Objective: This study aimed to determine the total phenolic content, correlation of it with antioxidant capacity, and peanut hulls as an antiproliferationon Henrietta Lacks (HeLa) cancer cells, which Indonesia has a serious problem in term of cervix cancer.Methods: Peanut hulls were extracted by Soxhlet extraction, ultrasound vibration, and reflux boiling to obtain the best extraction method. The totalphenolic content of the ethanol extract and the ethyl acetate fraction was determined using Folin–Ciocalteu method. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay was used to evaluated an antioxidant capacity of both samples. Identification of the isolated flavone was done by ultraviolet spectra and analyzed using liquid chromatography-mass spectrometer. Inhibition of proliferation of HeLa cancer cells was tested for the purified fraction using 3-(4,5-dimethyl thiazole-2-yl)-2, 5-diphenyltetrazolium bromide assay.Results: The results of total phenolic content determination giving 262 (ethanol extract) and 532 (ethyl acetate fraction) mg gallic acid equivalent/g extract, respectively. DPPH assay resulted antioxidant capacity with value of inhibitory concentration 50% (IC) was 36.36 (ethanol extract) and 18.68 (ethyl acetate fraction) µg/mL, respectively. Identification of isolated flavone resulted an apigenin and indicated moderate potency in inhibiting the proliferation of HeLa cancer cells with IC 50 value of 34 µg/mL. 50Conclusions: There was a correlation between the total phenolics with antioxidant capacity of the peanut hulls. The isolated flavone is predominated by apigenin. This isolated compound is potential as antioxidant and inhibiting the proliferation of HeLa cancer cells moderately.Keywords: Peanut hulls, Total phenolic, Antioxidant capacity, Flavone, Apigenin, Henrietta Lacks cancer cells.

Abstract Heracleum lasiopetalum Boiss, Kelussia odoratissima Mozaff., and Echinophora platyloba DC. belong the Apiaceae family. They are Iranian endemic plants. These three herbs have been used as food additives in traditional preparations such as pickles. Antioxidant activity (AA) of methanol extracts (ME) of the plants was evaluated by three assays, including DPPH, FRAP, and TEAC. From all three assays, comparing all the MEs for their IC50 and EC1 values, E. platyloba had the highest AA. Total phenolic content (TPC) of the extracts ranged from 74 to 120 mg TAE/g. The extract of H. lasiopetalum exhibited the highest TPC. The flavonoids content (FC) of the extracts ranged from 7.63 to 14.52 mg RE/g, from which the extract of E. platyloba had the highest flavonoids concentration. A positive correlation between the FC and AA in DPPH assay was found. A significant correlation was also found between the TPC and AA in FRAP assay. These results suggested that the level of AA in these plants varied in a great extent. Our results indicated that extract of E. platyloba could be an important dietary source of flavonoids compounds with high antioxidant capacity. In addition, E. platyloba can be used as an alternative preservative and natural flavor instead of synthetic ones in food industry (especially pickles)

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

This study aimed to investigate the phytochemical screening, total phenolic content, antioxidant and antimicrobial activities of Coleus forskohlii L. stem extract in Al-Baha area, Saudi Arabia. Stem samples were collected from Al-Baha area and air-dried followed by extraction with ethanol, petroleum ether, chloroform, ethyl acetate, and n-butanol. The extracts were then subjected to phytochemical screening, determination of total phenolic content, antioxidant and antimicrobial activities. Results showed the presence of flavonoids, alkaloids, tannins, terpenoids, steroids, saponins, and reducing sugars. Total phenolic content was significantly (P<0.001) higher in n-butanol extract (274.33±3.29 mg GAE/gm), followed by ethyl acetate extract (182.94±1.82 mg GAE/gm), ethanol extract (79.63±2.02 mg GAE/gm) and petroleum ether extract (73.38±3.07 mg GAE/gm), while the lowest content was in chloroform extract (60.06±2.12 mg GAE/gm). The antioxidant activity was significantly (P<0.001) higher in n-butanol extract (67.68±1.55%), followed by ethyl acetate extract (43.38±1.27%), ethanol extract (36.02±1.29%), petroleum ether extract (20.71±0.59%) and chloroform extract (19.73±0.74%). The antimicrobial activity showed that all microorganisms tested were resistant at the concentration of 25 and 50 mg/ml of plant extracts, whereas the concentrations of 100, 150 and 200 mg/ml showed varying activities against gram-negative (Escherichia coli, Pseudomonas aeruginosa), gram-positive (Staphylococcus aureus, Bacillus cereus) and Candida albicans. The study concluded that the stem extracts of C. forskohlli have promising pharmacological and biological activities that could be beneficial in pharmaceutical as well as food and medicinal industries.

