Siegesbeckia glabrescens (Compositae), an annual herb indigenous to Korean mountainous regions and has been eaten as a food in Korea. This study investigated ABTS, DPPH and nitric oxide (NO) radical-scavenging activities, and melanin production and TYR inhibitory effects-guided fractionation to identify therapeutic phytochemicals from S. glabrescens that can attenuate oxidation and melanogenesis in murine melanoma B16F10 cells. Nine compounds with inhibitory effects on melanin production, and TYR activity, and ABTS, DPPH, and NO radical scavenging activity were isolated from the 100% ethanol fraction from S. glabrescens. Among the nine compounds, kirenol (K), methyl ent-16α, 17-dihydroxy-kauran-19-oate (MDK) had strong inhibitory effects on melanin production and TYR activity with antioxidant effects. Western blot analysis revealed that K and MDK suppressed tyrosinase-related protein (TYRP)-1, TYRP-2 and microphthalmia-associated transcription factor (MITF) expression. Moreover, these two compounds inhibited intracellular reactive oxygen species (ROS) level in tert-butyl hydroperoxide (t-BHP)-treated B16F10 cells. Our results suggest that S. glabrescens containing active compounds such as K and MDK, which has antioxidant and antimelanogenesis effects, is the potent therapeutic and functional material for the prevention of oxidation-induced hyperpigmentation.

This study evaluated bioactivity-guided fractionation as a means to identify therapeutic phytochemicals from Pyracantha angustifolia that can attenuate melanogenesis and oxidation. Seven compounds with inhibitory effects on melanin production and tyrosinase (TYR) activity, and ABTS and DPPH radical-scavenging activities, which have not been reported as whitening materials, were isolated from the n-butanol fraction from P. angustifolia leaves (PAL). Among the seven compounds, p-hydroxybenzoic acid β-d-glucosylester (HG), and cimidahurinine (CH) had strong inhibitory effects on melanin production and TYR activity, as well as ABTS and DPPH radical-scavenging activities. Western blot analysis showed that HG and CH suppressed tyrosinase-related protein (TYRP)-1 and TYRP-2 expression. Moreover, HG and CH inhibited reactive oxygen species (ROS) generation in tert-butyl hydroperoxide (t-BHP)-treated B16F10 cells. These results suggest that P. angustifolia containing active compounds, such as HG and CH, is a potent therapeutic candidate for the development of hypopigmenting agents.

The phenolic compounds and radical scavenging activity of ethanolic extracts of five adzuki beans were evaluate according to cultivar. The predominant phenolic acids in five cultivars of adzuki bean were (+)-catechin and gallic acid. Antioxidant capacities were determined using 2,2-diphenyl-1-picrylhydrazyl and 2,2-azinobis (3-ethylbenothiazoline-6-sulphonic acid) diammonium salt, and reducing power was positively enhanced according to the total polyphenolic content. In a cell-based assay, adzuki beans showed cytoprotective effects against oxidative stress induced by tert-butyl hydroperoxide and inhibitory effects on the production of reactive oxygen species, except for the cultivar Vigna angularis var. nipponensis cv. Whinnarae. In conclusion, (+)-catechin was the predominant phenolic compound found in adzuki beans, but there were differences according to the cultivar. Overall, the adzuki bean cultivars showed different antioxidant activities and cytoprotective effects according to the concentration and composition of phenolic compounds.

Natural antioxidants can protect the human body from free radicals and retard the progress of many chronic diseases as well as lipid oxidative rancidity in foods. The role of antioxidants has protected effect against free radical damage that may cause many diseases including cancer. Primary sources of naturally occurring antioxidants are known as whole grains, fruits, and vegetables. Several studies suggest that regular consumption of nuts, mostly walnuts, may have beneficial effects against oxidative stress mediated diseases such as cardiovascular disease and cancer. The role of antioxidants has attracted much interest with respect to their protective effect against free radical damage that may cause many diseases including cancer. Juglans regia L. (walnut) contains antioxidant compounds, which are thought to contribute to their biological properties. Polyphenols, flavonoids and flavonols concentrations and antioxidant activity of Leaves, Stems and Nuts extract of Juglans regia L. as evaluated using DPPH, ABTS, Nitric acid, hydroxyl and superoxide radical scavenging activity, lipid peroxidation and total oxidation activity were determined. The antioxidant activities of Leaves, Stems and Nuts extract of Juglans regia L. were concentration dependent in different experimental models and it was observed that free radicals were scavenged by the test compounds in all the models.

