Immunosuppressant and steroid therapy for SLE have not shown satisfactory results. Another method of therapy that is being developed is vaccines and escalating dose immunotherapy using self-antigen. The aim of this study was to assess the balance of immune cells through the ratio of pro-inflammatory and anti-inflammatory cells and cytokines in SLE using self-antigen dsDNA therapy. Methods: Female Balb/c mice 6-8 weeks old separated randomly to negative control group and pristane induced lupus (PIL) mice group. PIL mice groups were injected pristane intraperitoneally. Twelve weeks after the injection, the mice were evaluated for clinical and serological manifestations (anti-dsDNA levels). Mice with lupus signs were divided into four groups; positive control group: PIL mice without EDI dsDNA therapy, treatment A: PIL mice with EDI dsDNA therapy dose I (0.01μg/ml, 0.1μg/ml, 1μg/ml), treatment B: PIL mice with EDI dsDNA therapy dose II (0.1μg/ml, 1μg/ml, 10μg/ml), and treatment C: PIL mice with EDI dsDNA therapy dose III (1μg/ml, 10μg/ml, 100μg/ml). dsDNA were injected once a week and the dose was increased every week. Samples were analyzed for active/inactive dendritic cells ratio, Th1/Th2 cells ratio, Th17/Treg cells ratio and IL-17/TGF-β levels ratio. Results: Escalating dose antigen specific therapy with dsDNA injection of third dose reduced active/inactive dendritic cells ratio (p=0.000), Th1/Th2 cells ratio (p=0.010), Th17/Treg ratio (p=0.004) and decrease IL-17/TGF- β levels ratio (p=0.004) significantly compared to positive control. Conclusion: Escalating dose antigen specific therapy with dsDNA injection of dose III was able to regulate balance ratio of inflammatory cells and cytokines in PIL mice thus the immune tolerance may improve compared to control groups.

Diabetes mellitus now becomes a global serious theat with continues encreasingly prevalence. Ethanol extract of squash fruit Chayotte (Sechium edule) is a plant that contains flavonoidsareanti-inflammatory. The objective of this study was to determine the effect of ethanol extract of Squash fruit Chayotte (Sechium edule) to Interleukin 6 level and increasedthe size ofthe diameter of the pancreas in STZ-induced hyperglycemic mice model. Goals : This research is experimental research uses Post test randomized controlled group design upon male mice, and divided into 4 groups; negative control group, positive control group is treated with streptozotocin at a dose of 60 mg/kg weight, and groups of therapy (dosage 100 gram/kg weight, 200 gram/kg weight). Methods : The measurement of the level interleleukin 6 (IL-6) used ELISA method and the description of histopatoligy pancreas. The data analysis of level IL-6 level uses One Way ANOVA (α = 0,005) and pancreas histology image used descriptive analysis. Result : The results showed a decrease in the levels of IL-6, although not significant if it’s compared to the positive control. The preparat histopathology pancreas observation increased diameter which showed widening of cell β of pancreas. Conclution : The conclusion of this research was that ethanol extract of squash fruit can reduce levels IL-6 and prevent the hiperglikemia mice pancreas damages

Spirulinasp.is an alga that has antioxidant and anti-inflammatory potential.Phycocyanin contained in Spirulina platensis can reduce tumor necrosis factor-? levels, neutrophils, and M2 macrophages in inflammation. This research aims to investigate the anti-inflammatory activity of S. platensisin a mice wound model. Thirty-two male Wistar rats were incised and divided into 4 groups. The first therapeutic group (X1) received S. platensisextract at a dose of 500 mg/kgBW/day, and the second therapeutic group received S. platensisextract at a dose of 750 mg/kgBW/day. The negative control group (C1) received a saline solution and the positive control group (C2) received diclofenac sodium 20mg/kgBW/day. Serum TNF-? levels examined by the Enzyme-Linked Immunosorbent Assaymethod. Neutrophils and M2 macrophages were calculated by tissue biopsy and hematoxylin-eosin (HE) staining. Data analysis was performed with ANOVA test and LSD Post-Hoc Test. The average levels of serum TNF-? on the 14-day were 394.50; 180.33; 2,980.33; and 607.42 pg/ml for X1, X2, C1 and C2, respectively. The average neutrophils number on the 14-day were 8.00; 6.83; 14.67; and 11.17 for X1, X2, C1 and C2, respectively. The average macrophage number in the 14-day was 15.50; 19.17; 6.33; and 11.17 for X1, X2, C1 and C2, respectively. We found significant differences between TNF-? levels, the number of neutrophils, and the number of M2 macrophages. The administration of S. platensisextract at a dose of 750 mg/ kgBW/day reduces serum TNF-? levels, neutrophil count, and increases M2 macrophages in skin incisional mice model.

