The ethanol extracts of root, bark and leaf of Bridelia ferruginea was investigated for antibacterial activity against clinical isolate of Staphylococcus aureus and Escherichia coli. The extracts had significant antibacterial activity in vitro at concentration of 25 mg/ml, 50 mg/ml, 100 mg/ml and 200 mg/ml and in vivo at dose of 50 mg/kg and 100 mg/kg. The root extract in vitro had the highest zone of inhibition, followed by the bark extract for both Staphylococcus aureus and Escherichia coli. The concentration of 200 mg/ml had the highest zone of inhibition in vitro. The minimum inhibitory concentration (MIC) showed a decreasing inhibitory effect of the plant extracts for both Staphylococcus aureus and Escherichia coli as the concentration decreases with root having 3.125 mg/ml, bark having 6.25 mg/ml and leaf having 25 mg/ml for Staphylococcus aureus and Escherichia coli. Likewise, the minimum bactericidal concentration (MBC) showed decreasing bactericide effects with decrease concentration with root having 12.5 mg/ml, bark having 12.5 mg/ml and leaf having 25 mg/ml for Escherichia coli while root had 6.25mg/ml, bark had 12.5mg/ml and leaf had 25mg/ml for Staphylococcus aureus. The in vivo investigation showed that the root and bark extract exhibited antibacterial activity on both Staphylococcus aureus and Escherichia coli at doses of 100mg/kg and 50mg/kg; the root extract had higher activity than the bark and root/bark combined. The dose of 100 mg/kg had the highest colonies reduction for Staphylococcus aureus and Escherichia coli in vivo. Preliminary phytochemical screening of root, bark and leaves of Bridelia ferruginea revealed the presence of tannins, flavonoids, carbohydrates, cardiac glycoside (root, bark and leaves), saponins (root and bark). The presence of tannins, saponins, flavonoid, cardiac glycoside and carbohydrate in the bark and root extracts of the plant indicates that the bark and root extracts were pharmacological importance

Abstract: Indonesia has a high diversity of flora and fauna in the world. Many types of plants can be used as a medicinal plant; one of them is yellow root (Arcengelisia flava Merr.) which has been known empirically by the Dayak community in Central Kalimantan as natural herbs to treat jaundice. This study aimed to determine the antibacterial effect of the yellow root bark extract against Staphylococcus aureus and Escherichia coli. This study was conducted in the Laboratory of Pharmacology and the Laboratory of Microbiology Faculty of Medicine University of Sam Ratulangi. This was an experimental laboratory study using the Kirby-Bauer modified technique. Yellow root bark extract was obtained from maceration process using ethanol 96%. The antibacterial effect was tested by using the well diffusion method. The results showed that distilled water, the negative control, did not generate inhibition zone around the wells. Ciprofloxacin, the positive control, had the greatest diameter of inhibitory zone. The average diameter of inhibition zone generated by ciprofloxacin was 39.23 mm against Staphylococcus aureus and 40.95 mm against Escherichia coli. Meanwhile, yellow root extract generated an average diameter inhibition zone of 12.27 mm against Staphylococcus aureus and 14.44 mm against Escherichia coli. Conclusion: Yellow root bark extract potentially had antibacterial effect against Staphylococcus aureus and Escherichia coli. Banyak jenis tanaman yang dapat dijadikan sebagai tanaman obat, salah satunya akar kuning (Arcangelisia flava Merr.) yang telah lama dikenal secara empiris oleh masyarakat dayak di Kalimantan Tengah sebagai tanaman herbal alami untuk mengobati penyakit kuning. Penelitian ini bertujuan untuk mengetahui efek antibakteri dari ekstrak batang akar kuning terhadap bakteri Staphylococcus aureus dan Escherichia coli dan dilakukan di Laboratorium Farmakologi, dan Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Sam Ratulangi. Jenis penelitian ini eksperimental laboratorium dengan metode modifikasi Kirby-Bauer. Uji efek antibakteri menggunakan metode sumuran. Esktrak batang akar kuning diperoleh dari proses maserasi dengan etanol 96%. Hasil penelitian memperlihatkan akuades sebagai kontrol negatif tidak menimbulkan zona hambat di sekitar sumur. Siprofloksasin sebagai kontrol positif memiliki diameter zona hambat yang paling besar. Rerata diameter zona hambat yang dihasilkan oleh siprofloksasin 39,23 mm terhadap Staphylococcus aureus dan 40,95 mm terhadap Escherichia coli. Ekstrak akar kuning menghasilkan rerata diameter zona hambat sebesar 12,27 mm terhadap Staphylococcus aureus dan 