<p class="MDPI17abstract"><strong>Objective: </strong>The study aimed investigated effect of microbial isolate levels and fermentation time on fermentability regarding ammonia (NH<sub>3</sub>) production, volatile fatty acids (VFA) and microbial protein production, dry matter digestibility and organic matter digestibility <em>in vitro</em>.</p><p class="MDPI17abstract"><strong>Methods: </strong>The experiment was conducted at the Laboratory of Nutrition and Feed Science, Faculty of Animal and Agricultural Sciences, Universitas Diponegoro. <em>In vitro</em> experiment was performed using a completely randomized design with a factorial pattern with 2 factors and 4 replications. The treatments were microbial isolate levels (1 and 3%) and fermentation time (14 and 28 days). The parameters observed included production of NH<sub>3</sub>, VFA, microbial protein and total protein as well as the digestibility of dry matter and organic matter. The data were analyzed based on analysis of variance and if there was a significant effect the data were further analyzed with Duncan’s Multiple Range Test.<strong></strong></p><p class="MDPI17abstract"><strong>Results: </strong>The amount of microbial isolate and fermentation time affected rumen ammonia production. On rumen microbial protein content, the amounts of microbial isolate and fermentation time had substantial impact (P&lt;0.05). The isolate level and fermentation time, however, had no interaction effect on VFA production, dry matter digestibility, or organic matter digestibility. The fermentation time influenced (P&lt;0.05) the production of VFA and the digestibility of dry matter, but the isolate level and fermentation time had no effect on total protein production or organic matter digestibility.<strong></strong></p><p><strong>Conclusions: </strong>Processing of palm fronds through fermentation using buffalo rumen cellulolytic microbial isolates increased nutrient values of palm fronds.</p>

ABSTRACT. Penelitian bertujuan mengetahui kualitas complete feed dengan level pelepah sawit fermentasi berdasarkan kecernaan bahan kering, bahan organik, produksi N-NH3, produksi volatile fatty acids (VFA) dan produksi biomassa protein mikrobia serta protein total secara in vitro. Materi yang digunakan adalah complete feed tersusun atas konsentrat dan pelepah sawit fermentasi dengan berbagai level yaitu 0, 10, 20 dan 30%. Rancangan percobaan yang digunakan adalah rancangan acak lengkap (RAL) dengan 4 perlakuan complete feed dengan level pelepah sawit fermentasi yang berbeda. Data diolah menggunakan sidik ragam yang dilanjutkan dengan uji beda wilayah berganda Duncan. Hasil penelitian menunjukkan bahwa complete feed dengan level pelepah sawit fermentasi yang berbeda berpengaruh nyata (p0,05) terhadap kecernaan bahan kering dan bahan organik, produksi N-NH3, produksi VFA, dan produksi protein total, sedangkan pada biomassa protein mikrobia tidak terdapat perbedaan yang nyata (p0,05). Rata-rata nilai kecernaan bahan kering pada perlakuan T0, T1, T2 dan T3 adalah 69,59; 71,9; 69,05; dan 62,58%. Rata-rata nilai kecernaan bahan organik pada perlakuan T0, T1, T2 dan T3 adalah 63,59; 63,15; 65,50; 52,66 %. Rata-rata produksi VFA pada perlakuan T0, T1, T2 dan T3 sebesar 105,8; 142,7; 136,4; dan 135,7 mM. Rata-rata produksi NH3, biomassa protein mikrobia dan produksi protein total pada perlakuan T0, T1, T2 dan T3 berturut-turut adalah 6,48mM, 15,04mg/ml;, 34,10mg/g; 7,36mM, 15,75mg/ml, 23,72mg/g; 8,18mM, 12,59mg/ml, 33,72mg/g); dan 6,60mM, 15,31mg/ml, 40,80mg/g. Berdasarkan hasil penelitian disimpulkan bahwa penggunaan pelepah sawit fermentasi dengan level 20% dalam complete feed menghasilkan produksi VFA, kecernaan bahan kering dan bahan organik yang cukup baik sehingga dapat menjadi pakan alternatif sumber serat pengganti rumput. (Digestibility, fermentability and in-vitro production of microbial protein on complete feed based on fermented palm frond) ABSTRAK. This study aimed to determine the quality of a complete feed containing fermented palm fronds based on the digestibility of dry matter, organic matter, N-NH3, VFA, microbial protein biomass, and total protein in vitro. The material used was complete feed composed of concentrates and fermented palm fronds at various levels, i.e., 0, 10, 20, and 30%. The experiment was conducted as a completely randomized design (CRD) with four complete feed treatments containing different levels of fermented palm fronds. The data were processed using analysis of variance, followed by Duncan’s multiple range test. The results demonstrated that the complete feed with different levels of fermented palm fronds had a significant effect (p0.05) on the digestibility of dry matter and organic matter, N-NH3 production, essential fatty acids production, and total protein production, whereas there was no significant difference (p 0.05) on microbial protein biomass. The average dry matter and organic matter digestibility values of T0, T1, T2, and T3 treatments were 69.59; 63.59, 71.9; 63.15, 69.05; 65.50, and 62.58%; 52.66% respectively. The average production of volatile fatty acids of T0, T1, T2, and T3 treatments were 105.8; 142.7; 136.4; and 135.7 mM. respectively, while the average N-NH3 production, microbial protein biomass, and total protein production of the T0, T1, T2, and T3 treatments were 6.48, 7.36, 8.18, 6.60 mM; 15.04, 75, 12.59, 15.31 mg/ml; and 34.10, 23.72, 33.72, 40.80 mg/g. In conclusion, the use of fermented palm fronds at a 20% level in complete feed gave the best result in the production of volatile fatty acids, improved digestibility of dry matter, and organic matter, so it can be used as an alternative feed to replace grass fiber.

