Objective: Comparative analysis of morphology and phytochemical constituents in different populations and morphotypes of Datura innoxia Mill. and Datura metel L. from Punjab plains.Methods: Morphological analyses of different populations have been done. Methanol extracts of seeds and roots of different populations have been prepared and total phenols, flavonoid contents were measured through spectrophotometry. Antioxidant activity was studied by 2, 2-diphenyl-1- picrylhydrazyl radical scavenging activity and total antioxidant capacity. Two major compounds, caffeic acid and chlorogenic acid were quantified by high-performance thin-layer chromatography (HPTLC) analyses.Results: Two morphotypes of D. metel were reported in the present study. Out of five different populations of D. metel and D. innoxia, the wild populations have more bioactive compound as compared to the cultivated ones from the detailed phytochemical investigation. Pharmacologically important two marker compounds chlorogenic acid and caffeic acid has been identified and quantified by HPTLC technique.Conclusion: Variation in terms of morphology and secondary metabolites exists among the different populations of Datura spp. Among the two plant parts studied, seeds have the maximum amount of bioactive metabolites and antioxidant activity. This study revealed that chlorogenic acid and caffeic acid are the potential polyphenolic compounds in Datura spp. It has been found that the antioxidant activity of plant is due to its polyphenol contents, which provides insight to various researchers to work on it as it imparts health benefit.

Aim of the study was to analyze the phytochemical constituents and estimation of total flavonoids and phenolic compounds of leaves and roots of the medicinal plant Lagascea mollis. Methanol extract of dried leaves and roots of L. mollis was prepared by using soxhlet apparatus. The extract prepared was tested forpreliminary qualitative phytochemical screening, followed by the quantitative estimation of total flavonoids and phenols by spectroscopy. The selected plant parts were found to contain alkaloids, flavonoids and terpenoids in both parts and saponin is present only in leaf. In quantitative estimation the reports revealedthe presence 0.017 and 0.013 mgRE/g extract and 458.91 and 704.50 mgGAE/g extract total flavonoids and phenolic compounds in leaf and root extract respectively. These major phytoconstituents present in this species may be accounted as factors for the medicinal importance of L. mollis.

Antibiotic resistance bacteria has become an increasing problem now today due to many factors. This study investigates the efficacy of Prismatomeris tetrandra K. Schum root extract as a new source of antibacterial activity for antibiotic resistant bacteria using agar well diffusion method. The results showed that S. aureus TISTR517 exhibited more sensitivity to P. tetrandra K. Schum root extract than other Gram-positive bacteria indicator strains. On the other hand, Gram-negative bacteria exhibited resistance to P. tetrandra K. Schum root extract. The study further showed the activity between P. tetrandra K. Schum root extract and gentamycin (10 µg), it revealed that MRSA142 was resistant to gentamycin (10µg) but sensitive to P. tetrandra K. Schum root extract. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) was evaluated by using S. aureus TISTR517 and MRSA142 as indicator strains. The MIC value was 0.59 mg/mL and 1.17 mg/mL for S. aureus TISTR517 and MRSA142, respectively. MBC assay demonstrated that the MBC value was 9.75 mg/mL and 150 mg/mL for S. aureus TISTR517 and MRSA142 respectively. The mode of action was investigated with the presence of P. tetrandra K. Schum root extract in the culture broth. The action of P. tetrandra K. Schum root extract was revealed of bacteriostatic activity due to the Optical density (OD) at 600 nm and Colony-Forming Units (CFU) of indicator strains were continuously decreased.

The ethanol extracts of root, bark and leaf of Bridelia ferruginea was investigated for antibacterial activity against clinical isolate of Staphylococcus aureus and Escherichia coli. The extracts had significant antibacterial activity in vitro at concentration of 25 mg/ml, 50 mg/ml, 100 mg/ml and 200 mg/ml and in vivo at dose of 50 mg/kg and 100 mg/kg. The root extract in vitro had the highest zone of inhibition, followed by the bark extract for both Staphylococcus aureus and Escherichia coli. The concentration of 200 mg/ml had the highest zone of inhibition in vitro. The minimum inhibitory concentration (MIC) showed a decreasing inhibitory effect of the plant extracts for both Staphylococcus aureus and Escherichia coli as the concentration decreases with root having 3.125 mg/ml, bark having 6.25 mg/ml and leaf having 25 mg/ml for Staphylococcus aureus and Escherichia coli. Likewise, the minimum bactericidal concentration (MBC) showed decreasing bactericide effects with decrease concentration with root having 12.5 mg/ml, bark having 12.5 mg/ml and leaf having 25 mg/ml for Escherichia coli while root had 6.25mg/ml, bark had 12.5mg/ml and leaf had 25mg/ml for Staphylococcus aureus. The in vivo investigation showed that the root and bark extract exhibited antibacterial activity on both Staphylococcus aureus and Escherichia coli at doses of 100mg/kg and 50mg/kg; the root extract had higher activity than the bark and root/bark combined. The dose of 100 mg/kg had the highest colonies reduction for Staphylococcus aureus and Escherichia coli in vivo. Preliminary phytochemical screening of root, bark and leaves of Bridelia ferruginea revealed the presence of tannins, flavonoids, carbohydrates, cardiac glycoside (root, bark and leaves), saponins (root and bark). The presence of tannins, saponins, flavonoid, cardiac glycoside and carbohydrate in the bark and root extracts of the plant indicates that the bark and root extracts were pharmacological importance

ABSTRACT: In the present study, antibacterial and antioxidant [1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity] activities of a weed, slender amaranth (Amaranthus viridis L.) were investigated. Extracts of different plant parts were prepared in n-hexane, chloroform and ethyl acetate. Antibacterial activity was measured by using 100 mg mL-1 concentration extracts against 4 deadly phytopathogenic bacterial species viz. Pseudomonas syringae Van Hall, Ralstonia solanacearum Smith, Erwinia carotovora (Jones), Holland and Xanthomonas axonopodis Hasse. In antioxidants assays, 10, 20 and 30 mg mL-1 extracts were used keeping DPPH as control. In these bioassays, ethyl acetate fraction of A. viridis leaf exhibited the best antibacterial and antioxidants activity. Ethyl acetate leaf fraction showed the highest inhibition zone diameter (IZD) where it caused 21 mm IZD against P. syringae and 19 mm IZD against E. carotovora. This extract also showed 22, 52 and 84% antioxidant activity at 10, 20 and 30 mg mL-1 concentrations, respectively. Previously there is no report available that describes antibacterial activity of root extract of A. viridis against P. syringae. Moreover, antioxidant activity of stem and root extracts in n-hexane, chloroform and ethyl acetate was investigated first time in the world. It was concluded that the biological activities observed during the present investigation may be due to the presence of bioactive constituents that can be harnessed as natural antibacterials and antioxidants.
