Osteoporosis is a bone disease characterized by reduced bone mass and damage to the microarchitecture of bone tissue. One of osteoporosis types is primary type-1 osteoporosis or postmenopausal osteoporosis due to estrogen deficiency. Calcium is an important mineral that plays a central role in the formation of bone strength structure. Jackfruit seeds (Artocarpus heterophyllus) contain isoflavones that can replace the role of estrogen in the body. Fortification of jackfruit seeds and high calcium milk can be a potential alternative for the treatment of postmenopausal osteoporosis. The purpose of this study was to determine the effect of drinking high calcium milk fortified with jackfruit seed extract (Artocarpus heterophyllus) on alveolar bone collagen fiber density of menopause induced rats. Twenty-five Sprague dawley rats were included in this study. Twenty Sprague dawley rats were ovariectomized, while 5 rats were left untreated (normal control group). Seven days after ovariectomy was performed, 5 rats were treated with fortification of jackfruit seed extract and high calcium milk, 5 rats were treated with jackfruit seed extract, 5 rats were treated with high calcium milk, and 5 rats were left untreated (ovariectomized control group). The treatments were done for 28 days. The rats were euthanized and the alveolar bone was taken and processed for a histological specimen. Alveolar bone collagen fibers were observed under a binocular microscope with 100 magnification. Kruskal-Wallis result showed there was a significant difference in the alveolar bone collagen fiber density among these groups (p<0.05). Mann-Whitney U Test showed a significant difference (p<0.05) in the alveolar bone collagen fiber density of the rats treated with fortification of jackfruit seed extract and high calcium milk compared to the group treated with jackfruit seed extract, that with high calcium milk, or ovariectomized control group. No significant difference in alveolar bone collagen fiber density was found between the group treated with fortification of jackfruit seed extract and high calcium milk and the normal control group (p>0.05). In conclusion, consuming high calcium milk fortified with jackfruit seed extract increases the density of alveolar bone collagen fibers.

Background: Carbonate hydroxyapatite (CHA) is used as a scaffold to repair bone resorption. Alveolar bone resorption in periodontitis caused by an infection requires the presence of an antibacterial to support bone regeneration. Human β-defensin 3 (HBD3) is an antimicrobial peptide. The local application of the HBD3 antimicrobial is beneficial to inhibiting drug resistance and protecting tissue regeneration against invasive bacteria. Purpose: This study aims to investigate the effect of the administration of a combination of   CHA with HBD3 on the collagen density of periodontitis rats (Sprague Dawley). Methods: This study was a true experimental study with a post-test control group design. Thirty-two Sprague Dawley animal models were randomly blind selected placed under anaesthetic, then a 2-mm silk ligature was attached as a ligation to the mandibular incisors for 14 days in order to generate periodontitis. The study subjects were divided into two groups, the group with CHA and CHA loaded HBD3 (CHA + HBD3) implantation. On days 7, 14, 21 and 28, four rats were taken randomly from each group for decapitation, followed by histological processing and examination with trichome Mallory staining. The data was analysed using the Kruskal Wallis test (p<0.05). Results: An increase in collagen density during the healing process was found. There was a significant difference between CHA and CHA+HBD (p=0.004 and p=0.008; p<0.05) in collagen density between the groups. Conclusion: The combination of CHA and HBD3 can enhance the collagen density in periodontitis Sprague Dawley rats, compared to CHA only groups.

DespiteEuphorbia hirtaL. ethnomedicinal benefits, very few studies have described the potential toxicity. The aim of the present study was to evaluate thein vivotoxicity of methanolic extracts ofE. hirta. The acute and subchronic oral toxicity ofE. hirtawas evaluated in Sprague Dawley rats. The extract at a single dose of 5000 mg/kg did not produce treatment related signs of toxicity or mortality in any of the animals tested during the 14-day observation period. Therefore, the LD 50 of this plant was estimated to be more than 5000 mg/kg. In the repeated dose 90-day oral toxicity study, the administration of 50 mg/kg, 250 mg/kg, and 1000 mg/kg/day ofE. hirtaextract per body weight revealed no significant difference (P>0.05) in food and water consumptions, body weight change, haematological and biochemical parameters, relative organ weights, and gross findings compared to the control group. Macropathology and histopathology examinations of all organs including the liver did not reveal morphological alteration. Analyses of these results with the information of signs, behaviour, and health monitoring could lead to the conclusion that the long-term oral administration ofE. hirtaextract for 90 days does not cause sub-chronic toxicity.