Telah dilakukan penelitian tentang aktivitas penstabil oksigen singlet dari hasil fraksinasi ekstrak metanol daun kayu manis (Cinnamomum burmanii) terhadap fotooksidasi asam askorbat dengan menggunakan cahaya dan eritrosin sebagai sensitiser. Serbuk daun kayu manis diekstraksi secara maserasi dengan metanol 95 % selama 3 x 24 jam, selanjutnya ekstrak yang diperoleh difraksinasi berturut-turut dengan pelarut n-heksan, etil asetat, butanol dan air. Penentuan kandungan total fenolik diukur dengan metode Folin-Ciocalteu dan aktivitas penstabil oksigen singlet diukur dengan laju penurunan konsentrasi asam askorbat. Hasilnya menunjukkan bahwa fraksi etil asetat memiliki kandungan total fenolik tertinggi yaitu 166,888 mg/g diikuti dengan fraksi butanol, n-heksan, dan air.  Fraksi etil asetat juga memiliki aktivitas penstabil oksigen singlet yang kuat, ditunjukkan dengan laju kerusakan fotooksidatif asam askorbat yang kecil yaitu -0,75 μg.mL-1.min-1A study of singlet oxygen quenching activity has been done from the fractination result of methanol extract of cinnamon leaf (Cinnamomum burmanii) against photo-oxidation of ascorbic acid by using light and erythrosine as sensitized. The cinnamon leaf powder was extracted by macerating with 95% methanol for  3 x 24 hours, then the extracts obtained was fractionated successively with n-hexane, ethyl acetate, butanol and water solvent. The determination of total phenolic content was measured by the Folin-Ciocalteu method and the singlet oxygen quenching activity was measured by the rate of decrease in ascorbic acid concentration. The results show that the ethyl acetate fraction has the highest total phenolic content of 166,888 mg/g followed by the fraction of butanol, n-hexane, and water, respectively. The ethyl acetate fraction also has a strong singlet oxygen quenching activity, indicated by a small photo-oxidative damage rate of ascorbic acid of                  -0.75 μg.mL-1.min-1

Objective: In present study different extracts of the leaves of plant Casearia tomentosa was investigated for the phytochemical screening, total phenolic content and various pharmacological activities like antioxidant, antidiabetic, antibacterial.Methods: Leaves of C. tomentosa were successively extracted by soxhlet extraction method using petroleum ether, chloroform, ethyl acetate and ethanol. The preliminary phytochemical investigation of different extracts was carried out according to standard methods. Antioxidant activity evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay and ferric reducing antioxidant potential (FRAP) method. Total phenolic content was evaluated by Folin-ciocalteau method. Antidiabetic activity estimated by α-amylase and α-glucosidase inhibitory assay while antibacterial activity was performed by disk diffusion method.Results: C. tomentosa leaves show the presence of various phytochemicals like alkaloids, flavonoids, carbohydrates, glycosides, protein etc. Results of antioxidant studies revealed that the ethanol extract possessed highest 2, 2-diphenyl-1-picryl-hydrazyl (IC50 31.87±0.65 μg/ml) activity, ferric reducing antioxidant power (43.78±0.65μM/ml, FRAP value=2.30). Among different extracts, ethanol extract shows highest (228.17±0.63 mg gallic acid equivalents (GAE)/gram of dry mass) amount of total phenolic content. The ethyl acetate extract exhibited highest inhibition on α-amylase (IC50 262.08±1.26 μg/ml) and α-glucosidase (IC50 459.25±1.80 μg/ml) enzymes. Results of antibacterial activity revealed that all extracts show inhibition against selected bacterial strains like Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumonia and Salmonella typhi.Conclusion: The present study suggests that Casearia tomentosa leaves can be used as a potential source of natural antioxidant and natural antidiabetic, antibacterial agents.

Objective: This study was to investigate the phytochemical screening, total phenolic content (TPC), and antioxidant activity of fractions of water, ethyl acetate, and n-hexane from Dendrophthoe pentandra (L.) Miq. and mistletoe of Moringa oleifera Lam.Methods: Simplicia extracted with 96% ethanol using maceration method. Liquid-liquid extraction was used for fractionation with water, ethyl acetate, and n-hexane as solvent. Phytochemical screening, determination of TPC, and antioxidant activity were performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method. The TPC was measured by Folin–Ciocalteu reagent and use gallic acid (GA) as a standard.Results: Water fraction contains saponin and polyphenols. Ethyl acetate fraction contains alkaloids, flavonoids, saponins, terpenoids, and polyphenols. N-hexane fraction only contains alkaloids and terpenoids. The TPC and antioxidant activity of fractions of water, ethyl acetate, and n-hexane were 9.42±1.16%, 21.35±1.86%, and 67.40±0.82% mg GA equivalent/g fraction and 29.46 μg/ml±0.99%, 7.08 μg/ml±0.39%, and 10.90 μg/ml±1.05%, respectively.Conclusion: Fraction of ethyl acetate exhibited the highest antioxidant activity and TPC. This study showed that there were significant correlations between TPC and antioxidant activity at significance level p<0.05.

