Phytochemical Analysis, In Vitro Antioxidant and Wound Healing Activities of Turbinaria ornata (Turner) J. Agardh from Gulf of Mannar, India

Abstract Turbinaria ornata, tropical brown algae found in the South pacific and Indian Ocean ecosystems. In accordance with recent studies, Turbinaria ornata J. has potent anti-inflammatory effects. Therefore, this study is aimed to explore the biological activities of ethanolic extract of Turbinaria ornata J by analyzing the presence of phytochemical components, antioxidant property, antimicrobial activity and the wound healing activity. From the results, phytochemical analysis of ethanolic extract of T. ornata showed the presence of alkaloids, saponins, oils, total phenolic and total flavanoid content were estimated to be 0.683 Abs and 0.433 Abs respectively. Anti-oxidant activity of the ethanolic extract of T. ornata extract showed remarkable DPPH radical scavenging activity of about 58.8% at 200μg/ml and total anti-oxidant activity of 0.257 absorbance at 100μg/ml concentration, as compared to that of their respective controls. The ethanolic extract of T. ornata exhibited the maximum zone of inhibition against the clinical pathogens like Pseudomonas aeuruginosa, Escherichia coli, Staphylococcus aureus, Candida albicans and Methicillin-resistant Staphylococcus aureus with their potent anti-microbial activity. Wound healing effects of the ethanolic extract of T. ornata was analysed by using Zebra fish model. The results showed the rapidand significant regeneration of the wounded caudal fin on day 14. Therefore, the preliminary results of this study strongly supports the ethanolic extract of T. ornata may be effective in wound healing and regeneration of the wounded tissues.

Objective: The objective of this study was to evaluate the nature of chemical constituents, total phenolics, total flavonoids, and antioxidant activity of Cleome gynandra and their functional groups with the help of phytochemical, Fourier-transform infrared spectroscopy (FTIR) analysis, colorimetric assay, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay. Methods: C. gynandra of the Cleomaceae family is an annual herb. The dried leaves were powdered and extracted using Soxhlet apparatus by different solvents. Preliminary phytochemical analysis was carried out to identify the phytoconstituents present in the extract of C. gynandra, FTIR spectrum was scanned at the range of 4000-400 cm−1. The extracts were subjected to the colorimetric assay in triplicate manner to quantitative determination of total phenolic and total flavonoid content. Gallic acid and rutin used as standards to determine the total phenolic content and total flavonoid content. Antioxidant activity was evaluated using DPPH radical scavenging method. Results: Phytochemical analysis of the ethanolic extract of C. gynandra revealed the presence of alkaloids, phenolics, saponins, steroids, flavonoids, cardiac glycosides, and tannins. FTIR spectrum showed intense bands at 3679.18, 3616.63, 3317.34, 2943.67, 1634.01, 1360.20, 1036.71, and 778.04 cm−1 corresponding to N-H2, O-H stretch, aliphatic C-H stretch, C=O, C-H benzene, C-O stretch, and C-Cl. The total phenolic content was found to be 8.39 ± 0.0952 mg gallic acid equivalent/g and 66.76 ± 0.0333 mg rutin equivalent/g. The DPPH radical scavenging activity of ethanolic extract was showed more scavenging activity compared to ethyl acetate and n-hexane fractions. Conclusion: The present research work creates a platform to screen many bioactive chemical constituents present in C. gynandra to treat various diseases.

ABSTRACTSolanum nigrum is a paramount in medicinal perspective and belongs to family Solanacea. From different parts of the plant significant pharmacological and biological activities have been reported previously.  In the present study, the ethanolic extract of leaves was investigated for phytochemical analysis, vitamin and mineral content. Phytochemical analysis of the leaves revealed the presence of phenols, alkaloids, flavonoids, glycosides, saponins, tannins, phytosterols and triterpenoides. The leaves extract was found to contain appreciable amounts of flavonoids and phenols. The extract showed the presence of vitamins such as ascorbic acid, folic acid, and niacinamide. It has been found that the leaves of Solanum nigrum is rich in minerals like copper, magnesium, manganese, vanadium, chromium, calcium, zinc, sodium, and potassium. The antioxidant (free radical scavenging) activity of the extract was determined against 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) ABTS, Nitric oxide(NO) and Superoxide Scavenging assays. Solanum nigrum leaves extract was found to be antioxidant in nature which is evident from DPPH, ABTS, NO, Superoxide radical scavenging assays. These findings suggest that Solanum nigrum possess biologically important phytoconstituents, significant amounts of vitamins and minerals. IN addition, S. nigrum leaves extract exert free radical scavenging potential in vitro.Key words: Medicinal plants; Solanum nigrum leaves; phytoconstiuents;Antioxidant.