14,44 mm terhadap Escherichia coli. Simpulan: Ekstrak batang akar kuning berpotensi memiliki efek antibakteri terhadap bakteri Staphylococcus aureus dan Escherichia coli. Zanthoxylum paracanthum Kokwaro (Rutaceae) is an endemic Kenyan and Tanzanian plant used in folk medicine by local populations. Although other Zanthoxylum species have been studied, only Z. paracantum stem extracts have been profiled, even though the roots are also used as herbal remedies. As root extracts may be another source of pharmaceutical compounds, the CH2Cl2/MeOH (1:1) root bark extract was studied in this report. Eight root bark compounds were isolated and their structural identities were confirmed by mass spectrometry (MS) and nuclear magnetic resonance (NMR) (using COSY, HSQC, NOESY and HMBC) analyses. The structural identities were determined as follows: the fatty acid—myristic acid (1); the sterol—stigmasterol (2); the lignan—sesamin (3); two β-carboline alkaloids—10-methoxycanthin-6-one (6) and canthin-6-one (7); and three phenanthridine alkaloids—8-acetonyldihydrochelerythrine (4), arnottianamide (5) and 8-oxochelerythrine (8). Some of these compounds were identified in the species for the first time. These compounds and the extract were then tested in vitro against methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli (ATCC 25922), Staphylococcus aureus (ATCC 29213) and Candida albicans (ATCC 10231) before tests for antiproliferative activity against the human breast cancer (HCC 1395), human prostate cancer (DU 145) and normal (Vero E6) cell lines were conducted. Minimum inhibition concentration values of 3.91, 1.95, 0.98 and 7.81 µg/mL against MRSA, S. aureus, E. coli and C. albicans, respectively, were recorded. Among the isolates, canthin-6-one was the most active, followed by 10-methoxycanthin-6-one. The root extract and some of the compounds also had antiproliferative activity against the HCC 1395 cell line. Stigmasterol and canthin-6-one had IC50 values of 7.2 and 0.42. The root bark extract also showed activity, at 8.12 µg/mL, against the HCC 1395 cells. Out of the chemical isolates, 10-methoxycanthin-6-one and canthin-6-one showed the strongest inhibition of the DU 145 cells. The root extract had significant antimicrobial and antiproliferative activities, supporting the traditional use of this plant in treating microbial infections and cancer-related ailments.

A new series ofN-{4-methyl-5-[4-(4-oxo-2-phenyl(1,3-thiazolidin-3-yl)]-5-sulfanyl(1,2,4-triazol-3-yl)-1,3-thiazol-2-yl }acetamide (7a-l) was synthesized in order to determine their antimicrobial activity and feasible structure–activity relationships. The compounds were synthesized in good yield and the structures of all newly synthesized compounds were established on the basis of their IR,1HNMR, and elemental analysis. The synthesized compounds were testedin vitroantibacterial activity againstStaphylococcus aureus,Escherichia coli,Pseudomonas aeruginosaandSalmonella typhiand antifungal activity againstAspergillus niger,Candida albicansby measuring the zone of inhibition in mm.

Abstract : Strychnos ligustrina stem has been empirically used by the people of West Nusa Tenggara and Bali in the treatment of malaria, tooth ache and diarrhea, but there is no scientific data that supports it. This study aims to determine and prove the antibacterial activity of Strychnos ligustrina methanol extract to pathogenic bacteria in vitro and in vivo. This research is an experimental study conducted by measuring the inhibition zone (mm) growth of pathogenic bacteria, determining minimum inhibitory concentration (MIC) and minimum killing concentration (MKC) in vitro, and determining the percentage of antibacterial activity of methanol extract of S. ligustrina stem in vivo. The experiment was conducted using 4 groups of concentrations of S. ligustrina stem methanol extract in an in vitro study of 25, 50, 75, and 100% with ciprofloxacin as a positive control and aquadest as a negative control. In vivo studies experiments were carried out using 6 treatment groups of test animals male mice Balb / c (Mus musculus). The in vitro test results showed that methanol extract of S. ligustrina stems was able to inhibit the growth of pathogenic bacteria with medium categories of clinical isolates of Staphylococcus aureus and categories of weaks to Klebsiella pneumonia and Escherichia coli isolates. The minimum inhibitory concentration (MIC) for S. aureus and K. pneumonia bacteria isolates was at a concentration of 25% while for E. coli isolates at a concentration of 30%. The methanol extract of the S. ligustrina stem