Abstract The aim of study was to evaluate different methods for subclinical endometritis (SCE) diagnoses and their accuracy to predict pregnancy. The study was performed on 51 postpartum cross breed dairy cows with pathological puerperium. Artificial insemination (AI) was done in spontaneous estrus. Endometrium samples were taken by cytotape (CT), cervical mucus (CM) samples by Metricheck and smears for crystallization degree were obtained at AI day. Blood serum samples for progesterone (P4) concentration determination were taken at AI day and 17 days later. Cows were examined by ultrasound 35 days after AI to confirm pregnancy. Percent (%) of polymorphnuclear leukocytes (PMNs) taken by CT were expressed by ROC curve (with cut-off point set at 1%). Out of the total of 51 cows involved in the study, 13.73% true positive and 41.17% true negative cows were found (54.90%). The content of CM sampled with Metricheck was altered in 11 cows (21.57%), and one of them was pregnant (1.96%). An average macroscopic CM value for pregnant cows was 0.06±0.24 vs. 0.39±0.79 for non-pregnant (p<0.05). An average value of CM crystallization in pregnant cows was 2.50±1.04 vs. 1.85±1.15 in non-pregnant (p<0.05). P4 concentration was not statistically different at AI day, but was statistically significantly higher 17 days later in pregnant cows (p = 0.002). CT alterations may be a reliable diagnostic tool for SCE diagnosis, while macroscopic CM alterations may be used for CE detection. P4 determination is beneficial in pregnancy diagnosis while CM crystallization degree is helpful in determination of best time for AI.

The objective of this study was to determine thiol/disulphide homeostasis (TDH) in infertile cows with subclinical endometritis (SCE). Endometrial cytological samples were collected using a cytobrush to diagnose SCE in 36 infertile cows. According to the results of the cytology examination, those with acute endometritis were classified as Group I (n = 20) and those with chronic endometritis were classified as Group II (n = 16). A control group was formed of heifers as Group III (n = 20). Blood samples were taken from each group on the day of diagnosis (day 0) to analyse TDH. In the cytology examination, both the Giemsa method and immunocytochemical staining were applied to determine chronic inflammation and activity status. In 55.55% (20/36) of the infertile cows with cytological endometritis, the inflammation was determined to be active, and in 44.44% (16/36) it had become chronic. The native thiol and total thiol levels were found to be statistically significantly lower in the acute (206.54 ± 8.30 μmol/L; 227.11 ± 9.30 μmol/L) and chronic SCE cases (225.15 ± 11.89 μmol/L; 247.96 ± 10.80 μmol/L) compared to the heathy control group (308.47 ± 13.59 μmol/L; 336.83 ± 15.5 μmol/L respectively) (P<0.001). Disulphide levels, disulphide/total thiol, native thiol/total thiol and disulphide/native thiol ratios were similar in all the groups (P>0.05). The diagnostic accuracy of native thiol, which can be used in the diagnosis of SCE, was 92.8%, that of total thiol was 89.3% and that of disulphide was 64.3% according to the ROC curve analysis. These results demonstrate that TDH is a reliable and sensitive indicator of oxidative stress in cow SCE, and that abnormal TDH might play a role in SCE pathogenic mechanisms. This is the first study to evaluate thiol/disulphide homeostasis in dairy cows with SCE as a new indicator of oxidative stress.

The action of the bovine placental lactogen (bPL) hormone on maternal metabolism is still poorly known. Some markers, such as the acute phase protein paraoxonase (PON1), are used as indicators of liver function and help to determine the metabolic condition during the transition period in dairy cows. The aim of this study was to evaluate the activity of paraoxonase (PON1) in the serum of peripartum dairy cows with different levels of bPL. Based on the plasma bPL concentration, 18 cows were divided equally into three groups: LOW ( < 2,68 ng bPL mL-1), MEDIUM (2,68–2,80 ng bPL mL-1), and HIGH ( > 2,80 ng bPL mL-1). The experiment was conducted between 21 days prepartum and 28 days postpartum. Serum samples were collected during the experiment for the determination of bPL concentrations and PON1 activity. The bPL concentration was significantly different between the experimental groups (P ? 0,0001) and the days of serum collection (P ? 0,0001). In the prepartum dairy cows, the PON1 levels were different between the groups (P ? 0,05) and the days of serum collection (P ? 0,05). Cows with high bPL concentration had lower serum PON1 activity (P ? 0,05), while cows with low hormone levels had higher enzyme activity (P ? 0,05). In the postpartum period, there was a significant difference between the days of serum collection (P ? 0,0001) and the interaction between groups and collections (P ? 0,01). The group with high concentrations of bPL had lower levels of PON1 (P ? 0,01), while the group with low bPL maintained higher concentrations of PON1 (P ? 0,01). It was concluded that the cows with higher concentrations of bPL in the prepartum period present a reduction in the serum activity of the PON1 enzyme during the peripartum period.

