An experiment was carried out during the year of 2010 - 2013 at Algarah farmer field (1740 m asl), under the aegis of Uttar Banga Krishi Viswavidyalaya, to evaluate the effect of different growth hormone on seed germination of jatamansi, and assess the effect of date of transplanting and plant geometry on its economic and biomass production under the mid hill condition of West Bengal. An experiment was with various growth regulators on Valeriana jatamansi revealed that maximum seed germination was observed with KNO3 (0.2%) (70.00±1.10), and significantly superior to rest of the hormonal treatments except Kinetin 200 ppm. This recorded 124 % more seed germination over the control. GA3 200 ppm took least time to start germination and was at par with Kinetin 200 ppm and KNO3 (0.2%) treated seed. Transplanting of seedling during the first week of July gave significant response on fresh aerial biomass (g/plant) at 6 months only. However, plant spacing gave significant response all the stage of observation, and maximum aerial biomass was recorded with spacing of 30 x 45 cm, and was at par with 30 x 30 cm plant spacing. Maximum underground biomass was recorded with July transplanting with 30 x 45 cm. Maximum underground rhizome biomass was recorded with 30 x 45 cm spacing, and was at par with 30 x 30 cm spacing at 9 and 18-month stage of observation. Root biomass failed to produce any significant response with time of transplanting. However, maximum root biomass was recorded with July transplanting. More root growth was recorded with 30 x 45 cm and was at par with 30 x 30 cm spacing during 15 and 18-month observation.

Asymbiotic seed germination of Eulophia nuda Lindl. was observed on Knudson C medium. About 90% seeds germinated within 8-10 weeks and formed green protocorms in 11-12 weeks. Effect of BA and IBA was studied on plantlet development from protocorms. BA shows the best results with respect to number and length of shoots. Maximum number (6.45±1.36) and length (3.90±0.99) was observed on MS medium supplemented with 4.44μM.BA. Maximum root growth was also observed on same medium (4.8±0.99 number of roots and 1.43±0.13cm length). The regenerated plantlets were successfully acclimatized and transferred to earthen pots. The results presented here show that in vitro seed germination and plantlet development in Eulophia nuda Lindl., an endangered orchid, can be achieved at a higher rate by this method.Int J Appl Sci Biotechnol, Vol 3(2): 243-247 DOI: http://dx.doi.org/10.3126/ijasbt.v3i2.12476

Valeriana jatamansi Jones (Syn. V. wallichii DC.) is an aromatic, medicinal herb used as a tranquilizer and in treating sleep disorders. Rhizome is mainly used to extract essential oil (EO) and valepotriates. High quality and economic yield of rhizomes are available in the third year of growth. Therefore, the cultivation of V. jatamansi is not picking up, and over-exploitation of this plant from wild habitats to meet the increasing demand of the pharmaceutical industry is the cause of threat to the genetic diversity of the species. Further, collections from the wild are heterogeneous, resulting in variable produce. The development of clonal lines can ensure uniform quality and yield of rhizome biomass. An effective clonal propagation method was standardized using different hormonal concentrations of naphthalene acetic acid (NAA) on apical shoot cuttings from the selected clone CSIR-IHBT-VJ-05 for different time durations and raised over various planting media. NAA treatment of 50 ppm concentration for 30 min was found optimum for root induction in apical shoots of V. jatamansi. Variations for EO composition within the clone were non-significant, while samples of the control population were variable. The best quality EO (patchouli alcohol ∼62%) was available during the third year of plant growth. A propagation technique for large-scale quality plant material (QPM) production has been standardized to reduce the stress over natural resources and promote V. jatamansi for use in the aromatic and pharmaceutical industry.

The experiment was conducted in the Agronomy Laboratory of Agrotechnology Discipline, Khulna University, Khulna from 29th August to 17th September, 2009. The experiment was laid out in a Completely Randomized Design (CRD) with two factors and three replications. The aim of the experiment was to study the efficacy of three osmopriming agents viz ., NaCl, KC1 and CaCb with control on seed germination and seedling growth of three T. aman rice varities viz., BRRI dhan40, BRRI dhan41 and BINA dhan7. Among the chemicals CaCb showed best as osmopriming agents and it enhanced all germination parameters. BRRI dhan41 showed the best performance. Better root length and shoot length were observed in control. Seeds of BRRI dhan41 when treated with NaCl showed the highest germination percentage (98.67%), germination energy (88%), germination speed (96.83%), but vigor index (10.63) was found highest in BRRI dhan40 when it was treated with KC1. BRRI dhan41 produced the largest root (8.56 cm) when seeds were treated with NaCl. BRRI dhan40 (10.51 cm) and BINA dhan7 (10.53 cm) produced the highest shoot length at controlled condition and CaCl2 treated seed, respectively.   DOI: http://dx.doi.org/10.3329/jbau.v10i1.12013   J. Seeds of three flue-cured tobacco (Nicotiana tabacum L.) varieties, Hicks Broadleaf, Yellow Gold, and Canadel, were soaked for 1, 2, 3 or 5 hours in a saturated solution of diethyl sulphate, a chemical mutagenic agent, to increase variability of plant characteristics. Diethyl sulphate had a detrimental effect on both extent and rate of germination; each decreased with length of soaking period. Seedlings from treated seed grew more slowly than controls.

