ANTIOXIDANT AND ANTIBACTERIAL ACTIVITIES OF RUTA ANGUSTIFOLIA EXTRACT

Ruta angustifolia was used in this study in order to evaluate the antimicrobial activity and antioxidant properties and its correlation with the polyphenolic content. Two Gram-positive bacteria (Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778) and two Gram–negative bacteria (Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 8739) were used to determine the antibacterial activity. Aqueous maceration extract was used for antioxidant activities and methanolic maceration extract was used for antibacterial activity. The antioxidant properties and activities were evaluated by using total phenolic content (TPC), total flavonoid content (TFC), DPPH free radical scavenging activity and beta-carotene bleaching method. Whereas, the antibacterial activity was examined using disc diffusion method against selected microorganism at concentration 1.0 mg/disc. The results showed the phenolic content of R. angustifolia extract was 18.89 g GAE/100 g extract while the flavonoid content was 14.170 g QE/100 g extract. R. angustifolia exhibited good radical scavenging with IC50 value of 2.04 mg/ml. The result for disc diffusion method showed no inhibition zone against all the strains of bacteria at 1.0 mg/disc concentration of the extract. Based on the results, it can be concluded that the R. angustifolia aqueous extract has the antioxidant properties and there is correlation between polyphenolic content of the extract with its antioxidant activity. However, R. angustifolia methanolic extract did not show any antibacterial activity.

Apart from being the primary source of food to other living things, plants also have medicinal value to treat various kinds of diseases. In recent years, it has been proposed that the extract from plants may be used as natural antioxidants which can help to prevent the generation of carcinogens in human body. In addition, plants also have antimicrobial agents to inhibit the growth of pathogenic microbes. This study was intended to investigate the antioxidant properties and antimicrobial activity of methanolic extract of Tinospora crispa stems extracted using soxhlet extraction method. The antimicrobial properties of T. crispa stems extract were tested using disc diffusion method against Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778, Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853, Candida albicans IMR C S23/11 A and Saccharomyces cerevisiae IMR S 617/068. The antioxidant properties of the extract were investigated by using Total Phenolics Content (TPC), Total Flavonoids Content (TFC), DPPH free radical scavenging and b–carotene bleaching assays.  The TPC value was 6.12 g GAE/100 g of dried extract while the TFC value was 55.58 g QE/100 g of dried extract. The IC50 of DPPH scavenging assay for the extract and ascorbic acid were 0.21 and 0.04 mg/mL, respectively. The average percentage of b–carotene bleaching assay was 38.3 % as compared to BHT, which was 45.1%.  The disc diffusion method showed no inhibition zone against all the strains of microorganisms at all concentrations of the extracts (0.5, 1.0, 2.5 and 5.0 mg/disc).

In vitro antibacterial activity of 21 plant methanolic extracts were investigated by disc diffusion method against gram-negative bacteria Escherichia coli. Amongst the extracts tested, the Datura stramonium L. and Ocimum basilicum L. showed significant antibacterial activity against the bacterial pathogen. D. stramonium showed the highest antibacterial activity followed by O. basilicum extract. Phytochemical analysis of extracts showed the presence of alkaloids, tannins, saponins, flavonoids, and phenols. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ABTS analysis of potent extract D. stramonium showed appreciable antioxidant activity. For DPPH and ABTS assay the highest antioxidant activity was observed at 60 mg ml-1 concentration with a percent inhibition of 71.66 and 77.19 with IC50 value 9.71 and 22.16 μg ml-1 respectively. Moreover, the radical scavenging activity of the extract was lower than that observed for the synthetic antioxidants butylated hydroxyanisole (BHA )and butylated hydroxytoluene (BHT). The total phenolic content of D. stramonium extract was found to be 0.592 mg GAE/gm which was assayed using Folin ciocalteau reagent. The results provide evidence that the extract of D. stramonium and O. basilicum can be further recommended in the treatment of the infections caused by E. coli; further D. stramonium is also a potential source of natural antioxidants.

Echinocystis lobata (Mich.) Torr. et Gray extracts were investigated for their bioactive compounds and their potential for natural antioxidant properties. In vitro antioxidant activity of methanol, ethanol, and water extracts were evaluated by 1,1-diphenyl-picrylhydrazyl (DPPH) radical scavenging activity. Total phenolic content (TPC) and total flavonoid content (TFC) of fruits and seeds were determined. TPC is a fairly constant value and varies weakly both among the different organs of Echinocystis lobata and among the geographical locations of the populations studied. Like other invasive species, Echinocystis lobata is superior to many other plants in terms of the total amount of flavonoids contained in its fruits and seeds. The dried fruits contain more flavonoids than the seeds. Seeds from Slovak populations have higher flavonoid content than those from Russian populations. The results demonstrated that Echinocystis lobata can be a good source of natural antioxidants and have a great potential for commercialization, especially in the pharmaceutical industry.

