Telah dilakukan penelitian tentang aktivitas penstabil oksigen singlet dari hasil fraksinasi ekstrak metanol daun kayu manis (Cinnamomum burmanii) terhadap fotooksidasi asam askorbat dengan menggunakan cahaya dan eritrosin sebagai sensitiser. Serbuk daun kayu manis diekstraksi secara maserasi dengan metanol 95 % selama 3 x 24 jam, selanjutnya ekstrak yang diperoleh difraksinasi berturut-turut dengan pelarut n-heksan, etil asetat, butanol dan air. Penentuan kandungan total fenolik diukur dengan metode Folin-Ciocalteu dan aktivitas penstabil oksigen singlet diukur dengan laju penurunan konsentrasi asam askorbat. Hasilnya menunjukkan bahwa fraksi etil asetat memiliki kandungan total fenolik tertinggi yaitu 166,888 mg/g diikuti dengan fraksi butanol, n-heksan, dan air.  Fraksi etil asetat juga memiliki aktivitas penstabil oksigen singlet yang kuat, ditunjukkan dengan laju kerusakan fotooksidatif asam askorbat yang kecil yaitu -0,75 μg.mL-1.min-1A study of singlet oxygen quenching activity has been done from the fractination result of methanol extract of cinnamon leaf (Cinnamomum burmanii) against photo-oxidation of ascorbic acid by using light and erythrosine as sensitized. The cinnamon leaf powder was extracted by macerating with 95% methanol for  3 x 24 hours, then the extracts obtained was fractionated successively with n-hexane, ethyl acetate, butanol and water solvent. The determination of total phenolic content was measured by the Folin-Ciocalteu method and the singlet oxygen quenching activity was measured by the rate of decrease in ascorbic acid concentration. The results show that the ethyl acetate fraction has the highest total phenolic content of 166,888 mg/g followed by the fraction of butanol, n-hexane, and water, respectively. The ethyl acetate fraction also has a strong singlet oxygen quenching activity, indicated by a small photo-oxidative damage rate of ascorbic acid of                  -0.75 μg.mL-1.min-1

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

Abstract Cadaba farinosa is used in traditional medicine for treatment of cancer, diabetes, and rheumatism. The research was aimed at evaluating the phytochemical and antioxidant activity of the extracts. The powdered stem bark was extracted successively with the aid of Soxhlet extractor using n-hexane, ethyl acetate, and methanol. The resulting extracts were concentrated on rotary evaporator and the percentage yields were calculated. The phytochemical and TLC profiles of the extracts were studied. The antioxidant activity of the extracts and ascorbic acid (standard) were determined using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical assay. The total phenolic content of the extracts and tannic acid (standard) were evaluated using Folin–Ciocalteu reagent. The percentage yields of n-hexane, ethyl acetate, and methanol extracts were found to be 1.19, 1.37, and 13.93%, respectively. The phytochemical screening revealed the presence of tannins, flavonoids, saponins, cardiac glycosides, alkaloids, and triterpenoids. The TLC profiles of the extracts revealed the presence of compounds as evidenced from their R f values. The total phenolic content of ethyl acetate and methanol extracts were found to be 135 and 112 mg, respectively. The free radical scavenging activity demonstrated by the extracts was comparable to ascorbic acid. The ethyl acetate extract had higher phenolic content and demonstrated the highest free radical scavenging with IC50 31.07 mg/mL. The results of research have provided strong preliminary evidence of antioxidant activity which may be because of phenolic compounds in the extracts.

Objective: This study aimed to determine the total phenolic content, correlation of it with antioxidant capacity, and peanut hulls as an antiproliferationon Henrietta Lacks (HeLa) cancer cells, which Indonesia has a serious problem in term of cervix cancer.Methods: Peanut hulls were extracted by Soxhlet extraction, ultrasound vibration, and reflux boiling to obtain the best extraction method. The totalphenolic content of the ethanol extract and the ethyl acetate fraction was determined using Folin–Ciocalteu method. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay was used to evaluated an antioxidant capacity of both samples. Identification of the isolated flavone was done by ultraviolet spectra and analyzed using liquid chromatography-mass spectrometer. Inhibition of proliferation of HeLa cancer cells was tested for the purified fraction using 3-(4,5-dimethyl thiazole-2-yl)-2, 5-diphenyltetrazolium bromide assay.Results: The results of total phenolic content determination giving 262 (ethanol extract) and 532 (ethyl acetate fraction) mg gallic acid equivalent/g extract, respectively. DPPH assay resulted antioxidant capacity with value of inhibitory concentration 50% (IC) was 36.36 (ethanol extract) and 18.68 (ethyl acetate fraction) µg/mL, respectively. Identification of isolated flavone resulted an apigenin and indicated moderate potency in inhibiting the proliferation of HeLa cancer cells with IC 50 value of 34 µg/mL. 50Conclusions: There was a correlation between the total phenolics with antioxidant capacity of the peanut hulls. The isolated flavone is predominated by apigenin. This isolated compound is potential as antioxidant and inhibiting the proliferation of HeLa cancer cells moderately.Keywords: Peanut hulls, Total phenolic, Antioxidant capacity, Flavone, Apigenin, Henrietta Lacks cancer cells.

