The objective of the present study was to determine genetic diversity among three geese color types using RAPD markers and hematological parameters. The overall mean, of the live weights, Hemoglobin, Hetrophil %, Lymphocyte %, Monocyte %, and H/L ratio were 3.006 (kg), 14.64, 36.896, 49.896, 2.233 and 0.736 respectively. The breed, sex and interaction between them have a significant effect on live body weight, Hemoglobin, and Monocyte %. Ten primers were used and six out of them were selected based on their number of bands (NB) and polymorphic characteristics. A total of 309 bands observed, ranged from 30 in primer OPB-07 to 54 bands in OPA-20. Five unique bands were found only in white goose, whereas the highest unique band was obtained in primer OPB-01 locus. Overall genetic distance among native geese arrived 64.122 and phylogenetic dendrograms showed that 3 clusters, the first cluster content only white geese (Male and Female) breed, the second one cluster is included piebald geese breed (Male and female) and the third one was including gray geese (Male and female) breed. It was concluded that the white geese was closer to piebald geese than to the gray geese breed. The high genetic distance (64.122%) and variation in phenotypic value such as live weight (2.375 to 3.600 kg/bird) for three native geese indicate that these native geese have a good amount of genetic resources to made genetically improvement in further and it means the three goose samples are independent breeds.

The aim of the study is to increase the morphological and functional status of cows depending on their physiologi-cal state. The morphological composition of blood is one of the main informative systems and has long attracted the attention of researchers. Blood, as a co-single tissue, has been studied well enough. Its indicators are the main fac-tors in determining the state of an animal health, as well as pointing out the presence of pathological processes in inside the organism. To determine the effect of complex use of STEMB (embryonic stimulator) and Uteromastin on the blood parameters of cows in comparison with individual administration three groups of pregnant cows (7.0-8.0 months) were formed separately, each with 20 heads in compliance with the method of groups-analogues (experimental-1, experimental-2, experimental-3). Animals in groups to be examined were provided the same con-ditions both in feeding and keeping. Animals of the first experimental group 25-30 days before calving were admin-istered STEMB drug hypodermic in the neck at a dose of 0.075 ml per 1 kg of live weight with an interval of 7 days three times, animals of the second experimental group were administered the drug Uteromastin 8-10 hours after calving intrauterine at a dose of 150 ml once. Cows of the third experimental group were administered drugs STEMB and Uteromastin in these doses (mixed). The influence of the complex use of drugs STEMB and Utero-mastin on morphological, biochemical and immunobiological parameters of blood of cows were studied on the 15th day after calving. The use of the drugs STEMB and Pteromalid complex improves cellular composition of the blood and indices of the blood serum, which enhances immunotoxicol system of organism of cows.

RAPD technique was used as a tool for assessing genetic diversity and varietal relationships among ten varieties of eggplant. Out of 21 primers screened four were selected. With these primers 76 clear and bright fragments were obtained of which 44 fragments considered polymorphic. The proportion of polymorphic loci and gene diversity values across all loci were 57.89% and 0.23, respectively. The UPGMA dendrogram based on genetic distance segregated the ten varieties of eggplant into two main clusters. Dohazari, Kazla, Nayantara and ISD-006 were grouped together in cluster I whereas Uttara, Islampuri, Khatkhatia, Singnath, BARI Begun-08 and Eggplant Line-083 into cluster II. Kazla and Nayantara variety pair was very close to each other with the highest intervarietal similarity index (92.54%) and lowest genetic distance (0.14). On the other hand, Khatkhatia and Nayantara pair was the lowest intervarietal similarity index (41.67%) with highest genetic distance (0.48). Therefore, identification of genetically distinct varieties using RAPD markers could be a potential tool for eggplant improvement. Key words: Eggplant, Polymorphism, Genetic relationship, RAPD D.O.I. 10.3329/ptcb.v19i2.5006 Plant Tissue Cult. & Biotech. 19(2): 119-126, 2009 (December)