The effect of molasses on ammoniated straw by using urea on dry and organic matter digestibility as in vitroABSTRACT. Aimed of this research was to find out the optimal level of molasses addition to improve quality, dry matter and organic matter digestibility of rice straw ammonization process. Materials used were rumen fluid of fistula cattle, grind of rice straw, water, urea and molasses. Research designed used Completely Randomized Design (CRD). As treatments were R0: rice straw 1000 g dry matter + 500 g water + 50 g urea + 0 percent of molasses, R1: R0 + 15 percent of molasses, R2: R0 + 30 percent of molasses. Urea and molasses dissolved in water and then entered into pollybag. All pollybag observe and let for 15 days, each treatment replicated 6 times. Variable measured were dry matter digestibility and organic matter digestibility. Research result showed that ammonization product of NH3, Acidity Level and crude fiber having decreased while crude protein content increased. Variance analysis indicated that treatments had significant effect (P0.05) on dry matter and organic matter digestibility. Orthogonal polynomial test indicated that level of molasses increase (P0.05) of dry matter and organic matter digestibility linearly. It can be concluded that addition up to 30 percent in ammoniating of rice straw using urea can improve quality of ammonization and increasing dry matter and organic matter digestibility.

Improving of rice straw quality and its effect on ability nutrient digestibility and rumen metabolism products of buffalo in-vitro with feces as inoculum source ABSTRACT.  This study was aimed to determine the effect of feeding  ammoniated rice straw plus concentrate on buffalo nutrient digestibility and rumen fermentation products by in vitro. The Research was carried out by using  experimental method, designed according to completely  randomized design (CRD). The source of inoculum was obtain from different feces of three  buffalos kept in  Datar Village of Purwokerto region fed  rice straw, rice straw plus concentrate and rice straw ammoniated plus concentrate with dry matter ratio of 80 : 20. The treatments tested consisted of three treatments, namely R0 =  control feed using rice straw; R1 = the use of rice straw plus concentrate with a ratio of  (DM basis) 80:20; R2 = the use of ammoniated rice straw plus concentrate with a ratio of (DM basis) 80:20. The treatments were repeated 7 times, so there were 21 experimental units. The Variables measured included total VFA, Ratio A/P, N-NH3, Microbial Protein Synthesis (MPS),   Dry Matter and Organic Matter Digestibility. The result of this study showed that the treatment had an effect significant (P0.05) on the concentration of VFA, Ratio A/P,  N-NH3,  Microbial Protein Synthesis (MPS), and Dry Matter and Organic Matter Digestibility. The HSD test showed that the highest production of  VFA,Ratio A/P, N-NH3, Microbial Protein Synthesis (MPS), Dry Matter and Organic Matter Digestibility were achieved at R2 followed by R1 and R0 respectively. The conclusion is that the ammoniated rice straw supplemented with concentrate can be recommended to be fed to buffalo