The focus of our study is to compare the anti-inflammatory effect of extracts prepared from Algerian mollusk called Helix ,on induced lung inflammation in rats: aqueous AE, HE and organic OE extracts. Animals were classified into six groups: control group T was instilled with NaCl (0.9%) in the trachea, negative control group was instilled with in trachea (5mg//Kg), positive control group was instilled with and treated with an anti-inflammatory drug, groups SAE, SHE and SOE were instilled with and treated with aqueous, and organic extracts respectively (4mg//Kg). 24h after instillation, we sacrificed animals, blood samples and fluid lavage (BALF) were collected. Lungs were for investigation. We measured activity (MPO) in BALF. Results showed induces a massive in lungs. HE and OE decreased leukocyte count in BALF and blood. AE increased leukocyte count in blood and decreased it in BALF. The three extracts decreased MPO activity in BALF, reduced number of inflammatory cells in the and protected lung from beads injuries. Both AE and HE reduced edema in lung. We conclude that treatment with Helix had an anti-inflammatory effect on inflammation.

Retrograde menstruation lead to I Kappa B Kinase (IKK) fosforilation in peritoneum macrophage and cause secretion of proinflammatory cytokine interleukin1β then stimulate endometriosis cell to produce Vascular Endothelial Growth Factor which lead to increasing of endometriosis lession seen as endometriosis implant area. Cytokine secretion was inhibited through prevention of NF-κB activation by dragon red fruit rind extract (Hylocereuspolyrhizus). The aim of this reserach is to know the effect of dragon red fuit rind extract with 0,25; 0,5; and 1 mg/g bodyweight dosage toward IL-1β, VEGF expression and implant area in endometriosis mice model. The design of this experiment was randomized post test only control group design.Endometrios mice model were made in 14 days and split into two group, positive control group and treatment group after two week negative control group and postive control group were given Na-CMC 0,5% solution consequetively, and treatment group were given dragon red fruit extract with different dosage. Signification number for IL-1β is p>0,05, signification number for VEGF is p>0,05, and implant area signification number is p>0,05. Administration of dragon red fruit rind extract can decrease IL-1β, VEGF, and implant area.

<p><strong><em>Background:</em></strong><em> Orthodontic tooth movement is a </em><em>interaction prosess</em><em> of resorption and deposition of bone remodeling. Orthodontic tooth movement by mechanical strength causes changes in alveolar bone. Osteocyte is an essential cell to respond bone remodelling. Hyperbaric Oxygen Therapy affects production of osteocyte because it can release Reactive Oxygen Species (ROS) and Nitrid Oxide (NO).  <strong>Purpose: </strong>To determine the difference number  of osteocyte in pressure and tension area during tooth movement by adjuvant of Hyperbaric Oxygen 2,4 ATA during 7 days starting on day 8 to day 14. <strong>Materials and Methods</strong>: This research used Completery Randomized Control Group Post Test Only Design. 36 cavia cobaya (male)  were divided into 3 groups randomly : the negative control groups, positive control group, and treatment group. Preparat staining used Hematoxylin Eosin (HE) and calculated on microscop 1000x with 20 field of view. Data analyses used one way ANOVA and LSD test then compared each area by using paired T test. <strong>Result:</strong> The data showed that the treatment group (P=10,67) tension area has the highest number of osteocyte than  negative control group (K-=3,67), positive control (K+=7,42). <p>Penelitian ini bertujuan untuk melihat pengaruh pemberian ekstrak etanol kulit petai (Parkia speciosa Hassk) terhadap penurunan kadar glukosa darah mencit jantan yang diinduksi aloksan. Hewan percobaan dibagi atas 5 kelompok diantaranya kelompok kontrol negatif, kelompok kontrol positif,dosis I (280 mg/kgBB mencit), dosis II (560 mg/kg BB mencit), dosis III (840 mg/kg BB mencit). Penelitian dilakukan selama 21 hari. Persentase penurunan kadar glukosa darah mencit jantan setelah diberikan ekstrak etanol kulit petai pada hari ke-21 adalah dosis I (77,52 %) lebih besar dibandingkan dengan dosis II (69,5 %) dan dosis III (73,37 %). Data yang diperoleh dianalisis dengan uji Two Way Anova dengan program SPSS 17. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak etanol kulit petai untuk tiga variasi dosis menyatakan perbedaan yang bermakna secara statistik terhadap penurunan kadar glukosa darah mencit jantan.</p><p><em>Petai (Parkia speciosa Hassk) has a compound β-sitosterol and stigmasterol that have efficacy to decreased blood glucose levels. This study aimed to determine the effect of ethanol extract of petai peel for decrease blood glucose levels of male mice induced by alloxan. Experimental animals were divided into 5 groups including negative control group, positive control group, the first dose (280 mg/kg in mice), the second dose (560 mg/kg in mice), the third dose (840 mg/kg in mice). The study was conducted for 21 days. After 21 days, the result found that the percentage of blood glucose levels after the male mice given the ethanol extract of petai peel was, the first dose (77.52%) biger than the second dose (69.5%) and the third dose (73.37%). The data obtained were analyzed by Two Way ANOVA using SPSS 17. The results showed that have signicantly difference between three dose variation of ethanol extract of petai peel in blood glucose levels.</em></p>