This investigation was aimed at exploring the genetic diversity and relationship among nine Brassica varieties, namely BARI Sharisha-12, Agrani, Sampad, BINA Sharisha-4, BINA Sharisha-5, BARI Sharisha-13, Daulot, Rai-5, Alboglabra using Random Amplified Polymorphic DNA (RAPD) markers. In total, 59 reproducible DNA bands were generated by four arbitrary selected primers of which 58 (98.03%) bands were proved to be polymorphic. These bands ranged from 212 to 30686 bp in size. The highest proportion of polymorphic loci and gene diversity values were 37.29% and 0.1373, respectively, for BARI Sharisha-12 and the lowest proportion of polymorphic loci and gene diversity values were 8.47% and 0.0318, 8.47% and 0.0382 for BINA Sharisha-4 and Rai-5, respectively. A dendrogram was constructed using unweighted pair group method of arithmetic mean (UPGMA). The result of cluster analysis indicated that the 9 accessions were capable of being classified into 2 major groups. One group consists of BARI Sharisha-12, Agrani, Sampad, Daulot, Rai-5, Alboglabra. where Daulot and Rai-5 showed the lowest genetic distance of 0.049. And another group contains BINA Sharisha-4, BINA Sharisha-5, and BARI Sharisha-1 3, where BINA Sharisha-5 and BARI sharisha-13 showed genetic distance of 0.071. Genetic diversity in 31 traditional Bangladeshi Aus rice genotypes were studied under transplanted condition through Mahalanobis D2 statistic for grain yield and yield contributing characters. The genotypes were grouped into five clusters. The inter-cluster distances were higher than intra-cluster distances indicating wider genetic diversity among the genotypes of different clusters. The intra-cluster distances were lower in all the cases reflecting homogeneity of the genotypes within the clusters. The cluster II contained the highest number of genotypes (08) and the cluster I and III contained the lowest (05). The highest intra-cluster distance was noticed for the cluster II and the lowest for cluster IV. The highest inter-cluster distance was observed between cluster II and V followed by cluster III and V, cluster I and V and the lowest between cluster I and III. Regarding inter-cluster distance, the genotypes of cluster V showed high genetic distance from all other clusters. The genotypes from cluster V could be hybridized with the genotypes of other characters of other clusters for producing transgressive segregants. The highest cluster means for yield, effective tiller number and grain length, were obtained from cluster I; whereas the lowest mean value for yield, culm length, plant height and grain breadth were found in cluster II. Therefore, genotypes under cluster I, cluster II and cluster V might be selected for future breeding program as parents for crossing to produce new recombinants with desired traits.Bangladesh J. Twenty six rice lines of PBRC (salt tolerant line-20) × BRRI dhan-29 were used to evaluate salinity tolerance at the seedling stage and tested for salt tolerance using RAPD markers. Salinity screening was done using hydrophonic system at the greenhouse following IRRI standard protocol. Among the studied line, ten were moderately salinity tolerant, nine susceptible and rest of the lines highly susceptible. For assessing genetic diversity and relationship of F3 rice lines including two parents were tested against PCR-based Random Amplified Polymorphic DNA (RAPD) technique using three arbitrary decamer primers; OPA02, OPC01, and OPC12. Selected three primers generated a total of 14 bands. Out of 14 bands, 12 bands (86.67%) were polymorphic and 2 bands (13.33%) were monomorphic. The Unweighted Pair Group Method of Arithmetic Means (UPGMA) dendrogram constructed from Nei’s (1972) genetic distance produced 2 main clusters of the 28 rice genotypes. Most of the moderately tolerant lines and PBRC (STL-20) (tolerant variety) were grouped in same cluster due to lower genetic distance, while maximum susceptible along with BRRI dhan29 (susceptible variety) showed higher genetic distance with PBRC (STL-20) and moderately tolerant lines. This result indicates that the lines which formed grouped together, they are less diversed. On the other hand the lines remain in different clusters or different groups, are much diversed. Thus RAPD perform a potentially simple, rapid and reliable method to evaluate genetic diversity and molecular characterization as well. Res. Agric., Livest. Fish.6(2): 215-225, August 2019

