Cytogenotoxic effects of ethanolic extracts of Annona crassiflora (Annonaceae)

AbstractInfusions of the leaves and seeds of Annona crassiflora Mart. are commonly employed in the treatment of diarrhoea, snakebites, tumours and disorders of the hair and scalp. The aim of the present study was to investigate the cytotoxic and genotoxic properties of ethanolic extracts of A. crassiflora by evaluating their effects on germination, root elongation, chromosome structure and the cell division of Lactuca sativa (lettuce). The experiment followed a randomized design involving the treatment of L. sativa seeds with ethanolic extracts from leaves and seeds of A. crassiflora applied at ten concentrations (0.0125, 0.025, 0.05, 0.1, 0.2, 0.4, 0.6, 0.8, 1.0, and 1.2 mg/L) and with five repetitions per treatment. Seeds of L. sativa exposed for 48 h to A. crassiflora leaf extract at concentrations ≥ 0.1 mg/L, or to seed extracts at concentrations ≥ 0.2 mg/L, showed germination percentages that were significantly (p < 0.05) lower than those of seeds exposed to aqueous ethanol control. Exposure of L. sativa seedlings to leaf (but not seed) extracts of A. crassiflora produced significant (p < 0.05) reductions in the mitotic indices of root meristem cells of lettuce and induced chromosome and nuclear abnormalities in the root cells. The presence of chromosome stickiness, bridges, fragments, laggard chromosomes and nuclear condensation were also observed. The cytogenetic effects observed suggest that folkloric medicines prepared with extracts of the leaves or seeds of A. crassiflora should be employed with caution.

In recent days, the thirst for the identification of the potential bioactive compounds from the natural sources like medicinal plants is on continuous demand. Among scientists and academicians, it has created many interdisciplinary platforms for research in establishing new drugs from the natural sources. According to many recent studies, Nigella sativa is believed to be the rich source of quinone, an effective bioactive compound with lots of medicinal values. The purpose of this study was to isolate and estimate the quinone in Nigella sativa seed extracts (aqueous and ethanol). Based on the qualitative and quantitative determination, the extracts were further focused for isolation of quinone from both aqueous and ethanolic  extracts of Nigella sativa. The isolated compound is identified by thin layer chromatography and purity is analyzed in High performance liquid chromatography. From the results we obtained, it was very clear that among the aqueous and ethanolic extracts of Nigella sativa, the ethanolic extract has been found with the highest quantity of quinine. This would be predicting that the ethanol extract of Nigella sativa may have good efficacy of pharmacological and therapeutic potentials like antidiabetic, anticancer, antimicrobial, anti-inflammatory properties when compared with the aqueous extract due to the presence of more quinone.

Analgesic, anti-inflammatory and anti-pyretic activities of n-hexane and aqueous ethanolic extracts of Malva neglecta in Swiss albino mice was evaluated. For each activity, the mice were divided into 4 groups: Group 1 served as control, Groups 2 and 3 were given n-hexane and aqueous ethanol extracts, respectively whereas Group 4 was treated with the standard drug. Analgesic activity was evaluated against acetic acid induced writhing, Eddy's hot plate method, and Formalin induced paw licking. Anti-inflammatory activity was evaluated by carrageenan-induced paw edema. The extracts were also examined for their anti-pyretic activities against yeast-induced pyrexia. Results showed that the n-hexane (p < 0.05) and aqueous ethanolic (p < 0.005) extracts of M. neglecta exhibited analgesic activity by reducing acetic acid-induced writhing, mean reaction time on hot plate model and formalin-induced paw licking in mice as compared to the control. The n-hexane extract (p < 0.05) as well as aqueous ethanolic (p < 0.005) extracts produced significant antiinflammatory activity as compared to the control. Both the n-hexane and aqueous ethanolic extracts revealed significant antipyretic activity (p < 0.005 and p < 0.05, respectively) in mice model. The results of the present study demonstrated that the n-hexane and aqueous ethanol extracts of M. neglecta possess analgesic, antiinflammatory and anti-pyretic activities. Bangladesh J. Mixing Crude Palm Oil (CPO) with high and lower Free Fatty Acid (FFA) is a technique to reduce FFA in CPO. Avocado Seeds Extract (ASE) was expected to reduce the FFA. This study was conducted to test the ability of ASE on reducing ALB in CPO. The variable in this study is ASE solution (0-50 ml). The whole treatment were conducted at same level of o temperatur 60 C, stirring speed (400 rpm) and reaction time (60 minutes). The research was employed non factorial Completely Randomized Design. Statistical analysis was performed using F Test along with Duncan Test. The results showed that addition of ASE can reduce FFA. The results of F Test showed that the addition of 25 ml ASE is the most effective for reduction ALB content (4.65 % to 0.22 %) in CPO.

Salinity can affect cell division and cause chromosomal abnormalities such as the formation of micronuclei, chromosome stickiness, c-mitosis and multipolar anaphases. Plants react to salt stress with morphological, biochemical, physiological, cellular and molecular adjustments. The aim of this study was to evaluate the effect of different leaf extract concentrations of purple lettuce on the cytogenetic activity of lettuce roots, cv. Regina, from seeds subjected to salt stress. Four extract concentrations of purple lettuce (0, 50, 100 and 150 g lettuce leaves L-1 water)and five concentrations of sodium chloride (0, 30, 60, 90 and 120 mM) were tested, constituting 20 treatments, with distilled water for a negative control. The analyses were of mitotic index (MI), phase index of mitosis (prophase, metaphase, anaphase and telophase) and the presence of chromosomal aberrations. Salt reduces the mitotic index and all index phases of the lettuce roots. The purple lettuce extract does not affect the mitotic index, reduces the cell index in prophase and increases the cells in telophase of lettuce roots. The purple lettuce extract and salt cause chromosomal abnormalities in lettuce root cells; however, a smaller number of mutations is found by applying 100 g L-1 extract.