Effects of cogongrass foliage and rhizome plus root residues on germination and shoot and root growth of barnyardgrass, bermudagrass, browntop millet, hemp sesbania, Italian ryegrass, and prickly sida were investigated in greenhouse experiments. Ground residues of dried cogongrass foliage and rhizomes plus roots were mixed separately with sterilized sand to obtain residue concentrations of 0, 0.25, 0.5, 1, 2, 4, and 8%. These residue concentrations were investigated on bermudagrass and Italian ryegrass, and the 8% residue concentrations were also evaluated on hemp sesbania, prickly sida, barnyardgrass, and browntop millet. Foliage and rhizome plus root residues at concentrations as low as 0.25% inhibited seed germination and shoot and root growth of all species except hemp sesbania. Germination of bermudagrass and Italian ryegrass was reduced by as much as 97% and shoot and root growth by as much as 94% at the highest residue concentrations. Rhizome plus root residues reduced germination and shoot and root growth of bermudagrass and Italian ryegrass more than foliage residues. Foliage and rhizome plus root residues reduced germination and shoot and root biomass of prickly sida, barnyardgrass, and browntop millet at similar levels. Results indicate that cogongrass tissue may contain allelochemicals that contribute to its invasiveness and extreme competitiveness.

A study was carried out on biomass, shoot density and leaf production variability in three Thalassia testudinum meadows under different environmental characteristics in Nuevitas Bay, Cuba, in different seasons. The first site has muddy-sandy sediments and it is affected by bottom trawl fishing; the second has sandy sediments and it is affected by waste-water discharges, and the third has sandy-muddy sediments and no human impacts are present. Leaf, rhizome, and root biomass, daily production of leaves, density of short shoots, and length and width of leaves were estimated five times in a year. Seasonal variations were observed, with higher values of leaf and rhizome biomass, short shoot density, and daily production of leaves occurring in spring and summer. Spatial differences seem to be related to the environmental characteristics of each site: Leaf daily production, leaf biomass and leaf length were lower, and root biomass was higher in the site where sediments are impacted by fishing with bottom trawls; short shoot density and rhizome biomass were higher in the nonaffected site. Lower values of root biomass appeared where waste-water discharges occurred.Se realizó un estudio de la variación de la biomasa, la densidad de vástagos y la producción de hojas en tres praderas de Thalassia testudinum con diferentes características ambientales en la bahía de Nuevitas, Cuba, en diferentes épocas del año. El primer sitio tiene sedimentos fango-arenosos y está afectado por pesquería de arrastre; en el segundo, el sedimento es arenoso y se vierten aguas residuales en su cercanía, y el tercero, con sedimentos arenoso-fangosos, no está afectado por impactos antropogénicos. La biomasa de hojas, rizomas y raíces, la producción diaria de hojas, la densidad de vástagos, y el largo y ancho de las hojas fueron estimadas cinco veces en un año. Se observaron variaciones estacionales, con los valores más altos de biomasa de hojas y de rizomas, densidad de vástagos y de la producción diaria de hojas en primavera y verano. Las variaciones espaciales parecen estar relacionadas con las características de cada sitio: La producción diaria de hojas, la biomasa de hojas y su longitud fueron menores, y la biomasa de raíces mayor, donde los sedimentos están impactados por la pesquería con arrastre; la densidad de vástagos y la biomasa de rizomas fueron mayores en el sitio no afectado por ninguna acción antrópica. Los menores valores de la biomasa de raíces aparecieron donde se realiza la descarga de residuales.

The effectiveness of solid matrix priming (SMP) and seed brushing was further evaluated by using an thermo-gradient table (Seed Processing, Holland) set at 10 different temperatures from 12 to 30 °C. Intact or brushed seeds of gourd (Lagenaria siceraria) were primed with Micorocel E (Celite Corp.) at 25 °C for 3 days in the mixture of 10 seed: 1 Microcel E: 3 water, by weight, and the primed seeds were dried again for long-term storage. SMP treatment significantly increased earlier seed germination at all temperatures. However, the difference in seed germination rate between intact and SMP-treated seeds was most pronounced at somewhat lower temperatures of 18-22 °C. SMP-treated seed showed about 20% final germination rate at 12 °C, whereas intact seeds did not germinate at all. Seed brushing treatment itself did not influenced the germination rate. However, brushing treatment before SMP treatment significantly increased the SMP effect. Combined use of chemicals in solution further increased the early germination. Details of various seed treatment methods will be presented.

Soil applications with poultry refuse, cocodust, vermicompost, ash, sawdust, khudepana, cowdung, solarized sand, Trichoderma harzianum and or with seed treatment by T. harzianum were evaluated against damping off disease complex of potato and chilli. All the treatments significantly reduced percent damping off over control. T. harzianum treated seed along with soil treatment with T. harzianum performed best in terms of seed germination, percent damping off reduction and enhanced growth characters than soil application with T. harzianum alone. The experiment indicates that seed treated with T. harzianum then sown in different soil amendment applied seed bed performed better in all parameters than only application with soil amendment. Among the different soil amendments, poultry refuse and vermicompost have promising impact on seed germination, reduction of percent damping off and growth of potato and chilli seedlings when applying along with T. harzianum.
