Screening of Antioxidant and α-Glucosidase Inhibitory Activities of Indian Medicinal Plants

Background and Objective: Chronic diseases are associated with low-grade inflammation and oxidative damage. Traditional medicines have been used to manage these disorders due to their high polyphenol content and potent antioxidant activity. We evaluated the in-vitro anti-diabetic and antioxidant potential of extracts of several medicinal plants namely, Mangifera indica, Terminalia arjuna, Moringa oleifera, Albizia lebbeck, Terminalia chebula and Hippophae rhamnoides. Methods: Total polyphenol, flavonoid, and saponin contents were estimated by standard methods. Antioxidant activity was measured using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay. The anti-diabetic potential was evaluated using in-vitro α-glucosidase inhibition assay. Results: Terminalia chebula was found to be the richest in both polyphenols (566.5 ± 21.9 μg Gallic acid equivalents/mg of dry weight) and flavonoids (190.67 ± 10.78 quercetin equivalents/mg of dry weight). Extract of Terminalia arjuna was the richest source of saponins (171.92 ± 12.48 μg saponin equivalents/mg of dry weight). All plant extracts showed potent anti-oxidant activity as reflected by their IC50 values in DPPH assay, with Albizia lebbeck (IC50 = 1.35 μg/ml) being the most potent. All plant extracts also showed potent anti-diabetic activity as inferred from their ability to inhibit α- glucosidase, the principal enzyme involved in the metabolism of dietary carbohydrates in the intestine. It was observed that all tested extracts were more potent (IC50 2.53 to 227 μg/ml) in comparison to the standard α-glucosidase inhibitor Acarbose (IC50=2.7 mg/ml). Conclusions: The plant extracts of Mangifera indica, Terminalia arjuna, Moringa oleifera, Albizia lebbeck, Terminalia chebula, and Hippophae rhamnoides possess potent antioxidant and α- glucosidase inhibitory potential and can aid in the management of postprandial hyperglycemia and oxidative damage.

Abstract Little is known about which secondary metabolites are responsible for inhibiting pathogenic bacteria and reducing the pro-oxidant effect on the leaves of four medicinal plants used as phytobiotic in animal production. The aim of this study was to evaluate the antimicrobial and antioxidant activity of four medicinal plants (Anacardium occidentale, Psidium guajava, Morinda citrifolia and Moringa oleifera.) in vitro. A total of six bacterial strains were inoculated, then minimum bactericidal concentration (MBC) was evaluated in fine powder and minimum inhibitory concentration (MIC) and MBC were determined on the aqueous extract. Also, the in vitro antioxidant activity was evaluated through 1,1-diphenyl-2-picryl-hydrazyl, as well as the main secondary metabolites were identified and quantified by chromatographic analysis. The results showed that Anacardium occidentale and Psidium guajava leaves had higher antimicrobial activity against all bacterial strains. In addition, Morinda citrifolia inhibited S. aureus in the aqueous extract, although without in vitro bactericidal effect, while Moringa oleifera leaf did not show antimicrobial effect. All plants showed antioxidant capacity, standing out Anacardium occidentale and Psidium guajava. Mainly the leaves of Anacardium occidentale showed high concentrations of quercetin 3-O-glucoside-7-O-rhamnoside, kaempeferol-7-O-glucoside, quercetin, caffeic acid, and cinnamic acid. Apparently, the antimicrobial and antioxidant activity are due to the main polyphenolic compounds identified in medicinal plants (mainly Anacardium occidentale and Psidium guajava); however, further studies are necessary to elucidate the exact mechanism.

The new SARS-CoV-2 belonging to the coronaviruses family has caused a pandemic affecting millions of people around the world. This pandemic has been declared by the World Health Organization as an international public health emergency. Although several clinical trials involving a large number of drugs are currently underway, no treatment protocol for COVID-19 has been officially approved so far. Here we demonstrate through a search in the scientific literature that the traditional Moroccan pharmacopoeia, which includes more than 500 medicinal plants, is a fascinating and promising source for the research of natural molecules active against SARS-CoV-2. Multiple in-silico and in-vitro studies showed that some of the medicinal plants used by Moroccans for centuries possess inhibitory activity against SARS-CoV or SARS-CoV-2. These inhibitory activities are achieved through the different molecular mechanisms of virus penetration and replication, or indirectly through stimulation of immunity. Thus, the potential of plants, plant extracts and molecules derived from plants that are traditionally used in Morocco and have activity against SARS-CoV-2, could be explored in the search for a preventive or curative treatment against COVID-19. Furthermore, safe plants or plant extracts that are proven to stimulate immunity could be officially recommended by governments as nutritional supplements.

