The feeding habits of adult Brachycera are understudied and may provide important context for understanding invasive pest biology, as with the polyphagous small fruit pest Drosophila suzukii. We developed molecular methods to study adult D. suzukii gut content in order to understand its feeding habits. We designed and verified two primer pairs specific for either blueberries or blackberries and used a qPCR melt curve analysis to determine whether we can detect the presence or absence of berry feeding by adult flies. In a laboratory assay, the blueberry fly meal DNA can be detected for longer periods than the blackberry meal DNA. Generally, female gut contents are less variable than male gut contents. We also tested recently emerged flies that were not fed as adults but developed as larvae in either blueberries or blackberries. Some adult flies from each fruit had detectable fruit DNA in their gut, which could be due to pupal meconium feeding after emergence. Next, we aimed to test the primers in the field to develop techniques to track fruit feeding by D. suzukii in its natural field environment. First, to identify the most appropriate collection method, we determined how long we could detect fruit DNA, using previously developed primers within D. suzukii gut preserved in four types of trap fluid in the laboratory. The likelihood of detecting blackberry DNA differed by day, trap fluid, and between sexes. For the blueberry primer, the possibility of detecting blueberry DNA differed by trap fluid only. Based on those results, we used RV antifreeze with a Scentry SWD lure in field trials at two research station locations, one containing blackberries and one with blueberries. We established transects away from each fruit planting and collected up to 120 total flies at each point along transects. There were no significant differences in the number of flies containing berry DNA among collection points along the transect in both locations. These results suggest that adult flies move between crop and non-crop habitats and may not be highly dependent on fruit food resources.

Abstract Spotted wing drosophila, Drosophila suzukii, is a serious invasive pest impacting the production of multiple fruit crops, including soft and stone fruits such as strawberries, raspberries and cherries. Effective control is challenging and reliant on integrated pest management which includes the use of an ever decreasing number of approved insecticides. New means to reduce the impact of this pest that can be integrated into control strategies are urgently required. In many production regions, including the UK, soft fruit are typically grown inside tunnels clad with polyethylene based materials. These can be modified to filter specific wavebands of light. We investigated whether targeted spectral modifications to cladding materials that disrupt insect vision could reduce the incidence of D. suzukii. We present a novel approach that starts from a neuroscientific investigation of insect sensory systems and ends with infield testing of new cladding materials inspired by the biological data. We show D. suzukii are predominantly sensitive to wavelengths below 405 nm (ultraviolet) and above 565 nm (orange & red) and that targeted blocking of lower wavebands (up to 430 nm) using light restricting materials reduces pest populations up to 73% in field trials.

Abstract Whole-culture extracts of Bacillus thuringiensis Berliner strains were assayed against larval and adult Drosophila suzukii (Matsumura), an important invasive pest of many thin-skinned soft fruit crops in North America. Of the 22 serovars tested versus larval D. suzukii , strains of Bacillus thuringiensis var. thuringiensis , kurstaki , thompsoni , bolivia , and pakistani caused high (75 to 100%) first-instar mortalities. Pupal mortality, measured as a failure of adults to emerge, varied with serovar. The first D. suzukii instar was the most susceptible of the three larval instars to B. thuringiensis var. kurstaki HD-1. Larval D. suzukii are shielded from crop treatments, as they develop under the skin of infested fruit, and adults would be a more vulnerable target for an efficacious strain of B. thuringiensis . Only one of the 21 B. thuringiensis serovars, var. thuringiensis , prepared as oral suspensions in sucrose for adult D. suzukii ingestion resulted in significant, albeit low mortality within 7 d. It is not a candidate for use in pest management, as it produces β -exotoxin that is toxic to vertebrates.

Drosophila suzukii (Matsumura, 1931) (Diptera: Drosophilidae), the spotted wing Drosophila (SWD) is a concern for small fruit and stone fruit growers. This invasive pest lays eggs in healthy fruits with a serrated ovipositor, resulting in considerable economic losses, mainly in berry crops. In Europe, it was first recorded in Switzerland in 2011, causing considerable damage in all small fruit crops, especially in later-developing crops (autumn raspberries, blackberries, blueberries and elderberries). The pest was found in all regions of the country, from low altitudes to the timberline. The range of host plants is very broad, not only affecting crops, but also wild fruits. Switzerland has implemented a strategy at the national level by combining an effective monitoring system with hygiene measures and mass trapping. Insecticide applications, usually based on spinosyns, are only considered as a last resort. In addition to this already operative strategy, innovative alternatives are considered, in particular the use of repellents or masking substances.

