Abstract Background: To explore the relationship between the pathological changes of colon, terminal ileum, lung, liver and kidney, and the changes of Bax, PCNA and PAF in a rat model of NEC.Methods: 140 neonatal SD rats were randomly divided into 2 groups. NEC rat model is the experimental group. From the 1st day to the 7th day,10 rats were sampled in each group for pathological examination of colon, terminal ileum, lung, liver and kidney tissue. The levels of Bax, PCNA and PAF were investigated by immunohistochemistry.Results: On the 1st day, the colon, terminal ileum, lung, liver and kidney showed inflammatory damage. On the 5th day, inflammatory injury was decreased. The inflammatory injury was nearly gone by the 7th day. There were differences in the time of apoptosis in intestine. In the intestine, the proliferation of weak to strong. Bax in liver and kidney showed marked apoptosis and apoptosis time increased in lung. PCNA was elevated in lung, liver and kidney, and the expression of PAF in lung and liver was increased.Conclusions: NEC can lead to secondary injury of different degrees in colon, terminal ileum, lung, liver and kidney, and the degree and time of injury and repair were different. In general, organ repair played a leading role on the 4th day after modeling.

NEC group was given hypoxia, cold stimulation and artificial feeding twice a day for 3 consecutive days. The control group was only fed normally. After modeling, From the 1st day to the 7th day, 10 rats were sampled in each group for pathological examination of colon, terminal ileum, lung, liver and kidney tissue. The levels of Bax, PCNA and PAF were investigated by immunohistochemistry. Results: Compared with the normal group, in the NEC group, on the 1st day, the colon, terminal ileum, lung, liver and kidney showed inflammatory damage. On the 5th day, the inflammatory injury was reduced. The inflammation disappeared on the 7th day. There were differences in the time of apoptosis in the intestine. In the intestine, the proliferation of PCNA was weak at first and then strong. Bax in liver and kidney showed marked apoptosis and apoptosis time increased in the lung. The expression of PCNA increased in lung, liver and kidney, and the expression of PAF increased in lung and liver. Conclusions: NEC can lead to secondary injury of different degrees in colon, terminal ileum, lung, liver and kidney, and the degree and time of injury and repair were different. In general, organ repair played a leading role on the 4th day after modeling.

SUMMARY 1. Differential centrifugation, in 0·25 m sucrose solution, has been performed with rat liver and kidney tissue to ascertain whether the yield and composition of the cytoplasmic fractions (mitochondrial, microsome and supernatant fractions) depend on the hormonal status of the animal. 2. After hypophysectomy the ribonucleic acid (RNA) of the mitochondrial fraction from liver underwent a decrease (in terms of body weight) which was sufficient to account for the fall in the RNA of the liver as a whole. There was also a decrease in the yield of the mitochondrial fraction. Administration of pituitary growth hormone (GH) to hypophysectomized rats not only restored to normal the amount of RNA in the mitochondrial fraction and the yield of that fraction, but also led to an apparent shift of RNA from the microsome fraction to the supernatant fraction. A further change observed after hypophysectomy, whether or not GH had been given, was a rise in the yield of the microsome fraction. Hypophysectomized rats given thyrotrophin (TSH) did not show significant cytoplasmic changes as found with untreated hypophysectomized rats, but it was not possible to conclude that TSH had actually reversed the effects of hypophysectomy. 3. Castrated rats showed no abnormalities in the yields of the liver cytoplasmic fractions or in the concentration of RNA in the fractions. Alloxan-diabetic rats showed a rise in the yield of the supernatant fraction from liver. 4. Untreated adrenalectomized rats showed a rise in liver deoxyribonucleic acid, a fall in the yield of the liver mitochondrial fraction, but not in the amount of RNA in that fraction, and a rise in the amount of RNA in the supernatant fraction. Replacement therapy with various adrenocorticoids was attempted, with only partial success. 5. In contrast with the RNA content, the phospholipid content of the liver cytoplasmic fractions was not, in general, dependent on hormonal status. 6. Determinations of the yield and composition (RNA and phospholipid) of the cytoplasmic fractions from kidney disclosed certain hormonal effects, differing from those observed with liver; for example, the kidneys from hypophysectomized rats furnished microsome fractions in lowered yield but with an increased concentration of RNA.

