Objective: Enterococcus faecalis, Gram-positive facultative anaerobe capable of invading the dentinal tubules, is resistant to several irrigating solutionsand intracanal medications. Motivated by the antibacterial effect exhibited by garlic extract against a wide variety of Gram-positive and Gram-negativebacteria, as well as anaerobic bacteria such as Lactobacillus and E. faecalis, we aimed in this study to analyze the efficacy of garlic extract against theviability of E. faecalis from clinical isolates of nonvital primary root canals.Methods: We used MTT assay to determine the viability of E. faecalis after exposure to increasing concentrations of garlic extract (10%, 25%, 50%,and 100%) and chlorhexidine (CHX) 2% as a positive control.Results: We performed the analysis using the Kruskal–Wallis test and the post hoc test for multiple comparisons at a 0.05 significance level. At allconcentrations, garlic extracts decreased the viability of E. faecalis. We found no significant differences between the viability values for 25%, 50%,and 100% garlic extracts (p>0.05); however, they were significantly different from the 10% extract and CHX 2% (p<0.05), both with a lower viabilityvalues.Conclusion: Our results showed that garlic extract was effective in decreasing the viability of E. faecalis.

Perawatan saluran akar adalah perawatan yang dilakukan untuk mempertahankan gigi agar dapatberfungsi di lengkung gigi selama mungkin. Penyebab dominan dari kegagalan perawatan endodontikadalah adanya bakteri di dalam saluran akar. Bakteri yang paling sering ditemukan pada isolasi darigigi yang mengalami kegagalan perawatan saluran akar adalah Enterococcus faecalis (E.faecalis).Salah satu jenis tanaman herbal yang memiliki sifat antibakteri yang tinggi adalah tanaman mimba(A.indica). Tujuan penelitian ini untuk menganalisis efek antibakteri ekstrak daun mimba (A.indica)terhadap pertumbuhan E. faecalis. Penelitian ini merupakan penelitian eksperimental laboratorisdengan desain penelitian true experimental posttest only control group. Penelitian dilalukan denganmenguji ekstrak daun mimba (A.indica) pada konsentrasi 10%, 20%, 40%, 60%, 80% dan 100%terhadap pertumbuhan E. faecalis yang dilakukan di Laboratorium Mikrobiologi Fakultas KedokteranHewan (FKH) Universitas Syiah Kuala Banda Aceh. Hasil uji fitokimia yang dilakukan menunjukkanbahwa ekstrak daun mimba (A. indica) positif mengandung triterpenoid, phenolic compound, tanin,steroid dan saponin yang berperan sebagai antibakteri. Hasil uji efek antibakteri ekstrak daun mimba(A. indica) menunjukkan adanya pembentukan zona hambat di sekitar kertas cakram pada setiapkonsentrasi. Hal ini membuktikan bahwa ekstrak daun mimba memiliki efek antibakteri yangmemiliki kemampuan dalam menghambat pertumbuhan E. faecalis. Uji statistik digunakanmenggunakan one way ANOVA. Kemudian dilakukan uji lanjut (Post hoc test) menggunakan LeastSignificant Difference untuk menganalisis perbedaan efek antibakteri ekstrak daun mimba (A. indica)dalam konsentrasi 10%, 20%, 40%, 60%, 80% dan 100% terhadap pertumbuhan E.faecalis.Kesimpulan penelitian ini ekstrak daun mimba (A. indica) berpotensi menghambatpertumbuhan E. faecalis.Kata Kunci: Azadirachta indica, Enterococcus faecalis, perawatan saluran akar.

Introduction. Cypress (Cupressus sempervirens L.) belongs to the family Cupressaceae. It is evergreen, and grows in Mediterranean region. The Cypress leaves and young branches are utilized to produce the essential oil. Cypress oil contain a number of components, in it α-pinene, Δ3-carene, α-terpinyl acetate, cedrol, α-terpinolene, β-myrcene, limonene, α-terpineolene, terpinen-4-ol, β-pinene, δ-cadinene and sabinene. The oil is used in therapy different diseases. It to have antimicrobial activity. Aim. The aim of the date was evaluation the susceptibility of anaerobic bacteria to Cypress oil. Material and methods. The anaerobic bacteria were isolated from patients. The 62 microorganisms, in it 36 strains of Gram-negative rods, 14 Gram-positive cocci and 12 Gram-positive rods, and 7 reference strains were tested. Susceptibility (MIC) was determined by means of plate dilution technique in Brucella agar supplemented with 5% defibrynated sheep blood, menadione and hemin. The Cypress oil was dissolved in DMSO and distilled water to obtain final following concentrations: 2.5, 5.0, 7.5, 10.0, 15.0 and 20.0 mg/ml. Inoculum containing 106 CFU per 1 ml was seeded with Steers replicator upon the agar with oil or without the oil (strains growth control). The agar plates was incubated in anaerobic condition in anaerobic jar in 37°C for 48 hrs. The MIC was interpreted as the lowest concentration of Cypress oil inhibiting the growth of tested bacteria. Results. The results indicated that from among Gram-negative rods the most susceptible to Cypress oil was the strains from genus Tannerella forsythia (MIC < 2.5-5.0 mg/ml), Bacteroides uniformis (MIC = 5.0 mg/ml), Bacteroides vulgatus and Porphyromonas asaccharolytica (MIC 5.0-7.5 mg/ml) and Porphyromonas levii (MIC = 7.5 mg/ml). The strains from genera Fusobacterium and of Bacteroides fragilis were the susceptible to 2.5-≥ 20.0 mg/ml. The Cypress oil was least active towards Prevotella and Parabacteroides strains (MIC ≥ 20.0 mg/ml).The tested Gram-positive cocci were more susceptible. The growth of the strains were inhibited by concentrations in ranges ≤ 2.5-7.5 mg/ml. The oil was minor active towards Gram-positive rods (MIC ≤ 2.5-20.0 mg/ml). Among the strains the genus of Actinomyces odontolyticus (MIC = 5.0 mg/ml) and Actinomyces viscosus (MIC ≤ 2.5-7.5 mg/ml) were the most susceptible. The growth of rods of Bifidobacterium breve was inhibited by concentrations 10.0 mg/ml. The data indicates that the Gram-negative rods were the less susceptible than Gram-positive bacteria to cypress oil. Conclusions. Among Gram-negative rods the most susceptible were the strains Tannerella forsythia, Bacteroides uniformis, Bacteroides vulgatus, Porphyromonas asaccharolytica and Porphyromonas levii. The oil was more active against Gram-positive cocci. Gram-positive anaerobic bacteria demonstrate the more susceptible to Cypress oil then Gram-positive rods.

