about decreasing bone density among them (p>0.10). The maximum difference of the mean of density decreasing was 1,906 g/cm2/mm between control and 2nd experiment groups. The increasing density mostly occurred in the 2nd experiment group. Conclusion: The combination of miacalcic, calcium lactate, and vitamin C are effective for inhibiting alveolar resorption, although statistically there was no significant difference about bone density decreasing. The sooner this treatment is given the better result will be achieved.Latar belakang: Pencabutan gigi menyebabkan resorpsi tulang alveolaris, dan akan mengurangi retensi geligi tiruan. Proses resorpsi tulang alveol pada osteoporosis mirip dengan proses resorpsi tulang pada penyembuhan luka bekas pencabutan. Miacalcic adalah obat utama untuk penderita osteoporosis. Kalsium dan vitamin D merupakan terapi yang rasional untuk meminimalkan resorpsi tulang. tujuan: Membuktikan apakah kombinasi miacalcic, kalsium laktat, and vitamin C juga efektif menghambat resorpsi tulang alveol pasca pencabutan. Metode: Sampel 33 wanita postmenopause yang sehat, terbagi secara acak ke dalam kelompok kontrol (tanpa perlakuan), kelompok eksperimen 1 (perlakuan mulai 3 bulan pasca pencabutan) dan kelompok eksperimen 2 (perlakuan mulai hari kedua pasca pencabutan). Perlakuannya yaitu: pemberian miacalcic semprot hidung, tablet kalsium laktat 500 mg dan vitamin C 100 mg setiap pagi, 10 hari dalam sebulan, selama tiga bulan. Foto sinar-X dari regio pasca pencabutan dibuat satu jam, 3 bulan, dan 6 bulan pasca pencabutan. Hasil: 6 bulan pasca-cabut, ada beda bermakna perihal selisih tebal bukolingual tulang alveol antar ketiga kelompok (p<0,05). Rerata penurunan ketebalan ini maksimal sebanyak 0.72 mm, antara kelompok kontrol dan kelompok eksperimen 2. Penurunan kepadatan tulang antar ketiga kelompok tidak bermakna (p>0,10). Beda maksimum rerata kepadatan tulang antara kelompok kontrol dan kelompok eksperimen 2 sebesar 1,906 g/cm2/mm. Peningkatan kepadatan terbanyak dialami anggota kelompok eksperimen 2. Kesimpulan: Kombinasi miacalcic, kalsium laktat, vitamin C efektif menghambat resorpsi tulang alveolaris, walaupun secara statistik beda penurunan kepadatan tidak bermakna. Makin awal pemberian perlakuan, hasilnya akan lebih baik.

