This study aimed to investigate the phytochemical screening, total phenolic content, antioxidant and antimicrobial activities of Coleus forskohlii L. stem extract in Al-Baha area, Saudi Arabia. Stem samples were collected from Al-Baha area and air-dried followed by extraction with ethanol, petroleum ether, chloroform, ethyl acetate, and n-butanol. The extracts were then subjected to phytochemical screening, determination of total phenolic content, antioxidant and antimicrobial activities. Results showed the presence of flavonoids, alkaloids, tannins, terpenoids, steroids, saponins, and reducing sugars. Total phenolic content was significantly (P<0.001) higher in n-butanol extract (274.33±3.29 mg GAE/gm), followed by ethyl acetate extract (182.94±1.82 mg GAE/gm), ethanol extract (79.63±2.02 mg GAE/gm) and petroleum ether extract (73.38±3.07 mg GAE/gm), while the lowest content was in chloroform extract (60.06±2.12 mg GAE/gm). The antioxidant activity was significantly (P<0.001) higher in n-butanol extract (67.68±1.55%), followed by ethyl acetate extract (43.38±1.27%), ethanol extract (36.02±1.29%), petroleum ether extract (20.71±0.59%) and chloroform extract (19.73±0.74%). The antimicrobial activity showed that all microorganisms tested were resistant at the concentration of 25 and 50 mg/ml of plant extracts, whereas the concentrations of 100, 150 and 200 mg/ml showed varying activities against gram-negative (Escherichia coli, Pseudomonas aeruginosa), gram-positive (Staphylococcus aureus, Bacillus cereus) and Candida albicans. The study concluded that the stem extracts of C. forskohlli have promising pharmacological and biological activities that could be beneficial in pharmaceutical as well as food and medicinal industries.

Objective: In present study different extracts of the leaves of plant Casearia tomentosa was investigated for the phytochemical screening, total phenolic content and various pharmacological activities like antioxidant, antidiabetic, antibacterial.Methods: Leaves of C. tomentosa were successively extracted by soxhlet extraction method using petroleum ether, chloroform, ethyl acetate and ethanol. The preliminary phytochemical investigation of different extracts was carried out according to standard methods. Antioxidant activity evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay and ferric reducing antioxidant potential (FRAP) method. Total phenolic content was evaluated by Folin-ciocalteau method. Antidiabetic activity estimated by α-amylase and α-glucosidase inhibitory assay while antibacterial activity was performed by disk diffusion method.Results: C. tomentosa leaves show the presence of various phytochemicals like alkaloids, flavonoids, carbohydrates, glycosides, protein etc. Results of antioxidant studies revealed that the ethanol extract possessed highest 2, 2-diphenyl-1-picryl-hydrazyl (IC50 31.87±0.65 μg/ml) activity, ferric reducing antioxidant power (43.78±0.65μM/ml, FRAP value=2.30). Among different extracts, ethanol extract shows highest (228.17±0.63 mg gallic acid equivalents (GAE)/gram of dry mass) amount of total phenolic content. The ethyl acetate extract exhibited highest inhibition on α-amylase (IC50 262.08±1.26 μg/ml) and α-glucosidase (IC50 459.25±1.80 μg/ml) enzymes. Results of antibacterial activity revealed that all extracts show inhibition against selected bacterial strains like Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia and Salmonella typhi.Conclusion: The present study suggests that Casearia tomentosa leaves can be used as a potential source of natural antioxidant and natural antidiabetic, antibacterial agents.

Cogon grass, Imperata cylindrica (L.) Räuschel, was studied for its flavonoid and phenol contents. A series of extracts of the underground (rhizome-root) parts was prepared by hot extraction using solvents of different polarities such as chloroform, methanol, and petroleum ether. The total flavonoid content of the plant extracts was determined based on the reaction with aluminum chloride, sodium nitrite, and sodium hydroxide. Quercetin was used as a standard reference. Chloroform extract showed highest content of flavonoids with 30.88 mg QE/g dry wt., followed by petroleum ether extract which was 22.05 mg mg QE/g dry wt., and methanol extract has the least value with 7.35 mg QE/g dry wt. The total phenolic content of the plant was estimated based on the reaction of Folin-Ciocalteu reagent using gallic acid as the standard reference. Again, the chloroform extract had the highest content with 7.54 mg GAE/g dry wt., while methanol extract contained 5.03 mg GAE/g dry wt., and petroleum ether extract contained 3.63 mg mg GAE/g dry wt. Our study shows that I. cylindrica is a good source of antioxidants.