We studied the urinary concentrating capacity in experimental hemochromatosis. Sprague-Dawley rats were randomized into iron (Fe)-loaded (injected sc with 1.2 g elemental iron/kg body weight as iron dextran) and pair-fed control groups. The urinary concentrating ability was studied after 10 months of iron loading. At basal condition, urine osmolality (Uosm) was significantly lower (P < 0.05) in the Fe-loaded rats compared with the control animals despite comparable urinary arginine-vasopressin (AVP) excretion in the two groups. Although 48-h water deprivation resulted in comparable rises in plasma concentration and urinary excretion of AVP in the two groups, maximal Uosm in the Fe-loaded animals was significantly lower than that seen in the control group (P < 0.01). Moreover, the observed urinary concentrating defect could not be corrected by pharmacological doses of exogenous AVP. There was no significant difference in renal chloride, sodium, calcium, or magnesium handling at either basal or sodium depleted states. Histologic studies showed marked iron deposition in the cortex and outer medulla accompanied by mild tubular atrophy particularly in the distal convoluted tubules. Thus, chronic experimental iron overload leads to nephrogenic diabetes insipidus marked by AVP-resistant urinary concentrating defect.

Gelam honey has been shown to exhibit antioxidant and anti-inflammatory activities in animal model. The aim of this study was to determine the effects of Gelam honey (Melaleuca cajuputi) on alveolar bone level in experimental periodontitis. Thirty male Sprague-Dawley rats were used in this study and randomly divided into four groups: ligated saline (LS), ligated honey (LH), nonligated saline (NLS), and nonligated honey (NLH). Fifteen days after supplementation with Gelam honey (3 g/kg), the rats were sacrificed and alveolar bone level was determined by radiography and histomorphometry. The number of osteoclasts was also calculated for all groups. Both radiographic and histomorphometric analyses showed that alveolar bone resorption was severely induced around the ligated molar in the LS and LH groups. There was no significant difference in alveolar bone level between the LS and LH groups. However, there was a nonsignificant reduction of osteoclast number by 15.2% in LH group compared to LS group. In the NLH group, there was less alveolar bone resorption and the number of osteoclasts was reduced by 13.2% compared to NLS group. In conclusion, systemically supplemented Gelam honey was shown to have the potential of reducing osteoclast activity in the experimental periodontitis rats, even though the effect on alveolar bone level was not well demonstrated and it warrants further research.

The objective of this work was to investigate the effect of orally administered genistein on the pharmacokinetics of imatinib and N-desmethyl imatinib in rats. Twenty-five healthy male SD (Sprague-Dawley) rats were randomly divided into five groups: A group (control group), B group (multiple dose of 100 mg/kg genistein for consecutive 15 days), C group (multiple dose of 50 mg/kg genistein for consecutive 15 days), D group (a single dose of 100 mg/kg genistein), and E group (a single dose of 50 mg/kg genistein). A single dose of imatinib is administered orally 30 min after administration of genistein (100 mg/kg or 50 mg/kg). The pharmacokinetic parameters of imatinib and N-desmethyl imatinib were calculated by DAS 3.0 software. The multiple dose of 100 mg/kg or 50 mg/kg genistein significantly (P<0.05) decreased theAUC0-tandCmaxof imatinib.AUC0-tand theCmaxof N-desmethyl imatinib were also increased, but without any significant difference. However, the single dose of 100 mg/kg or 50 mg/kg genistein has no effect on the pharmacokinetics of imatinib and N-desmethyl imatinib. Those results indicated that multiple dose of genistein (100 mg/kg or 50 mg/kg) induces the metabolism of imatinib, while single dose of genistein has no effect.

