Type B dihydrofolate reductase (dfrb) genes were identified following the introduction of trimethoprim in the 1960s. Although they intrinsically confer resistance to trimethoprim (TMP) that is orders of magnitude greater than through other mechanisms, the distribution and prevalence of these short (237 bp) genes is unknown. Indeed, this knowledge has been hampered by systematic biases in search methodologies. Here, we investigate the genomic context of dfrbs to gain information on their current distribution in bacterial genomes. Upon searching publicly available databases, we identified 61 sequences containing dfrbs within an analyzable genomic context. The majority (70%) of those sequences also harbor virulence genes and 97% of the dfrbs are found near a mobile genetic element, representing a potential risk for antibiotic resistance genes. We further identified and confirmed the TMP-resistant phenotype of two new members of the family, dfrb10 and dfrb11. Dfrbs are found both in Betaproteobacteria and Gammaproteobacteria, a majority (59%) being in Pseudomonas aeruginosa. Previously labelled as strictly plasmid-borne, we found 69% of dfrbs in the chromosome of pathogenic bacteria. Our results demonstrate that the intrinsically TMP-resistant dfrbs are a potential emerging threat to public health and justify closer surveillance of these genes.

Abstract Background: Microbiota carrying multiple antibiotic resistance genes (ARGs) and virulence factors (VFs) are posing increasing risks to public health. Particularly the rapid spread of human pathogenic bacteria (HPB) with antibiotic resistance is recognized as a top health issue. The occurrence and abundance of ARGs in aquaculture have been investigated following metagenomic approaches. However, few studies have investigated the antibiotic resistome and VFs and their HPB hosts in aquaculture. Moreover, the relationships between ARGs and VFs and their microbiome in aquaculture are poorly understood. Results: The profiles of the antibiotic resistome, VFs, and HPB in aquaculture in Southern China were investigated. In total, 492 subtypes of 24 ARGs types were detected. Multidrug ARGs were most predominant, followed by macrolide-lincosamide-streptogramin (MLS). Proteobacteria were the most predominant phylum carrying ARGs, followed by Firmicutes. Fifty-two HPB genera were detected. Firmicutes was the most abundant phylum, followed by Proteobacteria. Staphylococcus was the most abundant HPB genus. The samples contained 363 VFs, with Capsule being the most abundant. Seven HPB phyla, including 42 HPB genera, carried VFs, and the abundance of Bacillus was highest. The abundances of ARGs and VFs were highest in the sediment. However, the abundance of HPB was highest in shrimp guts and Staphylococcus was most abundant. Most ARGs were more prevalent on chromosomes than on plasmids. Source tracking analysis showed that the sediment was the greatest contributor to microbes carrying ARGs, VFs, and HPB in shrimp guts. Additionally, the water source contributed some of the HPB of shrimp guts. Conclusions: This study provides in-depth profiles of the abundances, diversity, distribution, and prevalence of ARGs, VFs, and their hosts HPB in aquaculture for the first time. Sediment was the most direct and important contributor to the ARGs, VFs, and HPB in the shrimp guts. The prevalence of HPB in aquaculture, particularly the high abundance of Staphylococcus in shrimp guts, poses potential risks to human health and food safety. Aquaculture water sources should be monitored and protected. The findings of this study provide a better understanding of the dissemination and hosts of ARGs and VFs for improving aquaculture management and public health surveillance.

ABSTRACTThe environmental microbiome harbors a vast repertoire of antibiotic resistance genes (ARGs) which can serve as evolutionary predecessors for ARGs found in pathogenic bacteria, or can be directly mobilized to pathogens in the presence of selection pressures. Thus, ARGs from benign environmental bacteria are an important resource for understanding clinically relevant resistance. Here, we conduct a comprehensive functional analysis of the Antibiotic_NAT family of aminoglycoside acetyltransferases. We determined a pan-family antibiogram of 21 Antibiotic_NAT enzymes, including 8 derived from clinical isolates and 13 from environmental metagenomic samples. We find that environment-derived representatives confer high-level, broad-spectrum resistance, including against the atypical aminoglycoside apramycin, and that a metagenome-derived gene likely is ancestral to an AAC(3) gene found in clinical isolates. Through crystallographic analysis, we rationalize the molecular basis for diversification of substrate specificity across the family. This work provides critical data on the molecular mechanism underpinning resistance to established and emergent aminoglycoside antibiotics and broadens our understanding of ARGs in the environment.

Complete genomes of microbial pathogens are essential for the phylogenomic analyses that increasingly underpin core public health laboratory activities. Here, we announce a BioProject (PRJNA556438) dedicated to sharing complete genomes chosen to represent a range of pathogenic bacteria with regional importance to Australia and the Southwest Pacific; enriching the catalogue of globally available complete genomes for public health while providing valuable strains to regional public health microbiology laboratories. In this first step, we present 26 complete high-quality bacterial genomes. Additionally, we describe here a framework for reconstructing complete microbial genomes and highlight some of the challenges and considerations for accurate and reproducible genome reconstruction.

