THE IMPACT OF THYME AND CAMOMILE FLOWER EXTRACTS ON TRYPSIN CATALYTIC ACTIVITY

The development of pathological processes in humans and animals is accompanied by an imbalance of regulatory mechanisms, in particular, proteinase enzymes and their regulators. Endogenous regulators, inhibitors and activators often fail to maintain the balance in the system “enzyme-substrate-inhibitor (activator)”, which leads to destructive inflammatory, immune reactions, and dyscrasia. Therefore, the search for unknown proteinase regulators is an important step towards the creation of new drugs to treat the diseases associated with changes in proteolysis processes. In this regard, we propose a hypothesis about the regulatory role of medicinal herbs that are widely used in both traditional and folk medicine. According to our assumption, the chemical compounds contained in medicinal herbs can both activate and inhibit proteolysis in cells. The aim of the research was to study the effects of the inflammatory process, the influence of the regulatory activity of aqueous thyme and chamomile extracts on the trypsin activity in the model experiments. Materials and Methods. In the studies the authors used crystalline trypsin, thyme, and chamomile flowers. They determined trypsin activity using the Anson method. Results. New information was obtained on the effect of an aqueous extract of medicinal herbs, in particular, thyme and chamomile flowers, on the activity of the proteolytic enzyme trypsin. This information can improve the herbs efficacy in the treatment of inflammatory processes. Conclusion. Thyme and chamomile flower extracts have an activating trypsin effect. The level of trypsin activation is proportional to the amount of extracts introduced into the incubation medium. Thyme extract has a more pronounced effect on trypsin activity. Развитие патологических процессов в организме человека и животных сопровождается разбалансировкой регуляторных механизмов, в частности состояния ферментов – протеиназ и их регуляторов. Эндогенные регуляторы, ингибиторы и активаторы зачастую не справляются со своей задачей по поддержанию баланса в системе «фермент-субстрат-ингибитор (активатор)», что приводит к возникновению деструктивных воспалительных, иммунных реакций, нарушению гомеостаза. Поэтому поиск пока еще неизвестных регуляторов протеиназ представляет собой важный шаг к созданию новых препаратов для лечения заболеваний, связанных с изменениями, происходящими в процессах протеолиза. В связи с этим нами предложена гипотеза о регуляторной роли лекарственных трав, широко используемых как в традиционной, так и в народной медицине. По нашему предположению, химические соединения, содержащиеся в лекарственных травах, вероятно, могут как активировать, так и ингибировать процессы протеолиза в клетках. Целью исследования было изучение воздействия в модельных опытах воспалительного процесса, влияния регуляторной активности водных экстрактов чабреца и ромашки на активность протеолитического фермента трипсина. Материалы и методы. В исследованиях использовали «Трипсин кристаллический», чабрец и цветки ромашки. Определение активности трипсина проводили по методу, разработанному Ансоном. Результаты. Получены новые сведения о влиянии водного экстракта лекарственных трав, в частности чабреца и цветков ромашки, на активность протеолитического фермента трипсина. Данная информация позволит повысить эффективность использования лекарственных растений в лечении воспалительных процессов. Выводы. Экстракты чабреца и цветков ромашки оказывают активирующее действие на протеолитический фермент трипсин. Уровень активации трипсина пропорционален количеству экстрактов, вносимому в инкубационную среду. Экстракт чабреца оказывает более выраженное влияние на активность трипсина. Background. Pharmaceutical risk sharing agreements (RSAs) are commonly used to manage uncertainties in costs and/or clinical benefits when new drugs are added to a formulary. However, existing mathematical models of RSAs ignore the impact of RSAs on clinical and financial risk. Methods. We develop a model in which the number of patients, total drug consumption per patient, and incremental health benefits per patient are uncertain at the time of the introduction of a new drug. We use the model to evaluate the impact of six common RSAs on total drug costs and total net monetary benefit (NMB). Results. We show that, relative to not having an RSA in place, each RSA reduces expected total drug costs and increases expected total NMB. Each RSA also improves two measures of risk by reducing the probability that total drug costs exceed any threshold and reducing the probability of obtaining negative NMB. However, the effects on variance in both NMB and total drug costs are mixed. In some cases, relative to not having an RSA in place, implementing an RSA can increase variability in total drug costs or total NMB. We also show that, for some RSAs, when their parameters are adjusted so that they have the same impact on expected total drug cost, they can be rank-ordered in terms of their impact on variance in drug costs. Conclusions. Although all RSAs reduce expected total drug costs and increase expected total NMB, some RSAs may actually have the undesirable effect of increasing risk. Payers and formulary managers should be aware of these mean-variance tradeoffs and the potentially unintended results of RSAs when designing and negotiating RSAs.

