Withdrawal effects of chronic androgenic anabolic steroid on hormonal andtesticular morphology were studied .Forty five male albino rats were divided into 1) Control 2)Chronic group and 3) Withdrawal group. Testoviron was injected at a dose of 400mg/kg bodyweight intramuscularly once in two weeks for 14 weeks, and then drug was withdrawan foranother 14 weeks. Testes were removed and fixed in 10% formalin and processed. Followingwithdrawal of AAS, testicular and relative testicular weight was restored to control. Increasedtubular count also returned near to normal while decreased diameter of seminiferous tubules,thickness of germinal epithelium, count and diameter of leydig cells were also restored nearto normal in withdrawal group when compared with the chronically treated group. Histologicalobservations also revealed that degenerated spermatogenic cells were returned to their normalappearance and oedematous vacuoles were reduced. Moreover, decreased level of reproductivehormones, i.e. FSH, LH and testosterone also returned to control level in withdrawal group.These results indicated that chronic AAS has substantial harmful effects on hormonal andtesticular morphology. However, these adverse effects gradually restored to normal followingwithdrawal from AAS.

Tramadol is a synthetic, centrally acting analgesic. It is the most consumed narcotic drug that is prescribed in the world. Tramadol abuse has dramatically increased in Egypt. Long term use of tramadol can induce endocrinopathy. So, the aim of this study was to analyze the adrenal insufficiency induced by long term use of tramadol in experimental animals and also to assess its withdrawal effects through histopathological and biochemical genetic study. Forty male albino rats were used in this study. The rats were divided into 4 groups (control group, tramadol-treated group, and withdrawal groups). Tramadol was given to albino rats at a dose of 80 mg/kg body weight for 3 months and after withdrawal periods (7–15 days) rats were sacrificed. Long term use of tramadol induced severe histopathological changes in adrenal glands. Tramadol decreased the levels of serum cortisol and DHEAS hormones. In addition, it increased the level of adrenal MDA and decreased the genetic expression of glutathione peroxidase and thioredoxin reductase in adrenal gland tissues. All these changes started to return to normal after withdrawal of tramadol. Thus, it was confirmed that long term use of tramadol can induce severe adrenal insufficiency.

Anticancer drugs like doxorubicin have been found to affect male gonads thereby leading to infertility. This study was conducted to evaluate the effects of doxorubicin over short, mid and long term on testes of male albino rats. Sixty male albino rats aged 6-8 weeks were taken for study. The rats were randomly divided into 3 groups of experimental (each group containing 10 rats) and 3 groups of control (each group containing 10 rats). The experimental groups were given a single dose of doxorubicin i.e. 10 mg/kg body weight intra-peritoneally and sacrificed after 3 different duration for each group (second week, eighth week and sixteenth week). All rats under 3 control groups were given a single intra-peritoneal dose of 2.5 ml/kg body weight normal saline and sacrificed with their respective experimental groups. Significant difference in diameters (p=0.029) and cross-sectional area (p=0.028) of seminiferous tubules was observed between short term experimental and short term control rats. For both between midterm experimental and midterm control group, and between long term experimental and long term control group, a significant difference in right testis weight (p<0.001 for both), left testis weight (p<0.001 for both), volume of testis (p<0.001 and p=0.038), diameter (p<0.001 for both) and area (p<0.001 for both) of seminiferous tubules was observed. As compared to short term experimental group, midterm experimental group and long term experimental group had significantly lower right testis weight (p<0.001 for both), left testis weight (p<0.001 for both), diameter of seminiferous tubule (p<0.001 for both) and cross-sectional area of seminiferous tubule (p<0.001 both). Cross-sections of the seminiferous tubules of all the control groups had normal architecture. However, there was progressive destruction of seminiferous tubules structure across the experimental groups. Objective: To study the effects of mobile phone induced Electromagnetic fields (EMFs) on testis of young mice. Settings: Department of Anatomy, College of Physicians and Surgeons Pakistan, Regional Centre, Islamabad. Period: January to June, 2008. Study Design: Experimental animal study. Materials & Methods: This study was conducted on two groups of young BALB-c (6 weeks of age) purchased from National Institute of Health Islamabad. These animals were divided into two groups control and treated, each consisting of twenty animals. The treated group was exposed for one month to mobile phone induced EMFs by placing a mobile phone in the floor of the cage. This phone was rung upon from any other line or cell phone twice daily for 15 minutes. The control group was kept under identical conditions except for mobile phone on the cage floor. Results and observations: Histological comparison of testis of the both group animals showed a significant increase, in the number of tubules with sperms in the lumen, increased sub capsular congestion of vessels, presence of vacuolation and giant cells in germinal epithelium and abnormal cells in the lumen of seminiferous tubules of the treated group. Conclusion: The results indicate altered testicular morphology of the EMFs exposed mice.

Vascular disease and heart attacks are a common phenomenon in the modern world, where lipid accumulates inside blood vessels and causes plug, eventually heart stroke and death. Utilize lipidlowering drugs are the most effective factors to prevent heart disease. Atorvastatin is the most important drugs that used in this field. The aim of this study was to investigate the adverse effects of atorvastatin (20 mg/kg/b.wt/day) on some organs of albino. 12 adult male albino rats weighing 200 ± 20 g were used and divided into two groups: First group served as control group, was given 0.72 ml distilled water/rat. The second group was subjected to treatments with atorvastatin in a dose of 20 mg/kg/b.wt/day as a single daily dose (0.72 ml contained 1.44 mg of atorvastatin) orally by gastric tube for 4 weeks. Histological alterations were examined, and the results of atorvastatintreated groups showed changes in the kidney sections, which include focal tubular edema and glomerular atrophy, also liver sections revealed foci of lymphocyte accumulation, cytoplasmic vacuolization, as well as sections of the testes showed degenerative changes in the seminiferous tubules. According to this study, we can conclude that atorvastatin-induced various deleterious changes in the histological structure of the testes, kidneys and cause mild liver alteration of adult male albino rat.

