This study was amid to  isolate  soil fungi from agriculture soil and study  their roles in the biodegradation of carbendazim fungicide at different concentrations were explored for two periods of incubation. Results showed the highest degradation rates of carbendazim at 4 part per million (ppm) concentration were observed by using single culture of Aspergillus niger (69.66% and 99.96% after 10 and 20 days, respectively).In the mixed cultures of Exerohilum sp., Fusarium sp., and A. niger, the maximum degradation rate (98.34%) was achieved at the same concentration after 10 days. The mixed culture of Exerohilum sp. and Fusarium sp. demonstrated the highest degradation rates of (92.14% and 55.74%) at 8 and 12 ppm concentrations, respectively, after 10 days of incubation.

This study focuses on the biodegradation of oxymatrine insecticide by some soil fungi isolated from four agriculture stations. The results showed that the highest degradation rate 94.66% was recorded by Ulocladium sp. at 10 days and A. niger recorded the lowest degradation rate 45.86%, while at 20 days Ulocladium sp. also showed the highest degradation rate 94.98% and the lowest degradation rate reached to 82.49% with A.niger. The mix (Exerohilum sp.+Ulocladium sp.) recorded the highest degradation rate of oxymatrine insecticide 90.22%, 88.51%, 85.34%  at 4, 8 and 12 ppm.The use of mixed isolates enhanced the biodegradation process. There is no study of oxymatrine biodegradation so this study is the first of its kind in the region which can be used as a baseline study for incoming studies


Rumen ciliates still have mysterious secrets and influences in ruminants. This study investigated the effect of transfaunation of pure and mixed cultures of rumen ciliates on physical clinical examination, selected serum parameters and milk profile in defaunated lactating dairy goats. A number of 8 Baladi native breed goats were randomly classified into two groups each one containing 4 goats. Pure culture group was transfaunated with 6 ml of pure culture of Holotricha spp., while mixed culture group was transfaunated with 6 ml of mixed culture of 81.85% Holotricha and 18.15% Ophryoscolex spp. once weekly for three consecutive weeks, after defaunation of both groups using 30 ml of 8% SLS for two consecutive days. Serum and milk samples were collected weekly for three successive weeks to study effect of type of ciliate culture, duration of transfaunation and their interaction. Results revealed that transfaunation of pure and mixed cultures of rumen ciliates had no effect on physical examination with minimal non-significant improvement of calcium, inorganic phosphorous, total protein and globulin in serum of defaunated goats. Transfaunation of pure or mixed cultures of rumen ciliates within three weeks could not improve significantly decreased milk fat % of defaunated goats without any effect on other measured milk profile parameters. It is concluded that further investigations on transfaunation without prior defaunation should be performed using different pure and mixed cultures of rumen ciliates for therapeutic and productive purposes.

A study was made of nitrogen-fixation byAzotobacter chroococcumalone in a medium containing dextrose (which it can utilize) and in mixture with a coliform organism on a medium containing no carbohydrate except starch, whichAzotobactercannot utilize unless it be hydrolysed by the coliform organism or some other agency.2. The amount of nitrogen fixed in the mixed cultures was found to be maximal at two temperatures, and a discussion is given of the causes thought to be operative in producing the double maximum.

