Salinity can affect cell division and cause chromosomal abnormalities such as the formation of micronuclei, chromosome stickiness, c-mitosis and multipolar anaphases. Plants react to salt stress with morphological, biochemical, physiological, cellular and molecular adjustments. The aim of this study was to evaluate the effect of different leaf extract concentrations of purple lettuce on the cytogenetic activity of lettuce roots, cv. Regina, from seeds subjected to salt stress. Four extract concentrations of purple lettuce (0, 50, 100 and 150 g lettuce leaves L-1 water)and five concentrations of sodium chloride (0, 30, 60, 90 and 120 mM) were tested, constituting 20 treatments, with distilled water for a negative control. The analyses were of mitotic index (MI), phase index of mitosis (prophase, metaphase, anaphase and telophase) and the presence of chromosomal aberrations. Salt reduces the mitotic index and all index phases of the lettuce roots. The purple lettuce extract does not affect the mitotic index, reduces the cell index in prophase and increases the cells in telophase of lettuce roots. The purple lettuce extract and salt cause chromosomal abnormalities in lettuce root cells; however, a smaller number of mutations is found by applying 100 g L-1 extract.

The mitotic index of the roots of pigeon pea can be the basis for determining the growth of pigeon pea. The purpose of this research was to determine the time of root cell division, to observe the mitotic phases, and to determine the mitotic index of pigeon pea root cells. The preparation of the pigeon pea was carried out for 4 days to grow the roots. The roots were cut off at 08.00, 08.15, and 08.30 WIT (Eastern Indonesian Time). The roots were cut 0.5-1cm. Carnoy’s solution was used as the fixative solution using the Squash technique. The prepared roots were then observed using an Olympus cx-22 microscope and an OptiLab camera with a magnification of 100x40. The data were descriptively analyzed to describe the images of mitotic phases and the mitotic index presentation in the root cells of pigeon pea. The results of this research showed that the cell division of the pigeon pea roots began at 08.00 WIT, which was marked by the presence of a lot of prophase. The next phases that appeared were prometaphase, metaphase, and anaphase which occurred from 08.15 to 08.30 with different numbers. The highest mitotic index occurred at 08.15, when most of the root cells underwent metaphase. This study succeeded in revealing that the optimum time for pigeon pea root cell division is 08.15 WIT. In the future, this research can help pigeon pea farmers in Southwest of Maluku to carry out vegetative reproduction which is closely related to this mitotic study.

The accentuated increase in the production of solid urban waste (SUW) and the consequent accumulation of leachate in landfills increase the risk of environmental contamination. Biomonitors are used to assess the toxicity of pollutants on living organisms. In this study, the pollutant potential of leachate from SUW from a deactivated landfill was evaluated by bioassays with Lactuca sativa L. and Lycopersicon esculentum Mill., and the relationship between chemical characteristics of the effluent and biological parameters was analyzed. The effluent was tested in its raw form and diluted in distilled water at concentrations of 75 and 50%. The percentage of germination, root growth (RG), mitotic index (MI), chromosomal abnormalities index (CAI), and micronuclei frequency (MCN) were analyzed. In the presence of effluent, germination and MI decreased, while RG, CAI and MCN frequencies increased in relation to the negative control (distilled water) for both species. Lead, iron and zinc presented negative relation with seed germination for both species, with RG of L. sativa and MI of L. esculentum, as well as a positive relation with MCN frequency in the studied species. Because of its larger chromosomes, L. sativa is a more suitable biomonitor of SUW leachate toxicity than L. esculentum. Even though the landfill is deactivated, it is necessary to treat this effluent, in order to minimize environmental impacts.

Medicinal plants used for generations one of which is crop cassava leaves. Cassava leaves is believed to cure rheumatism, gout, anemia, constipation, and to increase endurance. This study aims to determine the Effects Of Ethanol Leaf Extract Singkong (Manihot Utilissima Pohl) As Drug Alternatif Antirheumatic Of Pain On Mice. This study was an experimental study using 25 mice. These animals were divided into five groups where each group consisted of 5 mice. Cassava leaf extracts as a control test, flamar gel as a positive control, and distilled water as a negative control. Observations were made for 45 minutes using a hot excitatory response in the form of a decrease in a lick or jump after the treatments. The results showed a decline in the response of mice to lick the feet or jump to the heat stimuli were administered after administration of cassava leaf extract. Cassava leaf extract has an effect antirheumatic to pain in mice. Based on the analysis of SPSS best dose in this study is the first dose (0.65mg / 20gr mice BB).