Objective: This study aimed to determine the total phenolic content, correlation of it with antioxidant capacity, and peanut hulls as an antiproliferationon Henrietta Lacks (HeLa) cancer cells, which Indonesia has a serious problem in term of cervix cancer.Methods: Peanut hulls were extracted by Soxhlet extraction, ultrasound vibration, and reflux boiling to obtain the best extraction method. The totalphenolic content of the ethanol extract and the ethyl acetate fraction was determined using Folin–Ciocalteu method. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay was used to evaluated an antioxidant capacity of both samples. Identification of the isolated flavone was done by ultraviolet spectra and analyzed using liquid chromatography-mass spectrometer. Inhibition of proliferation of HeLa cancer cells was tested for the purified fraction using 3-(4,5-dimethyl thiazole-2-yl)-2, 5-diphenyltetrazolium bromide assay.Results: The results of total phenolic content determination giving 262 (ethanol extract) and 532 (ethyl acetate fraction) mg gallic acid equivalent/g extract, respectively. DPPH assay resulted antioxidant capacity with value of inhibitory concentration 50% (IC) was 36.36 (ethanol extract) and 18.68 (ethyl acetate fraction) µg/mL, respectively. Identification of isolated flavone resulted an apigenin and indicated moderate potency in inhibiting the proliferation of HeLa cancer cells with IC 50 value of 34 µg/mL. 50Conclusions: There was a correlation between the total phenolics with antioxidant capacity of the peanut hulls. The isolated flavone is predominated by apigenin. This isolated compound is potential as antioxidant and inhibiting the proliferation of HeLa cancer cells moderately.Keywords: Peanut hulls, Total phenolic, Antioxidant capacity, Flavone, Apigenin, Henrietta Lacks cancer cells.

Objective: This study was to investigate the phytochemical screening, total phenolic content (TPC), and antioxidant activity of fractions of water, ethyl acetate, and n-hexane from Dendrophthoe pentandra (L.) Miq. and mistletoe of Moringa oleifera Lam.Methods: Simplicia extracted with 96% ethanol using maceration method. Liquid-liquid extraction was used for fractionation with water, ethyl acetate, and n-hexane as solvent. Phytochemical screening, determination of TPC, and antioxidant activity were performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method. The TPC was measured by Folin–Ciocalteu reagent and use gallic acid (GA) as a standard.Results: Water fraction contains saponin and polyphenols. Ethyl acetate fraction contains alkaloids, flavonoids, saponins, terpenoids, and polyphenols. N-hexane fraction only contains alkaloids and terpenoids. The TPC and antioxidant activity of fractions of water, ethyl acetate, and n-hexane were 9.42±1.16%, 21.35±1.86%, and 67.40±0.82% mg GA equivalent/g fraction and 29.46 μg/ml±0.99%, 7.08 μg/ml±0.39%, and 10.90 μg/ml±1.05%, respectively.Conclusion: Fraction of ethyl acetate exhibited the highest antioxidant activity and TPC. This study showed that there were significant correlations between TPC and antioxidant activity at significance level p<0.05.

Paspalum scrobiculatum, commonly known as ‘Kodo’ is a millet variety found in India having medicinal and pharmaceutical importance. Crude extracts of chloroform, ethyl acetate, ethanol and methanol were evaluated for the presence of total phenolic content and total flavonoid content of the millet grains. Phytochemical screening was done for a qualitative basis of the study. Phytochemical tests showed the presence of phenolics and flavonoids in more polar solvent, methanol and ethanol. Carbohydrates and fats and oils were present in all four extracts.  The result of total phenolic content by Folin- ciocalteu method with gallic acid as standard at 765 nm ranged from 40.15 ± 0.17 to 175.94 ± 1.018 µg/100µg extract. Ethanol extract showed maximum phenolic content. The total flavonoid content at 510 nm using rutin as standard ranged from 4.88 ± 0.50 to 116.48 ± 1.57 µg/100µg extract. Highest flavonoid content was found in ethanol extract. Presence of good amount of phenolic and flavonoid content indicates presence of phytochemicals in kodo millet having good antioxidant potential.Keywords: Paspalum scrobiculatum, Phytochemical, TPC, TFC, Ethanol, etc.

Solanum sisymbriifolium Lam. (Solanaceae), commonly referred to as Kantikari (Bengali), sticky nightshade, or litchi tomato. Despite, the traditional use of this plant for several ailments, very little is known about the phytochemical and pharmacological content of the edible ripe berries of this medicinal plant. Ethanol and petroleum ether extract of the ripe and unripe fruits of Solanum sisymbriifolium were examined by using standard analytical methods to determine and compare their bioactive chemical constituents, total phenolic content, antioxidant, and cytotoxic potential. Quantitative phytochemical screening showed that the presence of medicinally active secondary metabolites like alkaloids, flavonoids, and saponins were detected in both ripe and unripe fruits. Antioxidant activity and total phenolic content were found to be more pronounced in ripe fruit extract, whereas unripe fruit extracts showed moderate cytotoxic activity. DPPH free radical scavenging method demonstrated that IC50 value of ethanol and petroleum ether extract of ripe fruits were 194.40μg/ml and 436μg/ml, and for unripe fruit the values were 689.82μg/ml and 1411.23μg/ml. In brine shrimp lethality bioassay, the LC50 value of unripe fruits was 230.6559μg/ml for ethanol extract and 314.9227μg/ml for petroleum ether extract. The LC50 value of the ethanol extract of ripe fruits was 456.1021μg/ml and for petroleum ether extract was 1,017.6848μg/ml. The results of this study showed that the litchi tomato not only has the potential to be a rich source of an edible compound due to the presence of profound phytochemical constituents but also can be used as a source of pharmacological references.

