The radioprotective effects of an acute administration of the isoflavone genistein (4?, 5, 7-trihydroxyflavone), have been investigated in the present study. Male mice were administered with different doses (100, 200, 300, and 400 mg/kg body weight) of genistein 24 hours prior to 8 Gy gamma irradiation and the 200 mg/kg dose of genistein was determined to offer the maximum survivability and was used as an optimum dose for further experimentation. The 0.5 ml dose of genistein (200 mg/kg) was administered intraperitoneally to 2 different groups of mice, 15 minutes and 24 hours prior to gamma irradiation. In the mice treated with genistein with the optimum dose 24 hours before irradiation, a significant increase in 30 day survival has been recorded in contrast to the mice treated with genistein 15 minutes before the irradiation. The longer survivability (i. e. 20% for a period of more than 30 days) has been observed in the 24 hour group as compared to that of 15 minutes (i. e. 20% for 22 days). Although the radioprotective effect of genistein was evident in both groups, it was of greater magnitude in the group with a longer interval, indicating thereby an efficacy with longer retention with the possible minimum toxicity, unlike hitherto known other radioprotective agents.

Целью настоящей работы стало исследование эффектов лактоферрина (Лф) человека у мышей после острого гамма-облучения в сублетальной дозе. Методы. Исследование было проведено на 2-2,5-месячных самцах мышей линии C57Bl/6. Животные из экспериментальных групп были подвергнуты общему воздействию гамма-излучения в дозе 7,5 Гр. Сразу после облучения и повторно через 24 часа после него часть животных получила инъекцию Лф (внутрибрюшинно, 4 мг на животное). Было изучено влияние Лф на выживаемость и среднюю продолжительность жизни мышей. Для оценки общей двигательной и исследовательской активности использовали тест «Открытое поле». Результаты. Введение Лф позволило увеличить выживаемость и среднюю продолжительность жизни облученных мышей в течение эксперимента. Происходила более быстрая нормализация динамики изменения массы тела. Кроме того, Лф оказал компенсаторное действие на исследовательскую активность облученных животных. The aim of this study was to investigate effects of human lactoferrin (Lf) in mice exposed to acute gamma-irradiation at a sublethal dose. Methods. C57Вl/6 2-2.5-month-old male mice were used for the experiments. Animals from experimental groups were exposed to whole-body gamma-radiation at a dose of 7.5 Gy. Some animals received an intraperitoneal injection of Lf (4 mg per animal) immediately and then at 24 hours after the irradiation. The effect of Lf on survival rate and life span was studied. The open field test was used to assess locomotor and research activity. Results. The Lf administration increased the survival rate and life span of irradiated mice during the experiment. The dynamics of body weight normalized faster. In addition, Lf exerted a compensatory effect on the research activity of irradiated animals.

Backgrounds and objectives. The wide use of paracetamol at high doses was found to alter sperm parameters especially sperm morphology, and thus its fertilizing capability. Therefore, the present study was designed to use different doses of paracetamol to identify its effect on sperm parameters and testosterone levels in adult male mice. Methods. Forty adult male albino mice were divided into four equal groups, the first group injected with distilled water, the three treated groups injected with different doses of paracetamol (20, 40, 80 mg/kg body weight /day) over a period of 42 days. All doses were given once daily via intraperitoneal injection. Results. The results showed that paracetamol causes a significant decrease in body weight, non-significance effect on sperm parameters at doses of 20 and 40 mg/kg, while it led to a significant effect on sperm parameters at a dose of 80 mg/kg. Also, there was no difference in testosterone level between control and the treated groups (20 and 40mg/kg). But it showed a significant decrease in testosterone level at dose 80 mg/kg treated groups. Conclusion. It is considered safe to use paracetamol at doses 20 and 40 mg/kg but the dose 80 mg/kg has adverse effects on sperm parameters and testosterone level.

