Abstract Objective This study was aimed at evaluating the toxicity and the antioxidant property of Cassia fistula stem bark extracts in zebrafish. Materials and Methods Crude aqueous and ethanol extracts of C. fistula stem bark were obtained following a standard solvent-based extraction method. The toxicity of these extracts on zebrafish embryonic development was determined and the LC50 values were calculated. Finally, the antioxidant property of C. fistula stem bark extracts was determined in arsenic-induced oxidative stress models of zebrafish. Results The aqueous extract of C. fistula stem bark showed a slightly larger LC50 value (213.6 ppm) compared with the ethanol extract (LC50 = 63.5 ppm), suggesting a lower toxicity of the aqueous extract. A significant reduction of reactive oxygen species (ROS) signal was observed in arsenic-exposed embryos treated with the aqueous extract, but not the ethanol extract, indicating that the antioxidant activity is present only in the aqueous extract of C. fistula stem bark. Conclusion Identification of antioxidants from natural sources is desirable because of increasing safety concerns associated with synthetic antioxidants. This study demonstrated that aqueous extract from C. fistula stem bark possesses antioxidant properties, which can be further characterized for mechanism of action and potential applications.

Ficus glumosa Delile (Moraceae), a reputed plant that is used in herbal medicine, is of high medicinal and nutritional value in local communities primarily ascribed to its phytochemical profile. Currently, there are hardly any fine details on the chemical profiling and pharmacological evaluation of this species. In this study, the flavonoids and phenolics contents of the ethanol extracts and four extracted fractions (petroleum ether (PE), ethyl acetate (EA), n-butanol, and water) of the stem bark of Ficus glumosa were firstly quantified. Further, their antioxidant and antiproliferative potentials were also evaluated. The quantitative determination indicated that the EA and n-butanol fractions possessed the highest total flavonoids/phenolics levels of 274.05 ± 0.68 mg RE/g and 78.87 ± 0.97 mg GAE/g, respectively. Similarly, for the 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric-reducing antioxidant power (FRAP) assays, the EA fraction exhibited high potency in both DPPH and ABTS+ scavenging activities with IC50 values of 0.23 ± 0.03 mg/mL, 0.22 ± 0.03 mg/mL, and FRAP potential of 2.81 ± 0.01 mg Fe2+/g, respectively. Furthermore, the EA fraction displayed high cytotoxicity against human lung (A549) and colon (HT-29) cancer cells. Additionally, the liquid chromatography coupled with electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) was employed in order to characterize the chemical constituents of the EA fraction of Ficus glumosa stem bark. Our findings revealed 16 compounds from the EA fraction that were possibly responsible for the strong antioxidant and anti-proliferative properties. This study provides edge-cutting background information on the exploitation of Ficus glumosa as a potential natural antioxidant and anti-cancer remedy.

ABSTRACT: The study is designed for the evaluation of in-vivo Hepato protective and in-vitro Anti oxidant activity of ethanol extracts from the bark of Shorea robusta (Dipterocarpaceae) by CCl4 induced hepatotoxicity in rats. Ethanol extracts from the bark Shorea robusta (EESR) was evaluated for hepatoprotective activity in rats by inducing liver damage by CCl4. The anti oxidant activity of EESR was assayed by various in-vitro antioxidant methods and activities were compared to standard ascorbic acid. Ethanol extracts at an oral dose 200mg/kg and 400mg/kg exhibited a significant (*p<0.005) protective effects by lowering the level of SGOT, SGPT, ALP, Serum bilirubin, total cholesterol and increasing the level of total proteins as compared to Silymarin (50mg/kg) used as positive control. The extracts exhibit significant anti oxidant activity in various in vitro anti oxidant models.  From these studies we are concluding that, the ethanolic extracts of S.robusta have potent hepatoprotective effects and have anti oxidant properties, hence can be used as a natural product against liver damage.KEY WORDS: Anti oxidant, Carbon tetra chloride,  Hepatoprotective,  Shorea robusta