Nypa fruticans Wurmb. (nipa palm) sap, locally known as air nira or tuak, is a sweet natural beverage in Malaysia with antioxidant potency beneficial for human health. However, nypa sap lacks scientific attention with no toxicity study has been established. Therefore, this study was performed to evaluate the acute toxicity of nypa sap in the female Sprague Dawley rats. Twenty-five female rats were randomly divided into one control group and four treated groups. Treated groups were orally administered with doses of 5, 50, 300, and 2000 mg/kg of nypa sap, while the control group had received normal saline solution. The animals’ vital signs and mortality were recorded daily at an interval of 30 min and continued up to 14 days. Their blood samples and organs were harvested for toxicity analysis to assess liver and kidney function, lipid profile, and full blood count. The acute toxicity test via measurement of the biochemical and haematological parameters had shown that there was no significant difference between the treated and control groups. However, the blood glucose level in the treated groups (at higher doses of 300 and 2000 mg/kg, respectively) was significantly decreased. A similar trend was recorded for mean corpuscular volume (MCV) albeit in the treated group of 50 mg/kg doses. Histopathological examination of the organs showed no signs of abnormality in both treated and untreated groups. Overall, the results showed that consumption of nypa sap is potentially safe with no acute toxic effect on the laboratory rat models.

Background : Coffee contains cafestol and kahweol which can increase LDL cholesterol levels. Effect of cafestol and kahweol with chlorogenic acids are antagonist in coffee. Chlorogenic acids is strong antioxidant in coffee. Unfiltered coffee contains more chlorogenic acids and cafestol than filtered coffee. The study aims to determine the differences of LDL cholesterol levels of Sprague Dawley rats in giving filtered and unfiltered coffee. Methods : a laboratory experimental study, with randomized pre-post test control group design, with a simple random sampling to 28 Sprague Dawley rats were divided into 3 groups. The first treatment was given unfiltered coffee 4x2.7 ml, the second treatment was given filtered coffee 4x2.7 ml and the control was given water 4x2.7 ml for 28 days. LDL cholesterol levels were determined by the homogenous enzyme method. Data were analyzed by paired t-test and Oneway ANOVA test at 95% confident level. Results : Changes levels of LDL cholesterol in filtered coffee (0.85 mg/dl) are higher than unfiltered coffee (0.15 mg/dl). However statistical test shows there is no significant difference of LDL cholesterol in filtered and unfiltered coffee (p>0.05). Conclusion : There is no significant difference of LDL cholesterol levels of Sprague Dawley rats in filtered and unfiltered coffee.

Objective: To evaluate the protective effects of vitamin E α-tocopherol isomer against the toxicity of cisplatin on sperm motility and morphology in Sprague Dawley rats. Material & Methods: Twenty-four rats were grouped into four groups (n=6). The control group (CN) was injected with normal saline, second group (CP) was injected with cisplatin, the third group (P1) was injected with cisplatin and vitamin E 50 mg/kgBW for 7 weeks P.O, the fourth group (P2) was injected with cisplatin and vitamin E 200 mg/kgBW for 7 weeks P.O. Vitamin E was given from 3 weeks before cisplatin injection and 4 weeks following cisplatin injection. At 7th week, all the samples were undergoing bilateral orchidectomy. Vitamin E that being used in this study was α-tocopherol isomer. Results: Cisplatin decreased motility and morphology of spermatozoa significantly against controls. Vitamin E 50 mg/kgBW and 200 mg/kgBW significantly increased motility of spermatozoa (p<0.05) compared to those in the cisplatin group only. Vitamin E 50 mg/kgBW, and 200 mg/kgBW did not have a significant difference in spermatozoa motility compare to control groups. Vitamin E 50 mg/kgBW and 200 mg/kgBW could increase the spermatozoa morphology significantly compare to those cisplatin only group. Vitamin E 50 mg/kgBW, and 200 mg/kgBW did not have a significant difference in spermatozoa morphology compared to control groups. Conclusion: α-tocopherol 50 mg/kgBW and 200 mg/kgBW provided a same protective effect against spermatozoa damage especially in motility and morphology aspect due to cisplatin exposure. Therefore, in this study it was more recommended to use α-tocopherol in 50 mg/kgBW dose than 200 mg/kgBW.