Abstrak: Penelitian ini telah dilaksanakan di Laboratorium Ilmu Nutrisi dan Makanan Ternak Program Studi Peternakan Fakultas Pertanian Universitas Syiah Kuala dan dilanjutkan dengan pengiriman sampel penelitian ke Laboratorium Ternak Perah Institut Pertanian Bogor (IPB) untuk pengujian kecernaan secara in vitro. Tujuan dari penelitian ini adalah menguji kecernaan secara in vitro complete feed berbahan dasar ampas sagu dengan teknik fermentasi yang berbeda.Pembuatan complete feed disusun sesuai dengan kebutuhan ternak domba. Penelitian ini menggunakan Rancangan Acak lengkap (RAL) dengan 3 perlakuan dan masing-masing perlakuan terdiri atas 5 ulangan. Perlakuan 1 (kontrol) adalah complete feed tanpa penambahan Saus Burger Pakan (SBP) dan tanpa fermentasi, perlakuan 2 adalah ampas sagu difermentasi dengan SBP selama 14 hari kemudian dicampur menjadi complete feed dan difermentasi hingga 21 hari, sedangkan perlakuan 3 adalah ampas sagu + complete feed kemudian difermentasi dengan SBP selama 21 hari. Parameter yang diamati dalam penelitian ini adalah pH, Bahan Kering (BK), Bahan Organik (BO), Koefisien Cerna Bahan Kering (KCBK), dan Koefisien Cerna Bahan Organik (KCBO). Hasil pengamatan menyimpulkan bahwa, teknik fermentasi yang berbeda berpengaruh sangat nyata (P0,01) terhadap semua parameter yang diamati, yaitu pH, BK, BO, KCBK dan KCBO. Dari hasil penelitian dapat disimpulkan bahwa nilai kecernaan complete feed berbahan dasar ampas sagu dipengaruhi oleh perbedaan teknik fermentasi. Digestibility Evaluation In Vitro Complete Feed Fermentation Sago Residues Based with Different Fermentation Techniques Abstract: This research was conducted at the Laboratory of Nutrition and Feed Studies, Program study Animal Husbandry, Faculty of Agriculture, University of Syiah Kuala and continued with the delivery of sample to the Laboratory of Dairy Cattle Institute Pertanian Bogor (IPB) for testing of in vitro digestibility. The purpose of this study was to test in vitro digestibility of complete feed made from  the residues of sago with different fermentation techniques. Complete feed was arranged based on the needs of sheep. This study used a complete randomized design (CRD) with 3 treatments and each treatment consisted of 5 replicates. Treatment 1 (control) was a complete feed without the addition of Saus Burger Pakan (SBP) and without fermentation, treatment 2 was sago residu  fermented with SBP for 14 days and then mixed into a complete feed and fermented up to 21 days, whereas treatment 3 was the sago residue + complete feed then fermented with SBP for 21 days. The parameters observed in this study were pH, Dry Matter (BK), Organic Matter (BO), Dry Matter digestibility coefficients (KCBK), and Organic Matter Digestibility Coefficient (KCBO). The result influence showed that, different fermentation technique was significantly (P 0.01) all parameters, namely pH, BK, BO, KCBK and KCBO. It can be concluded that the digestibility of complete feed made from sago residue influenced by differences in fermentation techniques

This research aims to improve the digestibility and nutritional value of rice straw as a ruminant animal feed and determine the effect of long soaking in sea water of rice straw on digestibility in vitro dry matter and organic matter. The used materials in this study are a bales machine, weighing scales, rapiah rope, poly bags, rice straw varieties Ciliwung obtained from rice fields in Bantimurung, sea water with 2.71% saline, as well as tools and chemicals for analysis of in vitro dry matter and organic matter digestibility with pepsin, calculus method. This study is done based on Randomized Complete  Design (RCD) with 5 treatments and 4 replications. Variance based on the long soaking treatment gives the nature of the response curve linear on the digestibility of dry matter and also to organic matter digestibility. The magnitude of the correlation length relationship soaking rice straw and with sea water for in vitro dry matter and organic matter digestibility that is 96.5% and 96.8% respectively. Based on the results of research and discussion can be concluded that rice straw is soaked with sea water 3-12 days producing dry matter digestibility and organic matter are higher than the rice straw that is not soaked with sea water. Soaking of the sea water for 3-12 days increase the digestibility of dry matter and organic matter.