Endosulfan is a worldwide used synthetic insecticide that has an important role on management of pests in agriculture. The present work was undertaken to determine the effect of endosulfan on the haematological and haemochemical parameters of albino rats. Rats were feed with 5 mg/kg body weight endosulfan in mixed food stuff for 42 days. The studies were conducted on sexually matured male rats covering five groups of animals with control. Total counts of erythrocytes and haemoglobin were decreased and leucocytes were increased in treated group. Differential counts of leucocytes showed significant increase in basophils and monocytes. The levels of serum glucose, urea, creatinine and bilirubin increased significantly, suggesting that the synthetic insecticide had remarkable toxic effects on the haematological and biochemical parameters in the experimental animals. Abstract Background Sildenafil (Viagra) is widely used for the treatment of erectile dysfunction under various etiologies. Unfortunately, chronic administration of sildenafil negatively affects fertility. The aim of the present study is to investigate the improvement role of sesame on the testis alterations caused by long-term administration of sildenafil on albino rats. Results Light microscopically observations showed that overdoses of sildenafil had provoked tubular and interstitial histological alterations. Tubular degeneration and abnormality in the germinal epithelium of the seminiferous tubules included spermatocyte degeneration and arrest of spermatogenesis. Sperm count and motility were negatively affected by sildenafil. These alterations were improved by sesame feeding. Conclusion Sesame intake improve the side effects of prolonged treatment with sildenafil significantly. These results demonstrate the pivotal role of sesame that may improve male infertility.

ABSTRACT The amount of testosterone required for quantitative maintenance of spermatogenesis has been re-evaluated using techniques aimed at minimizing the synthesis of testosterone after removal of the testis. Adult male rats were treated with ethane dimethane-sulphonate (EDS) to destroy the Leydig cells, and were supplemented with 25, 5 or 1 mg testosterone esters by injection every 3 days for 21 days. Serum hormone levels, testicular morphology and spermatogenesis were assessed and the intratesticular levels of testosterone compared in testes either removed under ether anaesthesia and placed in liquid nitrogen (right testis) or removed after collection of blood and placed in ice (left testis). Data for testosterone-supplemented rats were compared with those for control rats and rats treated with EDS alone. All doses of testosterone suppressed LH and FSH levels in serum to within the hypophysectomized range, and Leydig cell regeneration in EDS-treated rats was prevented completely. Treatment of EDS-injected rats with 25 or 5 mg testosterone maintained testicular weight, the number of germ cells and the diameter of seminiferous tubules at stage VII within or above the control range, although there was a significant increase in the number of degenerating pachytene spermatocytes at stage VII with 5 but not 25 mg testosterone; none of these parameters was maintained at control levels by a dose of 1 mg testosterone. Levels of testosterone in testosterone-supplemented rats differed little between testes collected in ice and liquid nitrogen, but in controls and rats treated with EDS alone, testosterone levels were overestimated by 75 and 27% respectively when comparing testes collected in ice with those collected in liquid nitrogen. This suggests that synthesis of testosterone does occur after removal and cooling of the testes. Using the data for testes collected in liquid nitrogen, the present results suggest that intratesticular levels of testosterone need to be maintained at 24–46% of control values (i.e. higher than previously shown) for quantitative maintenance of spermatogenesis, although it is argued that even these values probably still represent underestimates. The effects of methanolic extract of Dracaena arborea on mean testicular weight, mean cauda epididymal sperm reserve, and testicular morphology were evaluated. A total of sixty mature male Sprague Dawley rats were divided into three equal groups. The first group (A) received distilled water while the other two groups (B and C) received orally the methanolic leaf extracts of Dracaena arborea in two doses (100&nbsp;and 500 mg/kg, respectively) daily for 84 days. Following oral administration of the extract, mean testicular weight, mean cauda epididymal sperm reserve, and testicular morphology were determined on days 28, 42, 56, 70, and 84. The extract produced a significant and dose-dependent increase (P &lt; 0.05) in the sperm number. There was also a significant increase (P &lt; 0.05) in the mean testicular weights on days 70 and 84 of the extract administration. The testicular morphology remained unchanged while further testicular histology examination revealed increased spermatogenesis. It was concluded that the methanolic leaf extract of D. arborea has fertility enhancing properties. &nbsp;

Cadmium is an industrial pollutant that may exert specific toxic effect on mammals. The aim of study was to investigate the protective effect of Cinnamon on male reproductive system of albino rat induced by cadmium toxicity. Forty male albino rats were divided into 4 groups. 1st  Group administrated saline and kept as control.2nd Group administrated 200mg/kg / day Cinnamon extract. 3rd Group administrated 20mg/kg/day cadmium chloride.4th Group administrated 200mg/kg/day Cinnamon extract and 20mg/kg/day cadmium chloride orally for 8 weeks. . Our results revealed significant decrease in testosterone hormone level, sperm viability, and significant increase in sperm abnormality. An increase in expression of Nrf2 gene was recorded. Pathological changes in testes showed focal degeneration with loss of spermatogenic series in the seminiferous tubules. All of the above mentioned results were significantly improved in 4th group. In conclusion, Cinnamon extract has a protective effect on the testicular damage induced by cadmium chloride.
