Bio-oils from biomass pyrolysis can be a resource for upgrading to chemicals or fuels. Here, for the first time, we compare the composition of bio-oils produced from two feedstocks (wheat straw, softwood) in pyrolysis units of different mode of operation (continuous—rotary kiln vs. batch) using Fourier transform ion cyclotron resonance mass spectrometry (FTICR MS) in different ionization modes (APPI (+), ESI (+/−)). Our results demonstrate that the pyrolysis unit design had only a minor influence on the composition of bio-oils produced from low-mineral containing wood biomass. Yet, the wheat straw-derived bio-oil produced in the continuous unit comprised lower molecular weight compounds with fewer oxygen-containing functional groups and lower O/C and H/C ratios, compared to bio-oils from batch pyrolysis. Longer residence time of vapours in the heated zone in the rotary kiln and a higher mineral content in wheat straw resulted in increased catalytically-mediated secondary reactions that favoured further bio-oil decomposition. This work shows for the first time that it is possible to produce distinct bio-oils without the need for external catalyst addition, by matching reactor type/design and feedstock.

AbstractDespite broad application of different analytical techniques for studies on organic matter of chondrite meteorites, information about composition and structure of individual compounds is still very limited due to extreme molecular diversity of extraterrestrial organic matter. Here we present the first application of isotopic exchange assisted Fourier transform ion cyclotron resonance mass spectrometry (FTICR MS) for analysis of alkali extractable fraction of insoluble organic matter (IOM) of the Murchison and Allende meteorites. This allowed us to determine the individual S-containing ions with different types of sulfur atoms in IOM. Thiols, thiophenes, sulfoxides, sulfonyls and sulfonates were identified in both samples but with different proportions, which contribution corroborated with the hydrothermal and thermal history of the meteorites. The results were supported by XPS and thermogravimetric analysis coupled to FTICR MS. The latter was applied for the first time for analysis of chondritic IOM. To emphasize the peculiar extraterrestrial origin of IOM we have compared it with coal kerogen, which is characterized by the comparable complexity of molecular composition but its aromatic nature and low oxygen content can be ascribed almost exclusively to degradation of biomacromolecules.

ABSTRACT Staphylococcus aureus is an important human pathogen whose success is largely attributed to its vast arsenal of virulence factors that facilitate its invasion into, and survival within, the human host. The expression of these virulence factors is controlled by the quorum sensing accessory gene regulator (Agr) system. However, a large proportion of clinical S. aureus isolates are consistently found to have a mutationally inactivated Agr system. These mutants have a survival advantage in the host but are considered irreversible mutants. Here we show, for the first time, that a fraction of Agr-negative mutants can revert their Agr activity. By serially passaging Agr-negative strains and screening for phenotypic reversion of hemolysis and subsequent sequencing, we identified two mutational events responsible for reversion: a genetic duplication plus inversion event and a poly(A) tract alteration. Additionally, we demonstrate that one clinical Agr-negative methicillin-resistant S. aureus (MRSA) isolate could reproducibly generate Agr-revertant colonies with a poly(A) tract genetic mechanism. We also show that these revertants activate their Agr system upon phagocytosis. We propose a model in which a minor fraction of Agr-negative S. aureus strains are phase variants that can revert their Agr activity and may act as a cryptic insurance strategy against host-mediated stress. IMPORTANCE Staphylococcus aureus is responsible for a broad range of infections. This pathogen has a vast arsenal of virulence factors at its disposal, but avirulent strains are frequently isolated as the cause of clinical infections. These isolates have a mutated agr locus and have been believed to have no evolutionary future. Here we show that a fraction of Agr-negative strains can repair their mutated agr locus with mechanisms resembling phase variation. The agr revertants sustain an Agr OFF state as long as they exist as a minority but can activate their Agr system upon phagocytosis. These revertant cells might function as a cryptic insurance strategy to survive immune-mediated host stress that arises during infection.