Delonix elataL. (Ceasalpinaceae), is widely used by the traditional medical practitioners of Karnataka, India, to cure jaundice, and bronchial and rheumatic problems. The objective of this study was to screen thein vitroantioxidant and hepatoprotective activity of the stem bark extracts against CCl4-induced liver damage in rats. Among different stem bark extracts tested, the ethanol extract (DSE) has shown significantin vitroantioxidant property in radicals scavenging, metal chelating, and lipid peroxidation inhibition assays. HPLC analysis of the DSE revealed the presence of known antioxidant molecules, namely, gallic acid, ellagic acid, coumaric acid, quercetin, and rutin. Bioassay-guided fractionation of DSE has resulted in the isolation and characterization of quercetin. DSE and quercetin have shown significant prophylactic effects by restoring the liver function markers (AST, ALT, ALP, serum bilirubin, and total protein) and antioxidant enzymes (SOD, CAT, GPx, and GST). These results were proved to be hepatoprotective at par with silymarin and well supported by the histological observations of liver sections with distinct hepatic cells, and mild degree of fatty change and necrosis. The results indicated that the DSE and quercetin were significant for prophylactic activity against CCl4-induced liver damage in rats. This activity could be attributed to the antioxidant constituents in the DSE and hence justified the ethnomedicinal claims.

Uapaca guineensis Mull. Arg. (Euphorbiaceae) commonly called false mahogany and “Ewe akun” in the western part of Nigeria grows in humid localities, mixed evergreen forest and sea-level up to 1100 m altitude. The study was designed to isolate Betulinic acid (3?-hydroxylup-20-(29)-en-28-oic acid) which is a naturally occurring pentacyclic triterpenoid that exhibited antibacterial, antimalarial, anti-inflammatory, anthelmintic and antioxidant properties from the stem bark and root of Uapaca guineensis. The air-dried and pulverized stem bark and root of U. guineensis (0.5 kg each) were separately extracted with aqueous ethanol (80%) for 72 hour at room temperature, filtered, concentrated on rotatory evaporator at 37 oC and then stored in a desiccator. The ethanol extracts of the stem bark was partitioned successively with n-hexane, dichloromethane and methanol, while ethanol extract of the root was chromatographed directly. The dichloromethane and methanol extracts of the stem bark was also chromatographed. These results in the formation of whitish powder that are UV inactive and also gave single spot each with cerium sulphate. They are coded as DBG-D13 (25.0 mg), MBG-Q3 (6.1 mg) and ERG-E7 (20.0 mg) for dichloromethane, Methanol and Ethanol extracts respectively. The structure was unequivocally established by 1HNMR, 13CNMR, 2D-NMR, IR, UV and MS spectra analysis as well as by comparison of the spectra data with previously reported values. The dichloromethane, methanol and the ethanol extracts all yielded betulinic acid in substantial quantity. The isolated compound is a confirmation to existing information and an addition to the library of organic compounds.

Objective: The objective of the present study was to evaluate the hepatoprotective effect of mixture of stem bark, leaf and root extract of Bauhinia acuminata L. using carbon tetrachloride induced hepatotoxicity model. Method: Powdered mixture materials were extracted with distilled water using soxhlet apparatus. The dried extracts were subjected to preliminary phytochemical analysis and the extracts were evaluated for acute oral toxicity by OECD guidelines 423.The aqueous extract at a dose level 200 mg/kg and 400 mg/kg were selected and administered to albino rats for hepatoprotective screening. Hepatotoxicity was induced by Carbon tetrachloride (1 ml/kg with CMC). Silymarin (100 mg/kg p. o) was used as the standard. Result: The preliminary phytochemical examination showed the presence of Alkaloids, Glycosides, Saponins, Flavanoids, Tannins, Aminoacids, Tryptophan, Quinones, Terpinoids, Starch, Vitamin C and Carbohydrates. Furthermore, the acute toxicity study results showed that the extracts were found to be safe up to 2000 mg/kg b. wt. The extract dose dependently shows hepatoprotective potential by restoring the elevated biochemical parameters. Conclusion: The results obtained from the study indicates that mixture of stem bark, leaves and root extract of Bauhinia acuminata L. possess promising hepatoprotective activity. The activity might be due the presence of the phytoconstituents including Alkaloids, Glycosides, Saponins, Flavanoids, Tannins, Aminoacids, Carbohydrates Terpinoids, Starch, Vitamin C, Tryptophan and Quinones in the extract. Further studies are required to identify the active principle responsible for the hepatoprotective activity.