This study was performed to clarify the inhibitory effects of cycloheterophyllin on melanin synthesis. In order to elucidate the inhibitory effects of cycloheterophyllin on the B16F10 cell line, cell viability, messenger ribonucleic acid (mRNA) expressions, tyrosinase activity assay, and melanin production assay were measured. The effects of cycloheterophyllin on tyrosinase-related protein 1 (TYRP1)/TYRP2/tyrosinase (TYR)/microphthalmia-associated transcription factor (MITF) mRNA expressions and melanin content were determined. Quantitative real-time RT-PCR showed that cycloheterophyllin decreased the mRNA expression level of TYRP1/TYRP2/TYR/MITF genes and melanin production contents than α-MSH-treated B16F10 cells. The tyrosinase activity assay revealed that cycloheterophyllin decreased the melanin production in the B16F10 cells. These data show that cycloheterophyllin increases the whitening effects in the B16F10 cells; thus, cycloheterophyllin is a potent ingredient for skin whitening. Thus, further research on the mechanism of action of cycloheterophyllin for the development of functional materials should be investigated.

Abstract Purpose To characterize and optimize the productivity of melanin using an extremotolerant actinobacterium, Dietzia schimae NM3, for the first time. Methods An extracellular brown pigment produced by D. schimae NM3 in the nutrient broth and cheese whey medium by adding L-tyrosine. The extracted melanin was analyzed by UV-visible, HPLC, and FTIR assays. The radical scavenging activity (by DPPH) and sun protection factor (SPF) of the extracted melanin were measured. The melanin cytotoxicity was assayed by MTT and chromate biosorption was measured by atomic absorption spectroscopy. Finally, melanin production by D. schimae NM3 was optimized by response surface methodology (RSM) using Box-Behnken design in the whey medium. Result The purified melanin showed similar peak to the standard melanin (SIGMA) at 3.5 min in HPLC, and C=O bands, NH2, CH, C-N, and aromatic groups by FTIR. The radical scavenging activity (by DPPH) and SPF of the extracted melanin were obtained 188.9% and 20.22, respectively. Using MTT assay, the melanin revealed non-toxic effect on the normal human fibroblast (HFB) cell culture. The melanin yield 790 mg l−1, and tyrosinase activity 3400 U ml−1 were obtained in the medium contained whey powder [5% (w v−1)], L-tyrosine 2.5 g l−1, CuSO4 0.013 g l−1, and pH 10.5, incubated at 32 °C for 3 days. The ANOVA results indicated significant P-value, model F-value, and probability, with insignificant lack of fit. After optimization with mono-factors, the nutrient broth came up with melanin yield as 1.2 g l−1 and tyrosinase activity as 4040 U ml−1. Conclusion This is the first report of melanin production by D. schimae NM3 and this natural melanin showed valuable biological properties such as high antioxidant activity and radioprotection (SPF) and the biocompatibility to human cell line.

Kedelai adalah produk pangan bernilai gizi tinggi yang banyak dikonsumsi oleh masyarakat Indonesia. Pada penelitian ini, kedelai dimodifikasi dengan cara perkecambahan. Komponen zat gizi akan berubah menjadi senyawa-senyawa yang lebih sederhana selama proses perkecambahan sehingga menjadi lebih mudah dicerna bagi tubuh manusia. Selain itu, perkecambahan juga dapat meningkatkan kapasitas antioksidan. Tujuan dari penelitian ini adalah menentukan kondisi perkecambahan untuk mengoptimalkan kapasitas antioksidan dari biji kedelai varietas Wilis. Hasil penelitian menunjukkan bahwa kondisi terbaik untuk mengoptimalkan kapasitas antioksidan adalah dengan cara perkecambahan pada suhu 25°C selama 15 jam dengan rasio biji kedelai dan air 1:3 tanpa adanya penambahan garam. Kapasitas antioksidan kecambah kedelai var. Wilis pada 100 ppm memiliki radical scavenging activity sebesar 9,69% dengan scavenging ternormalisasi 0,65% dengan total fenol 2,1 mg GAE/g pada biji dan 2,9 mg GAE/g pada kecambah. Dari hasil identifikasi GC-MS ditemukan senyawa-senyawa antioksidan yang terdapat pada biji kedelai dan kecambah kedelai berupa benzoic acid, 2-methoxy-4-vinylphenol, 2,5-dihydroxytoluene, pyrogallol 1,3-dimethyl ether, 2,5-dihydroxytoluene, pyrogallol 1,3-dimethyl ether, hexadecanoic acid methyl ester dan methyl oleate serta terdapat juga senyawa-senyawa antioksidan baru yang muncul setelah proses perkecambahan yaitu neophytadiene, campesterol, stigmasterol, delta-tocopherol dan gamma-tocopherol.  ABSTRACT: Soybean is a high nutrition food product that is largely consumed by Indonesians. In this research, the soybean will be modified by going through the germination process. Complex nutrition would be changed into substances that were more simple during the germination process. Therefore, it would be easier for human to digest it. Moreover, this process might also increase the antioxidant capacity of the soybean itself. The aim of this research is to find out the most suitable germination condition in order to acquire the optimum antioxidant capacity of Wilis variety soybean. The result shows that the best condition to optimize the antioxidant capacity is by performing the germination process at 25°C for 15 hours with soybean to water ratio being 1:3 without any salt addition. The antioxidant capacity of Wilis soybean at 100 ppm has 9,69% of radical scavenging activity, with 0,65% being normalized. The total phenolic compound found in the beans was 2,1 mg GAE/g, whereas 2,9 mg GAE/g were found in the sprout. Based on the GC-MS identification, the antioxidant compounds that are found in both the soybean and sprout are known to be benzoic acid, 2-methoxy-4-vinylphenol, 2,5-dihydroxytoluene, pyrogallol 1,3-dimethyl ether, 2,5-dihydroxytoluene, pyrogallol 1,3-dimethyl ether, hexadecanoic acid methyl ester, and methyl oleate. In addition, there are also several new antioxidant compounds that emerge after the germination process, which are neophytadiene, campesterol, stigmasterol, delta-tocopherol, and gamma-tocopherol. The aim of this study was to optimize the extraction of antioxidant compounds from Schinus areira leaves using  ultrasound assisted extraction and response surface methodology. The effect of sonication time and plant material:solvent ratio were used to optimize the recovery. Results showed that a high recovery of antioxidant compounds from leaves of three different S. areira specimens was achieved under optimized conditions. The leaf extracts obtained displayed a DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical scavenging activity analogous to the well-known antioxidant trolox  (EC50 = 23-46 vs 36.1 µg/mL, respectively). In addition, these extracts showed a good potency to eliminate superoxide and nitric oxide-radicals as well as a moderate antimicrobial activity against gram positive Staphylococcus aureus and Enterococcus faecalis and yeast. HPLC chromatography analysis of the three S. areira leaf extracts showed different high contents of kaempferol-3-O-rutinoside, quercetin-3-O-galactoside and 3-O-caffeoylquinic acid. The results showed that the S. areira leaf extracts contained a high amount of antioxidant phenolic compounds, which might be a valuable source to be used as additives in plant-based foods.