In this study, we were interested in comparing the influence of different extraction methods on antioxidant and antibacterial activities of <em>V.amygdalina</em> leaves extracts. The extracts were also analyzed for their total phenolic and flavonoid content.  The extraction methods used maceration, soxhlet, and fractionation, the evaluation of antioxidant activity using DPPH (1,1-diphenyl-2-picryl-hydrazyl) method and antibacterial activity by agar diffusion method. The total phenolic and flavonoids are determined by the spectrophotometric method.  Ethanol extract by fractionating on this method showed the highest antioxidant properties compared to other extracts with IC<sub>50 </sub>170 μg/mL in category potent antioxidant with total phenolic content 28.83 ± 2.62 mg GAE/g, and the total flavonoid content 18.78 ± 0.15 mg QE/g. Evaluation of antibacterial properties the extracts of <em>V. amyigdalina</em> showed moderate antibacterial activity against all bacteria tests with inhibition zone  6.1 ± 0.1 to  9.4 ± 0.9 mm at variation concentration 62.5 – 1000 μg/mL. The fractionation method using ethanol solvent is the best for the extraction of antioxidant compounds from <em>V. amygdalina</em>.

Mistletoes are being used for food or beverage and for variety of medicinal purposes. In Nepal, mistletoes are traditionally used as fodder and as remedy to cure various ailments. In this study, methnolic extracts of seven common mistletoe species (Helixanthera ligustrina, Macrosolen cochinchinensis, Scurrula elata, S. parasitica, Viscum album, V. articulatum and V. liquidambaricola) were analyzed for their phytochemical constituents and antioxidant potential. The total flavonoid content (TFC) and total phenol content (TPC) were quantitatively determined by aluminum chloride colorimetric and folin-cioalteau reagent methods, respectively. The TFC was found highest for V. album (31.5 ± 2.3 mg QE/g) and lowest for M. cochinchinensis (24.9 ± 2.3 mg QE/g). Similarly, highest total phenolic content was found in S. parasitica (32.9 ± 2.5 mg GAE/g) and lowest in V. album (20.6 ± 2.1 mg GAE/g). The antioxidant activity measured by DPPH radical scavenging assay was found best for S. parasitica (with lowest IC₅₀ value; 26.0 ± 0.7 µg/ml) among the studied mistletoes. Overall analysis provides scientific evidence in favor of indigenous use of these mistletoes as a source of fodder, food and for medicinal purposes

The current investigation is intended to evaluate the content of phytochemical constituents and antioxidant potential of hydroalcoholic extracts of stem and root of Grewia serrulata DC (HAESGS & HAERGS) and leaf and bark of Grewia Nervosa (Lour.) panigrahi (HAELGN & HAEBGN). Initially, all the extracts at different concentrations were estimated for their total phenolic content and total flavonoid content. The study was further extended for their antioxidant potential evaluation using various in vitro methods such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical scavenging assays. The total phenolic content (mg gallic acid equivalent per gram of extract) was high in HAELGN (170.82±0.19) and HAERGS (123.00±0.48) than HAESGS (111.2±0.26) and HAEBGN (119.60±0.23). The total flavonoid content (mg quercetin equivalent per gram) is greater in HAERGS (71.24±0.50) and HAESGS (65.68±0.27) than HAELGN (55.82±0.35) and HAEBGN (62.38±0.45). The IC50 values (µg/ml) of different plant extracts inferred that DPPH radical scavenging activity is greater in HAELGN (42.91±0.88) and HAEBGN (53.87±0.35) than HAESGS (126.73±1.20) and HAERGS (88.87±1.25). However, hydroxyl and superoxide radical scavenging activity is more in HAERGS (135.41±1.19 & 88.00±1.42) and HAELGN (172.28±1.91 & 108.163±1.09) than HAESGS (237.3±1.65 & 110.074±0.87) and HAEBGN (204.7±1.04 & 125.54±1.07). The results of present comprehensive analysis demonstrated that both the plants Grewia serrulata DC and Grewia Nervosa (Lour.) panigrahi possess high phenolic, flavonoid contents and potential antioxidant activity, and could be used as a valid source of natural antioxidants and might be utilized for pharmacological screening of various therapeutic activities. Plant extracts are an important alternative to antibiotics, which are ever more restricted because of their developing microbial resistance and some adverse effects that have been observed following frequent application. The aim of the present study was to determine the antifungal and antioxidant activity of the methanolic extracts of Acorus calamus, Chlorella vulgaris, Lemna minuta and Scenedesmus dimorphus. The antifungal activity of the extracts against strains of Aspergillus flavus, Aspergillus parasiticus, Aspergillus ochraceus, Aspergillus niger, Aspergillus carbonarius, Fusarium graminearum, Fusarium oxysporum, Penicillium chrysogenum and Alternaria alternata was evaluated via the agar well diffusion method. The antioxidant activity of the extracts was measured through the determination of three parameters—total phenolic content, total flavonoid content and radical scavenging potential (determined through UV/Vis analysis). A. calamus extracts had the highest antimicrobial activity against eight fungal strains, followed by the C. vulgaris, L. minuta and S. dimorphus extracts, which were inhibitory against two to three strains. Among the extracts from the species studied, the extract from S. dimorphus showed the highest antioxidant potential, as determined via the DPPH (1,1’-diphenyl-2-picrylhydrazil-radical) method. This correlated to its high total phenolic and flavonoid content. From A. calamus and L. minuta, methanolic extracts were obtained that exhibited similar values of the aforementioned parameters, followed by C. vulgaris extracts, which showed the lowest antioxidant activity. Based on the Pearson correlation coefficients, the impacts of the total phenolic content and the total flavonoid content on radical scavenging capacity are similar, and flavonoids were a significant part of the total phenolic compounds extracted from the plant materials studied.

