Introduction. Mother of thousands (Kalanchoe daigremontiana) in folk medicine is applied in the form of compresses fresh, crushed leaf, or juice of the leaves of inflammation of the skin, burns and wound healing. Aim. The aim of the study was to compare the anti-inflammatory and antioxidant properties of juices obtained from the leaves of K. daigremontiana, as well as to determine the content of the polyphenols and flavonoids. Material and methods. Compared the anti-hyaluronidase activity and antioxidant potential (DPPH and FRAP) of juices from K. daigremontiana using a spectrophotometric methods. Moreover, total phenolic content and total flavonoid content were determined by spectrophotometric methods, with the Folin-Ciocalteu reagent (FC) and AlCl3 reagent respectively. Results. The obtained results indicate that the juice prepared from the leaves is characterized by a higher content of active compounds and shows a higher anti-inflammatory and antioxidant activity than the commercial preparation. Conclusions. Juices from K. daigremontiana could be helpful in the treatment of skin diseases.

Abstract Background This study was focused on the measurement of anticancer properties of six medicinal plants from western Nepal in three cell lines; HeLa, Hep3B, and HCT116, and anti-inflammatory properties in RAW 264.7 cell line through NO, PGE2, and TNF-α production. In addition, the phytochemical screening, total phenolic, flavonoid content, and antioxidant properties were evaluated. Results The qualitative phytochemical analysis revealed the presence of different secondary metabolite and range of total phenolic and total flavonoid content. The highest antioxidant activities were observed in Bergenia pacumbis against both DPPH (IC50 = 25.97 ± 0.19 μg/mL) and ABTS (IC50 = 14.49 ± 0.40 μg/mL). Furthermore, the highest antiproliferative effect against cervical, liver, and colon cancer cell lines were observed in Melia azedarach as IC50 values of 10.50, 5.30, and 1.57 μg/mL respectively, while the strongest anti-metastatic potential on liver cancer cell line was found in Pleurospermum benthamii. In addition, P. benthamii showed the most potent anti-inflammatory effect in RAW264.7 murine macrophage cells. Conclusion This study provided the evidence for M. azedarach and P. benthamii to have great anticancer potential and finding builds the enough scientific backgrounds in future to isolate and purify the bioactive compounds for further applications.

Microsechium helleri(Cucurbitaceae) has been used in ethnopharmacological as a lotion to prevent hair loss, diuretic and cathartic, in the region of central Veracruz, Mexico is used as antidiabetic. The antioxidant properties of the hexanic (EHex), chloroformic (ECHCl3) and ethanolic (EEtOH) extracts, were evaluated by 2,2diphenyl-1-pychrylhydrazyl (DPPH) test, the Ferric Reducing/Antioxidant Power (FRAP) and the total phenolic content test. The anti-inflammatory effect was evaluated in the acute ear edema induced with phorbol 12-myristate 13-acetate (TPA) in mouse and the hypoglycemic and cardioprotective effects of the EEtOH were determined in rats. The EEtOH was the most active in the antioxidant potential DPPH test and the ECHCl3was the best in the FRAP assay and the total polyphenols content. In the anti-inflammatory assay, the ECHCl3showed the most activity. The EEtOH had the decreased the glucose levels and reduced myocardial damage. The results support the use of this plant in folk medicine in Mexico as antioxidant, anti-inflammatory, hypoglycemic and cardioprotective.

Apoptosis is one of markers considered in drug design including in treating cancers. Conventional treatments for cancer cause various side effects. One of possible alternatives is honey, an antioxidant driven by its phenolic and flavonoids content. This study aims to observe the effects of Malaysian Acacia honey in inhibiting breast cancer cell growth through apoptosis. Antioxidant properties of the honey were measured using total phenolic content (TPC) and total flavonoid content (TFC). Then, antiproliferative activity of the honey were observed using MTT assay. The honey exhibited IC50 at concentration of 5.5% (v/v). Further studies using TUNEL and live cell view revealed that Acacia honey induced apoptosis after 6 hours of treatment. Cell shrinkage, which is one of the apoptotic features, was observed as early as 2 hours, followed by the formation of apoptotic bodies within 6 hours of the honey treatment. Details of mechanism and actual compounds involved in displaying the results are being intensively studied.