Euphorbia nivulia (EN) one of the members of Euphorbiaceae family, is a medicinal plant of Cholistan Desert (Punjab, Pakistan) that is traditionally used for a number of diseases. The plant is enriched with many phyto-constituents including flavonoids, triterpenes and polyphenols. In present study, crude extract as well as various fractions were assessed for phytochemical analysis, HPLC profiling, radical scavenging property and total phenolic and flavonoid contents. Hydro-alcoholic (70%) crude extract of EN was subjected to fractionation using different solvents including n-hexane, chloroform, butanol and aqueous. After performing the phytochemical screening and HPLC profiling, antioxidant activity was estimated by using DPPH and FRAP assays. Total flavonoid and phenolic contents were also estimated. HPLC analysis confirmed the presence of flavonoids and polyphenols like quercitin, gallic acid, caffeic acid, vanillic acid, benzoic acid, chlorogenic acid, syringic acid and ferulic acid in crude extract as well as butanol and aqueous fractions. Results revealed that butanol fraction showed maximum phenolic (143.26±2.65 mg/g GA/g) and crude extract showed maximum flavonoid (69.80±1.212 mg/g Q) contents. Maximum antioxidant potential was displaced by butanol fraction which was IC50=0.04±0.02 by DPPH and 1193.77±12.4 μmol TE/ml by FRAP respectively. Current study is the first information about the HPLC profiling of phenolic compounds in EN and its antioxidant potential that could be a step forward towards ethno-pharmacological based phyto-medicine.

The ethanolic extract of the leaves of Cassia occidentalis (Co) were subjected to phytochemical analysis by standard qualitative analysis and the invitro antioxidant activity was evaluated by determination of total antioxidant capacity, 1.1- diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, hydrogen peroxide (H2O2) radical scavenging activity, superoxide scavenging activity and Ferric reducing anti oxidant potential (FRAP). The analyses revealed that the ethanolic extract of Co was able to efficiently scavenge the free radicals in a dose dependant manner. The results were compared with the standard antioxidant ascorbic acid. The results have shown that crude ethanolic extract of the leaves of Co showed excellent antioxidant activity due to the presence of bioactive compounds namely alkaloids, betacyanin, cardiac glycosides, coumarins, flavonoids, phenols, steroids, saponins, tannins, terpenoids, anthraquinones and emodins.

A method for the separation and quantification of acteoside by reverse-phase high-performance liquid chromatography (HPLC) was developed and validated. Hot water and ethanolic extracts of Osmanthus fragrans leaves and flowers were analyzed for acteoside content. Excellent linearity was obtained, with an r2 higher than 0.999. The precision, specificity, and accuracy of our method were excellent, suggesting that it can be conveniently used for the quantification of acteoside in the crude extract of O. fragrans. The hot water and ethanol extracts were analyzed, and their biological activities were tested. The extraction yields, marker (acteoside) contents, and antioxidant activities of the leaf and flower extracts were analyzed. The antioxidant activity was confirmed by measuring the 2,2-diphenyl-2-picrylhydrazyl radical scavenging activity, reducing power, and total phenolic content. The acteoside content tended to be higher in the 100% ethanol extract of O. fragrans compared to those with the other extraction conditions tested. Overall, almost all extracts prepared with ethanolic solvents tended to produce better antioxidant activity than those prepared with hot water. These results suggest that the ethanolic extract of O. fragrans could serve as a potential antioxidant and anti-inflammatory pharmaceutical source, and our validated method would be useful for the quality control of O. fragrans extracts.

Background: According to the Iranian Traditional Medicine (ITM) references, Platanus orientalis L. possesses wound healing properties. Herein, we developed different topical formulations based on the ethanolic extract of P. orientalis leaves and evaluated its wound healing effects through an in vivo model. Methods: Hydroalcoholic extract of the leaves was obtained from ethanol 80% and it was evaluated for DPPH radical scavenging activity, total phenolic and flavonoid contents as well as the presence of tannins. Different topical formulations including ointment (D-O) and polymer film (D-F), were prepared and an in vivo test was run for 14 days in an excision wound model consisting of 5 groups of 6 rats. Results: Our results indicated the higher efficacy of D-O comparing with D-F, as wound surface area remarkably reduced within 14 days post-injury. Also, histological features including epitheliogenesis score, neovascularization, and collagen density indicated the potential wound healing effect of D-O. Conclusion: Wound healing properties of the ethanolic extract of P. orientalis leaves depended on the type of formulation and D-O was found to be much more potent than D-F, from reducing wound surface area, maximum epitheliogenesis score, proper neovascularization pattern, and early type I collagenization points of view.