has no killing power against the pathogenic bacteria tested. Antibacterial activity in vivo was able to inhibit the growth of S. aureus pathogenic bacteria by 6.60% (at 25% concentration), 8.62% (at 50% concentration), and 17.31% (at 100% concentration), against K. pneumonia was 11.85% (at 25% concentration), 51.21% (at 50% concentration), and 65.92% (at 100% concentration), against E. coliat 19.18% (at concentration 25%), 29.98% (at 50% concentration), and 40.88% (at 100% concentration). Methanol extract of S. ligustrina stem proved to have antibacterial activity in vitro and in vivo. Key words: Srychnos ligustrina, pathogenic bacteria, antibacterial, in vitro, in Vivo. Abstrak : Strychnos ligustrina secara empiris  telah digunakan oleh masyarakat Nusa Tenggara Barat dan Bali dalam pengobatan penyakit malaria, sakit gigi, dan diare, tetapi belum ada data ilmiah yang mendukung. Penelitian ini bertujuan untuk menentukan dan membuktikan aktivitas antibakteri ekstrak metanol batang bidara laut terhadap bakteri patogen secara in vitrodan in vivo. Penelitian ini merupakan penelitian eksperimental yang dilakukan dengan mengukur zona hambat (mm) pertumbuhan bakteri patogen, menentukan konsentrasi hambat minimum (KHM) dan konsentrasi bunuh minimum (KBM) secara in vitro, serta menentukan persentase aktivitas antibakteri ekstrak metanol batang bidara laut secara in vivo. Percobaan dilakukan menggunakan 4 kelompok konsentrasi ekstrak metanol batang bidara laut pada penelitian in vitro yaitu 25, 50, 75, dan 100% dengan ciprofloxacin sebagai kontrol positif serta aquadest sebagai kontrol negatif. Pada penelitian in vivo percobaan dilakukan menggunakan 6 kelompok perlakuan hewan uji mencit jantan galur Balb/c (Mus musculus). Hasil uji in vitro menunjukkan ekstrak metanol batang bidara laut mampu menghambat pertumbuhan bakteri patogen dengan kategori sedang terhadap Staphylococcus aureus isolat klinis dan kategori lemah terhadap Klebsiella pneumonia dan Escherichia coli isolat klinis. Nilai konsentrasi hambat minimum (KHM) untuk isolat bakteri S. aureus dan K. pneumoniae adalah pada konsentrasi 25% sedangkan untuk isolat E. coli pada konsentrasi 30%. Ekstrak metanol batang bidara laut tidak memiliki daya bunuh terhadap bakteri patogen yang diuji. Aktivitas antibakteri secara in vivo mampu menghambat pertumbuhan bakteri patogen S. aureus sebesar 6,60% (pada konsentrasi 25%), 8,62% (pada konsentrasi 50%), dan 17,31% (pada konsentrasi 100%), terhadap K. pneumonia sebesar 11,85% (pada konsentrasi 25%), 51,21% (pada konsentrasi 50%), dan 65,92% (pada konsentrasi 100%),   terhadap E. coli sebesar 19,18% (pada konsentrasi 25%), 29,98% (pada konsentrasi 50%), dan 40,88% (pada konsentrasi 100%). Objectif : L’objectif de ce travail était d’évaluer in vitro l’activité antimicrobienne des extraits de feuilles et tige de Jatropha multifida sur la croissance de Candida albicans, Escherichia coli et Staphylococcus aureus, puis d’évaluer in vivo la toxicité de cette plante. Méthodologie et résultats : Les méthodes de diffusion en milieu gélosé et de microdilution en milieu liquide ont été utilisées pour évaluer l’effet antimicrobien. Une étude en subaigüe était réalisée afin d’explorer les effets toxiques de l’extrait aqueux des feuilles. Les résultats des tests antimicrobiens montrent une activité des extraits de feuilles et tige de J. multifida sur la croissance des souches utilisées avec des diamètres de zones d’inhibition allant de 8 à 25 mm et des concentrations minimales inhibitrices (CMI) variant de 0,039 mg/mL à 1,25 mg/mL à l’exception des souches de E. coli qui sont résistantes aux extraits de la tige. L’administration en subaigüe de l’extrait aqueux des feuilles de J. multifida à la dose de 600 mg/kg entraîne une perte significative de poids chez les souris. Conclusion et applications des résultats : Les extraits aqueux, éthanolique et hydroéthanolique des feuilles et tige de J. multifida possèdent d’activité antimicrobienne et pourraient être utilisés dans le traitement des Candidoses à C. albicans et des infections à S. aureus. Mais l’essai de toxicité subaigüe montre que l’extrait aqueux de la plante serait toxique. Des études toxicologiques approfondies restent donc nécessaires sur ces extraits afin de mieux élucider leur inocuité. Methodology and Results: The agar well-diffusion and the NCCLS broth microdilution methods were used to assess the antimicrobial effect. A subacute study was carried out to explore the toxic effects of the aqueous extract of the leaves. The results of the antimicrobial tests show an activity of the extracts of leaves and