Indikator klinis penyakit Diabetes Melitus adalah hiperglikemia. Mimba (Azadirachta indica A. Juss) merupakan salah satu tumbuhan yang berpotensi sebagai alternatif obat herbal hiperglikemia. Penelitian ini bertujuan untuk menganalisis pengaruh ekstrak etanol daun mimba terhadap struktur histologis pankreas pada tikus hiperglikemia. Penelitian ini mengunakan Rancangan Acak Lengkap (RAL) dengan jumlah tikus 24 ekor yang dibagi menjadi 6 kelompok perlakuan dan 4 kali ulangan. P0 (kontrol normal) adalah kelompok tikus normal yang diberi akuades, P1 (kontrol negatif) adalah tikus hiperglikemia yang diberi akuades. P2 (kontrol positif) adalah kelompok tikus hiperglikemia yang diberi glibenklamid dosis 2,25 mg/kg BB. P3, P4, dan P5 adalah kelompok tikus yang diberi ekstrak etanol daun mimba dosis 100, 200, dan 400 mg/kg BB. Data dianalisis dengan ANOVA pada signifikansi 95%. Hasil penelitian menunjukkan bahwa perlakuan ekstrak etanol daun mimba dosis 100, 200, dan 400 mg/kg BB tidak memberikan pengaruh nyata terhadap bobot pankreas, diameter, luas, dan densitas pulau Langerhans (P>0,05). Skoring struktur pulau Langerhans berdasarkan uji Mann-Whitneymenunjukkan hasil beda nyata pada kelompok  tikus yang diberi daun mimba (P≤0,05). Kesimpulan dari penelitian ini, pemberian ekstrak daun mimba dosis 400mg/kg BB menunjukkan adanya perbaikan morfologi pulau Langerhans. The clinical indicator of Diabetes mellitus was hyperglycemia. Azadirachta indica A. Juss was a plant has the potential to alternative medicine for hyperglycemia. The study was to analyze the ethanol neem leaf extract effect on histological structure of hyperglycemic rat pancreas. This study used a completely randomized design (CRD) with 24 rats were divided into 6 treatment groups and 4 replications. P0 (control) was a normal rats group were given distilled water, P1 (negative control) was a hyperglycemic rats group were given distilled water. P2 (positive control) was a hyperglycemic rats were given 2.25 mg/kg BW of glibenclamide. P3, P4, and P5 were rats were given 100, 200, and 400 mg/kg BW of ethanolic neem leaf extract.The data analyzed by ANOVA at 95% significance showed the treatment of 100, 200, and 400 mg/kg BW ethanolic neem leaf extract had no significant effect on the pancreatic weight, diameter, area, and density of Langerhans islet (P> 0.05). The score of Langerhans islet structure based on the Mann-Whitney test showed significant differences in the groups of mice given neem leaves (P≤0.05). Treatment of 400mg/Kg BW neem leaf extract showed an improvement in the morphology of the islets of Langerhans.

Patients withdiabetes mellitus (DM) continues to grow because prosperity and people's lifestyles.Treatment of diabetes often usei njections of insulin and oral antidiabetic drugs. Thetreatment has no side effects. Therefore, it is necessary to find effective drugs using plants thatred betel leaf (Piper crocatum Ruiz & Pav.). Red betel leaf contains flavonoids which are antioxidants. This study aims to determine the effect and dose of extract of red betel leaf (Piper crocatum Ruiz & Pav.) The most effective agains blood glucose in mice(Mus musculusL.) male induced sucrose.This study was an experimental study. The research was conducted in October 2015 in the Division of Laboratory Animal and Zoology Department of Biology, State University of Padang. The subject of research in the form of mice (Mus musculus L.) males totaled 24 tails. The design used was completely randomized design (CRD) with 6 treatments and 4 repetitions. The treatment is given as follows: treatment I: the diabetes control without any treatment given, treatment II: as a negative control (sucrose 3 g/kg bw), treatment III: sucrose+suspension of red betel leaf extract (dosage 0,7 g/kg bw), treatment IV: sucrose+suspension of red betel leaf extract (dosage 1,4 g/kg bw), treatment V: sucrose+suspension of red betel leaf extract (dosage of 2,1 g/kg bw) and treatment VI: sucrose+suspension extracts red betel leaf (dosage 2,8 g/kg bw).The results showed that the extract of red betel leaf (Piper crocatum Ruiz & Pav.) at a dose of 0,8 g/kg bw 1,4 g/kg bw 2,1 g/kg bw and 2,8 g/kg bw can lowers blood glucose in mice. However, the most appropriate dose in lowering blood glucose in mice (Mus musculus L.) at 2,8 g/kg bw in mice.

