The aim of this study is to isolate the pathogenic fungi from selected infected vegetable plants in the local area of kushtia, Bangladesh. Some of the most devastating and harmful pathogenic fungi are Phytophthora infestans and Aspergillus nigerwhich we were isolated from edible vegetables potato and onion respectively by using Potato dextrose agar (PDA) using morphological and fungal characteristics. After the morphological study, we observe that Phytophthora infestans have Sporangia shape spores with the white color colony and aspergillus niger has produced black colonies with wooly smooth-walled colonies of conidia. Those fungi create tremendous yield loss subsequently decrease the market value and nutritional value of vegetables. These observed characteristics have similarities with the previous research of another scientist. But further study is needed to confirm our identified isolates. Because of the Covid-19 pandemic, we are unable to study more in the laboratory.

This research was conducted to determine the fungal pathogens responsible for post harvest losses of pineapple sold at Wudil and Yen lemo markets.  Two samples of pineapples were purchased twice a week from both Wudil and Yanlemo markets for four months. The samples were investigated for the presence of fungal pathogen using standard microbiological methods. The methods involve mounting small portion of pineapple in the plate containing Potato dextrose agar to isolate the fungi. Three fungal pathogens belonging to Aspergillus species were isolated, and Aspergillus niger had the highest frequency of occurrence of (50%). Followed by A. flavus with (27%). The A. fumigatus had the lowest frequency of occurrence of (23%). The differences between the fungal isolates recorded were significantly different (P<0.05) between the two markets, where higher fungal isolates were recorded at Yanlemo market 159 (40.6%) and Wudil 38 (9.71%). The study showed that the post harvest losses of pine apple in the two markets are attributed to fungal infection. Therefore, safe guarding the two markets from debris and dumps of rotten fruits and vegetable may assist in reducing fungal inoculums in the two markets.

A total of 14 different fungal species belonging to 7 genera were isolated from the upland and lowland soils of Usmanu Danfodiyo University, Sokoto, between October to December, 2015. The mycoflora were isolated using dilution plate technique on Potato dextrose agar amended by 1% streptomycin. Identification was made microscopically using the lacto-phenol cotton blue method and macroscopically by comparing the cultural and morphological features with the help of authentic fungal manual and taxonomic key. The identified species are; Aspergillus niger, A. flavus, A. fumigatus, A. nidulans, A. oryzae, Alternaria longifes, Fusarium oxysporum, F. solani, F. mangifera, Rhizopus stolonifer, R. orizae, Saccharomyces cerevisae, Trichoderma harzianum and Trichophytum quallinum. The highest number of fungi (50 isolates) were obtained from lowland uncultivated soils (D), followed by the upland uncultivated soils (B) with about 34 strains, and the least (22 isolates) were obtained from upland cultivated soils (A) out of the total 132 isolates. Variations between sites were statistically analyzed.

Abstract This study aimed to isolation and identification of pathogenic fungi from Shatt – al-Arab River in Basra city, Fourteen water samples were collected from different area from Shatt-al-Arab River (AL Ashar, AL Tnoma, AL Makal, AL Qurna, AL Karma, AL Jabiluh, AL-Hartha), from October to December in 2017, with 250 ml volume, this samples centrifuged at 5000 rpm for 10 min at room temperature, the floating was removed and then take the precipitate and pour directly into the center of the media of SDA and PDA and then incubation in a temperature range25-27c for 4 days after that the growth on the media made pure culture and each fungi species diagnosed based on the cultural and microbiological phenotypes, smear prepared with lacto phenol cotton blue stain and the results show 57.1% of growth was Aspergillus niger, 85.7% Aspergillus flavus and 42.8% was Aspergillus candidus and 14.2% was Rhizopus, while the results show 42.8% of growth was Penicillium..