The technique of Z-contrast in STEM offers the possibility to determine the local concentration of macromolecules like lipids, proteins or DNA. Contrast formation depends on the atomic composition of the particular structure. In the case of DNA, its phosphorous content discriminates it from other biological macromolecules. In our studies, sections of E. coli, the dinoflagellate Amphidinium carterae and Euglena spec. cells were used which were obtained by cryofixation followed by freeze-substitution into acetone with 3% glutaraldehyde. The samples were then embedded either in Lowicryl HM20 at low temperature or in Epon at high temperature. Sections were coated on both sides with 30Å carbon.The DF- and the inelastic image have been recorded simultaneously with a Cryo-STEM. This Cryo-STEM is equipped with a highly dispersive Electron Energy Loss Spectrometer. With this instrument pure Z-contrast can be achieved either with a Filtered DF-image divided by the inelastic image or, as is used in this paper, by dividing the conventional DF-image by an inelastic image which has been recorded with an inelastic detector whose response is dependent on the total energy loss of the inelastically scattered electrons.

Objectives: Hysterosalpingography and laparoscopy both are the diagnostic methods for assessment of female infertility.  The present study was to compare the evaluation of hysterosalpingography (HSG) versus laparoscopy in determination of tubal factors in female infertility. Methods: Detailed assessment, physical examination and clinical investigations were performed in all 100 infertile female with age 20 years to 40 years. All patients were advised to perform digital HSG. Patients with an abnormal HSG underwent laparoscopy without delay, whereas in patients with a normal HSG, laparoscopy was performed three months after HSG. HSG is best scheduled during the 2nd -5th day interval immediately following the end of menstruation, to minimize risk for infection, avoid interference from intrauterine blood and clot, and to prevent any possibility that the procedure might be performed after conception. Results: Data was analysed by using IBM SPSS version 23 software.  All data was tabulated and percentages were calculated. Mean ± standard deviation was observed. Conclusions: Diagnostic laparoscopy is the gold standard in diagnosing tubal pathology and other intra-abdominal causes of infertility. Other hand, Hysterosalpingography is a frequently utilized diagnostic tool in the assessment of tubal status and detection of uterine anatomical defects in infertility. Hysterosalpingography and laparoscopy are not alternatives but complimentary investigations. But, inadequacy of hysterosalpingography (HSG) in determining the state of tubal patency, emphasizes the need for laparoscopy. Laparoscopy provides both a panoramic view of the pelvic reproductive anatomy and a magnified view of pelvic organs and peritoneal surfaces. Pain is defined as a disagreeable sensory and emotional experience related to a tissue or potential lesion. Paracetamol (Acetaminophen) is the most used non-morphine analgesic. For the determination of paracetamol we developed and validated the high performance liquid chromatography (HPLC) analysis using a Dionex Ultimate 3000 liquid chromatograph equipped with a multidimensional detector. After determining the optimum conditions of analysis (80/20 water / acetonitrile mobile phase, flow rate 1.0 mL / min, detection wavelength 245 nm) we validated the method following the following parameters: linearity of response function, linearity of results, limit (LD = 0.66 mg / mL) and quantification limit (LQ = 2.00 mg / mL), and precision. The method of determining paracetamol by HPLC was applied to 30 samples of serum collected from patients who had pain and were treated with paracetamol.

