The cuneate nucleus (CN) is among the first sites along the neuraxis where proprioceptive signals can be integrated, transformed, and modulated. The objective of the study was to characterize the proprioceptive representations in CN. To this end, we recorded from single CN neurons in three monkeys during active reaching and passive limb perturbation. We found that many neurons exhibited responses that were tuned approximately sinusoidally to limb movement direction, as has been found for other sensorimotor neurons. The distribution of their preferred directions (PDs) was highly non-uniform and resembled that of muscle spindles within individual muscles, suggesting that CN neurons typically receive inputs from only a single muscle. We also found that the responses of proprioceptive CN neurons tended to be modestly amplified during active reaching movements compared to passive limb perturbations, in contrast to cutaneous CN neurons whose responses were not systematically different in the active and passive conditions. Somatosensory signals thus seem to be subject to a 'spotlighting' of relevant sensory information rather than uniform suppression as has been suggested previously.

Das Spektrum der schlafmedizinischen Erkrankungen umfasst eine Reihe von speziellen neurologischen Erkrankungen, die sich durch eine hohe Prävalenz kennzeichnen oder die Lebensqualität stark beeinträchtigen. Dem praktisch tätigen Arzt kommt eine Schlüsselrolle in der Erkennung und Behandlung dieser Erkrankungen zu und sollte deshalb mit den wichtigsten schlafmedizinischen neurologischen Erkrankungen vertraut sein. Im Folgenden werden als wichtigste Vertreter, das Restless Legs Syndrom (mit oder ohne Periodic Limb Movement im Schlaf), Bewegungsstörungen im Schlaf (v. a. nonREM- und REM-Schlaf-Parasomnien), epileptische Anfälle im Schlaf sowie die Narkolepsie vorgestellt. Die Narkolepsie gilt zwar als seltene Erkrankung, ermöglicht aber als Modellerkrankung physiologische bzw. pathophysiologische Vorgänge der Schlaf-/Wachregulation zu verstehen. Eine zunehmende Bedeutung gewinnt auch die REM-Schlafverhaltensstörung, die als Frühzeichen einer neurodegenerativen Erkrankung (z. B. Synukleinopathien wie Parkinson-Erkrankung) auftreten kann. Eine frühzeitige Diagnose eröffnet hier die Möglichkeit in Zukunft neuroprotektive Substanzen einzusetzen.

Abstract The experiment reported here was aimed at determining whether the level of brain activity can be related to performance in trained subjects. Two tasks were compared: a temporal and a linguistic task. An array of four letters appeared on a screen. In the temporal task, subjects had to decide whether the letters remained on the screen for a short or a long duration as learned in a practice phase. In the linguistic task, they had to determine whether the four letters could form a word or not (anagram task). These tasks allowed us to compare the level of brain activity obtained in correct and incorrect responses. The current density measures recorded over prefrontal areas showed a relationship between the performance and the level of activity in the temporal task only. The level of activity obtained with correct responses was lower than that obtained with incorrect responses. This suggests that a good temporal performance could be the result of an efficacious, but economic, information-processing mechanism in the brain. In addition, the absence of this relation in the anagram task results in the question of whether this relation is specific to the processing of sensory information only.

Die Komorbidität von Depression und Schlafstörungen ist hoch. Dabei kann die Klärung, welche Störung der anderen vorausgegangen ist, bzw. der Ausschluss einer organischen Ursache, für die weitere Behandlung von großer Wichtigkeit sein. Als besonders häufige körperliche Ursache für Schlafstörungen, die wiederum eine depressive Symptomatik bedingen können, kommt dem Periodic-Limb-Movement-, dem Restless-Legs- sowie dem obstruktiven Schlafapnoesyndrom eine besondere Bedeutung zu. Eine ausführliche Medikamenten- und Alkoholanamnese kann Hinweise auf eine substanzinduzierte Insomnie geben. Schlafanstoßende Antidepressiva können neben verhaltenstherapeutischen Maßnahmen einen positiven Effekt auf die affektive Störung wie auch die Insomnie haben.

Selective attention is a fundamental cognitive function that guides behavior by selecting and prioritizing salient or relevant sensory information of our environment. Despite early evidence and theoretical proposal pointing to an implication of thalamic control in attention, most studies in the past two decades focused on cortical substrates, largely ignoring the contribution of subcortical regions as well as cortico-subcortical interactions. Here, we suggest a key role of the pulvinar in the selection of salient and relevant information via its involvement in priority maps computation. Prioritization may be achieved through a pulvinar- mediated generation of alpha oscillations, which may then modulate neuronal gain in thalamo-cortical circuits. Such mechanism might orchestrate the synchrony of cortico-cortical interaction, by rendering neural communication more effective, precise and selective. We propose that this theoretical framework will support a timely shift from the prevailing cortico- centric view of cognition to a more integrative perspective of thalamic contributions to attention and executive control processes.