Phytochemical analysis of C. alatavicus revealed the presence of phenols, flavonoids, anthocyanins, carotenoids, amino acids and carbohydrates. The flavonoid, amino acids and carotenoid contents were higher in aerial part (1.50%, 7.49% and 9.78mg%, respectively) than in bulb (0.43%, 3.88% and 0.91 mg%, respectively). Total phenolic content (TPC), total antioxidant (TAA), 2.2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and antibacterial activities of water, methanol, ethanol and dichloromethane extracts from aerial part and bulb were tested. TPC ranged from 13.63 to 72.29 mg gallic acid equivalents (GAE)/g extract. The maximum TAA were observed in ethanol (61.34%) and methanol extracts (46.13%) from aerial part with a high TPC (72.29 and 62.37 mgGAE/g extract, respectively). Ethanol extracts from aerial part and bulb had good scavenger of DPPH radicals (65.5% and 54.08%, respectively) with an IC50 387 and 447 µg/ml. Ethanol extract from aerial part was most effective against gram-positive bacterial strains S. aureus, B. subtilis and B. cereus. Biological activities of the extracts were correlated with the TPC. It can be deduced that ethanol and methnol extracts of C. alatavicus contains useful potent bioactive compounds with antioxidant and antimicrobial activities.

Aster scaber is a wild vegetable cultivated in Korea and is known to contain phytochemicals with various biological activities. The potential antioxidant and anti-obesity effects of A. scaber and their mechanism are yet to be reported. We evaluated the total phenolic, flavonoid, and proanthocyanidin contents and oxygen radical absorbance capacity of A. scaber ethanolic extract (ASE), and analyzed the major phenolic compounds of ASE. Antioxidant activity was measured at the chemical level through 2,2-diphenyl-1-picrylhydrazyl (DPPH), reducing power assay, 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), and fluorescence recovery after photobleaching (FRAP) assay. In addition, it was measured in vitro through inhibition of Reactive oxygen species (ROS) production in 3T3-L1 adipocyte, and inhibition of lipid accumulation was also evaluated. ASE reduced the expression of enzymes involved in the production of ROS and increased the expression of antioxidant enzymes that reduce increased ROS levels. They also reduced the expression of adipogenesis transcription factors that regulate adipocyte differentiation in relation to ROS production, inhibited the expression of lipogenesis-related genes related to fat accumulation through AMP-activated protein kinase (AMPK) activity, and increased expression of lipolysis-related genes. Thus, ASE containing CGA (chlorogenic acid) inhibits ROS production in 3T3-L1 adipocytes, owing to its strong antioxidant activity, and inhibits lipid accumulation caused by oxidative stress. The extract can be used as a potential functional food material for reducing oxidative stress and obesity.

Abstract Curcuma longa is an important dietary plant which possess several pharmacological activities, including antioxidant, antimicrobial, anti-inflamatory, anticancer and anti clotting etc. The aim of the present study was to determine the phenolic profile of Curcuma longa and in vitro antioxidant and antidiabetic activities. In HPLC chromatogram of Curcuma longa rhizome extract 15 phenolic compounds were identified namely Digalloyl-hexoside, Caffeic acid hexoside, Curdione, Coumaric, Caffeic acid, Sinapic acid, Qurecetin-3-D-galactoside, Casuarinin, Bisdemethoxycurcumin, Curcuminol, Demethoxycurcumin, and Isorhamnetin, Valoneic acid bilactone, Curcumin, Curcumin-O-glucuronide respectively. The ethanolic extract displayed an IC50 value of 37.1±0.3 µg/ml against alpha glucosidase. The IC50 value of DPPH radical scavenging activity was 27.2 ± 1.1 μg/mL. It is concluded that ethanolic extract of Curcuma long is rich source of curcumin and contain several important phenolics. The in vitro antioxidant and alpha glucosidase inhibitory effect of the plant justifies its popular use in traditional medicine.

Medicinal plants are the potent source of biologically active compounds and have always been of field of interest for the effective chemotherapeutic agents and offering a broad spectrum of activity with greater emphasis on preventive action. The objective of this study was to screen the phytochemicals, estimate the content of phenolic, flavonoids and alkaloids compounds and determines the antioxidant capacity of the rhizomes of Kaempferia galanga Linn, Kaempferia parviflora and Kaempferia pulchra (K. galanga, K. parviflora and K, pulchra, Zingiberaceae). Qualitative analysis of various phytochemical constituents and quantitative analysis of total phenol, flavonoids and alkaloids were determined by the well-known test protocol available in the literature. The ethanolic extract of rhizomes of K. galanga, K. parviflora and K, pulchra was studied for antioxidant activity on different in vitro models namely 1,1-diphenyl, 2-picryl hydrazyl (DPPH) assay and Nitric oxide (NO) radical scavenging method. Phytochemical analysis of ethanolic extract of all three plants revealed the presence of flavonoids, alkaloids, saponins, phenolics, carbohydrate, and tannin. The total phenolic, flavonoids and alkaloids content of ethanolic extract of K. galanga, K. parviflora and K, pulchra rhizomes were 0.813, 1.146; 1.047, 1.237, 0.285; 0.755, 0.822, 0.975/100mg respectively. All extracts showed dose dependent free radical scavenging property in the tested models, which was comparable to that of ascorbic acid. The present study concluded that the crude extract of K. galanga, K. parviflora and K, pulchra rhizomes is a rich source of secondary phytoconstituents which impart significant antioxidant potential. It is expected that the important phytochemical properties recognized by our study in the indigenous medicinal plants will be very useful in the curing of various diseases when taken along with our food.