: Childhood obesity has assumed epidemic proportions and is currently one of the most widespread public health problems. Many are the factors involved in the pathogenesis of excess weight with interactions between genetic, environmental and biological factors and therefore, also the therapeutic approach must be multidisciplinary and multidimensional. In this review of the literature, we report the contiguity of childhood obesity with eating disorders and the importance of involving the family context in order to induce stable lifestyle changes, both in relation to dietary and nutritional habits, but also in increasing physical activity. Finally, among the therapeutic options, although for selected cases, pharmacotherapy and bariatric surgery can be used as treatment strategies.

Protestant beliefs have made several significant contributions to conservatism, both in the more abstract realm of ideas and in the arena of political positions or practical policies. First, they have sacralized the established social order, valued and defended customary hierarchies; they have discouraged revolt or rebellion; they have prompted Protestants to view the state as an active moral agent of divine origin; and they have stressed the importance of community life and mediating institutions such as the family and the church and occasionally provided a modest check on an individualistic and competitive impulse. Second, certain shared tenets facilitated this conjunction of Protestantism and conservatism, most often when substantial change loomed. For example, common concerns of the two dovetailed when revivals challenged the religious status quo during the colonial Great Awakening, when secession and rebellion threatened federal authority during the Civil War, when a new type of conservatism emerged, and dismissed the older sort as paternalistic, when the Great Depression opened the door to a more intrusive state, when atheist communism challenged American individualism, and, finally, when the cultural changes of the 1960s undermined traditional notions of the family and gender roles. Third, certain Christian ideas and assumptions have, at their best, served to heighten or ennoble conservative discourse, sometimes raising it above merely partisan or pragmatic concerns. Protestantism added a moral and religious weight to conservative beliefs and helped soften the harshness of an acquisitive, sometimes cutthroat, economic order.

The emergence or re-emergence of viruses with epidemic and/or pandemic potential, such as Ebola, Zika, Middle East Respiratory Syndrome (MERS-CoV), Severe Acute Respiratory Syndrome Coronavirus 1 and 2 (SARS and SARS-CoV-2) viruses, or new strains of influenza represents significant human health threats due to the absence of available treatments. Vaccines represent a key answer to control these viruses. However, in the case of a public health emergency, vaccine development, safety, and partial efficacy concerns may hinder their prompt deployment. Thus, developing broad-spectrum antiviral molecules for a fast response is essential to face an outbreak crisis as well as for bioweapon countermeasures. So far, broad-spectrum antivirals include two main categories: the family of drugs targeting the host-cell machinery essential for virus infection and replication, and the family of drugs directly targeting viruses. Among the molecules directly targeting viruses, nucleoside analogues form an essential class of broad-spectrum antiviral drugs. In this review, we will discuss the interest for broad-spectrum antiviral strategies and their limitations, with an emphasis on virus-targeted, broad-spectrum, antiviral nucleoside analogues and their mechanisms of action.

There exists a normative representation of family meals in contemporary Western societies which is promoted as imperative through public health programs, larger discourses and by some studies in the nutritional and public health research fields. Family meals, also called domestic commensality, are represented as convivial events and are associated with positive health and wellbeing outcomes but there is minimal evidence to show they are beneficial for family members and it is not known which aspect of the family meal could be responsible for these alleged benefits. This normative family meal image is based on a representation of the family as a peaceful unit exempt from external constraints. This narrative literature review of qualitative studies of family meals seeks to put forward the underlying premises of this representation and compare it with reports about actual practices. The results emphasize that eating together is still practiced and remains valued by family members, which is in contrast to discourses lamenting the decline of the family meal. However, the valorisation and recurrence of family meals depends on class, gender and cultural positions. There is a gap between the norm of healthy or convivial and achievable family meals, which can reinforce the so-called “mental load” and “emotion work” of those in charge of feeding the family and heighten inequalities within the household. In fact, there are many challenges to family meals which originate from external constraints or are inherent aspects of family life. The results from this review suggest that we should focus on family meals by taking into account the food work surrounding it and focussing on the interactional aspects of family meals. Ethnographic methods allow the researcher to observe the diversities and complexities of commensality as well as family dynamics and, in doing so, could provide more realistic representations of eating within the family.

AbstractThe ‘family house’ has played a major role within the urbanisation processes that have been transforming the Italian landscape since the 1960s. It is a common feature of the widespread settlements that are part of what has been labelled the ‘diffuse city’ and was the subject of numerous studies during the 1990s. More than 20 years later, this paper returns to the topic of the Italian family house using a renewed methodological approach to describe relevant changes. The hypothesis here is that in order to grasp the tensions affecting ‘family houses’ in today’s context of demographic transition and increased imbalances between dynamic and declining areas, and to contemplate their future, the qualitative gaze adopted by scholars in the 1990s must be integrated with other investigative tools, focusing on demographic change, uses, and the property values of buildings. Using this perspective, the paper provides a series of ‘portraits’ rooted in four meaningful territorial contexts, portraits which may help scholars to redefine their imagery associated with family house and be useful for dedicated building policies.