AbstractDrosophila suzukii flies cause economic losses to fruit crops globally. Previous work shows various Drosophila species are attracted to volatile metabolites produced by individual fruit associated yeast isolates, but fruits naturally harbour a rich diversity of yeast species. Here, we report the relative attractiveness of D. suzukii to yeasts presented individually or in combinations using laboratory preference tests and field trapping data. Laboratory trials revealed four of 12 single yeast isolates were attractive to D. suzukii, of which Metschnikowia pulcherrima and Hanseniaspora uvarum were also attractive in field trials. Four out of 10 yeast combinations involving Candida zemplinina, Pichia pijperi, M. pulcherrima and H. uvarum were attractive in the laboratory. Whilst a combination of M. pulcherrima + H. uvarum trapped the greatest number of D. suzukii in the field, the efficacy of the M. pulcherrima + H. uvarum combination to trap D. suzukii was not significantly greater than traps primed with volatiles from only H. uvarum. While volatiles from isolates of M. pulcherrima and H. uvarum show promise as baits for D. suzukii, further research is needed to ascertain how and why flies are attracted to certain baits to optimise control efficacy.

The feeding habits of myctophid larvae of Symbolophorus californiensis were examined in the southern transition region of the western North Pacific where the main spawning and nursery grounds of S. californiensis are formed. This species is a key component of the pelagic ecosystems of this region, and their larvae attain one of the largest sizes among myctophids. To analyse gut contents larvae, including most life history stages after yolk-sac absorption (3.7 to 22.2 mm body length (BL)), were collected in the upper 100 m layer in 1997 and 1998. Feeding incidence was higher during the day than at night (53.1–92.3% versus 0–5.6%), and daytime feeding incidence increased gradually with larval growth. Larvae fed mainly on copepods of various developmental stages. Larvae of S. californiensis showed an ontogenetic change in their diet: larvae ≤7.9 mm BL (i.e. preflexion stage) fed mainly on copepod eggs and nauplii, while the larvae ≥8 mm BL consumed mainly calanoid copepodites such as Pseudocalanus and Paracalanus spp. In the largest size-class (16–22.2 mm BL), the furcilia stage of euphausiids was also an important prey item. There was an increase in the average prey size with growth in larvae ≤11.9 mm BL, while the number of prey eaten positively correlated with growth in larvae ≥12 mm BL. The trophic niche breadth also increased with larval growth, which would ensure a wide range of available food resources for the larger size-class larvae.

The role of the host plant in the development of larval diapause in the millet stem borer, Coniesta ignefusalis (Hampson) was investigated in northern Ghana in 1996 and 1997. Surveys conducted in farmers' fields in the Guinea and Sudan savannah revealed that of all the upland cereals grown, the insect survived the dry season only in stalks and stubble of pearl millet, Pennisetum glaucum and late sorghum, Sorghum bicolor. This observation was confirmed by results from field trials conducted at the Manga Research Station. In these studies, C. ignefusalis larvae entered diapause only in late millet and late sorghum, with a higher incidence in the former. The insect neither attacked nor entered diapause in maize planted during the same period as the other crops. Results from controlled experiments showed that diapause incidence in the preferred host, millet, was higher in older than in younger plants, suggesting that host plant maturation is a key factor influencing the development of larval diapause in C. ignefusalis.

AbstractOriginally from Asia,Drosophila suzukii(Matsumura, 1931, Diptera:Drosophilidae) is presently a global pest of economically important soft-skinned fruits. Also commonly known as spotted wingDrosophila(SWD), it is largely controlled through repeated applications of broad-spectrum insecticides. There is a pressing need for a better understanding of SWD biology and for developing alternative environmentally-friendly methods of control. The RNA-guided Cas9 nuclease has revolutionized functional genomics and is an integral component of several recently developed genetic strategies for population control of insects. Here we have developed transgenic strains that encode three different terminators and four different promoters to express Cas9 in both the soma and/or germline of SWD. The Cas9 lines were evaluated through genetic crossing to transgenic lines that encode single guide RNAs targeting the conserved X-linkedyellowbody andwhiteeye genes. We find that several Cas9/gRNA lines display very high editing capacity. Going forward, these tools will be instrumental for evaluating gene function in SWD and may provide tools useful for the development of new genetic strategies for control of this invasive species.

Tropical high mountain lakes show unique environmental conditions where chironomids play an important role in ecosystem functioning. The characteristics of these environments could favor diet overlap and therefore a high interspecific competition. This study described the dietary habits of chironomid genera, identified whether the dietary habits were specialized or generalist, and analyzed the diet overlap in the genera. Chironomidae larvae were collected from four lakes of the Chingaza paramo during the dry season, between April and May of 2 016. The feeding habits of larvae were evaluated by analyzing gut contents following standard methods. Each genus was assigned to trophic guilds (carnivore, detritivore and algivore) and the diet overlap was estimated using the Pinka's index. A total of 1 003 individuals were collected and nine genera were identified. Larvae consumed mainly fine particulate organic matter (FPOM), algae, macrophyte fragments, macroinvertebrates, and animal tissues. FPOM was the main feeding resource of detritivores. The analysis of diets showed a high affinity of each genus for a single trophic guild and most of the genera were generalist in the use of resources. For all lakes, high levels of diet overlap were observed among genera and trophic guilds, mainly among detritivores. Our results suggested that Chironomidae larvae of these lakes presented well differentiated trophic habits, and showed a moderate diet overlap within detritivores and carnivores.
