The Estrogen was very beneficial hormone for men and women. Isoflavon compound was natural estrogen source which contain three main compound in chemical form, the aglycones, daidzein and genistein. They were found only 0,25% on plant, exclusively on leguminous plant. An alternative method have been required to overcome these problem, for example, through plant cell culture technic with heavy metal elicitor. The aims of this research were to study the effect of various concentration of Al3+ elicitor and age of culture for callus growth and accumulation of daidzein and genistein from in vitro Phaseolus vulgaris L. And Phaseolus aureus Roxb callus culture. Stable plant callus culture were established and subculture for three times approximately 4 weeks. The calli were elicitated by Al3+ elicitor with 0, 50, 150, 300 µM concentrations and incubated on 4, 6, and 8 weeks. Qualitative data were determined based in changed calli colour, TLC plate and value of Rf. Quantitative data were determined based in fresh and dry weight of the calli and the content of daidzein and genistein compound. Quantitative data were analized using Anova and continued with DMRT 95%.  The results showed that the callus growth more influenced by age of culture. The sixth weeks led to higher fresh and dry weight. The Colour of Calli had been changed from white yellowness become brown at the eighth weeks. The texture of Calli have no difference between control and treatment of elicitor, that was friable until the end of treatment. The Calli with addition of elicitor Al3+ with 150 µM concentrations had significantly highest of fresh weight than control, they were over 0,463 g. The Content of daidzein and genistein on Phaseolus vulgaris L. had significantly increased in addition of Al3+ elicitor with 150 µM concentration. But, the content of daidzein and genistein on Phaseolus aureus Roxb. had significantly increased in addition of Al3+ elicitor with 300 µM concentration.   Keywords: Phaseolus vulgaris L., Phaseolus aureus Roxb., elicitor Al3+, Daidzein, Genistein, Callus Growth.

Phaseolus vulgaris L. (common bean) is a leguminous species that is an important dietary component due to its high content of proteins, unsaturated fatty acids, minerals, dietary fibers and vitamins. Due to the high content of polyphenols, several biological activities have been described for bean extracts, making it possible to include P. vulgaris among food with beneficial effects for human health. Moreover, more than 40,000 varieties of beans have been recognised with different nutraceutical properties, pointing out the importance of food biodiversity. In this work, we describe for the first time the chemical composition and biological activity of a newly recognized Italian variety of P. vulgaris grown in a restricted area of the Tuscany region and named “Fagiola di Venanzio”. Fagiola di Venanzio water extract is rich in proteins, sugars and polyphenols and displays antioxidant, anti-inflammatory and antiproliferative activities in in vitro assays on colon cancer cellular models. Our data indicate that this variety of P. vulgaris appears to be a promising source of bioactive compounds and encourage more in-depth studies to better elucidate the implications of its consumption for public health.

Nine cultivars of Phaseolus vulgaris L. were grown in nutrient solution to study the effect of boron on growth and mineral composition. Data obtained in thie study allowed for the following conclusions: (1) high levels of boron affected plant height, root length, dry weight of tops, dry weight of root, and total dry weight; (2) regression analysis was used to point out differential behaviour among cultivars in relation to boron concentration in nutrient solution; (3) the best mineral concentration in the plant tissue was obtained with application of 0,5 ppm of boron in the nutrient solution.

Lepidium sativum L. is a rich source of polyphenols that have huge medicinal and pharmaceutical applications. In the current study, an effective abiotic elicitation strategy was designed for enhanced biosynthesis of polyphenols in callus culture of L. sativum. Callus was exposed to UV-C radiations for different time intervals and various concentrations of melatonin. Secondary metabolites were quantified by using high-performance liquid chromatography (HPLC). Results indicated the total secondary metabolite accumulation of nine quantified compounds was almost three fold higher (36.36 mg/g dry weight (DW)) in melatonin (20 μM) treated cultures, whereas, in response to UV-C (60 min), a 2.5 fold increase (32.33 mg/g DW) was recorded compared to control (13.94 mg/g DW). Metabolic profiling revealed the presence of three major phytochemicals, i.e., chlorogenic acid, kaemferol, and quercetin, in callus culture of L. sativum. Furthermore, antioxidant, antidiabetic, and enzymatic activities of callus cultures were significantly enhanced. Maximum antidiabetic activities (α-glucosidase: 57.84%; α-amylase: 62.66%) were recorded in melatonin (20 μM) treated callus cultures. Overall, melatonin proved to be an effect elicitor compared to UV-C and a positive correlation in these biological activities and phytochemical accumulation was observed. The present study provides a better comparison of both elicitors and their role in the initiation of physiological pathways for enhanced metabolites biosynthesis in vitro callus culture of L. sativum.

SUMMARYIn a glasshouse study, the pattern of flower and pod production at the various flowering nodes of Phaseolus vulgaris (cv. 344) was mapped out. Soft X-radiation techniques were also used to study the development of retained and abscissed pods at the various nodes while varying levels of flower or pod removal were used to study the modification to pod development at each flowering node following organ removal.The period of flower opening over the whole plant spanned an average of 12 days and 90% of the pods retained to maturity were formed from flowers opened within 4 days of anthesis. The terminal inflorescence produced most of the flowers on the plant (25%), but nodes 2 and 3 developed the bulk (50%) of the pods retained. Only 30% of the open flowers on the plant formed pods that were retained to maturity.Pod elongation was initially very rapid in the pods to be retained, following successful fertilization, with the latter period of pod growth devoted almost entirely to seedfilling. A slow growth rate characterizes the development of abscission in Phaseolus pods and the number of seeds per pod is determined later in pod growth.Flower or pod removal prolonged flowering and led to longer leaf retention and more branches on the main stem. The number of mature pods was lower on the treated plants and the more severe the removal the larger were these treatment effects. Plants compensated for fewer pods by having more seeds per pod but these were not enough to prevent significant depression of seed dry weight per plant by the most severe removals. Flower or pod removal led to a shift of emphasis in pod production by the plant from nodes 2 and 3 to other areas of the plant where natural wastage of flower and young pods is high.
