Src kinases play fundamental roles in several crucial cell processes. Their activity is tightly regulated by conformational transitions between the active and the inactive forms, which are carried out by complex protein structural rearrangements. Here, we present an in-depth study of such structural transitions coupling extensive all-atoms molecular dynamic simulations coupled to an algorithm able to drive the system between defined conformational states. Our results, in line with the available experimental data, confirm the complexity of such a process indicating the main molecular determinants involved. Moreover, the role of an Src inhibitor—able to bind to the protein inactive state—is discussed and compared with available experimental data.

Аннотация: О роли разминки перед тренировкой и соревнованиями написано много научных статьей и диссертаций. Ещё больше о значимости физической подготовки спортсменов, в том числе и шахматистов. В то же время авторы считают, что вопросу физической подготовки шахматистов и в частности разминке в научной и научно-методической литературе уделяется недостаточно внимания. В своей статье авторы приводят собственные экспериментальные данные о влиянии физических упражнений (разминки) на умственную работоспособность студентов-шахматистов БГУ и как следствие на спортивные результаты команды. AbstractATP-independent chaperones are usually considered to be holdases that rapidly bind to non-native states of substrate proteins and prevent their aggregation. These chaperones are thought to release their substrate proteins prior to their folding. Spy is an ATP-independent chaperone that acts as an aggregation inhibiting holdase but does so by allowing its substrate proteins to fold while they remain continuously chaperone bound, thus acting as a foldase as well. The attributes that allow such dual chaperoning behavior are unclear. Here, we used the topologically complex protein apoflavodoxin to show that the outcome of Spy’s action is substrate specific and depends on its relative affinity for different folding states. Tighter binding of Spy to partially unfolded states of apoflavodoxin limits the possibility of folding while bound, converting Spy to a holdase chaperone. Our results highlight the central role of the substrate in determining the mechanism of chaperone action.

The objective of the present study has been to assess the influence that the sex of the aggressor and the sex of participant have on the perception of three types of psychological violence in young couples. A total of 693 young people, ranging from 17 years to 25 years, were randomly assigned six different scenarios in which situations of psychological violence between young heterosexual couples were described and where the sex of the aggressor and the types of psychological violence varied. The results of our research revealed that differences in the perception of violence are observed based on the sex of the aggressor, the sex of the participant, and the type of psychological violence, independently, as well as in the interaction of the three variables. This result is relevant as it points to the need for further in-depth study into situations that could contribute to justifying violence.

Natural flavor and fragrance market size is expected to grow steadily due to the rising consumer demand of natural ingredients. This market request is guided by the general opinion that the production of natural compounds leads to a reduction of pollution, with inherent advantages for the environment and people’s health. The biotransformation reactions have gained high relevance in the production of natural products. In this context, few pieces of research have described the role of microalgae in the oxidation of terpenoids. In this present study, we questioned the role of microalgal based oxidation in the synthesis of high-value flavors and fragrances. This study investigated the role of three different microalgae strains, Chlorella sp. (211.8b and 211.8p) and Chlorococcum sp. (JB3), in the oxidation of different terpenoid substrates: α-ionone, β-ionone, theaspirane and valencene. Unfortunately, the experimental data showed that the microalgal strains used are not responsible for the substrate oxidation. In fact, our experiments demonstrate that the transformation of the four starting compounds is a photochemical reaction that involves the oxygen as oxidant. Even though these findings cast a shadow on the use of these microorganisms for an industrial purpose, they open a new possible strategy to easily obtain nootkatone in a natural way by just using an aqueous medium, oxygen and light.

Clinical and experimental data suggest that stress contributes to the pathology of epilepsy. We review mechanisms by which stress, primarily via stress hormones, may exacerbate epilepsy, focusing on the intersection between stress-induced pathways and the progression of pathological events that occur before, during, and after the onset of epileptogenesis. In addition to this temporal nuance, we discuss other complexities in stress-epilepsy interactions, including the role of blood-brain barrier dysfunction, neuron-glia interactions, and inflammatory/cytokine pathways that may be protective or damaging depending on context. We advocate the use of global analytical tools, such as microarray, in support of a shift away from a narrow focus on seizures and towards profiling the complex, early process of epileptogenesis, in which multiple pathways may interact to dictate the ultimate onset of chronic, recurring seizures.

The paper analyzes the course of events and the international context of Сarpatho- Ukrainian state’s rise and defeat in terms of role and impact of the leading European actors at the end of the 1930’s. Based on an in-depth study of the wide range of literature, documents and relevant archives, the author highlights the role of Carpatho-Ukraine, which for a short period happened to be at the epicenter of the geopolitical interests of the states whose actions or inaction fueled the warmongers. The revival and strengthening of a number of European countries at that historical period deteriorized international relations. The erosion of the Versailles-Washington bases intensified the contradictions in the foreign policy between the victorious and vanquished states. The defeated countries reinforced their positions, since the victors, who were expanding their military might at the expense of Czechoslovak Republic, including Transcarpathia, were becoming more submissive in accommodating territorial claims. This situation largely updated the approaches to the Ukrainian question. The variability of the ways to solve it largely depended on the situational decisions of the Third Reich political leadership.

Critical theory is a paradigm that promotes viewpoints that are alternative and, at times, contrary to mainstream beliefs and dictates. In 2012, I adopted this perspective to review the role of ethnography and surmised that the data which arise from this research approach, which I described as an in-depth study of cultures, can be used to discipline and control these groups. In this edition of Qualitative Health Research, another author has critiqued this position. In this article, I review this critique, reiterate my position, update the data I used for my 2012 article, and highlight how I navigate what I feel is a tension between critical theory and practice.

AbstractEven though Italy officially joined the IAU in 1921, Italian astronomers were involved in its birth as early as 1919, when Annibale Riccò, Director of the Astrophysical Observatory of Catania, proposed to the IAU Committee to hold its first General Assembly in Rome. This contribution will analyze the role played by Italian astronomers in the development of the IAU from its foundation to the Second World War. The recent project of reordering of the astronomical historical archives in Italy permits for the first time a more in-depth study of the relations between Italian astronomers and the international scientific community.