stems of J. multifida on the growth of the strains used with diameters of inhibitory zones ranging from 8 to 25 mm and minimum inhibitory concentrations (MIC) varying from 0.039 mg/mL to 1.25 mg/mL exception E. coli strains which are resistant to extracts from the stem. Subacute administration of the aqueous extract of the leaves of J. multifida at a dose of 600 mg/kg leads to a significant loss of weight in the mice. Conclusion and application of findings : The aqueous, ethanolic and hydroethanolic extracts of the leaves and stem of J. multifida have antimicrobial activity and could be used in the treatment of Candidiasis and bacterial infections due respectively to C. albicans and S. aureus. But the subacute toxicity test shows that the aqueous extract of the plant would be toxic. Extensive toxicological studies therefore remain necessary on these extracts in order to better elucidate their safety. Alafia barteri (Apocynaceae) is a climbing shrub having white or pink flowers. Traditionally, it has been used to treat diseases like malaria, sickle cell anemia, and eye infections. This research is focused on investigating the antioxidant and anti-arthritic activities of the aqueous and ethanol leaf and root extract of Alafia barteri plant in vitro. In-vitro antioxidant methods used were 2, 2 -diphenyl-1-picrylhydrazyl assay, reducing power activity and hydrogen peroxide scavenging assay while the anti-arthritic activity was studied using the assay method of protein denaturation. Results revealed that aqueous and ethanol root extracts scavenge free radicals, thus inhibiting damage caused by oxidative stress in arthritis while the ethanol extracts of both the leaf and roots had good anti-arthritic activities as seen in its ability to decrease protein denaturation.

Este trabalho reporta o potencial antibacteriano in vitro e toxicológico in vivo das folhas do jambo (Syzygium malaccense) frente a zebrafish (Danio rerio) adulto (ZFa). As folhas de jambo foram submetidas a desidratação (35 ± 2°C) por 24 horas, trituração e posterior extração de metabólitos por decocção, infusão e maceração com água destilada. Os extratos obtidos foram liofilizados e submetidos a análise de atividade antibacteriana in vitro frente a Gram-negativas (Escherichia coli ATCC 25922, Salmonella Enteritidis IAL 1132) e Gram-positivas (Listeria monocytogenes ATCC 19115 e Staphylococcus aureus ATCC 27664), bem como ao potencial toxicológico in vivo frente ao ZFa. O extrato obtido por infusão se mostrou mais promissor, pois apresentou concentração mínima bactericida (CMB) e concentração mínima inibitória (CMI) com maior potencial frente às gram- positivas (CMB - 6,25 e CMI - 6,25 mg/ml), bem como às gram-negativas (CMB - 25,0 e 3,125 e CMI - 3,125 mg/ml). Todos os extratos testados não se mostraram tóxicos frente ao zebrafish adulto e não alteraram o sistema locomotor dos mesmos. Desta forma, conclui-se que o extrato aquoso das folhas do jambo vermelho (Syzygium malaccense) obtido por infusão é seguro e pode ser utilizado como conservante natural com maior ação antibacteriana. Este trabalho nos conduz a novos estudos de isolamento e caracterização de princípios bioativos.

Background: Sclerocarya birrea is widely used in Nigerien communities for medicinal purposes to treat several gastrointestinal diseases including diarrhea. To lend credibility to its traditional use against diarrhea, laboratory studies were conducted. Objective: The aim of this study is to investigate the antibacterial activity and the phytochemical constituents of the crude extracts of root, bark and leaf of S. birrea. Materials and Methods: The collected different plant parts were air dried, powdered and separately extracted with ethanol and methanol. The alkaloid, flavonoid, saponin and tannin contents in all the plant parts were estimated using standard methods. The total and serially diluted fractions of the extracts were tested for antibacterial activity against selected enteropathogens by agar well diffusion and deep-well microdilution method. Results: Phytochemical screening revealed the presence of flavonoid, saponin and tannin in all the plant extracts. The extracts from the different parts showed varied antibacterial activity against the test bacteria. The bark extracts showed superior activity against Escherichia coli (zone of inhibition = 17 mm) and Salmonella typhi (zone of inhibition = 20 mm) at 200 mg/mL. Conclusion: The presence of important phytochemical groups and the antibacterial potential of alcoholic extracts of S. birrea could permit to justify its traditional usage against diarrhea.