Introduction Lymphatic filariasis causes long term morbidity and hampers the socio-economic status. Apart from the available treatments and medication, control of vector population Culex quinquefasciatus Say through the use of chemical insecticides is a widely applied strategy. However, the unrestrained application of these insecticides over many decades has led to resistance development in the vectors. Methods In order to determine the insecticide susceptibility/resistance status of Cx. quinquefasciatus from two filariasis endemic districts of West Bengal, India, wild mosquito populations were collected and assayed against six different insecticides and presence of L1014F; L1014S kdr mutations in the voltage-gated sodium channel gene was also screened along with the use of synergists to evaluate the role of major detoxifying enzymes in resistance development. Results The collected mosquito populations showed severe resistance to insecticides and the two synergists used–PBO (piperonyl butoxide) and TPP (triphenyl phosphate), were unable to restore the susceptibility status of the vector thereupon pointing towards a minor role of metabolic enzymes. kdr mutations were present in the studied populations in varying percent with higher L1014F frequency indicating its association with the observed resistance to pyrethroids and DDT. This study reports L1014S mutation in Cx. quinquefasciatus for the first time.

In the article, the bryophytes of the upper reaches of the Western Bug River, which is physically and geographically located within Male Polissya, partly Roztochia, and to a minor extent in the Gologoro-Voronyatsky denudo-structural hills, have been studied. Based on our survey, a list of the bryophytes has been compiled for the first time. Ecological features, substrate preferences and life forms of the bryophytes have been analysed. According to the ecological features, subheliophytes (30.9%) and hemisciophytes (30.9%) predominate in the spectrum of heliomorphs; mesophytes (29.7%), hygromesophytes (21.2%) and xeromesophytes – in the spectrum of hydromorphs (19.4%); cold-tolerant species (59.4%) – in the spectrum of thermomorphs. Based on the analysis of the substrate preferences of the bryophytes, the following groups were identified: epigeans (116 species), epixils (56 species), epiphytes (46 species), epiliths (43 species), aquatic (22 species). The prevailing life forms are turf (30.3%), rough mat (18.2%), weft (15.2%), tuft (10.3%) and smooth mat (9.7%). 3 species that are officially recognised as rare and 16 species that are recognized as regionally rare have been found. In the group of bryophytes associated with wetland ecosystems, 2 officially rare and 6 regionally rare species were found in the study area. Given the large areas of drained land in Lviv Region, these species are of particular value, especially in the context of conservation of the biodiversity and protection of the valuable natural areas in accordance with the Development Strategy of Lviv Region by 2027.

The aim: To study the association between A/A, G/A, A/A genotypes, alleles A, G of the SNP rs17216473 of the gene that encodes ALOX5AP and the risk of myocardial infarction within the Ukrainian population. Materials and methods: PCR in real time and the analysis to discriminate alleles were used. The statistical processing was carried out by χ2 criteria and by χ2 criteria with Yates correction. Results: For the first time the SNP rs17216473 of gene that encodes ALOX5AP has been established to be statistically significantly associated with the risk of myocardial infarction in Ukrainian population. The connection with genotype A/A was opposite to that with genotype G/G. That is, A/A contribution to myocardium infarction has been statistically significant whereas, G/G has been statistically significantly associated with the absence of myocardial infarction. G/A genotype has not been statistically significantly associated with myocardial infarction. It has also been established a statistically significant connection exists between the risk of myocardial infarction and the presence of allele A (minor allele) of the polymorphism. Allele G, however, has a statistically significant association with the absence of myocardial infarction. All humans-homozygotes with the minor allele A had suffered from myocardial infarction. In the control group, humans-homozygotes with the minor allele A were not found. Conclusions: Summarizing our obtained results, we assume the carriers of G/G genotype to have a minimal risk of myocardial infarction onset, the carriers of G/A genotype to have a moderate risk and the carriers of A/A to have a great risk.