Prunus domestica L. is a member of the Rosaceae family that shows many biological activities including antioxidant, antimicrobial, antihaemolytic, anti-inflammatory, hepatoprotective activity and many other activities. In the current study, we evaluated nutritive value, phytochemical screening, total phenolic content and antioxidant activity by DPPH and FRAP method for the different extracts obtained by successive soxhlet extraction using the different solvents based on their polarity. Results show that it is a good source of energy. Phytochemical screening revealed the presence of many secondary metabolites which include alkaloids, carbohydrates, glycosides, protein, steroids and terpenoids, fixed oils and fat as well as phenolic compounds. The highest total phenolic content was found in the ethyl acetate fraction. Highest antioxidant activity by DPPH method is reported in ethyl acetate fraction (IC50 =1837.399±0.377µg/ml) while the ferric reducing antioxidant power was maximum for diethyl ether (56.032±0.985µM/ml FRAP value = 0.325±0.002).

The methanol, petroleum ether, chloroform, ethyl acetate, 1-butanol and water extracts were obtained by extraction of mountain germander (Teucrium montanum L). The total phenolic content in extracts was measured by Folin-Ciocalteu method. The 1-butanol extract had the highest phenolic content (296.00 mg/g). High performance liquid chromatography (HPLC) was employed to define qualitative and quantitative content of phenolic acids in mountain germander extracts. The largest number of phenolic acids were determined in ethyl acetate and 1-butanol extracts, while these acids were not present in petroleum ether extract. The highest content of phenolic acids (28.619 mg/g) had ethyl acetate extract and gentisic acid (14.432 mg/g) was its major component. Despite of a large number of phenolic acids in 1-butanol extract their content was only 3.740 mg/g.

Parkia speciosa (Fabaceae),  much grow in South Sumatera. The seed of P. speciosa used as traditional medicine for  diabetes mellitus therapy. Another parts of P speciosa is suspected to have the same chemical compounds and potency as the seed. Based on phytochemical screening of leaf and rind of P speciosa have secondary metabolites as flavonoid, phenolic and terpenoid.  This research aims to know effectiveness of alpha glucosidase inhibitory effect of ethanol extract of rind, leaf and  seed of  p. speciosa and its correlation to total phenolic content of the extracts. The inhibitory activity of the alpha glucosidase enzyme was measured at l 405 nm. The result showed that there was correlation between effectiveness of inhibitory  and total phenolic content of the extract, that is the higher of  the total phenolic content will caused the greater of enzymatic inhibition of extract. The  IC50 of alpha glucosidase inhibitory effect of ethanol extract of rind, leaf and seed of P speciosa are 4,596 ppm, 54,341 ppm, dan 67,425 ppm and the total Phenolic content of the extract are 138,15 mgGAE/g, 59,25 mgGAE/g, dan 36,25 mgGAE/g respectively.

Plant natural products have a long history of being a rich source of neuroactive drugs.The total phenolic and flavonoid content, and antioxidant activity of ethanol, chloroform and aqueous extracts of AlgerianMedicagoSativaL, leaves were measured to discover new potential sources of Algerian natural antioxidants and neuroactives. All extracts were non-neurotoxic up to 1 mg/ ml for 24 h exposure. The total phenolcontent of M.Sativa extracts ranged between 38 and 69 mg GAE per g extracts. All of the extracts displayed antioxidant capacityassessed by two methods, but at different levels depending on the concentration and the extraction solvent used. The overall antioxidant potency was in the order water > chloroform> ethanol extracts, and the phenolic content exhibited a positive correlation with the antioxidant activity of M. Sativa extracts.Despite a weaker antioxidant activity, the ethanol extract elicited a calcium spike and suppressed the KCl-induced depolarisation response in neurons, indicating effect on ligand-and/or voltage gated calcium channels. In contrast, the chloroform and water extractsdisplayed no apparent direct functional effects. In conclusion, the antioxidant and functional properties related to phenolic content of Algerian Medicago Sativa L. in water and ethanol extracts, respectively.