AbstractSpring wheat varieties with the Sm1 gene for resistance to wheat midge, Sitodiplosis mosellana (Géhin) (Diptera: Cecidomyiidae), were compared with susceptible wheat (Triticum Linnaeus; Poaceae) with respect to sources of yield loss and reduction in market value from wheat midge feeding damage. Four resistant varietal blends (90% Sm1 wheat plus 10% susceptible refuge) and four susceptible cultivars were grown in replicated experiments at eight locations in western Canada. Frequencies and 1000-kernel weights of undamaged and midge-damaged seeds were assessed before harvest by dissecting samples of ripe spikes, and after harvest in samples of cleaned grain. Spike data were used to estimate yield losses from reduced weight of damaged seeds and loss of severely damaged seeds (⩽8 mg) at harvest. Among midge-damaged seeds in spikes, few were severely damaged in resistant varietal blends, whereas most were severely damaged in susceptible cultivars. Cleaned, harvested grain of resistant varietal blends and susceptible cultivars had similar frequencies of midge damage and were assessed similar market grades. The primary benefit of midge-resistant wheat was reduced yield loss due to seed damage by wheat midge larvae. Resistant wheat did not protect against loss of market grade, but market value could increase due to larger yields.

Growth and virulence stimulations of sublethal doses of fungicides on plant-pathogenic fungi and oomycetes have been reported and the stimulatory effects are potentially relevant to plant disease management. Sclerotinia sclerotiorum is one of the most devastating and economically important necrotrophic fungal phytopathogens, capable of infecting more than 400 species of plants worldwide. In order to study stimulatory effects of sublethal doses of fungicides on S. sclerotiorum, 55 dimethachlon-sensitive isolates and 3 dimethachlon-resistant isolates of S. sclerotiorum were assayed to determine effects of sublethal doses of dimethachlon on mycelial growth rate on potato dextrose agar (PDA) media and virulence on oilseed rape plants. Results showed that all 3 dimethachlon-resistant isolates and 13 of the 55 sensitive isolates exhibited stimulatory responses to sublethal doses of dimethachlon. Dimethachlon-resistant isolates grew significantly (P < 0.05) faster on PDA media amended with dimethachlon at 0.5 to 4 μg/ml than on fungicide-free PDA media. As for virulence on detached leaves of oilseed rape plants, lesion diameters of dimethachlon-resistant isolates after growth on PDA media amended with dimethachlon at 0.5 to 2 μg/ml were significantly larger (P < 0.05) than the control. The maximum stimulatory effects were 42.40 to 59.80%. In pot experiments, for both dimethachlon-sensitive and -resistant isolates, significant (P < 0.05) virulence stimulations were observed after spraying with dimethachlon at a concentration of 2 μg/ml. After growing on dimethachlon-amended PDA media, H2O2 sensitivity of S. sclerotiorum decreased significantly (P < 0.05) compared with the nonamended PDA control.