This study is aimed at analyzing the anticancer properties of papaya leaf extract, specifically the inhibition of cell proliferation and apoptotic induction through nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) and p53 pathways. Twenty-five mice (Mus musculus), aged 2 months and weighing 20–30 g, was injected with 0.5 mg dexamethasone for 7 days. The mice were then injected intracutaneously with 1 ml of HeLa cells (8 × 106 HeLa cells/microliter). The mice were divided into five groups (5 each): negative control (P1) (5% CMC-Na, sodium carboxymethyl cellulose), treatment II (225 mg/kg BW (body weight) papaya leaves methanol extract), treatment III (450 mg/kg BW), treatment IV (750 mg/kg BW), and treatment PV (2 mg alcohol anticancer drug). Papaya leaf extract treatments were applied for 2 weeks. Then, the tumor tissue was isolated for hematoxylin and eosin staining. Immunohistochemical imaging was used to detect Ki-67, caspase-3, NF-κB, and p53 expression. Further analysis was undertaken using the ImmunoRatio software program. The results indicated that administration of papaya leaf methanol extract significantly increased the expression of NF-κB and p53 at a dose of 450 mg/kg BW. Our results also showed that the mice treated with 450 mg of papaya leaf extract per kg of BW (P3) had the largest increase of caspase-3 expression compared to the negative control group. Papaya leaf ethanol extract decreased the cancer cell proliferation index and increased apoptosis of cancer cells in animal models of cervical cancer; it may also work to increase NF-kB expression and expression of the p53 gene.

Immunomodulator is a compound that can increaase the imuno system. One of the plants that have immunomodulator’s activity is Waru Leaf (Hibiscus tiliaceus). the purpose of this research was to test the effect of immunomodulator by extract of Waru Leaf ethanol on rat male. The activity of immunomodulator was determined by using digital pletysmometer by measuring the differences between the last leg swelling’s volume and the first leg swelling’s volume. The treatment group were divided into 5 groups. Each group consistof 5 rats CMC-Na 0,5% (negative control), Stimuno®  32,5 mg/kgBW (positive control), dose of EEDW 50, 100 and 200 mg/kgBW, and bacteria E.coli as antigen. The results slowed that distribution of EEDW dose 200 mg/kgBW can give the effect of immunostimulant by swelling enthancement compared by CMC-Na 0,5 %. EEDW 200 mg/kgBW that have activity comparable with Stimuno®  32,5 mg/kgBW. Thus, it is concluded that of Waru Leaf extract has immunomodulator effects on delayed-type hypersensitivity response of rat male.

Abstract Background Salt stress threatens crop yields all over the world. Many NAC transcription factors have been reported to be involved in different abiotic stress responses, but it remains unclear how loss of these transcription factors alters the transcriptomes of plants. Previous reports have demonstrated that overexpression of OsNAC45 enhances salt and drought tolerance in rice, and that OsNAC45 may regulate the expression of two specific genes, OsPM1 and OsLEA3–1. Results Here, we found that ABA repressed, and NaCl promoted, the expression of OsNAC45 in roots. Immunostaining showed that OsNAC45 was localized in all root cells and was mainly expressed in the stele. Loss of OsNAC45 decreased the sensitivity of rice plants to ABA and over-expressing this gene had the opposite effect, which demonstrated that OsNAC45 played an important role during ABA signal responses. Knockout of OsNAC45 also resulted in more ROS accumulation in roots and increased sensitivity of rice to salt stress. Transcriptome sequencing assay found that thousands of genes were differently expressed in OsNAC45-knockout plants. Most of the down-regulated genes participated in plant stress responses. Quantitative real time RT-PCR suggested that seven genes may be regulated by OsNAC45 including OsCYP89G1, OsDREB1F, OsEREBP2, OsERF104, OsPM1, OsSAMDC2, and OsSIK1. Conclusions These results indicate that OsNAC45 plays vital roles in ABA signal responses and salt tolerance in rice. Further characterization of this gene may help us understand ABA signal pathway and breed rice plants that are more tolerant to salt stress.

SUMMARYIn this study, mitotic effects of gamma rays on Ekiz 1 variety belonging to Helianthus annuus L. (2n= 34) in the M0 (first irradiated seeds), M1 and M2 generations have been investigated. Seeds (M0) were irradiated with gamma rays at 10, 20, 30, 40 and 50 kR doses. Percentage of total abnormalities in the M0, M1 and M2 generations increased parallel to the increasing dose of radiation. These abnormalites have been observed as C-metaphase, chromosome stickiness, laggards and bridges with or without fragment. Mitotic index (M.I.) in the M0, M1 and M2 generations has decreased parallel to the dose increase. When the generations are compared, both the amounts of decrease in mitotic index and in the percentage of mitotic abnormalities were mostly observed in M0.

Background: Antimicrobial resistance is the main concern worldwide to combat infectious. Over the years studies on leaf extracts Tinospora cordifolia have demonstrated the potent role its antibacterial property. The current study is an attempt to test its antibacterial property against Escherichia coli cell division. Material and methods: Phytochemical screening assay of T. cordifolia leaf extract was done using standard procedure and the results showed the presence of alkaloid, carbohydrate, terpenoid, steroid, tannin, amino acid, flavonoid and glycoside components. Results: HPLC analysis revealed the presence of berberine in T. cordifolia leaf extract. Further E. coli cells were treated with berberine to study its efficacy in inhibiting cell division. Antibacterial assay was performed by using disc diffusion method. Conclusion: Among aqueous, methanolic, ethanolic, chloroform, hexane and acetone extract only methanolic extract showed zone of inhibition.