Turmeric (Curcuma longa L.), a member of Zingiberacaeae, is used for Thai traditional medicine, flavoring, preservative, and coloring agent. In Southern Thailand, turmeric is a famous ingredient for food recipes. Not only is the ripened rhizome used for cuisine, but also the young rhizomes. Previous studies have indicated that a variety of turmeric products from the ripened rhizome are valuable source of antioxidant compounds. However, there are ambiguous data regarding the level of antioxidant activity of the young rhizome. The aim of this study was to investigate the level of total phenolic content (TPC) and antioxidant activity of the young rhizomes by comparing to the ripened rhizome. TPC and antioxidant activity of aqueous and 80% methanolic extracts from the young turmeric grown in Southern Thailand were estimated using the Folin-Ciocalteu method and DPPH free radical scavenging activity (DPPH-RSA) assay, respectively. Results showed that the concentration of TPC determined in methanolic extract (19.80±0.25 mg GAE/ g extract) from the young rhizome was significantly higher than the corresponding aqueous extract (18.38±0.41 mg GAE/ g extract). The aqueous and methanolic extracts exhibited significant inhibition in DPPH-RSA with the half-maximal inhibitory concentration (IC50) value of 5.88±0.70 mg/mL and 3.00±0.31 mg/mL, respectively. However, in comparison with the ripened rhizome, TPC and DPPH-RSA of the young rhizome were lower. These results indicate that the young rhizome grown in Southern Thailand could be a source of antioxidant compounds, but the ripened rhizome provides better antioxidant properties than the young rhizome.

The methanol extract of the leaves of Buddleja asiatica Lour. (Loganiaceae) showed antioxidant activity toward the well known in vitro antioxidant tests such as total antioxidant capacity by the phosphomolybdenum method, free radical scavenging activity by the 1,1- diphenyl-2-picrylhydrazyl scavenging assay (DPPH assay) and hydrogen peroxide scavenging methods. Due to the high scavenging activity of the n-butanol successive fraction toward DPPH and H2O2 (SC50 = 11.99 and 18.54 μg/ml, respectively), this extract was subjected to chromatographic separation and isolation. Four non-phenolic compounds were isolated and identified on the basis of spectroscopic and chemical analyses: 1-O-ß-D-glucopyranosyl- 2-methoxy-3-(2-hydroxy-triaconta-3,12-dienoate)-glycerol (1), 3-O-[α-l-rhamnopyranosyl- (1→4)-ß-d-glucopyranosyl-(1→3)]-[ß-d-glucopyranosyl-(1→2)]-ß-d-fucopyranosyl-olean- 11,13(18)-diene-3ß,23,28-triol (2), 3-O-[α-l-rhamnopyranosyl-(1→4)-ß-d-glucopyranosyl- (1→4)-ß-d-glucopyranosyl-(1→3)]-ß-d-fucopyranosyl-olean-11,13(18)-diene-3ß,23,28-triol (3), and 3-O-[α-l-rhamnopyranosyl-(1→4)-ß-d-glucopyranosyl-(1→3)]-[ß-d-xylopyranosyl- (1→2)]-ß-d-glucuronopyranosyl-acid-olean-11,13(18)-diene-3ß,23,28-triol (4). The four compounds were evaluated as antioxidant agents using the three antioxidant bioassay tests.