Pendahuluan: Pencabutan gigi merupakan prosedur yang sering dilakukan di kedokteran gigi. Setelah pencabutan gigi akan dihasilkan suatu perlukaan. Proses penyembuhan luka dapat dipercepat pada kondisi tertentu. Salah satu bahan alami yang dapat membantu proses penyembuhan luka adalah getah tanaman betadine (Jatropha multifida L.). Penelitian bertujuan untuk menganalisis efektivitas getah tanaman betadine terhadap penyembuhan luka dan terhadap tanda-tanda infeksi pasca pencabutan gigi Metode: Desain penelitian studi eksperimental laboratorium dengan rancangan penelitian post-test only control group design menggunakan 30 ekor tikus Sprague-Dawley. Teknik pengambilan sampel yaitu convenience sampling. Sampel dibagi menjadi 2 kelompok yaitu kelompok perlakuan dan kelompok kontrol. Kelompok perlakuan diberikan getah tanaman betadine dan kelompok kontrol diberikan asam traneksamat secara oral menggunakan sonde lambung. Tunggu selama 4 jam setelah pemberian getah tanaman betadine dan asam traneksamat. Anestesi pada tikus menggunakan ketamin 50 mg/kg berat badan secara intramuskular kemudian dilakukan pencabutan pada gigi tikus. Pengamatan dilakukan dengan melihat kriteria klinis pada hari 1,3,7 dan diperhatikan sampai luka sembuh serta lihat tanda-tanda infeksi. Analisis data dilakukan dengan uji normalitasShapiro Wilik. Hasil penelitian menunjukkan bahwa data berdistribusi tidak normal. Analisis data dilanjutkan menggunakan uji statistik mann whitney. Hasil: Terdapat perbedaan signifikan penyembuhan luka soket pasca pencabutan gigi setelah diberikan getah betadine dan asam traneksamat dengan nilai p=0,037 (p<0,005). Simpulan: Pemberian getah tanaman betadine terbukti lebih efektif terhadap proses penyembuhan luka soket pasca pencabutan gigi dibandingkan dengan pemberian asam traneksamat. Kata kunci: tikus Sprague-Dawley; penyembuhan luka; pencabutan gigi; getah batang betadine ABSTRACTIntroduction: Tooth extraction is a procedure often performed in dentistry. Tooth extraction will always cause injuries. However, the wound healing process can be accelerated under certain conditions. One of the natural ingredients that can accelerate the wound healing process is betadine (Jatropha multifida L.) plant sap. The study was aimed to analyzed the effect of betadine plant sap on wound healing and signs of infection after tooth extraction. Methods: Experimental laboratory study design with post-test only control group design was conducted towards 30 Sprague-Dawley rats. The sampling technique was convenience sampling. The sample was divided into two groups, the treatment group and the control group. The treatment group was administered with betadine plant sap, and the control group was administered with tranexamic acid orally using a gastric probe, then waited 4 hours after. The anaesthesia was then performed using 50 mg/kg body weight of ketamine intramuscularly. The extraction was performed after. Observations was conducted at the clinical criteria on days 1, 3, and 7 and continue to be monitored until the wound heals. Then, the signs of infection were observed. Data analysis was carried out using the Shapiro Wilk normality test. The results showed that the data was not normally distributed. Thus, data analysis was continued using the Mann Whitney statistical test. Results: The results showed a significant difference in the healing of socket wounds after tooth extraction after being administered with betadine sap and tranexamic acid with a value of p=0.037 (p<0.005). Conclusions: Administration of betadine plant sap is proven to be more effective in accelerating the healing process of socket wounds after tooth extraction than tranexamic acid. Keywords: Sprague-Dawley rats; wound healing; pencabutan gigi; getah batang betadine

Background. This study examines the impact of one-time direct application of haemostatic agent zeolite–bentonite powder to wounded skin on the healing process in rats. Materials and Methods. 24 male Sprague-Dawley rats were randomly allocated into two groups (n=12): (1) the rats whose wounds were washed only with sterile normal saline (NS-treated) and (2) those treated with zeolite–bentonite compound (ZEO-treated). The wound was circular, full-thickness, and 2 cm in diameter. At the end of the 12th day, six animals from each group were randomly selected and terminated. The remaining rats were terminated after 21 days. Just after scarification, skin samples were excised and sent for stereological evaluation. Results. The results showed a significant difference between the two groups regarding the length density of the blood vessels and diameter of the large and small vessels on the 12th day after the wound was inflicted. Besides, volume density of both the dermis and collagen bundles was reduced by 25% in the ZEO-treated rats in comparison to the NS-treated animals after 21 days. Conclusions. One-time topical usage of zeolite–bentonite haemostatic powder on an animal skin wound might negatively affect the healing process through vasoconstriction and inhibition of neoangiogenesis.

DespiteEuphorbia hirtaL. ethnomedicinal benefits, very few studies have described the potential toxicity. The aim of the present study was to evaluate thein vivotoxicity of methanolic extracts ofE. hirta. The acute and subchronic oral toxicity ofE. hirtawas evaluated in Sprague Dawley rats. The extract at a single dose of 5000 mg/kg did not produce treatment related signs of toxicity or mortality in any of the animals tested during the 14-day observation period. Therefore, the LD 50 of this plant was estimated to be more than 5000 mg/kg. In the repeated dose 90-day oral toxicity study, the administration of 50 mg/kg, 250 mg/kg, and 1000 mg/kg/day ofE. hirtaextract per body weight revealed no significant difference (P>0.05) in food and water consumptions, body weight change, haematological and biochemical parameters, relative organ weights, and gross findings compared to the control group. Macropathology and histopathology examinations of all organs including the liver did not reveal morphological alteration. Analyses of these results with the information of signs, behaviour, and health monitoring could lead to the conclusion that the long-term oral administration ofE. hirtaextract for 90 days does not cause sub-chronic toxicity.