ABSTRACT Pterocarpus santalinus, listed in CITES Appendix II, is an endangered timber species as a result of illegal harvesting due to its high value and commercial demand. The growing demand for P. santalinus and timbers with the morphologically similar Pterocarpus tinctorius has resulted in confusion as well as identification problems. Therefore, it is of vital importance to explore reliable ways to accurately discriminate between P. santalinus and P. tinctorius. In this study, the method of direct analysis in real time and fourier transform ion cyclotron resonance mass spectrometry (DART-FTICR-MS), combined with multivariate statistical analysis, was used to extract chemical information from xylarium wood specimens and to explore the feasibility of distinguishing these two species. Significant differences were observed in their DART-FTICR-MS spectra. Orthogonal partial least square-discriminant analysis (OPLS-DA) showed the highest prediction, with an accuracy of 100%. These findings demonstrate the feasibility of authenticating wood types using DART-FTICR-MS coupled with multivariate statistical analysis.

A flavonoid was isolated from the fronds of Asplenium ruta-muraria and A. altajense (Aspleniaceae) collected in the Altai Mountains and adjacent area. The compound was identified as kaempferol 3- O- β-[(6″′- E-caffeoylglucopyranosyl)-(1→3)-glucopyranoside]-7- O- β-glucopyranoside (1) by UV, 1H and 13C NMR spectroscopy, LC-MS, and acid and alkaline hydrolyses. Another flavonoid (2) was isolated from A. altajense, as a minor compound, together with 1 and identified as deacylated compound 1, i.e. kaempferol 3- O-laminaribioside-7- O-glucoside. They were found in nature for the first time.

The aim of this study is to find the optimal pretreatment conditions and hydrolysis in order to obtain a high yield of bioethanol from wheat straw. The pretreatments were performed with different concentrations of sulphuric acid 1, 2 and 3% (v/v), and were followed by an enzymatic hydrolysis that was performed by varying the solid-to-liquid ratio (1/20, 1/25 and 1/30 g/mL) and the enzyme dose (30/30 µL/g, 60/60 µL/g and 90/90 µL/g Viscozyme® L/Celluclast® 1.5 L). This mix of enzymes was used for the first time in the hydrolysis process of wheat straws which was previously pretreated with dilute sulfuric acid. Scanning electron microscopy indicated significant differences in the structural composition of the samples because of the pretreatment with H2SO4 at different concentrations, and ATR-FTIR analysis highlighted the changes in the chemical composition in the pretreated wheat straw as compared to the untreated one. HPLC-RID was used to identify and quantify the carbohydrates content resulted from enzymatic hydrolysis to evaluate the potential of using wheat straws as a raw material for production of cellulosic ethanol in Romania. The highest degradation of lignocellulosic material was obtained in the case of pretreatment with 3% H2SO4 (v/v), a solid-to-liquid ratio of 1/30 and an enzyme dose of 90/90 µL/g. Simultaneous saccharification and fermentation were performed using Saccharomyces cerevisiae yeast, and for monitoring the fermentation process a BlueSens equipment was used provided with ethanol, O2 and CO2 cap sensors mounted on the fermentation flasks. The highest concentration of bioethanol was obtained after 48 h of fermentation and it reached 1.20% (v/v).

ABSTRACTThe growth of GaAs from triethylgallium (TEG) and trimethylgallium (TMG) with tertiarybutylarsine (tBAs), triethylarsenic (TEAs), and trimethylarsenic (TMAs), has been investigated by using a reactor equipped with a recording microbalance for in situ rate measurements. Rate data show that the growth with these precursors is dominated by the formation of adduct compounds in the gas lines, by adduct related parasitic gas phase reactions in the heated zone, and by the surface reactions. A model is proposed for the competition between deposition reactions and the parasitic gas phase reactions. Model predictions are in very good agreement with experimental data for all combinations of precursors except for TEG/TMAs where extensive gallium droplet formation is observed at low temperatures. Growth of reasonable quality GaAs with Hall mobilities of 7600 cm2/Vs at 77 K using TEG and tBAs is reported for the first time.