<p>The post-harvest fungi is one of the greatest cause of various diseases. Particularly, in fungus that infects seeds, the toxic substance called aflatoxin is found. This has caused us to wonder: Has fungi been pathogenic fungi since the beginning of creation? The purpose of this research is to study: 1) The Place of Fungi in God’s Original Creation and the Origin of Pathogenic Fungi, 2) Characterization of post-harvest fungus on corn seeds, 3) Aspergillus sp.’s Structure as Proof of God’s Wonderful Original Creation and Providence, and 4) The factors that contribute to the growth of fungi in post-harvest corn seeds and our responsibility. We conclude that in the beginning, fungi were a part of God’s originally good and perfect creation. These fungi were created by God with the structure and function to support its operation in fulfilling God’s creative purpose. The complex structure of Aspergillus niger and Aspergillus flavus that infected the corn seeds showed God’s providence after the Fall. Pathogenic fungi were the result of the Fall of man into sin. Their growth is caused by the failure to give proper care to the crops. But this should not discourage us since the Bible tells us that God is continually working to restore His creation. Therefore, we are called to be stewards of His creation, to develop and preserve whatever is entrusted in our hands, including the crops for God’s glory and under His authority until He has fully renewed and restored everything.</p><p><strong>BAHASA INDONESIA ABSTRACT:</strong> Jamur pasca panen merupakan salah satu penyebab terbesar timbulnya berbagai penyakit, khususnya disebabkan oleh jamur yang menginfeksi biji-bijian dan menghasilkan substansi beracun disebut dengan aflatoksin. Keadaan ini menimbulkan pertanyaan apakah jamur patogenik ada sejak awal penciptaan? Penelitian ini bertujuan untuk mempelajari: 1) Posisi jamur patogenik di awal penciptaan dan asal mula jamur patogenik, 2) karakteristik jamur pasca panen pada biji jagung, 3) struktur <em>Aspergillus</em> sp sebagai bukti dari providensi Allah akan ciptaanNya yang sangat luar biasa, 4) faktor-faktor yang berkontribusi terhadap pertumbuhan jamur pasca panen biji jagung dan Tanggung jawab kita. Dari hasil penelitian ini disimpulkan bahwa pada mulanya, jamur merupakan bagian ciptaan Allah yang sungguh amat baik dan sempurna. Struktur yang kompleks dari <em>Aspergillus niger</em> dan <em>Aspergillus flavus </em>yang menginfeksi biji jagung menunjukkan providensi Allah setelah kejatuhan dosa. Pertumbuhan jamur patogenik merupakan gambaran kejatuhan manusia ke dalam dosa yaitu kegagalan manusia untuk memberikan pemeliharaan yang memadahi pada biji-bijian pasca panen. Namun demikian, keadaan ini tidak mematahkan semangat kita karena Alkitab mengatakan bahwa Allah masih terus bekerja untuk memulihkan ciptaanNya. Oleh karena itu, diharapkan kita semua menjadi pelayan atas ciptaanNya untuk mengembangkan dan menjaga apa yang dipercayakan kepada kita di bawah otoritasNya termasuk biji-bijian untuk memuliakan Allah hingga Dia secara penuh memperbaharui dan memulihkan segala sesuatu</p>

Crop waste or cacao (Theobroma cacao L.) processing waste that doesn’t handle with care can polluting the environment around the farm. Rind of cacao can be used for composting, but it can be less effective because the nutrient of fresh cacao rind too complex and the ratio of C/N too high. This research intention to knows the potential of Aspergillus niger to degrade cellulose on waste cacao rind who have high ratio of C/N, with combination inoculum concentration and incubation time. The methods of this research begin with isolation and identification of A. niger from litter of cacao field, then the in vitro test done by CMCA (Carboxyl Methyl Cellulose Agar) selective media, and then the cacao rind was testing by in vivo method. The results of this research shown that A. niger can degrade cellulose by in vitro ways, it can be seen by the clear zone that formed on CMCA selective media. The in vivo test on cacao rind shown decreasing of C/N ratio between 18,940%-40,623%. Combination of A3B3 treatment (15 mL inoculum concentration, 28 days incubation time) indicate the lowest C/N ratio 18,940%.

In this study, the purification and characterization of an extracellular enzyme form Aspergillus niger was performed. With an optimized protocol, it was conducted a 42.6-fold purification with a yield of 26.2%. The purified lipase had a monomeric molecular weight of 40.5kDa and an isoelectric point of 6.01, and its maximum enzyme activity could be achieved at 40°C and pH 7.5-9.0. The enzyme could be activated by Ca2+, Mg2+ and Fe2+, while its activity could be inhibited by Zn2+ and Cu2+. Additionally, organic compounds exerted an inhibitory effect on the enzyme activity in a descending order of methanol, ethanol, DMSO, EDTA, acetone. Meanwhile, the specificity analysis of the enzyme indicated a preference to tributyrin and vegetable oils as well as long-chain fatty acid methyl esters (C12-C18). Most importantly, this enzyme could successfully transform deoxynivalenol (DON). Using HPLC analysis,it was detected a biotransformation rate of more than 70%.The liquid chromatography-mass spectrometry (LC-MS) analysis showed that the molecular weight of the transformation product was 18.0 larger than that of DON, indicating that DON could be hydrolyzed by the enzyme. Overall, the proposed method here provides a new avenue for reducing the toxicity of DON, which appears to have a wide application outlook for DON biotransformation.