The aim of this study was to compare the vertical fracture resistance of roots obturated with different root canal filling materials and sealers. Crowns of 55 extracted mandibular premolar teeth were removed to provide root lengths of 13 mm. Five roots were saved as negative control group (canals unprepared and unfilled). Fifty root canals were instrumented and then five roots were saved as positive control group (canals prepared but unfilled). The remaining 45 roots were randomly divided into three experimental groups (n=15root/group) and obturated with the following procedures: in group 1, glass ionomer-based sealer and cone (ActiV GP obturation system); in group 2, bioceramic sealer and cone (EndoSequence BC obturation system); and in group 3, roots were filled with bioceramic sealer and cone (Smartpaste bio obturation system). All specimens were tested in a universal testing machine for measuring fracture resistance. For each root, the force at the time of fracture was recorded in Newtons. The statistical analysis was performed by using Kruskal-Wallis and post hoc test. There were no significant differences between the three experimental groups. The fracture values of three experimental and negative control groups were significantly higher than the positive control group. Within the limitations of this study, all materials increased the fracture resistance of instrumented roots.

Methanol poisoning is a hazardous intoxication characterized by visual impairment and formic acidemia. The therapy for methanol poisoning is alcohol dehydrogenase (ADH) inhibitors to prevent formate accumulation. Ranitidine has been considered to be an inhibitor of both gastric alcohol and hepatic aldehyde dehydrogenase enzymes. This study aimed at testing ranitidine as an antidote for methanol acute toxicity and comparing it with ethanol and 4-methyl pyrazole (4-MP). This study was conducted on 48 Sprague-Dawley rats, divided into 6 groups, with 8 rats in each group (one negative control group [C1], two positive control groups [C2, C3] and three test groups [1, 2 and 3]). C2, C3 and all test groups were exposed to nitrous oxide by inhalation, then, C3 group was given methanol (3 g/kg orally). The three test groups 1, 2 and 3 were given ethanol (0.5 g/kg orally), 4-MP (15 mg/kg intraperitoneally) and ranitidine (30 mg/kg intraperitoneally), respectively, 4 hours after giving methanol. Rats were sacrificed and heparinized, cardiac blood samples were collected for blood pH and bicarbonate. Non-heparinized blood samples were collected for formate levels by high performance liquid chromatography. Eye balls were enucleated for histological examination of the retina. Ranitidine corrected metabolic acidosis (p = .025), decreased formate levels (p = .014) and improved the histological findings in the retina induced by acute methanol toxicity.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole. Objective: The aim of this work was to study the biochemical characteristics of coconut milk and its antigenic effect on the Balb/c mice immunized with α-lactalbumin protein, as well as its consequences on the structure of the intestinal epithelium.Methods: To achieve the objective of the study, an electrophoresis was realised on a polyacrylamide gel to determine various proteins contained in coconut milk. In addition, Lowry’s method was used to determine the amount of proteins in the formula. The antigenicity of coconut milk in sera was also studied using the Enzyme-Linked Immunosorbent Assay (ELISA) method. For the histological study, 21 w-old mice Balb/c were used and distributed in three groups of 7 mice each. Group 1, received a standard feed with no treatment (Negative control), group 2 and 3 received respectively a standard feed (Positive control) and coconut milk for a period of 28 d after being immunized with α- lactalbumin.Results: Analysis of the data revealed that the rate of proteins of cow’s milk is higher than that of the coconut milk ( p0.01). However, after carrying out the electrophoresis analysis, the coconut milk showed the absence of intact proteins. The anti α-Lactalbumin IgG titers significantly increased in positive control groups that received coconut milk (p<0.0001). Moreover, there was an increase of the intestinal villi height of mice fed with coconut milk, in the structure level of their intestinal epithelium compared to the negative control group.Conclusion: The findings of the study provide the evidence that coconut milk is a possible alternative to the cow’s milk formula in case of allergy.