The present study included detection of genetic variability, and identification of genetic relationship and finding a fingerprint of the ten clinical isolates related to Trichophyton rubrum using RAPD and ISSR markers. The experiment was carried out and the results performed using six primer of the RAPD markers  these primers showed 239 amplified  band , out of  these band 90 of them was considered as a main band, and 149 was Polymorphic band , the largest number of bands was 30 in the isolate TR6  and less number of bands in the isolate TR7.   The results clear the value of genetic diversity based on RAPD analysis the lowest value of genetic diversity (0.13005) between the isolate TR3 and TR9while the highest genetic diversity (0.55941) was between the isolates TR5 and TR8. The analysis of the relationship shows that there are three main groups: the first group include isolate TR8, while the second group included three isolates are isolate TR2, isolate TR10 and isolates TR3, The third group included three subgroups included the first isolate TR1,isolates TR4 and second isolate TR3 and isolate TR9 and the third included the isolate TR5 and isolates TR6.The results of ISSR experiments: the use six a primers in the of the ISSR showed 192 bands, in the isolate  of Trichophyton rubrum, two of these primers showed monomorphic bands (in number and location) and six primers showed monomorphic and polymorphic bands, while one showed only polymorphic bands among Trichophyton rubrum isolates. And the largest number of bands was 24 in the TR5 and less number of bands 16 in isolate TR3 and isolate TR8 were finding DNA fingerprint to isolate TR1, isolate TR5, and isolate TR3.    The ISSR markers showed lowest genetic polymorphism was (0.05561) between the isolates TR2 and isolate TR7 and the largest genetic distance was (0.40501) between the isolate TR4 and isolate TR8. The analysis of the relationship of genetic showed that there are three groups key first included isolates and only one isolate TR8 and the second involving isolates TR2 and isolate TR7, isolate TR4 and isolate TR3, while the group included three sub-groups, the first included isolate TR1 and isolate TR6, and the second involving isolate TR5, isolate TR10 and isolate TR9. The results confirmed the efficiency markers of the RAPD and ISSR in contrast, find a genetic fingerprint to the isolates of Trichophyton rubrum, and these markers differ in mechanical detection contrast, and coverage is Genome. Therefore Complementary to each other, although the ISSR markers were more efficient in terms of the number of binding  sites in each type of the isolates of Trichophyton rubrum and so can the initiator of the discovery of a much larger area from  Genome.

Genetic variability among the genotypes of any species could be utilized for its improvement. PCR-based Random Amplified Polymorphic DNA (RAPD) technique was used to determine the genetic diversity and relationship among 10 cucumber varieties and genotypes. Five decamer primers were used to amplify genomic DNA and the primers yielded a total of 54 bands of which 36 bands were polymorphic and 18 bands were monomorphic. The UPGMA dendrogram based on Nei’s (1972) genetic distance indicated segregation of 10 cucumber varieties and genotypes into two main clusters. Variety Joti alone grouped in cluster 1 while variety Green Master, Shahi-50, Shikha, Shila, Shital, Naogaon-5, Shohag-50, Giant Long and genotype CS-043 grouped in cluster 2. Variety Shila was very close to variety Shital with the least genetic distance (0.1712). The highest genetic distance (0.5352) was found between Joti and Naogaon-5. Random amplified polymorphic DNA (RAPD) markers are widely used for evaluating the genetic relationship of crop germplasm. Five different landraces of yacon (Smallantus sonchifolius (Poepp. and Hendl.) H. Robinson; Asteraceae) collected in various countries and showing different morphological traits were investigated using a total of 61 decamer primers. A total of 282 RAPD markers were scored and 28.7% of them were polymorphic at least within landraces. RAPD markers generated by one primer (OBP14) discriminated between all landraces. Markers were used to calculate genetic similarity coefficient and to build a dendrogram representing the genetic relationship between analysed landraces. The results suggest that RAPD markers could be used as a reliable tool to perform fingerprinting studies in Smallantus sonchifolius genome. This is the first report on the use of RAPDs to evaluate genetic distance and to distinguish between different landraces in yacon.