Diabetic dyslipidemia and hyperglycemia contribute to excessive reactive oxygen species (ROS) production, leading to deleterious complications, such as nephropathy, atherosclerosis and cardiac dysfunction, and target major organs in the body. The aim of this study was to investigate the effect of caffeic acid (CA) on mouse weight and survival, serum level of fasting blood glucose (FBG), serum lipid parameters and atherogenic indices, oxidative damage in blood, liver and kidney tissue, pathophysiological changes and their function markers in healthy and alloxan-induced type 1 diabetic mice. Diabetes was induced in mice with a single intravenous injection of alloxan (75 mg kg−1). Two days later, CA (50 mg kg−1) was given intraperitoneally for seven days in diabetic mice. Diabetes affected glucose level, lipid profile, hematological and biochemical parameters, induced DNA damage and apoptotic/necrotic death in whole blood cells, liver and kidney, leading to weight loss and a decreased lifespan. CA treatment of diabetic mice revealed a protective effect on the liver and kidney, hypoglycemic and hypolipidemic properties and high protection against atherogenic outcomes. The obtained results suggest that CA is a safe and potent agent against diabetes that acts as an effective antioxidant in reducing serum glucose, lipid profile and atherogenic indices, leading to increased lifespan in mice.

OBJECTIVE: To verify whether the ileal exclusion interferes with liver and kidney functional changes secondary to extrahepatic cholestasis.METHODS: We studied 24 rats, divided into three groups with eight individuals each: Group 1 (control), Group 2 (ligation of the hepatic duct combined with internal biliary drainage), and Group 3 (bile duct ligation combined with internal biliary drainage and exclusion of the terminal ileum). Animals in Group 1 (control) underwent sham laparotomy. The animals of groups 2 and 3 underwent ligation and section of the hepatic duct and were kept in cholestasis for four weeks. Next, they underwent an internal biliary bypass. In Group 3, besides the biliary-enteric bypass, we associated the exclusion of the last ten centimeters of the terminal ileum and carried out an ileocolic anastomosis. After four weeks of monitoring, blood was collected from all animals of the three groups for liver and kidney biochemical evaluation (albumin, ALT, AST, direct and indirect bilirubin, alkaline phosphatase, cGT, creatinine and urea).RESULTS: there were increased values of ALT, AST, direct bilirubin, cGT, creatinine and urea in rats from Group 3 (p < 0.05).CONCLUSION: ileal exclusion worsened liver and kidney functions in the murine model of extrahepatic cholestasis, being disadvantageous as therapeutic procedure for cholestatic disorders.

In November 2018, a horse owner from the Czech Republic experienced the loss of two of his stallions in consequence of probable poisoning. The affected animals were kept in the stable only. The mares, which were grazing on pasture during the day, were not affected. All the animals were fed the same type of feed. The day before the animals became sick, between 15:00 to 19:00 h the stable was not secured against the entry of strangers. When the mares were brought back to the stable and all animals were closed and locked for the night, no signs of health issues were observed. In the following morning, the two stallions were found in lateral recumbency with seizure attacks. Despite symptomatic treatment (infusion with saline, flunixine meglumine, activated charcoal) and applications of myorelaxants, the symptoms persisted until the afternoon, and the owner agreed with euthanasia. Based on the clinical signs and pathological examination, poisoning by pesticides was suspected. The results of toxicological analyses revealed that carbofuran, metaldehyde, and strychnine were not responsible for the poisoning as they were not detected in the samples. The only positive finding was a high aluminium concentration in the stomach content, liver and kidney samples. Based on the observed symptoms and the high aluminium concentration, it was presumed that aluminium phosphide could be the source of poisoning. As there was no evidence of the possession of such product by the horse breeder and the stable stayed unsupervised for a few hours, intentional poisoning cannot be excluded.
