AbstractGrapevine trunk diseases (GTD) have a variety of symptoms and causes. The latter include fungal species from the family Diaporthaceae. The aim of our study was to determine Diaporthaceae species present in the woody parts of grapevines sampled from 12 vine-growing coastal and continental areas of Croatia. The fungi were isolated from diseased wood, and cultures analysed for phenotype (morphology and pathogenicity) and DNA sequence (ITS1, 5.8S, ITS2). Most isolates were identified as Phomopsis viticola, followed by Diaporthe neotheicola and Diaporthe eres. This is the first report of Diaporthe eres as a pathogen on grapevine in the world, while for Diaporthe neotheicola this is the first report in Croatia. Pathogenicity trials confirmed Phomopsis viticola as a strong and Diaporthe neotheicola as a weak pathogen. Diaporthe eres turned out to be a moderate pathogen, which implies that the species could have a more important role in the aetiology of GTD.

Anise (Pimpinella anisum L.) is an important medicinal spice plant that belongs to the family Apiaceae. Anise seeds are rich in essential oils and this is a reason why anise production in Serbia has increased over the last decade. During a routine health inspection on anise seeds collected from three localities in the province of Vojvodina (Mošorin, Veliki Radinci and Ostojićevo) during 2012 and 2013, it was found out that Fusarium spp. were a commonly observed fungi. The presence of Fusarium fungion the seed samples ranged from 3.75-13.75%. The aim of this study was to isolate and identify the strains of Fusarium species present on anise seed samples as it is necessary that commercially used anise seeds are completely free of Fusarium. Based on morphological, microscopic characteristics and a molecular identification by sequencing of TEF gene, the presence of the following species was confirmed on the anise seeds: F. tricinctum, F. proliferatum, F. equiseti, F. oxysporum, F. sporotrichoides, F. incarnatum and F. verticillioides. According to our knowledge and research, this is the first report of F. tricinctum and F. sporotrichoides as pathogens on anise seeds in the world. All seven isolates of Fusarium species are pathogenic to the anise seedlings, while the most virulent species were F. oxysporum, F. tricinctum and F. incarnatum.

In September 2010, during survey of diseased grapevines (Vitis vinifera L.) in vineyards at localities Zmajevac (BZ), Orahovica (SO), Cilipi (KC), and Novalja (PN), symptoms characteristic of grapevine trunk diseases (GTD) (3) were observed, showing on cross-sectioned cordons and trunks as brown, wedge-shaped perennial cankers and/or dark streaking of the wood. In Croatia, these symptoms were traditionally associated with Eutypa Tul. & C.Tul. and with fungi from Diaporthaceae (2). From affected grapevines (cvs. Grasevina, Pinot bijeli, Malvazija dubrovacka, and Gegic), samples of symptomatic cordons and trunks were collected (n ≥ 35). To isolate the causal agents from the samples, woodchips of symptomatic tissue, surface-sterilized in 2% sodium hypochlorite for 2 min, were placed on potato dextrose agar amended with streptomycin sulphate (50 μg/ml) and incubated for 7 days at 25°C in darkness. A percentage of samples (72, 15, 27, and 54% from BZ, SO, KC, and PN, respectively) yielded fungal colonies with abundant aerial mycelium, initially white, but turning olivaceous grey after 5 days. From these colonies, monohyphal isolates were obtained and pycnidial formation stimulated by cultivation on 2% water agar with stems of plant species Foeniculum vulgare Mill. at 25°C under diffuse light for 3 weeks. Pycnidia contained conidia that were hyaline, unicellular, ellipsoid with round apices and truncated bases, and thin walled with smooth surface. Dimensions of conidia (n ≥ 50) were (12.8) 15.3 ± 1.4 (17.6) × (5.4) 6.3 ± 0.8 (7.6) μm, with length/width ratio (2.0) 2.5 ± 0.5 (3.2). Based on morphological data, species Neofusicoccum parvum (Pennycook & Samuels) Crous, Slippers & A.J.L. Phillips was suspected (1). For molecular identification, isolates BZ330, SO334, KC342, and PN121 were used for PCR to amplify internal transcribed spacer region and partial translation elongation factor 1-alpha gene, using primers ITS5/ITS4 and EF1-728F/EF1-986R, respectively. Obtained sequences were shown to be identical between the four isolates (GenBank: KF296318, KF296319) and when compared with sequences for reference N. parvum isolate CMW9080 (AY236942, AY236887) they showed >99% homology, confirming the isolates as species N. parvum. Pathogenicity tests were done by inoculation of detached green shoots (GS) and lignified canes (LC) (n = 5) of grapevine cv. Skrlet by either mycelial plugs of the same four isolates, or sterile agar plugs for the controls. Inoculated GS were kept in flasks with sterile water in a glasshouse for 10 days, and LC in humid dark chambers for 30 days, at 25°C. Resulting vascular necrosis measured 62 to 81 mm (GS) and 215 to 246 mm (LC), but was absent on controls. Koch's postulates were satisfied by successful reisolation of N. parvum only from plants inoculated with mycelial plugs. N. parvum has been recognized as a serious grapevine pathogen, causing similar symptoms worldwide (3). To our knowledge, this is the first report of N. parvum associated with GTD in Croatia, and due to its relatively high incidence at surveyed localities, it could present considerable threat, particularly for neighboring vine growing regions. Abstract This paper describes a trematode isolated from the air sacs of a common blackbird (Turdus merula) by the authors in Qaemshahr city, Mazandaran province, north of Iran. The specimens were in the best form to make whole-mounts for species identification. The identified species was Morishitium vagum (Morishita 1924) Witenberg 1928, belonging to the family Cyclocoelidae Stossich 1902, according to morphologic and morphometric characters, as explained in references. This is the first report of a species belonging to the family Cyclocoelidae in Iran, and the first report of this type-species isolated from Turdus merula in the world.