Chickpea (Cicer arietinum L.) is an important pulse crop grown and consumed all over the world. It is a good as well as the cheapest source of protein, soluble, insoluble fibers, vitamins, potassium and phosphorus. The present research was done to check the efficacy of gamma irradiation on chickpea’s proximate components and how the physical mutagen helped in Salmonella inactivation without any significant change in the proximate components of chickpea. The samples were treated with three different doses of gamma radiation (0.5, 1.0 and 2.0 kGy). Screening and evaluation of native micro flora on chickpea was performed and the viable counts of the microbes detected on samples of chickpea were Bacillus subtillis, Escherichia coli, Streptococcus pyogenes, Staphylococcus aureus and Salmonella sp. which were confirmed by biochemical test (API strips) before and after gamma irradiation. Results showed that 2 kGy is the optimum dose for chickpea at which complete elimination of Salmonella was recorded with no significant effect on sensory property as well as the proximate content of Cicer arietinum L.

Abstract Objectives Curcuma supplementation has been investigated to prevent or treat obesity. However, evidence suggests development of pancreatic steatosis as well. In this context, the aim of this study was to investigate the effect of different doses of curcumin supplementation on weight, biochemical profile, and histological analysis of the pancreas and liver of mice fed a high-fat diet. Methods This was an experimental, longitudinal and randomized study. Fifty C57BL/6 male mice, thirty days age, were separated into five groups: 1. Standard diet (n = 10); 2. High-fat diet (n = 10); 3. High-fat diet plus 50 mg of curcumin/kg of body weight (n = 10); 4. High-fat diet plus 250 mg of curcumin/kg of body weight (n = 10); 5. High-fat diet plus 500 mg of curcumin/kg of body weight (n = 10). Group 1 was fed a standard control diet (AIN 93 G), group 2 was fed a purified high-fat diet (AIN 93 HF 60%) and both groups received only the vehicle (carboxymethyl cellulose - CMC 1%) by gavage. Mice from groups 3, 4 and 5 were fed a purified high-fat diet (AIN 93 HF 60%) plus curcumin at different doses (50, 250 and 500 mg/kg of body weight diluted in 1% CMC) by gavage for twelve weeks. All groups received food and water ad libitum. At the end of the experimental period, we analysed lipid profile, blood glucose, insulin, histology of the pancreas and liver. ANOVA one way and Kruskal-Wallis analysis were performed and a value of P ˂ 0.05 was used to denote statistical significance. Results Curcumin supplementation did not improve weight and biochemical profile. Additionally, histological changes were not observed at any dose of supplementation. Pancreatic or hepatic steatosis was not evidenced in high-fat diet groups and also in the groups who received curcumin, suggesting no toxic effects at the different doses of supplementation provided. Conclusions Our results suggest that curcumin supplementation has no beneficial effect on weight gain prevention and biochemical profile, regardless of the dose administered. The aim of the study was to investigate the effects of Moringa oleifera aqueous seed extract on live body weight, gonadal and extragonadal dimensions and sperm reserves of Yankasa rams. Twenty five apparently healthy Yankasa rams aged 1-2 years and weighing 19.0 ± 2.1 Kg were used for the study. The rams were randomly selected into five groups: A, B, C, D and E with five rams in each group as treatment and control groups respectively. Groups A - D were given oral dose of Moringa oleifera aqueous seed extract at a dose rate of 1000, 2000, 3000, 4000 (mg/kg), respectively while group E was given 10 ml/kg water orally, daily for five months. Live body weight, gonadal and extragonadal reserves were determined according to standard techniques. The results showed a significant increase in live body weight in the months of April to June among rams treated with different doses of Moringa oleifera aqueous seed extract compared with the control group. The control group showed no significant differences in the body weight, gonadal and extragonadal dimensions and sperm reserves. In conclusion, the treatment of Yankasa rams with Moringa oleifera aqueous seed extract increased live body weight, but had no significant effects on gonadal and extragonadal dimensions and sperm reserves in Yankasa rams. Therefore, it is recommended that M. oleifera aqueous seed extract can be used at doses of 2000mg/kg to 3000mg/kg in Yankasa rams for optimum gain in live body weight.