Hodgkin lymphoma (HL) is a lymphoid-type hematologic disease that is derived from B cells. The incidence of this lymphoid malignancy is around 2–3/100,000/year in the western world. Long-term remission rates are linked to a risk-adapted approach, which allows remission rates higher than 80%. The first-line treatment for advanced stage classical HL (cHL) widely used today is doxorubicin, bleomycin, vinblastine, and dacarbazine (ABVD) or escalated bleomycin, etoposide, doxorubicin, cyclophosphamide, vincristine, procarbazine, and prednisone (BEACOPPesc) chemotherapy. Randomized studies comparing these two regimens and a recently performed meta-analysis have demonstrated consistently better disease control with BEACOPPesc. However, this treatment is not the standard of care, as there is an excess of acute hematological toxicities and therapy-related myeloid neoplasms. Moreover, there is a recurrent controversy concerning the impact on overall survival with this regimen. More recently, new drugs such as brentuximab vedotin and checkpoint inhibitors have become available and have been evaluated in combination with doxorubicin, vinblastine, and dacarbazine (AVD) for the first-line treatment of patients with advanced cHL with the objective of tumor control improvement. There are still major debates with respect to first-line treatment of advanced cHL. The use of positron emission tomography-adapted strategies has allowed a reduction in the toxicity of chemotherapy regimens. Incorporation of new drugs into the treatment algorithms requires confirmation.

Glycolysis is a crucial metabolic process in rapidly proliferating cells such as cancer cells. Phosphofructokinase-1 (PFK-1) is a key rate-limiting enzyme of glycolysis. Its efficiency is allosterically regulated by numerous substances occurring in the cytoplasm. However, the most potent regulator of PFK-1 is fructose-2,6-bisphosphate (F-2,6-BP), the level of which is strongly associated with 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase activity (PFK-2/FBPase-2, PFKFB). PFK-2/FBPase-2 is a bifunctional enzyme responsible for F-2,6-BP synthesis and degradation. Four isozymes of PFKFB (PFKFB1, PFKFB2, PFKFB3, and PFKFB4) have been identified. Alterations in the levels of all PFK-2/FBPase-2 isozymes have been reported in different diseases. However, most recent studies have focused on an increased expression of PFKFB3 and PFKFB4 in cancer tissues and their role in carcinogenesis. In this review, we summarize our current knowledge on all PFKFB genes and protein structures, and emphasize important differences between the isoenzymes, which likely affect their kinase/phosphatase activities. The main focus is on the latest reports in this field of cancer research, and in particular the impact of PFKFB3 and PFKFB4 on tumor progression, metastasis, angiogenesis, and autophagy. We also present the most recent achievements in the development of new drugs targeting these isozymes. Finally, we discuss potential combination therapies using PFKFB3 inhibitors, which may represent important future cancer treatment options.

Abstract Background Salvadora persica is an endangered medicinal plant due to difficulties in its traditional propagation. It is rich in bioactive compounds that possess many pharmaceutical, antimicrobial activities and widely used in folk medicine. The current study aims at in vitro propagation of Salvadora persica and the application of different nanoparticles (NPs) to induce the synthesis of bioactive and secondary metabolites within the plant. The cellular and genetic responses to the application of different NPs were evaluated. Results The impact of nanoparticles NPs (ZnO, SiO2, and Fe3O4) on callus growth of Salvadora persica and the production of its active constituent benzyl isothiocyanate was examined, regarding some oxidative stress markers, antioxidant enzymes, and genetic variabilities. An encouraging impact of 0.5 mg/l ZnO NPs on benzyl isothiocyanate production was shown reaching up to 0.905 mg/g callus fresh weight in comparison to 0.539 mg/g in control callus. This was associated with decreasing hydrogen peroxide content and increasing superoxide dismutase and peroxidase activities. The deposition of the NPs on cellular organelles was detected using a transmission microscope. Fifteen Inter-Simple Sequence Repeats (ISSR) primers detected an overall, 79.1% polymorphism among different treatments. A reduction in genomic DNA template stability (GTS) was made and was more pronounced in higher doses of different NPs. Conclusion This study is a stepping stone in developing a productive protocol for in vitro production of benzyl isothiocyanate from Salvadora persica using NPs as a valuable anticancer compound.