Grapevine trunk diseases (GTD) are caused by several fungal species and are major limiting factors to vineyard productivity and profitability in all viticulture areas. This study is aimed at addressing the gap in the knowledge with regards to measuring the long-term benefits of post-pruning fungicide application on trunk diseases incidence and crop yield in grape production systems. It also calculated the net economic benefit of implementing such practice over the vineyard lifespan. We selected a newly planted commercial table grape vineyard in the California desert and divided it in two blocks. In one block, the registered fungicide thiophanate-methyl was mechanically applied on pruning wounds for six consecutive years, while the other half remained untreated. Our results showed a significant lower GTD incidence and vine replants in treated blocks combined with a significant increase of total and marketable fruit. Potential annual economic benefits of applying fungicide on pruning wounds appear to be in the range of $8500–$12,500 per hectare annually in a 50–75% disease control scenario.

In this work, the chemical composition of Rubia tinctorum root hydromethanolic extract was analyzed by GC–MS, and over 50 constituents were identified. The main phytochemicals were alizarin-related anthraquinones and flavoring phenol compounds. The antifungal activity of this extract, alone and in combination with chitosan oligomers (COS) or with stevioside, was evaluated against the pathogenic taxa Diplodia seriata, Dothiorella viticola and Neofusicoccum parvum, responsible for the so-called Botryosphaeria dieback of grapevine. In vitro mycelial growth inhibition tests showed remarkable activity for the pure extract, with EC50 and EC90 values as low as 66 and 88 μg·mL−1, respectively. Nonetheless, enhanced activity was attained upon the formation of conjugate complexes with COS or with stevioside, with synergy factors of up to 5.4 and 3.3, respectively, resulting in EC50 and EC90 values as low as 22 and 56 μg·mL−1, respectively. The conjugate with the best performance (COS–R. tinctorum extract) was then assayed ex situ on autoclaved grapevine wood against D. seriata, confirming its antifungal behavior on this plant material. Finally, the same conjugate was evaluated in greenhouse assays on grafted grapevine plants artificially inoculated with the three aforementioned fungal species, resulting in a significant reduction in the infection rate in all cases. This natural antifungal compound represents a promising alternative for developing sustainable control methods against grapevine trunk diseases.

Dark discolorations of the vascular vessels of 3-year-old potted plants of Asturian grapevine cultivar (Northern Spain), were observed during pruning. These symptoms can be associated to fungal trunk diseases that, in the last decades, are affecting young vineyards all over the world. Cross sections from root, trunk and canes of 19 young grapevine plants were analyzed for the presence of pathogenic fungi associated with these diseases. Non-pathogenic fungi were isolated from both asymptomatic and symptomatic samples, showing that dark discolorations, in some cases, were a consequence of abiotic causes. Regarding pathogenic fungi, Cylindrocarpon spp. colonies were the most frequent, isolated mainly from roots and from asymptomatic tissue. Botryosphaeria spp. colonies were mostly isolated from trunk and from sections with dark discolorations generated by pruning. Phaeoacremonium spp. was isolated from all the organs (roots, trunk and canes). Only one colony of Libertella spp. was isolated. These results suggest the need of a standard protocol, combining treatments and management activities, to be performed in nurseries to limit the spread of these diseases.

Abstract Grapevine trunk diseases (GTDs) are destructive and important economically with worldwide distribution. In this survey 233 fungal isolates were obtained from grapevine cultivars showing trunk diseases symptoms in Kurdistan Province, Iran. Based on sequences data and morphology 24 species belong to 20 genera were characterized. Botryosphaeriaceae, Alternaria, Sporocadaceae and Phaeoacremonium members were the most prevalent identified fungal groups. At the species level Botryosphaeria dothidea, Alternaria malorum, Phaeoacremonium aleophilum and Acremonium sclerotigenum were the most frequent identified species. All species are new records in Kurdistan Province. Clonostachys rosea and Neoscytalidium novaehollandiae are new records on grapevine in Iran. Acremonium sclerotigenum, Alternaria chlamydosporigena, Ascochyta herbicola and Paecilomyces formosus are new records on grapevine around the world. In phylogenetic analyses based on LSU, ITS, TEF-1α and TUB2 sequence data four pestalotioid species belong to Sporocadaceae were identified. Of these, three species are new for science and introduced here as Seimatosporium marivanicum, Sporocadus kurdistani and Xenoseimatosporium kurdistanicum. Furthermore, three new combinations in Sporocadus are proposed.

Morphological characters of spores of <em>Acaulospora rehmii</em> and <em>Gigaspora margarita</em> (Glomeromycota) were described and illustrated. Spores of the two species were found in field-collected mixtures of rhizosphere soil and roots collected in Poland. Attempts to produce spores in trap cultures succeeded only with <em>G. margarita</em>. All attempts to establish one-species cultures of the two fungi failed. <em>Gigaspora margarita</em> was for the first time found in Poland and this paper is the first report of the occurrence of <em>A. rehmii</em> in Europe. The known distribution of the two fungal species in the world is also presented.