20-HETE (20-Hydroxyeicosatetraenoic acid), is a cytochrome P450 (CYP) 4A-derived arachidonic acid metabolite. 20-HETE has been linked to both pro-hypertensive (via increased vasoconstriction, vascular remodeling and vascular injury of renal microvessels) and anti-hypertensive (inhibiting ion transport in the distal nephron) functions. In this study we examined the effect of 20-SOLA (2,5,8,11,14,17-hexaoxanonadecan-19-yl-20-hydroxyeicosa-6(Z),15(Z)-dienoate), a water soluble antagonist of the actions of 20-HETE on renal hemodynamics and sodium (Na) excretion in Cyp4a14 knockout (CYP4a14-/-) male mice. The CYP4a14-/- male mice display hypertension accompanied by increased vascular 20-HETE levels. Administration of 20-SOLA (10mg/kg/day in drinking water) normalized blood pressure (BP) in male Cyp4a14-/- mice at day 10 of treatment (124±1 vs. 153±2 mmHg in untreated male Cyp4a14-/- mice; p<0.05). The normalization of blood pressure was accompanied by transient increase in the urinary sodium excretion in the Cyp4a14-/- male mice (8.3±0.7 vs. 5.8±0.5 μmol/g body weight/day; p<0.05). Importantly, 20-SOLA increased glomerular filtration rate (GFR) of Cyp4a14-/- mice (2.38±0.05 vs. 1.88±0.18 μL/min/mg kidney weight, p<0.05) as opposed to no changes observed in the wild type (WT: (2.26±0.18 vs. 2.33±0.20μL/min/mg kidney weight). Evaluation of the renal blood flow (RBF) by laser Doppler flowmetry showed that treatment with 20-SOLA increased the RBF in Cyp4a14-/- mice by 12.3±4%, which remained unaltered in the WT. Additionally, the pressure-induced myogenic tone of isolated preglomerular microvessels was significantly elevated in Cyp4a14-/- mice; 20-SOLA treatment prevented the increase in myogenic responses. The natriuretic response to an isotonic saline loading challenge (10% of body weight, IP) was significantly attenuated in the Cyp4a14-/- mice as compared to the WT (35.5±2.8 vs. 57.4±8.3 percentage of Na load, p<0.05); this was corrected by 20-SOLA (61.7±5.7 percentage of Na load, p<0.05). These results confirm that 20-SOLA normalizes blood pressure of Cyp4a14-/- male mice and demonstrates that this is associated with increases in GFR, RBF and natriuresis.

Overweight/obesity has been suggested as a risk factor for asthma development, and prospective studies have confirmed that high body weight precedes asthma symptoms. However, the nature of the association between overweight/obese status and asthma remains unclear. Animal models of obesity-related asthma are very useful for understanding disease pathophysiology. Although C57/B6J mice are the most widely used animal model for researching obesity-related asthma, gender differences are not always taken into consideration. Therefore, to explore the effect of gender on the development of obesity-related asthma, both female and male C57/B6J mice were used in this study. The mice were fed with a high-fat diet or a low-fat diet as control. Body weight, body length, liver weight, and Lee’s Index were used to evaluate obesity status, and lung histology, lung inflammatory cells infiltration, and inflammatory cytokines in bronchoalveolar lavage fluid (BALF) were examined for asthma evaluation. We found that the mean body weight of male mice on a high-fat diet gradually increased and was significantly higher than control male mice on a low-fat diet ( P < 0.01), while no significant differences were found between female mice at the end of 12 weeks of feeding. Furthermore, the obese asthma group female and male mice exhibited significantly high inflammatory cells infiltration than normal weight or obese female and male mice ( P < 0.01). However, the obese asthma group presented higher Neu infiltration, Th1 cytokine, and interferon gamma (IFNγ) concentrations in BALF than the asthma group in both the genders ( P < 0.01). In conclusion, both female and male mice are suitable for the obesity-related asthma model, although male mice might be more stable. Besides, obesity-related asthma is not Th2 type asthma.