Objective: This study was to investigate the phytochemical screening, total phenolic content (TPC), and antioxidant activity of fractions of water, ethyl acetate, and n-hexane from Dendrophthoe pentandra (L.) Miq. and mistletoe of Moringa oleifera Lam.Methods: Simplicia extracted with 96% ethanol using maceration method. Liquid-liquid extraction was used for fractionation with water, ethyl acetate, and n-hexane as solvent. Phytochemical screening, determination of TPC, and antioxidant activity were performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method. The TPC was measured by Folin–Ciocalteu reagent and use gallic acid (GA) as a standard.Results: Water fraction contains saponin and polyphenols. Ethyl acetate fraction contains alkaloids, flavonoids, saponins, terpenoids, and polyphenols. N-hexane fraction only contains alkaloids and terpenoids. The TPC and antioxidant activity of fractions of water, ethyl acetate, and n-hexane were 9.42±1.16%, 21.35±1.86%, and 67.40±0.82% mg GA equivalent/g fraction and 29.46 μg/ml±0.99%, 7.08 μg/ml±0.39%, and 10.90 μg/ml±1.05%, respectively.Conclusion: Fraction of ethyl acetate exhibited the highest antioxidant activity and TPC. This study showed that there were significant correlations between TPC and antioxidant activity at significance level p<0.05.

PENENTUAN TOTAL FENOLIK DAN AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DARI TEPUNG PELEPAH AREN (Arenga pinnata) ABSTRAK Tujuan penelitian ini untuk menentukan total fenolik dan aktivitas antioksidan fraksi etil asetat dari tepung pelepah aren (Arenga pinnata). Tepung pelepah aren diekstraksi maserasi menggunakan etanol hasil redestilasi cap tikus kemudian di partisi menggunakan pelarut petroleum eter, etil asetat dan aquades. Penentuan kandungan total fenolik menggunakan metode Folin-Ciocalteu dan aktivitas antioksidan menggunakan metode serapan radikal 1,1-difenil-2-pikrilhidrazil (DPPH). Didapati kandungan total fenolik fraksi etil asetat sebesar 172.857 ± 4.762 mg/kg dan aktivitas penangkal radikal bebas fraksi etil asetat sebesar 90.573 µg/mL. Hasil ini menunjukan bahwa fraksi etil asetat berpotensi sebagai antioksidan alami. Aren’s midrib powder extracted with maceration method with cap tikus redistillated ethanol and then partitioned with solvents such as petroleum ether, ethyl acetate and distillate water. The determination of phenolic content was using Folin-Ciocalteu method and the antioxidant activity was done using diphenylpycrylhydrazyl (DPPH) radical scavenging. Total phenolic value of ethyl acetate fraction is 172.857 ± 4.762 and IC50 value of ethyl acetate fraction is 90.573 µg/mL. This result shows ethyl acetate fraction as natural antioxidant potential. Penelitian ini bertujuan untuk,menentukan kandungan total fenolik, dan mengetahui tingkat toksisitas ekstrak muharang bawine (Dendrophthoefalcate (Lf) Etinggsh) dengan Metode BLST (Brine shrimp lethality test) dari beberapa pelarut.Uji toksisitas dengan menggunakan udang A. salina leach.Serbuk daun muharang bawine diekstraksi dengan cara maserasi bertingkat dengan menggunakan pelaut n-heksan, etil asetat dan metanol selama 3 x 24 jam sehingga diperoleh ekstrak kental. Penentuankandungan total fenolik diukur dengan metode Folin-Ciocalteu dan nilai LC50 dihitung dengan menggunakan SPSS 15.0.Kandungan total fenolik tertinggi terdapat pada hasil maserasi bertingkat ekstrak metanol, diikuti dengan hasil maserasi bertingkat ekstrak etil asetat dan hasil maserasi bertingkat ekstrak n-heksan. Hasil LC50 paling terbaik terdapat pada hasil maserasi bertingkat ekstrak metanol, kemudian diikuti dengan hasil maserasi bertingkat ekstrak etil asetat dan hasil maserasi bertingkat ekstrak n-heksan. ABSTRACT This study aims to determine the total phenolic content, and determine the toxicity level of muharang bawine extract (Dendrophthoe falcate (LF) Ettingsh) with the BLST (Brine Shrimp Lethality Test) method of several solvents. Toxicity test using A. salina Leach shrimp. Leaf Extract Muharang Bawine was extracted by multilevel maceration using n-hexane, ethyl acetate and methanol for 3 x 24 hours for thick extracts. Determination of total phenolic content was measured by the Folin-Ciocalteu method and LC50 valueswere calculated using SPSS 15.0. The highest total phenolic content was found in the results of maceration of methanol extract multilevel, followed by maceration results of multilevel ethyl acetate extract and maceration results of n-hexane extract. The best LC50 results were found in the results of maceration of methanol extract multilevel, then followed by maceration results of multilevel extract of ethyl acetate and maceration results of n-hexane extract.