PENENTUAN TOTAL FENOLIK DAN AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DARI TEPUNG PELEPAH AREN (Arenga pinnata) ABSTRAK Tujuan penelitian ini untuk menentukan total fenolik dan aktivitas antioksidan fraksi etil asetat dari tepung pelepah aren (Arenga pinnata). Tepung pelepah aren diekstraksi maserasi menggunakan etanol hasil redestilasi cap tikus kemudian di partisi menggunakan pelarut petroleum eter, etil asetat dan aquades. Penentuan kandungan total fenolik menggunakan metode Folin-Ciocalteu dan aktivitas antioksidan menggunakan metode serapan radikal 1,1-difenil-2-pikrilhidrazil (DPPH). Didapati kandungan total fenolik fraksi etil asetat sebesar 172.857 ± 4.762 mg/kg dan aktivitas penangkal radikal bebas fraksi etil asetat sebesar 90.573 µg/mL. Hasil ini menunjukan bahwa fraksi etil asetat berpotensi sebagai antioksidan alami. Aren’s midrib powder extracted with maceration method with cap tikus redistillated ethanol and then partitioned with solvents such as petroleum ether, ethyl acetate and distillate water. The determination of phenolic content was using Folin-Ciocalteu method and the antioxidant activity was done using diphenylpycrylhydrazyl (DPPH) radical scavenging. Total phenolic value of ethyl acetate fraction is 172.857 ± 4.762 and IC50 value of ethyl acetate fraction is 90.573 µg/mL. This result shows ethyl acetate fraction as natural antioxidant potential. Gynura bicolor(Compositae) which is widely used by the locals as natural remedies in folk medicine has limited scientific studies to ensure its efficacy and nontoxicity. The current study reports the total phenolic content, antioxidant capacity, cytotoxicity, and acute oral toxicity of crude methanol and its fractionated extracts (hexane, ethyl acetate, and water) ofG. bicolorleaves. Five human colon cancer cell lines (HT-29, HCT-15, SW480, Caco-2, and HCT 116), one human breast adenocarcinoma cell line (MCF7), and one human normal colon cell line (CCD-18Co) were used to evaluate the cytotoxicity ofG. bicolor. The present findings had clearly demonstrated that ethyl acetate extract ofG. bicolorwith the highest total phenolic content among the extracts showed the strongest antioxidant activity (DPPH radical scavenging assay and metal chelating assay), possessed cytotoxicity, and induced apoptotic and necrotic cell death, especially towards the HCT 116 and HCT-15 colon cancer cells. The acute oral toxicity study indicated that methanol extract ofG. bicolorhas negligible level of toxicity when administered orally and has been regarded as safe in experimental rats. The findings of the current study clearly established the chemoprevention potential ofG. bicolorand thus provide scientific validation on the therapeutic claims ofG. bicolor.

Fruits are high in polyphenols which are compounds associated with the protection against diseases such as diabetes and cancer. However, food processing including canning can leads to the loss of polyphenol in the fruits. Thus, the aim of this study was to evaluate the total phenolic content (TPC) and antioxidant capacity of canned fruits commercially available in the local supermarkets in Malaysia. The TPC was determined by using Folin-Ciocalteu method while the antioxidant capacity was evaluated by using 2,2-diphenyl-1-picrylhydrazyl radical (DPPH assay) and Ferric Reducing Antioxidant Power (FRAP assay). Five types of different canned fruits with the total of 21 samples including longan, lychee, rambutan, pineapple and orange were selected for the analysis. The samples were analyzed in two forms which were, the homogenized (fruits and syrup) and syrup samples. Canned pineapples have the highest TPC for both homogenized (95.16±30.16 mg GAE/100 g) and syrup sample (108.62±33.88 mg GAE/100 g). For antioxidant capacity, canned pineapple also had the highest value for the homogenized sample (41.79±4.20 umol TE/100 g) while lychee was the highest (46.84±12.81 umol TE/100 g) for syrup sample assessed by DPPH assay. For FRAP assay, lychee was highest in antioxidant capacity for both homogenized sample (40.61±10.55 umol TE/100 g) and syrup sample (33.58±7.56 umol TE/100 g). A positive and significant (P < 0.001) correlation was found between TPC and antioxidant capacity (DPPH and FRAP). In conclusion, canned pineapple and lychee were the highest sources of polyphenol as compared to other types of canned fruits. Further investigation is warranted to determine the specific polyphenol present in the canned fruits.

Prunus domestica L. is a member of the Rosaceae family that shows many biological activities including antioxidant, antimicrobial, antihaemolytic, anti-inflammatory, hepatoprotective activity and many other activities. In the current study, we evaluated nutritive value, phytochemical screening, total phenolic content and antioxidant activity by DPPH and FRAP method for the different extracts obtained by successive soxhlet extraction using the different solvents based on their polarity. Results show that it is a good source of energy. Phytochemical screening revealed the presence of many secondary metabolites which include alkaloids, carbohydrates, glycosides, protein, steroids and terpenoids, fixed oils and fat as well as phenolic compounds. The highest total phenolic content was found in the ethyl acetate fraction. Highest antioxidant activity by DPPH method is reported in ethyl acetate fraction (IC50 =1837.399±0.377µg/ml) while the ferric reducing antioxidant power was maximum for diethyl ether (56.032±0.985µM/ml FRAP value = 0.325±0.002).