Aim. The aim of this study was to compare the effects of sildenafil and trimetazidine on bilateral cavernosal nerve injury-induced oxidative damage and fibrotic changes in cavernosal tissue in rat model.Material and Methods. A total of 32 male Sprague-Dawley rats were randomly divided into 4 groups; each group consist 8 rats (control, BCI, BCI + TMZ, and BCI + sildenafil groups). Tissue superoxide dismutase (SOD), malondialdehyde (MDA), and protein carbonyl (PC) levels were determined biochemically and distribution of cavernosal fibrosis density among groups was performed histopathologically.Results. Tissue SOD levels in BCI group were significantly lower than the control group (P<0.05). Tissue MDA and PC levels in BCI group were significantly higher than the control group (P<0.05). TMZ and sildenafil administration significantly increased tissue SOD levels (P<0.05) and reduced tissue MDA and PC levels (P<0.05). Histologically, the degree of cavernosal fibrosis and collagen density was higher in BCI group in comparison to control, TMZ-treated, and sildenafil-treated groups.Conclusion. BCI caused oxidative damage and increased cavernosal fibrosis in rat penis. TMZ and sildenafil treatment decreased oxidative damage and reduced the degree of fibrosis in penile tissue due to BCI.

We studied the urinary concentrating capacity in experimental hemochromatosis. Sprague-Dawley rats were randomized into iron (Fe)-loaded (injected sc with 1.2 g elemental iron/kg body weight as iron dextran) and pair-fed control groups. The urinary concentrating ability was studied after 10 months of iron loading. At basal condition, urine osmolality (Uosm) was significantly lower (P < 0.05) in the Fe-loaded rats compared with the control animals despite comparable urinary arginine-vasopressin (AVP) excretion in the two groups. Although 48-h water deprivation resulted in comparable rises in plasma concentration and urinary excretion of AVP in the two groups, maximal Uosm in the Fe-loaded animals was significantly lower than that seen in the control group (P < 0.01). Moreover, the observed urinary concentrating defect could not be corrected by pharmacological doses of exogenous AVP. There was no significant difference in renal chloride, sodium, calcium, or magnesium handling at either basal or sodium depleted states. Histologic studies showed marked iron deposition in the cortex and outer medulla accompanied by mild tubular atrophy particularly in the distal convoluted tubules. Thus, chronic experimental iron overload leads to nephrogenic diabetes insipidus marked by AVP-resistant urinary concentrating defect.

Background: Damage to bone tissue resulting from tooth extraction will cause alveolar bone resorption. Therefore, a material for preserving alveolar sockets capable of maintaining bone is required. Hydroxyapatite Gypsum Puger (HAGP) is a bio-ceramic material that can be used as an alternative material for alveolar socket preservation. The porous and rough surface of HAGP renders it a good medium for osteoblast cells to penetrate and attach themselves to. In general, bone mass is regulated through a remodeling process consisting of two phases, namely; bone formation by osteoblasts and bone resorption by osteoclasts. Purpose: This research aims to identify the effects of HAGP scaffold application on the number of osteoblasts and osteoclasts, as well as on the width of trabecular bone area in the alveolar sockets of rats. Methods: This research used Posttest Only Control Group Design. There were three research groups, namely: a group with 2.5% HAGP scaffold, a group with 5% HAGP scaffold and a group with 10% HAGP scaffold. The number of samples in each group was six. HAGP scaffold at concentrations of 2.5%, 5% and 10% was then mixed with PEG (Polyethylene Glycol). The Wistar rats were anesthetized intra-muscularly with 100 mg/ml of ketamine and 20 mg/ml of xylazine base at a ratio of 1:1 with a dose of 0.08-0.2 ml/kgBB. Extraction of the left mandibular incisor was performed before 0.1 ml preservation of HAGP scaffold + PEG material was introduced into the extraction sockets and suturing was performed. 7 days after preparation of the rat bone tissue, an Hematoxilin Eosin staining process was conducted in order that observation under a microscope could be performed. Results: There were significant differences in both the number of osteoclasts and osteoblasts between the 2.5% HAGP group, the 5% HAGP group and the 10% HAGP group (p = 0.000). Similarly, significant differences in the width of the trabecular bone area existed between the 5% HAGP group and the 10% HAGP group, as well as between the 2.5% HAGP group and the 10% HAGP group (p=0.000). In contrast, there was no significant difference in the width of the trabecular bone area between the 2.5% HAGP group and the 5% HAGP group. Conclusion: The application of HAGP scaffold can reduce osteoclasts, increase osteoblasts and extend the trabecular area in the alveolar bone sockets of rats.