Objectives: The goals of this research were to observe antioxidant properties from different parts of super red dragon fruit (Hylocereus costaricensis) using two antioxidant testing methods which were 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS).Methods: Antioxidant activities were determined using DPPH and ABTS assays, total phenolic content (TPC) using Folin–Ciocalteu reagent, flavonoid content by Chang’s method.Results: Inhibitory concentration 50% (IC50) of DPPH scavenging activity of all of the extracts in the range of 2.69 μg/ml was −94.17 μg/ml. The ethyl acetate peel extract of super red dragon fruit expressed the highest TPC (4.56 g GAE/100 g) and the highest total flavonoid content (12.63 g QE/100 g). TPC in flesh extract of super red dragon fruit had a negative and significant correlation with their IC50 of ABTS. The IC50 of DPPH and IC50 of ABTS of flesh extract of super red dragon fruit showed positive and significant correlation.Conclusion: All different parts extracts of super red dragon fruit (except n-hexane flesh extract) were categorized as a very strong antioxidant by DPPH method. Phenolic compounds in flesh extract of super red dragon fruit were the major contributor in antioxidant activities by ABTS method. DPPH and ABTS showed linear results in antioxidant activities of super red dragon fruit flesh extract.

This study compared the mineral content and bioactive properties of flowers and fruit coming from wild elderberry plants with those of flowers and fruit harvested from elderberry cultivars grown in an orchard. Elderberry fruit and flowers were analyzed for the content of selected minerals, phenolic compounds, and anthocyanins, as well as for antioxidant activity. Mineral content was determined by the atomic absorption spectrometry method, while antioxidant activity and the content of polyphenols and anthocyanins were determined by spectrophotometric methods. Flowers were found to contain more total ash and much higher content of most of minerals, except magnesium which was present in high concentrations in fruit. Fruit showed significantly higher antioxidant activity than flowers, whereas the total phenolic content varied depending on the growing location or cultivar. The material obtained from selected cultivars growing in an orchard had higher antioxidant activity and polyphenol and anthocyanin content than the material obtained from wild plants. Fruit of the ”Haschberg” cultivar and flowers of the ”Sampo” cultivar had the best bioactive properties of the studied samples.

Chinese bayberry (Myrica rubra Sieb. et Zucc.) is an economically important medicinal plant with multiple uses. Two varieties ‘Dongkui Oriental Pearl’ (Dongkui for short) and acuminata ‘Nakai’ (Nakai for short) were used to compare and evaluate the antioxidant activities of hydroethanolic extracts of the fruit using ultrasonic and stirring extraction methods. Dongkui bayberry fruit extract (BFE) prepared using the ultrasonic method exhibited a significantly higher value for the total phenolic content (TPC) and had lower 50% inhibitory concentration (IC50) values of scavenging activities against 1,1-diphenyl-2- picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid (ABTS), and hydrogen peroxide (H2O2), as well as reducing power compared to the other treatment. The TPC of the BFE was significantly correlated with its DPPH, ABTS, and H2O2 radical-scavenging and reducing power activities. Dongkui BFE at a concentration of 0.25 mg/mL exhibited significantly greater protection of RAW 264.7 mouse macrophages against H2O2-induced damage and lipopolysaccharide (LPS)-stimulated nitric oxide production by macrophages, and it displayed remarkable inhibitory effects compared to the other extracts using the ultrasonic extraction method. Furthermore, compared to the Nakai BFE, macrophages exposed to the Dongkui BFE by the ultrasonic extraction method significantly inhibited LPS-induced production of tumor necrosis factor-α at a concentration of the extract of 0.25 mg/mL. The antioxidant properties and anti-inflammatory and protective effects of BFE prepared by stirring and ultrasonic methods are discussed for the first time in this study.   ********* In press - Online First. Ruta angustifolia was used in this study in order to evaluate the antimicrobial activity and antioxidant properties and its correlation with the polyphenolic content. Two Gram-positive bacteria (Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778) and two Gram–negative bacteria (Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 8739) were used to determine the antibacterial activity. Aqueous maceration extract was used for antioxidant activities and methanolic maceration extract was used for antibacterial activity. The antioxidant properties and activities were evaluated by using total phenolic content (TPC), total flavonoid content (TFC), DPPH free radical scavenging activity and beta-carotene bleaching method. Whereas, the antibacterial activity was examined using disc diffusion method against selected microorganism at concentration 1.0 mg/disc. The results showed the phenolic content of R. angustifolia extract was 18.89 g GAE/100 g extract while the flavonoid content was 14.170 g QE/100 g extract. R. angustifolia exhibited good radical scavenging with IC50 value of 2.04 mg/ml. The result for disc diffusion method showed no inhibition zone against all the strains of bacteria at 1.0 mg/disc concentration of the extract. Based on the results, it can be concluded that the R. angustifolia aqueous extract has the antioxidant properties and there is correlation between polyphenolic content of the extract with its antioxidant activity. However, R. angustifolia methanolic extract did not show any antibacterial activity.