This experiment investigated various substrate growth of bamboo stems microbes (BSM) in rice straw fermentation and its effect on dry matter digestibility (DMD), organic matter digestibility (OMD), NH3-N and VFA concentration. Materials were rumen fluid of beef cattle from the slaughterhouse. Basal diets consisted of rice straw and concentrate with ratio of 40 : 60 (% dry matter) were administered to five treatments namely P0 = untreated rice straw (control), P1 = Ammoniated rice straw,  fermented rice straw using microbial bamboo stems grown on ammoniated rice straw (P2), rice bran (P3) and cassava waste (P4). The experiment used in vitro method was designed with Completely Randomized Design and the data were analyzed variance. The results showed that the treatment had no significant effect on dry matter digestibility (DMD), organic matter digestibility (OMD), NH3-N and VFA concentration (P>0,05). The fermentation process of microbial bamboo stems in rice straw with a level of 10% with different substrate did not able to  increase the digestibility and products fermentation, but there tends that the rice bran and substrates can increase DMD and N-NH3 concentration

In Indonesia, forage is difficult to obtain in the dry season, while the agricultural waste very abundant and potentially pollute the environment. Use of agricultural waste as feed animal is very advantage to obtain cattle nutrients need and alleviate pollution. An experiment was conducted to evaluate the nutrient quality of fermented complete feed based on soybean straw (CFS), compared with forage (F) or forage and concentrate with 4:1 ratio (FC). Proximate analysis was conducted to three kind of feed, then nutrients digestibility were conducted by in-vitro analyzed. Each kind of feed consisted of 4 replicates. The results showed that CFS had higher ash and crude protein content and less crude fiber than FS and F. CFS had dry matter digestibility (DMD), organic matter digestibility (OMD) and total digestible nutrients (TDN) significantly (p< 0.05) higher than F but not significantly different with FC. There are not significantly different of NH3 production among the three kinds of feed. The conclusion was CFS had highest nutrient content. CFS and FC had digestible nutrient that higher than F. The quality of CFS can still be improved by adding N digestible for rumen microbe growth.Key words: complete feed, soybean straw, nutrient digestibility

The research aims to evaluate calsium soap protection as feed supplement based on dry matter<br />digestibility, organic matter digestibility and pH in vitro in the rumen and post rumen. Calsium soap<br />was produced from sardine fish oil and soybean meal, through two protection methods. It was<br />saponification and microencapsulation. This research used Split Subject Repeated Measurement<br />Design with 3 treatments (calcium soap, soybean meal and pangola grass) which each treatment was<br />repeated 5 times. The observation kinetics of dry matter digestibility, organic matter digestibility, and<br />pH rumen fluid were done in incubation times (2, 4, 8, 12, 24 and 48 hours respectively). The first<br />results of the research showed that in the first step (rumen) has dry matter digestibility of calcium<br />soaps: 38.65; 40.00; 42.66; 46.32; 54.40 and 51.40% respectively; organic matter digestibility: 39.44;<br />41.27; 44, 31; 46.32; 56.62 and 52.50% respectively. Kinetics of pH rumen fluid: 6.89; 6.79; 6.78;<br />6.74; 6.71, and 6.88 respectively. In the second step (post rumen), the digestibility of calsium soap<br />production has dry matter digestibility of 74.85% and organic matter of digestibilty of 72.94%. The<br />results could be concluded that in the rumen, calcium soap is the most microbial degradation resistant<br />compared the others and it can be degraded and digested in the abomasum to the small intestine.<br />Therefore pH of rumen fluid is optimal, so it does not interfere with the fermentation in the rumen.<br />Keywords: calsium soap, in vitro, dry matter digestibily, organic matter digestibility, pH value