ABSTRACT Resistance of Gram-positive pathogens to first-line antimicrobial agents has been increasing in many parts of the world. We compared the in vitro activities of torezolid with those of other antimicrobial agents, including linezolid, against clinical isolates of major aerobic and anaerobic bacteria. Torezolid had an MIC90 of ≤0.5 μg/ml for the Gram-positive bacterial isolates tested and was more potent than either linezolid or vancomycin.

Objective: The aim is to estimate the effect of curcumin and rosemary as antibacterial agents among dental caries cases. Methods: Samples of saliva were randomly collected from 40 patients in Al-Furat General Hospital who attended the hospital from July to September 2018. Swabs were cultured on blood agar at 37°C for 24 h and then subcultured in mannitol salt agar and trypticase soy broth at 37°C for 24 h. Different concentrations of aqueous extract curcumin solution (0.1, 0.3, 0.5, and 1 mg/ml) and rosemary solution (1 g/ml) were prepared and added to the bacterial culture. Later, minimum inhibition zones of the bacterial cultures were determined. Results: The results showed that there were 25 cases of Streptococcus mutans, 10 cases of Staphylococcus aureus, 3 cases of anaerobic bacteria, and 2 cases of normal flora among 40 culturing swabs of bacteria. Aqueous extract of curcumin showed antibacterial effect with concentrations (0.1, 0.3, 0.5, and 1 mg/ml) against oral bacteria; nevertheless, these bacteria were resistant to the aqueous extract of rosemary with concentration 1 g/ml. Conclusion: It can be concluded that curcumin can be an effective antibacterial agent against dental caries disease and its effect increases positively in relation to its concentration. On the other hand, rosemary with 1 g/ml concentration did not show any effect on oral bacteria.

PURPOSE: The aim of this study was to evaluate, in vitro, the quality of calcium hydroxide [Ca(OH)2] paste filling (Ultracal, Ultradent) associated or not with Ca(OH)2-containing gutta-percha points (Calcium Hydroxide Plus PointsTM, Roeko) in curved root canals. MATERIAL AND METHODS: One hundred and twenty roots of extracted human teeth, randomly divided into three curvature ranges (mild - 0 to 14º; moderate - 15º to 29º; severe - >30º) were used. After chemomechanical preparation, the roots were assigned to 4 groups (n=30), according to the technique of intracanal dressing placement: group 1 - Ca(OH)2 paste was applied with a lentulo spiral; group 2 - Ca(OH)2 paste was applied with a lentulo spiral and a Ca(OH)2 point was inserted into the canal; group 3 - Ca(OH)2 paste was applied with a NavitipTM tip (supplied with Ultracal system); group 4 - Ca(OH)2 paste was applied with a NavitipTM tip and a Ca(OH)2 point was inserted into the canal. The roots were cleared and the quality of apical third filling was assessed by a calibrated experienced examiner. The specimens were examined under stereomicroscopy and scored 1 to 4 (i.e., from inadequate to complete root canal filling). The results were analyzed statistically by ANOVA and Duncan's post hoc test at 5% significance level. RESULTS: There were no statistically significant differences (p>0.05) among the curvature degrees in groups 1, 3 and 4. Severely curved roots in group 2 presented bordering significance (p=0.05). The groups that associated the use of Ca(OH)2 paste and points (2 and 4) showed better apical filling than the other groups, but this difference was statistically significant (p<0.001) only for roots with severe curvature. CONCLUSION: According to the results of this study, the curvature degree did not influence the quality of filling. The techniques that used Ca(OH)2-containing gutta-percha points yielded better filling of the apical third in roots with severe curvature.