Abstract Raceophenidol in feed at concentration levels intended for growth promotion of poultry can be estimated by a curvature inversion measurement related to the ultraviolet spectrum of derived p-methylsulfonylbenzaldehyde. The drug can be accurately measured at the 0.0005% level. Collaborative studies on the method showed an average value of 94 ± 10% of claim. The method is recommended for adoption as official, first action.

Background/Aim. The vertical dimension of occlusion is a very important parameter for proper reconstruction of the relationship between the jaws. The literature describes many methods for its finding, from the simple, easily applicable clinically, to quite complicated, with the use of one or more devices for determination. The aim of this study was to examine the possibility of determining the vertical dimension of occlusion using the vocals ?O? and ?E? with the control of values o btained by applying cognitive functions. Methods. This investigation was performed with the two groups of patients. The first group consisted of 50 females and 50 males, aged 18 to 30 years. In this group the distance between the reference points (on top of the nose and chin) was measured in the position of the mandible in the vertical dimension of occlusion, the vertical dimension at rest and the pronunciation of the words ?OLO? and ?ELE?. Checking the correctness of the particular value for the word ?OLO? was also performed by the phonetic method with the application of cognitive exercises when the patients counted from 89 to 80. The obtained difference in the average values i n determining the vertical dimension of occlusion and the ?OLO? and ?ELE? in the first group was used as the reference for determining the vertical dimension of occlusion in the second group of patients. The second group comprised of 31 edentulous persons (14 females and 17 males), aged from 54 to 85 years who had been made a complete denture. Results. The average value obtained for the vertical dimension of rest for the entire sample was 2.16 mm, for the word ?OLO? for the entire sample was 5.51 mm and for the word ?ELE? for the entire sample was 7.47 mm. There was no statistically significant difference between the genders for the value of the vertical dimension at rest, ?ELE? and ?OLO?. There was a statistically significant difference between the values f or the vertical dimension at rest, ?OLO? and ?ELE? for both genders. There was a statistically significant correlation between the value for the vertical dimension at rest, ?OLO? and ?ELE?, for both groups of subjects. Conclusion. Determining the vertical dimension of occlusion requires 5.5 mm subtraction from the position of the mandible in pronunciation of the word ?OLO? or 7.5 mm in pronunciation of the word ?ELE?.

Bovine leukemia virus (BLV) is a retroviral infection that disrupts the immune function of infected animals. It is widespread among U.S. dairy cattle. In this pilot study, the average total IgA and IgM concentrations in milk, saliva, and serum samples from BLV ELISA-positive (ELISA+) dairy cows were compared against samples from BLV ELISA-negative (ELISA−) cows using the Kruskal–Wallis test (with ties). The results from ELISA+ cows were also stratified by lymphocyte count (LC) and proviral load (PVL). In milk and saliva from ELISA+ cows, the average total IgA and IgM concentrations were decreased compared to ELISA− cows, although this was only statistically significant for saliva IgM in cows with low PVL (p = 0.0424). Numerically, the average total IgA concentrations were 33.6% lower in milk and 23.7% lower in saliva, and the average total IgM concentrations were 42.4% lower in milk and 15.5% lower in saliva. No significant differences were observed in the total serum IgA concentrations, regardless of PVL and LC. The total serum IgM from ELISA+ cows was significantly decreased (p = 0.0223), with the largest decreases occurring in the highest PVL and LC subgroups. This pilot study is a first step in investigating the impact of BLV on mucosal immunity and will require further exploration in each of the various stages of disease progression.

Abstract A simple, accurate and precise RP-HPLC method was developed for the simultaneous determination of chloroquine, pyrimethamine and cetirizine hydrochloride concentrations in bulk drug and human serum. The assay was performed using a mobile phase of methanol: water (70:30) at pH of 2.8 ± 0.05 on the Purospher C-18 column with UV detection at 230 nm and rosuvastatin used as an internal standard. The retention times observed for chloroquine, pyrimethamine and cetirizine hydrochloride were 3.5, 2.5 and 5.5 minutes, respectively. The method was found to be specific for the assayed drugs showing a linear response in the concentration range of 1–100 μg mL−1 with coefficients of determination values of (r = 0.999). The method was developed and validated according to ICH guidelines. The method was used to monitor the serum samples and was found to be sensitive for therapeutic purposes, showing the potential to be a useful tool for routine analysis in laboratories.