The production and stability of two novel myco-flocculants produced by river water fungus (RWF) were investigated. Screening tests were conducted to find suitable nutrients, pH, nutrient concentration, inoculum dose, and stability for two myco-flocculants L. squarrosulus (RWF5) and S. obclavatum (RWF6). The strains showed good flocculating activity in reducing turbidity of kaolin suspension while malt extract was used as nutrient source. Supernatants of RWF5 and RWF6 were able to reduce turbidity from 900±10 NTU to 46 NTU (95%) and 195 NTU (78%), respectively. In order to enhance the production, optimization of cultivation conditions were studied using a one-factor-at-a-time (OFAT) method. L. squarrosulus (RWF5) reduced 96% of turbidity at optimum conditions, comprising of 0.1% (w/v) malt extract, 3% (v/v) inoculum dose, and initial pH 7.0 for 6 days. The results of the compatible mixed culture showed good flocculation activity at 88% compared to a single culture of S. obclavatum at 78%. On the other hand, L. squarrosulus showed better turbidity reduction in the single culture rather than the mixed culture. The stability of L. squarrosulus and S. obclavatum supernatants showed excellent turbidity reduction over a wide pH range of 4-8 with the maximal flocculation rate of 96% and 90%, respectively, at pH 7.0. They also exhibited high turbidity removal ability in a temperature range of 4 oC – 55 oC for 24h with a maximum turbidity removal rate of 96% (RW5) and 87% (RW6) at 25 oC. Time stability of the L. squarrosulus supernatant showed good turbidity removal potential at above 90% at room temperature (28± 2 oC) and 85% at low temperature (4 oC) for 12 days. The high flocculating rate of the myco-flocculants and their good stability under wide range of temperature indicated their potentiality as biodegradable flocculants for water and wastewater treatment industry. ABSTRAK: Keberhasilan dan kestabilan dua myco-gumpalan baharu oleh kulat air sungai (RWF) telah dikaji. Ujian penapisan telah dijalankan untuk mencari nutrien sesuai, pH, kepekatan nutrien, dos inokulum dan kestabilan dua myco-gumpalan L. squarrosulus (RWF5) dan S. obclavatum (RWF6). Rantaian menunjukkan aktiviti gumpalan yang baik dalam mengurangkan kekeruhan air sungai dengan penggantungan koalin di mana ekstrak malt telah digunakan sebagai sumber nutrien. Larutan supernatan RWF5 dan RWF6, masing-masing mampu mengurangkan kekeruhan dari 900±10 NTU kepada 46 NTU (95%) dan 195 NTU (78%). Bagi meningkatkan pengeluaran, keadaan optimum bagi menggalakkan hasil telah diselidiki menggunakan kaedah Satu Faktor pada Tiap Masa (OFAT). Pada takat optimum, L. squarrosulus (RWF5) mengurangkan 96% kekeruhan, ianya terdiri daripada ekstrak malt 0.1% (w/v), dos inokulum 3% (v/v) dan pH awal 7.0 selama 6 hari. Keputusan kultur campuran yang sesuai menunjukkan aktiviti penggumpalan yang baik pada 88% berbanding kultur sendirian S. obclavatum pada 78%. Pada waktu sama, L. squarrosulus menunjukkan pengurangan kekeruhan yang lebih baik dalam kultur sendirian berbanding kultur campuran. Kestabilan larutan supernatan L. squarrosulus dan S. obclavatum menunjukkan pengurangan kekeruhan yang sangat baik pada pH yang luas iaitu 4-8 dengan kadar maksimum kekeruhan pada 96% dan 90%, pada pH 7.0 masing-masing. Keduanya menunjukkan kebolehan penyingkiran kekeruhan yang tinggi pada skala suhu 4 oC – 55 oC selama 24 jam dengan kadar nyah kekeruhan maksimum pada 96% (RW5) dan 87% (RW6) pada suhu 25 oC. Kestabilan masa larutan supernatan L. Squarrosulus menunjukkan potensi penyingkiran kekeruhan terbaik atas 90% pada suhu bilik (28± 2 oC) dan 85% pada suhu rendah (4 oC) selama 12 hari. Kadar gumpalan yang tinggi oleh myro-gumpalan dan kestabilan yang baik pada julat suhu yang luas menunjukkan potensinya sebagai agen biodegradasi gumpalan kepada air dan industri rawatan loji air.