Objective: The purpose of the present investigation was to analyze the effets of zidovudine on the postimplantation embryo and fetus.Methods: Pregnant Sprague-Dawley rats were given various doses (10 mg/kg, 30 mg/kg, 150 mg/kg) of zidovudine or saline by an endotracheal tube during the period of embryogenesis (days 6–8, 9–11, 6–11 postconception). The animals were sacrificed on days 18–19 of pregnancy, and their fetuses were removed by hysterotomy. Autopsies under low (15×) and high (40×) power light microscopy were performed on all fetuses.Results: There was no statistically significant difference among the groups with respect to maternal weight gain. There were more pregnancy resorptions in the group receiving high-dose zidovudine (150 mg/kg/day) throughout embryogenesis than in the control group (P = 0.001, respectively). Four major structural anomalies were noted among the 689 fetuses examined, but zidovudine was not associated with an increased frequency of congenital anomalies in rats when it was administered in doses similar to, 3-, and 15-fold higher than the regimen recommended for adult humans. The drug, however, was embryocidal in the high-dose group (P = 0.002).Conclusions: These findings are consistent with previous studies of preimplantation mouse embryos that demonstrated an embryocidal effect on preimplantation conceptuses. In summary, post-implantation embryonic zidovudine exposure was associated with significantly increased pregnancy losses (resorptions and intrauterine deaths).

Cedera sel yang luas pada luka bakar melepaskan banyak mediator inflamasi. Respon tubuh yang berlebihan pada keadaan tersebut akan memperberat kerusakan sel yang sudah terjadi. Nurtrisi (lemak) yang cukup seperti omega 3 dapat membantu proses penyembuhan luka serta mencegah infeksi pada luka supaya tidak menjadi luka kronis. Maka dari itu peneliti memilih judul penelitian tentang efektifitas kombinasi suplementasi omega 3 dengan restriksi kalori terhadap penyembuhan luka bakar pada tikus Sprague Dawley, ditinjau dari VEGF dan Re-epitelisasi. Untuk mengetahui tingkat efektifitas suplementasi Omega 3 dan retriksi kalori pada penyembuhan luka bakar. Peneliti dalam melakukan penelitian menggunakan metode penelitian eksperimental dengan menggunakan 1 kelas kontrol dan 3 kelas perlakuan tikus Sprague Dawley, yang diinduksi luka bakar kemudian diberi perlakuan, dan diamati secara berkala. Hasil penelitian didapatkan perbedaan yang signifikan antara kelompok kontrol dan kelompok perlakuan yang di uji dengan one way ANOVA mempunyai nilai p = 0,000 (p < 0,05), dan uji Mann-Whitney didapatkan perbedaan yang signifikan antara kelompok perlakuan 1, 2, 3, dan kelompok kontrol p = 0,025; 0,004; 0,004; 0,004; 0,004, nilai p < 0,05. Dan uji Games-Howell ditunjukkan pada kolom rerata ± SD jika memuat huruf yang  berbeda berarti ada perbedaan yang bermakna (p-value <0.05) K1 = 23,3587a ± 0,57556; P1 = 17,1015b ± 2,60108; P2 = 124,1503c ± 15,30168; P3 = 237,5053d ± 0,78263. Kesimpulan yang peneliti dapat bahwa kombinasi suplementasi omega 3 dengan retriksi kalori berpengaruh signifikan terhadap penyembuhan luka bakar pada tikus Sprague Dawley. To determine the effectiveness of Omega 3 supplementation and calorie restriction when healing burns. The researchers using experimental research methods with 1 control and 3 training classes of Sprague Dawley mice, which were induced by burns were then observed periodically. The results showed a significant difference between the control group and the treatment group tested by one way ANOVA having a value of p = 0,000 (p <0.05), and the Mann-Whitney test found a significant difference between the treatment groups 1, 2, 3, and the control group p = 0.025; 0.004; 0.004; 0.004; 0.004, p value <0.05. And the Games-Howell test is shown in the mean column ± SD if it contains different letters it means there is a significant difference (p-value <0.05) K1 = 23.3587a ± 0.57556; P1 = 17.1015b ± 2.60108; P2 = 124,1503c ± 15.30168; P3 = 237.5053 d ± 0.78263. The researchers concluded that the combination of omega-3 supplementation with calorie retention had a significant effect on burn healing in Sprague Dawley rats.