The aim of this study was to evaluate the total phenolic and flavonoid content, and the in vitro antioxidant, anti-inflammatory, antibacterial, antifungal, antimalarial, cytotoxicity, and antiprotozoal activities of the Algerian plant Cytisus villosus Pourr. (Syn. Cytisus triflorus L’Hérit.). Additionally, the radioligand displacement affinity on opioid and cannabinoid receptors was assessed for the extracts and isolated pure compounds. The hydro alcoholic extract of the aerial part of C. villosus was partitioned with chloroform (CHCl3), ethyl acetate (EtOAc), and n-butanol (n-BuOH). The phenolic content of the C. villosus extracts was evaluated using a modified Folin–Ciocalteau method. The total flavonoid content was measured spectrometrically using the aluminum chloride colorimetric assay. The known flavonoids genistein (1), chrysin (2), chrysin-7-O-β-d-glucopyranoside (3), and 2″-O-α-l-rhamnosylorientin (4) were isolated. The antioxidant activities of the extracts and isolated compounds were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DDPH) and cellular antioxidant activity (CAA) assays. The plant extracts showed moderate antioxidant activity. EtOAc and n-BuOH extracts showed moderate anti-inflammatory activity through the inhibition of induced nitric oxide synthase (iNOS) with IC50 values of 48 and 90 µg/mL, respectively. The isolated pure compounds 1 and 3 showed good inhibition of Inducible nitric oxide synthase (iNOS) with IC50 values of 9 and 20 µg/mL, respectively. Compounds 1 and 2 exhibited lower inhibition of Nuclear Factor kappa-light-chain-enhancer of activated B cells (NF-κB) with IC50 values of 28 and 38 µg/mL, respectively. Furthermore, the extracts and isolated pure compounds have been shown to exhibit low affinity for cannabinoid and opioid receptors. Finally, n-BuOH extract was a potent inhibitor of Trypanosoma brucei with IC50 value of 7.99 µg/mL and IC90 value of 12.61 µg/mL. The extracts and isolated compounds showed no antimicrobial, antimalarial nor antileishmanial activities. No cytotoxic effect was observed on cancer cell lines. The results highlight this species as a promising source of anti-inflammatory and antitrypanosomal agents.

Polyphenols are naturally occurring compounds found largely in fruits and vegetables. The antioxidant properties of these polyphenols including total phenolic content (TPC), total flavonoid content (TFC), tannin content, 1,1-diphenyl-2-picrylhydrazyl free radical (DPPH), 2,2′-azinobis-(3-ethylbenzo-thiazoline-6-sulfonic acid) (ABTS) scavenging abilities and ferric ion reducing antioxidant power (FRAP) were measured among sixteen (16) plant foods (mango, blueberry, strawberry, black carrot, raspberry, dark grapes, garlic, ginger, onion, cherry, plum, apple, papaya, peach, pear and apricot) by modifying, standardising and translating existing antioxidant methods using a 96-well plate reader. Eighteen targeted phenolic acids and flavonoids were characterised and quantified using high-performance liquid chromatography-photometric diode array (HPLC-PDA) and verified by modifying an existing method of liquid chromatography coupled with electrospray-ionisation triple quadrupole time-of-flight mass spectrometry (LC-ESI-QTOF/MS). While most of these compounds were accurately detected by the HPLC-PDA at a low concentration, a few polyphenols in low concentrations could be only be characterised using the LC-ESI-QTOF/MS method. Our results showed that mango possessed the highest overall antioxidant activity, phenolic acid and flavonoid content among the selected fruits. Factor analysis (FA) and Pearson’s correlation tests showed high correlations among ABTS, DPPH, FRAP and phenolic acids, implying the comparable capabilities of scavenging the DPPH/ABTS free radicals and reducing ferric ions from the antioxidant compounds in the samples. Phenolic acids contributed significantly to the antioxidant activities, and flavonoids contributed more to tannin content based on the correlations. Overall, methods modified and standardized in this study can provide better understanding of high throughput technologies and increase the reliability of antioxidant data of different plant foods.

Purpose: Milk thistle (Silybum marianum (L.) Gaertn) has been used for centuries as a natural remedy for liver and gall bladder / biliary tract diseases. Silymarin is the purified and standardized extract from milk thistle seeds. The purpose of the present study is to present a comparative analysis of the total phenolic substances content and the antioxidant effect of silymarin and three different types of Silybum marianum extracts (i.e. ethanol extract, water extract and lyophilized extract). Methods: Total phenolic and flavonoid content of extracts was analysed and the antioxidant activity was assessed by determining their free radical scavenging ability by DPPH method. Results: Total phenolic content is the highest in silymarin– 134.12±3.22 mg GAE/g, followed by lyophilized water extract – 51.74±2.10 mg GAE/g. Silymarin has also been found to have the highest antioxidant activity among all extract samples (338.03 VCEAC µmol /g), followed by S.marianum lyophilised extract (123.05 VCEAC µmol/g). Conclusions: A strong correlation between total phenolic and flavonoid content in the extracts and their antioxidant activity has been found.