The objective of this study was to identify the effect of L. plantarum and S. cerevisiae mixed culture inoculation into cocoa pods silage on chemical composition and in vitro digestibility. The four treatments were: 1 kg freshly harvested cocoa pods without inoculants as control (K); K + L. plantarum (KLp); K + S. cerevisiae (KSc); and K + L. plantarum and S. cerevisiae mixture (KLp+Sc) 0.1% dry matter, Cassava meal were added in all treatments. Each treatment was replicated 3 times, and then fermented for 21 days. Parameters observed in current study were gas test production, ruminal fermentation parameter, and in vitro digestibility. The collected data were analyzed by one-way analysis of variance and followed by Duncan’s new Multiple Range Test for data with significant differences. Result showed that the mixed culture Lp+Sc inoculation increased (P<0.05) chemical quality of cocoa pods by reducing fibre fraction and increase NFE contents, increased degradation rate, degradation theory, reduced rumen pH, and propionate acid production, without affecting acetate to propionate ratio, microbial protein synthesis, and digestibility of cocoa pod silage.

No Brasil, a adição de fitases em ração animal tem sido usada para minimizar os efeitos antinutricionais doácido fítico, além de diminuir os impactos ambientais e a adição de fósforo na ração. Este estudo teve comoobjetivo avaliar a produção de fitases e caracterizar, bioquimicamente, as enzimas produzidas por fungosisolados na região do Alto Paranaíba, no estado de Minas Gerais. Foram utilizados quatro isolados de Rhizopussp., obtidos a partir de amostras de amendoim e dois isolados de Aspergillus niger, um isolado de Trichodermasp., um isolado de Xylaria sp. e um isolado de Fusarium sp., obtidos no banco de culturas do laboratório deMicrobiologia da Universidade Federal de Viçosa (UFV), campus Rio Paranaíba. Os isolados de Rhizopus sp.,Thichoderma sp. e Fusarium sp. não apresentaram atividade enzimática significativa, enquanto os isolados deA. niger e Xylaria sp., por apresentarem atividade enzimática significativa, foram selecionados para a etapa decaracterização bioquímica. Na fermentação no estado líquido, um dos isolados de A. niger apresentou maioratividade enzimática (0,89 U.mL-1) em 216 h, enquanto na fermentação no estado sólido, o isolado de Xylariasp. apresentou maior atividade enzimática (2,88 U.g-1) em 264 h. A fitase de Xylaria sp. apresentou pH ótimoigual a 2,5, temperatura ótima de 45 °C e manteve 91% de atividade residual, após ser mantida a 50 °C durante15 min., enquanto a fitase de A. niger apresentou pH ótimo igual a 3, temperatura ótima de 60 °C e manteve68% de atividade residual, após ser mantida a 70 °C durante 15 min. Os resultados indicam que as enzimasapresentam potencial para aplicação industrial.

ABSTRACT: The aim of this study was to evaluate the nutritional diversity of Brachiaria ruziziensis clones through chemical composition and in vitro kinetics of ruminal fermentation. Twenty three clones of Brachiaria ruziziensis were used (15, 16, 46, 174, 411, 590, 651, 670, 768, 776, 844, 859, 950, 965, 970, 975, 1067, 1093, 1296, 1765, 1806, 1894 and 1972) and Brachiaria ruziziensis cv. ‘Kennedy’, Brachiaria brizantha cv. ‘Marandu’ and Brachiaria decumbens cv. ‘Basilisk’ as controls within 27 days of harvesting. The experimental design used randomized blocks with 26 treatments (genotypes) and three replications. Evaluation of the nutritional divergence was performed using principal components analysis, based on the following discriminatory variables: in vitro dry matter digestibility (IVDMD), neutral detergent fiber (NDF), lignin, crude protein (CP), degradation rate of non-fibrous carbohydrates (KdNFC) and degradation rate of fibrous carbohydrates (KdFC). The evaluation of the nutritional diversity of Brachiaria genotypes was based on the two main components (IVDMD and NDF), which explains 96.2% of the total variance Variables of lower contribution to the discrimination of the clones were as degradation rates of the fibrous and non-fibrous carbohydrates. In the agglomerative hierarchical grouping analysis, five distinct groups were identified, where V group, formed by clones 46, 768 and 1067 have higher values of IVDMD compared to the other clones.