AbstractThe establishment of colonies along the Hellespont by inhabitants of Ionia, Athens and Lesbos is well-known from historical texts. Recently, stratified contexts at Troy as well as other surveys and excavations have yielded new information about the chronology and material markers of Archaic period settlements in the Troad and the Gallipoli peninsula. The archaeological evidence for colonisation in this region is not clearly seen until the late seventh to early sixth century BC when there is a dramatic change in the material culture. Destruction evidence from Troy indicates that the new settlers probably entered a weakened and depopulated region in the second half of the seventh century BC. The Ionian colonists transplanted their pottery traditions and started production of East Greek style ceramics in the Troad. Neutron Activation Analysis of Wild Goat style ceramics found at Troy offers further confirmation for the existence of Hellespontine Wild Goat style ceramic production centres. The Wild Goat style examples from Troy help to define the characteristics of the Hellespontine group, as well as the chronology and impact of colonisation in this area.

In chronic kidney disease inflammatory processes and stimulation of immune cells result in overproduction of free radicals. In combination with a reduced antioxidant capacity this causes oxidative stress. This review focuses on current pathogenic concepts of oxidative stress for the decline of kidney function and development of cardiovascular complications. We discuss the impact of mitochondrial alterations and dysfunction, a pathogenic role for hyperuricemia, and disturbances of vitamin D metabolism and signal transduction. Recent antioxidant therapy options including the use of vitamin D and pharmacologic therapies for hyperuricemia are discussed. Finally, we review some new therapy options in diabetic nephropathy including antidiabetic agents (noninsulin dependent), plant antioxidants, and food components as alternative antioxidant therapies.

A wave motion compensating platform has the function of compensating the ship’s generalized heave motion (a coupling result of roll, pitch and heave). It can decrease the impact of ship motion on some sea works and equipments. The hydraulic mechanism of platform system has the characteristics of nonlinear and big inertia. In order to compensate generalized heave motion effectively, an adaptive predictive control policy is used for controlling the hydraulic mechanism. Based on equal-dimension and new information, an automation regressive model can get adaptive multi-step prediction. The model parameter estimation based on the least square algorithm is easy to blow up and be unstable when the system has random noise. To improve the problem solution, a damped recursive least square algorithm is proposed to estimate the parameters on line. For the short regulation time, strong anti-disturbance ability and great robustness, a nonlinear PID controller whose gain parameters vary with errors is suitable for controlling the hydraulic mechanism. Using the collected experimental data, the simulations suggest that adopting the above adaptive predictive control policy to control hydraulic mechanism is able to decrease the generalized heave amplitude of wave motion compensating platform.

This article provides an analysis of scientific research on the life and activities of the first Russian charge d’ affaires and consul general in Korea Karl I. Waeber, shows the specific contribution of scholars to the study of the professional biography of this outstanding diplomat. Despite the fact that the activity of K.I. Weber in Korea is partially reflected in the works of Boris D. Pak and Bella B. Pak on the history of Russo-Korean relations, as well as in several separate articles, the first special monographic work on this topic belongs to the pen of the author of this article. The monographic research focuses on a detailed coverage of the tasks, goals facing Waeber in Korea, the specific forms and conditions for their implementation, the impact he exerts on the course of the Russian government towards Korea; analysis of the most complex international circumstances, against the background of which he made certain decisions.   This article contains answers to T.M. Simbirtseva and S.V. Volkov’s critical remarks regarding some of the information and photographic documents given in the work concerning K.I. Waeber and the accusations against the author of the article in connection with the publication in Germany in the summer of 2021 of Dr. S. Braezel's photobook "Pictures of the life of a diplomat between Europe and East Asia: Karl von Waeber (1841-1910)". The author of the article drew attention to some erroneous judgments in the article by T.M. Simbirtseva and S.V. Volkov, formed due to ignorance and bias, analyzed and refuted the most unfounded accusations, clarified the position regarding new information about K.I. Waeber.

Purpose The purpose of this study is to investigate whether the STOXX Europe Christian price index (SECI) follows the premise of efficient market hypothesis (EMH). Design/methodology/approach The study used daily data of SECI for the period of 15 years as its launch date i.e. 31 December 2004 to 31 December 2019. Data are analyzed by taking a full-length sample and fixed-length subsample. For subsample, the data are divided into five subsamples of three years each. Subsample analysis is important for analyzing time varying efficiency of the series, as the market is said to follow EMH if it is being efficient throughout the sample. Both type of samples is examined through linear tests including autocorrelations test and variance ratio (VR) test. Findings Tests applied conclude that SECI is weak-form efficient, which means that the prices of the index include all the relevant past information and immediately react to new information. Hence, the investors cannot earn abnormal returns. Originality/value Religion-based indices grasped the attention of investors, policymakers and academic researchers because of increased concern over ethics in business. Though the impact of religion on the economy have been studied in many ways but the efficiency of religion-based indices have been less explored. The current study is primary in its nature as it analysis the efficiency of SECI. This index is important to explore because Christianity is the world’s top religion with 2.3 billion followers around the globe.
