DEVELOPMENT OF COMPOSITION, TECHNOLOGY AND ANALYSIS OF GRANULES WITH LARYNX TREE EXTRACT LIQUID

The purpose of the study was to substantiate the optimal composition, development of technology and methods for analyzing a new dosage form - granules with hydrangeas with a liquid tree extract. Optimal auxiliary substances for granule production were experimentally selected and process parameters were determined: fractional composition, abrasion, disintegration, mass loss during drying, bulk density. It was found that for all indicators granules with hydrangea tree extract liquid meet the requirements of GF XIV. Process diagram of granules manufacturing by wet granulation method has been developed. The TLC method made it possible to identify rutin, quercetin, luteolin, chlorophyll, coumarin. The method of differential spectrophotometry was tested for the method of quantitative determination of the sum of flavonoids. The proposed quantification methodology has been found to be suitable for analytical purposes. The content of the sum of flavonoids in the granules in terms of rutin is about 0,0225%. The relative error of the technique does not exceed ± 2,5%.

Biologically active sulfur-containing compounds (BASC) exhibit pronounced antioxidant properties. Glutathione reduced (GSH) occupies a particular position among these compounds. It represents a key link in the 3 antioxidant systems of the body from the existing four. Based on the foregoing, a GSH-based dosage form with antioxidant properties was proposed. The aim of this study is to work out a model granulated dosage form based on GSH and methods of its analysis by means of pre-column derivatization with ortho-phthalic aldehyde.Materials and methods.GSH and granulated dosage form based on GSH obtained by wet granulation were used as the object of the study. Quantitative evaluation of GSH content in the obtained granules was carried out using pre-column derivatization by the method of reversed-phase high-performance chromatography (RP HPLC). Ortho-phthalic aldehyde was used as a derivatizing agent. A diode-array detector was used to detect the resulting derivative. Ortho-phthalic aldehyde was used as a derivatizing agent. A diode-matrix detector was used to find out the resulting derivative.Results.In the course of the work, a model dosage form was created – granules based on GSH. By reference to the recommendations on the dosage of the drug, the concentration of the active substance was selected. Lactose was chosen as an auxiliary component. Physical and technological characteristics of a model sample of granules with GSH and lactose as a filler were studied. A method of quantitative determination of GSH in granules using pre-column derivatization with ortho-phthalic aldehyde was developed and validated by HPLC. The method of quantitative determination of GSH in granules with the use of pre-column derivatization by ortho-phthalic aldehyde by HPLC was developed and validated.Conclusion.The developed granulated dosage form meets the requirements given in the pharmacopoeial item “Granules” according to the analyzed indicators. Using the validation evaluation it was established, that the developed methods for the quantitative determination of GSH in granules is correct, precise and specific.

Chamomilla recutita is used in scientific medicine. Tripleurospermum inodorum (L.) Sch. Bip. is widely spread among possible impurities of Chamomilla recutita (L.) Rauschert. This plant is perspective for establishment into scientific medicine but it can’t change Chamomilla recutita (L.) Rauschert. Purpose of research: development of a method for the quantitative determination of the amount of flavonoids in terms of rutin in Tripleurospermum inodorum’s flowers using differential spectrophotometry. Materials and methods. The samples prepared in different parts of Russia were used as objects of research. (2017 – 2020). Spectral studies were carried out in the wavelength range of 350-430 nm with a step of 1 nm using a spectrophotometer SF-2000. Results. To determine the analytical wavelength, the UV spectra of alcohol extraction of Tripleurospermum inodorum’s flowers were studied. Maximum of absorption was noticed at wavelength 370 nm. The differential spectrum of the same extraction with an aluminum chloride solution of 2% in 96% alcohol has a maximum at a length of 410±2 nm, which coincides with the maximum of the Standard Sample (SS) of rutin. The largest number of flavonoids is extracted by 70% alcohol. The maximum optical density and the highest output of the number of flavonoids from the raw material is observed at a degree of grinding of 2 mm with a single extraction for 60 minutes. In the conditions of complexation, the optimal ratio of the volume of the test solution and aluminum chloride with a solution of 2% in 96% alcohol is the ratio of 1:1. The stability of the complex with an aluminum chloride solution of 2% in 96% alcohol is observed in 40 minutes after the start of the reaction and retains it for 30 minutes. Findings. The method is developed and the parameters of the quantitative determination of the amount of flavonoids in Tripleurospermum inodorum flores are determined in terms of rutin using differential spectrophotometry.

BACKGROUND: Pugun tano extract had been studied for its effect as hepatoprotector. However, the usage of the plant in the form of extract has a limitation, especially if the extract is consumed by the people due to the unpleasant taste and odour. Then, the extract needs to be transformed into a particular dosage form, such as a capsule. But before the capsule can be produced, a preformulation study of pugun tano extract into a granule mass in capsule need to be evaluated. AIM: The study aimed to formulate the ethanolic extract of pugun tano (Curanga fel-terrae (Lour.) Merr) as granule mass in the capsule dosage form. METHODS: The pugun tano ethanolic extract was formulated in several steps included preparation of dry extract using coating method with polyvinylpyrrolidone (PVP) and granule mass production. The excipients used for the granule mass were lactose granules (made with tapioca starch using wet granulation), corn starch (made with 3 concentrations of 5% (F1), 7.5% (F2) and 10% (F3)), talcum, magnesium stearate, methylparaben, and propylparaben. The granule mass was evaluated for the bulk density, tapped density, inter-particle porosity, Carr’s index, Hausner ratio, angle of repose, and flowability. RESULTS: The results showed that all of the formulae passed the requirement of the preformulation test. The bulk density of the granule mass was 0.79 – 0.86 g/ml; the tapped density was 0.88 – 0.90 g/ml; the inter-particle porosity was 0.03 – 0.14; the Carr’s index was 2.71 – 11.94%; the Hausner ratio was 1.09-1.12; the angle of repose was 26.10 – 28.90°; and the flowing time was 5.97 – 6.63 seconds. All of the formulae showed good flowability and free-flowing properties. CONCLUSION: It is concluded that the obtained formula

The article is devoted to the assessment the error in determining the mass of a sample bulk materials when using the values of their bulk density. Precisely the accuracy of measurement results is necessary in many fields of activity to ensure the efficiency and safety of technological processes, as well as improving product quality. The relevance of this work is associated with the assessment of the inevitable deviations of the bulk density the same material in a given volume as a result of an uncontrolled fractional composition of a sample of bulk material. As a consequence of these deviations, an error arises in determining the mass of a sample of a given volume, even if this volume is an uniquely determined quantity. The paper presents the results of studies of the influence of the fractional composition of bulk materials on the determination of their mass in the indirect measurement method. A significant effect of the fractional composition of bulk materials on the error of the indirect method for determining the mass of the sample was revealed. It is shown, that there is a need for fractional control of the composition of bulk materials in the development of control operations of technological processes, as well as restrictions on the use of bulk density with increased requirements for the accuracy of determining the mass of a sample.