 Objective: The objective of the present work is to study the in vitro anti-inflammatory and antioxidant activity of medicinal plants. The extent and correlation between anti-inflammatory and antioxidant activity have been studied.Method: Methanolic and aqueous extracts of five medicinal plants, namely, Ficus racemosa, Aloe vera, Cannabis sativa, Datura stramonium, and Calotropis gigantean have been taken for in vitro anti-inflammatory and total antioxidant activity.Result: The study showed that the inhibition of protein (albumin) denaturation was maximum in aqueous extract of A. vera with 97.55±1.45%. Proteinase inhibitory action of different plant extracts showed significant action and was found to be maximum in aqueous extract of D. stramonium with 87.89±2.58%. Heat-induced hemolysis showed that maximum inhibition was with aqueous extract of F. racemosa with 90.72±3.33%. When hypotonicity-induced hemolysis activity was done it was found maximum in methanolic extract of C. gigantea with 90.58±3.04%. Anti-lipoxygenase activity was found maximum in methanolic extract of F. racemosa with 94.05±4.24%. When total antioxidant activity was done, it was found highest in F. racemosa (4.38±0.546 mM equivalent of ascorbic acid/g tissue).Conclusion: An overall strong positive correlation between anti-inflammatory and antioxidant activity was observed, indicating that antioxidant activity of the plant species studied might be responsible for their anti-inflammatory property. Further work needs to be undertaken to fully elucidate the antioxidants responsible for anti-inflammatory action and to develop better herbal drug formulations.

The present work had the objective of producing liqueurs from mango peels (varieties “Haden” and “Tommy Atkins”) by processes of alcoholic maceration and maceration with pectinase, as well as to evaluate bioactive compounds by reversed-phase high-performance liquid chromatography coupled to diode array detection and fluorescence-detection (RP-HPLC/DAD/FD) and in vitro antioxidant activity (AOX), for by-product potential reuse. Alcoholic maceration in wine ethanol (65% v/v) produced liqueurs with higher phytochemical and AOX content. Maceration with pectinase resulted in liqueurs with higher quercetin-3-O-glucopyranoside content. In relation to mango varieties, Haden liqueurs presented higher bioactive content than Tommy Atkins liqueurs. The liqueurs presented high antioxidant activity. The main bioactive compounds found were flavanols (epicatechin-gallate, epigallocatechin-gallate), flavonols (quercetin-3-O-glucopyranoside and rutin), and phenolic acids (gallic acid, o-coumaric acid, and syringic acid). The present study showed that the production of liqueur enabled the recovering of an important part of the bioactive content of mango peels, suggesting an alternative for the recovery of antioxidant substances from this by-product.

<P>Background: As natural herbs and medicinal plants extracts are widely used as alternative treatments for different parasitic diseases, some have been tested on Cryptosporidium either in vitro or in vivo. This study assessed the prophylactic and therapeutic treatments' effect of Moringa oleifera (M. oleifera) leaves methanol extract on immunosuppressed-Cryptosporidium infected mice. Methods: The evaluation was carried out by Cryptosporidium oocysts count in fecal samples, histopathological changes in the intestinal tissues, determination of IFN-γ level in mice sera and measuring the antioxidant activity in the intestinal tissues. Results: Prophylactic treatment by M. oleifera extract lowered Cryptosporidium oocysts shedding from immunosuppressed-infected mice although there was no complete elimination of the parasite. However, therapeutic treatment induced a significant reduction in Cryptosporidium oocysts counts by 91.8% higher than that of the drug control (nitazoxanide) group (77.2%). Histopathologically, the intestinal tissues from immunosuppressed-Cryptosporidium infected mice showed loss of brush border with severe villous atrophy and extensive necrosis. M. oleifera prophylactic treatment induced a moderate improvement of the pathological changes. However, the villi in M. oleifera therapeutic treated mice retained their normal appearance with minimal inflammatory cells. It was observed that M. oleifera extract induced a significant upregulation of IFN-γ in both prophylactic and therapeutic treated groups compared to that of the infected untreated group. In addition, M. oleifera leaves extract exhibited a significant antioxidant activity by reducing the levels of Nitric Oxide (NO) and Malondialdehyde (MDA) and increasing Superoxide Dismutase (SOD) level in the intestinal tissues compared to those of the infected and drug controls. Conclusion: M. oleifera leaves extract has potent prophylactic and therapeutic activities against infection with Cryptosporidium.</P>