Prunus domestica L. is a member of the Rosaceae family that shows many biological activities including antioxidant, antimicrobial, antihaemolytic, anti-inflammatory, hepatoprotective activity and many other activities. In the current study, we evaluated nutritive value, phytochemical screening, total phenolic content and antioxidant activity by DPPH and FRAP method for the different extracts obtained by successive soxhlet extraction using the different solvents based on their polarity. Results show that it is a good source of energy. Phytochemical screening revealed the presence of many secondary metabolites which include alkaloids, carbohydrates, glycosides, protein, steroids and terpenoids, fixed oils and fat as well as phenolic compounds. The highest total phenolic content was found in the ethyl acetate fraction. Highest antioxidant activity by DPPH method is reported in ethyl acetate fraction (IC50 =1837.399±0.377µg/ml) while the ferric reducing antioxidant power was maximum for diethyl ether (56.032±0.985µM/ml FRAP value = 0.325±0.002).

One of the traditionally used herbs is the byproduct of the maize plant, the 10-20 cm long corn silk which from the female maize flowers. The aim of this study is to evaluate the total phenolic content, antioxidant activity, and cytotoxicity activity of corn silk. The corn silk was minced and was extracted with methanol-water (80 % v/v), methanol, ethanol, ethyl acetate, and hexane by using the maceration method. The total phenolic content (TPC) of corn silk was determined to assess the presence and level of phenolic compounds in each sample. The antioxidant activities of all corn silk extracts were determined via DPPH method, and MTT assay was used to study the viability of the cells after the cells were treated with corn silk extracts at different time intervals. The highest phenolic content was exhibited by the methanol extract. The EC50 value for methanol-water (80 % v/v), methanol, ethanol, ethyl acetate and hexane extracts were 251 μg/ ml, 300 μg/ ml, 330 μg/ ml, 550 μg/ ml and 1736 μg/ ml respectively. The MTT assay, the lowest IC50 values at 24 and 48 hours intervals, was exhibited by methanol-water extract (104 μg/ ml). In contrast, methanol (308 μg/ ml) was found with the highest IC50 value for all 24, 48, and 72 hours intervals. At 72 hours interval, ethyl acetate (88 μg/ ml) shown the lowest IC50 value. This study suggested that corn silk could be potentially used as a source of antioxidant and can further evaluate for cancer studies.

A raw extract and four extractive fractions were obtained from Cedrus brevifolia (Cyprus cedar) bark. They were all studied regarding the phenolic content and profile using spectrophotometry and HPLC-DAD-ESI-MS. The antioxidant activity was investigated using in vitro assays: DPPH and ABTS radicals scavenging and reducing power assays. The ethyl acetate fraction had the highest total phenolic and proanthocyanidin contents; a taxifolin-O-hexoside, catechin, epicatechin and procyanidin oligomers (three dimers, two trimers) were identified in this fraction. The ethyl acetate fraction was found to possess the highest DPPH and ABTS radicals scavenging effects (EC50=13.9 ± 0.3 and 2.3 ± 0.0 μg/mL, respectively) and reducing capacity (EC50=9.1 ±0.1 μg/mL). Antioxidant effects were highly correlated with total phenolic and proanthocyanidin contents (r=0.89-0.99). These results suggest that Cedrus brevifolia bark is a new source of antioxidants.

The aim of the current research was to investigate and compare the total phenolic content and antioxidant capacity of sweet pepper cultivars at different development and maturation stages, in order to optimize the beneficial effects. For that purpose, three important sweet pepper cultivars, namely Dolmy-F1, Yahoo-F1 and Florinis-NS-700, were cultivated in a greenhouse. Their total phenolic content, ascorbic acid content and antioxidant properties were assessed at different development and maturation stages. In the aforementioned cultivars, the total phenolic content ranged from 345.2 to 602.1, 404.9 to 794.5, and 795.7 to 2220.3 μg GAE g−1 FW respectively. The ascorbic acid content ranged from 236 to 957, 258 to 1157, and 410 to 1550 μg AA g−1 FW respectively. The highest antioxidant activity was noted at the red maturity stage. Particularly the cultivar Florinis NS 700 was found to possess higher total phenolic, flavonoid phenol, non-flavonoid phenol, ascorbic acid contents, and greater antioxidant capacity, compared to the other cultivars. The results of our study recommend the consumption of the sweet peppers at red maturity stage, for achieving the maximum health-beneficial effects.