Fungal food spoilage plays a key role in the deterioration of food products, and finding a suitable natural preservative can solve this problem. Therefore, antifungal activity of green mandarin (Citrus reticulata) essential oil (GMEO) in the vapor phase against the growth of Penicillium (P.) expansum and P. chrysogenum inoculated on wheat bread (in situ experiment) was investigated in the current research. The volatile compounds of the GMEO were analyzed by a gas chromatograph coupled to a mass spectrometer (GC–MS), and its antioxidant activity was determined by testing free radical-scavenging capacity (DPPH assay). Moreover, the disc diffusion method was used to analyze the antifungal activity of GMEO in in vitro conditions. The results demonstrate that the Citrus reticulata EO consisted of α-limonene as the most abundant component (71.5%), followed by γ-terpinene (13.9%), and β-pinene (3.5%), and it displayed the weak antioxidant activity with the value of inhibition 5.6 ±0.7%, which corresponds to 103.0 ±6.4 µg TEAC.mL-1. The findings from the GMEO antifungal activity determination revealed that values for the inhibition zone with disc diffusion method ranged from 0.00 ±0.00 (no antifungal effectiveness) to 5.67 ±0.58 mm (moderate antifungal activity). Finally, exposure of Penicillium strains growing on bread to GMEO in vapor phase led to the finding that 250 μL.L-1 of GMEO exhibited the lowest value for mycelial growth inhibition (MGI) of P. expansum (-51.37 ±3.01%) whose negative value reflects even supportive effect of the EO on the microscopic fungus growth. On the other hand, GMEO at this concentration (250 μL.L-1) resulted in the strongest inhibitory action (MGI: 54.15 ±1.15%) against growth of P. chrysogenum. Based on the findings it can be concluded that GMEO in the vapor phase is not an effective antifungal agent against the growth of P. expansum inoculated on bread; however, its antifungal potential manifested against P. chrysogenum suggests GMEO to be an appropriate alternative to the use of chemical inhibitors for bread preservation.

Propolis is one of the natural products produced by kelulut bees and is still not widely used. The type of stingless bee that is the prima donna in the community is Heterotrigona itama. This study aims to determine the phytochemical content of the n-hexane fraction of Heterotrigona itama bee propolis collected from Kutai Kartanegara, East Kalimantan. The n-hexane fraction was obtained from the methanol extract of H. itama propolis by the liquid-liquid partition method. After obtaining the n-hexane fraction, the research continued with a qualitative phytochemical test to identify the compound and determine total phenolic. Antibacterial activity was determined by the agar well diffusion method with a serial concentration in Escherichia coli bacteria. Qualitative phytochemical analysis in the form of color changes showed that the n-hexane fraction of H. itama propolis contained flavonoids, alkaloids, saponins, and tannins. Based on the results, the total phenolic content of the n-hexane fraction sample was 490 mgGAE/100 g. It caused the n-hexane fraction to have lower phenolic content than the methanol extract (792 mg GAE100 g). Furthermore, this result indicated that the non-polar fraction was not substantial enough to extracted phenolic compounds. It correlated to the antibacterial activity of the n-hexane fraction, which was very weak (2  mm ± 1.5) at  200µg/mL concentration.

This study was based on screening antibacterial activity of the ethanol extract ofBaeckea frutescensL. against MRSA clinical isolates, analyzes the potential antibacterial compound, and assesses the cytotoxicity effect of the extract in tissue culture. Leaves ofBaeckea frutescensL. were shade dried, powdered, and extracted using solvent ethanol. Preliminary phytochemical screening of the crude extracts revealed the presence of alkaloids, flavonoids, steroids, terpenoids, phenols, and carbohydrates. The presence of these bioactive constituents is related to the antibacterial activity of the plant. Disc diffusion method revealed a high degree of activity against microorganisms. The results confirm thatBaeckea frutescensL. can be used as a source of drugs to fight infections caused by susceptible bacteria.
